1.Isolation,expansion and identification of fetal rat neural stem cell

Objective:To culture neural stem cell (NSC) in vitro,observe its biological property and provide basis for further study on differentiation.Methods:NSC of rat fetus were isolated and cultured.Single-cell clone culture was used to confirm the character of self-renewing.Ability of multi-directional differentiation and specific antigen, nestin,were detected by immunocytochemistry (ICC).BrdU was used to show cell stage in division.Effect of combination of EGF,bFGF on proliferation of rat NSC were evaluated.Results:The cultured NSC showed multipotential of differentiation into neurons and glial cells,as well as the ability of self-renewing.Cells expressed nestin.The population of the NSC expanded quickly and the total number of the cell increased 2.45?10~4 folds after 15 times' passage.The property of NSC was stable after long culture and preservation.Combination of EGF and bFGF stimulated cell proliferation than used singly.Conclusion:The cultured NSC express nestin, have the ability of self-renewing and multi-directional differentiation.Combination of EGF and bFGF could promote the proliferation of rat NSC.
2.Expression of Vascular Endothelial Growth Factor and Monocyte Chemoattractant Protein-1 in Patients with Psoriasis

Objective To study the production and distribution of vascular endothelial growth factor (VEGF) and monocyte chemoattractant protein 1 (MCP-1) in the skin and sera of patients with psoriasis and their roles in the pathogenesis of psoriasis. Methods Levels of VEGF and MCP-1 in sera were measured using double antibody sandwich enzyme linked immunosorbent assay(ELISA) in patients with psoriasis. The expression and distribution of VEGF and MCP-1 in psoriatic lesions, non lesional skin and normal controls were detected by immunohistochemical method. Results ①The expression of VEGF in lesional skin and non lesional skin of psoriasis patients was higher than that in normal controls. Levels of MCP-1 were increased in lesional skin than those in non lesional skin and normal controls.②Serum levels of VEGF were significantly increased in psoriasis as compared with normal control. There was no significant difference in serum levels of MCP-1 between patients and controls.③There was no correlation between the expression of MCP-1 or VEGF of psoriatic lesion and PASI index. Conclusion Overexpression of two cytokines, VEGF and MCP-1, known to promote new blood vessel formation may contribute to the pathogenesis of psoriasis.
3.DIRECTIONAL INDUCEMENT OF SCIATIC NERVE DERIVED DIFFUSIBLE FACTORS ON REGENERATING AXONS OF ADULT FROGS

Objective To study the effects of directional inducement of factors produced by injured sciatic nerve on the regenerating axons of adult frog in vivo. Methods Conditioned medium with fraction of 30kD to 100kD was obtained from sciatic nerve of adult frog and pumped out and formed a gradient of concentration toward the end of central nerve stump of cutaneous pectoris nerve with different distances and angles by miniosmotic pumps.After 28\|35 days,the chest tissue was dissected and stained with immunocytochemistry to show the regenerated motor\|axons and the directional value is analyzed. Results The regenerated motor axons grew obviously towards the tip of the cannula with high concentration of CM.The directional value is 0\^84 whereas the value is 0\^04 with random direction.The average directional value between the two groups is significant and is not influenced by the distances and angles of the cannula to the nerve stump. Conclusion Damaged peripheral nerve can produce diffusible active factors with molecular weight between 30kD to 100kD,which induce the directional outgrowth of regenerating motor axons from sectioned nerve stump of adult frogs in vivo. 5.Analysis of related factors of compliance to medical order behavior in patients with chronic prostalitis

Objective To analyze the related factors of medical order compliance in patients with chronic prostatitis. Methods 482 patients with chronic prostatitis were investigated by using the National Institutes of Health chronic prostatitis symptom index(NIH-CPSI)、Eysenck personality qusetionnaire( EPQ)and the self-de-signed medical order compliance questionnaire and the patients' disease characteristics and personality were also analyzed.The related factores of medical order compliance in patients were analyzed through hierarchical multiple regression analysis. Results Patients with chronic pwstatitis were younger and their disease condition was seri-ous. Their personality was introverted. They had many psychological problems, such as anxiety, depression, sus-pect, et al. All these were related with patients' compliance. Conclusions Attention should be paid to the patients' personality, and psychotherapy should be given to patients with introverted personality to improve pa-tients' compliance,so that the treatment effect will be increased for patients with chronic prostatitis.
8.Thought on development of minimally invasive gastrointestinal surgery in China.

Laparoscopic surgery for gastrointestinal diseases had a remarkable progress in the past 20 years. Laparoscopic surgery for colon cancer is adopted worldwide by increasingly surgeons based on the evidence from randomized controlled studies. Minimally invasive surgery is also adopted as alternative procedure for early gastric cancer. The standardization and spreading of the minimally invasive technique for gastrointestinal surgery is also being implemented efficiently in China. However, how to innovate the new technique and improve the new instrumentation for minimally invasive surgery is still a problem which we have to confront in the current situation. But we believe that with the great efforts of gastrointestinal surgeons, the widespread adoption of minimally invasive gastrointestinal surgery would surely take place in the near future.

9.The different negative pressure values of the effect on skin graft

Objective To study the effect of different negative pressure values of negative pressure wound therapy (NPWT) effect on skin graft .Methods Eighty patients were selected ,who underwent chronic wound bed and preparedto treat with skin graft in surgical ward of emergency department in the Second Affiliated Hospital of Chongqing Medical University from January 2012 to July 2012 .Eighty patients were divided into 8 groups ,group A were treated with packing pressure after skin graft ,group B~ H were treated with continuous NPWT ,pressures were -10 ,-20 ,-30 ,-40 ,-50 ,-60 and -70 kPa ,respectively .Whether they were in comfortable situation at second day after surgery were observed .Flap survival area at Seventh day after surgery was meas‐ured .Results The patient comfort ,flap survival area ,wound healing time of group B to H were higher than those of group A (P<0 .05) .Patient comfort tended to decrease with the increasing negative pressure values between -10 - -40 kPa(P<0 .05) .Flap survival area ,healing time of group B ,C were higher than those of group D to F(P<0 .05) .Conclusion Application of NPWT technology in wound ,-10 kPa negative pressure value can not only improve wound blood circulation ,but also ensure patient be comfortable .
