1.Cloning and expressing of vacA gene related segment with vacuolating activity of Helicobacter pylori

Objective:To obtain the antigen of recombinant vacuolating segment of Helicobacter pylori vacuolating cytotoxing by gene-engineering.Methods:The objective gene was cloned into PQE-30,sequenced and expressed using PCR and recombinant techniques.After expression,the antigenicity of recombinant protein was analyzed by Western blotting.Results:DNA sequence analysis identified that the sequence was integrated.Compared with standard strain AF361700,1.5% of the cloned gene was mutated,one amino acid radicle was changed.SDS-PAGE revealed the molecular weight of objective recombinant protein was 27 000 as it was predicted;the level of expression product was over 30%.Western blotting results showed the objective protein could be recognized by anti-serum against HP.Conclusion:This recombinant protein has perfect antigenicity,so it may be used as a protective antigen in preventing HP infection.
2.Clinical analysis of inflammatory bowel disease in ten general hospitals of Kunming city

Objective To retrospectively investigate hospitalized patients with inflammatory bowel disease (IBD) in Kun-ming ,to understand their characteristics of clinical data in recent years .Methods Ten three-tertiary general hospitals in Kunming were selected and clinical information of 1 029 patients diagnosed IBD from January 2007 to December 2013 were reviewed ,inclu-ding gender ,age ,course of disease ,symptoms ,diagnosis ,treatment and outcomes .Results This study showed that there was an up-ward tendency of IBD patients in recent 7 years in Kunming .Among the 1 029 IBD patients ,853 patients had ulcerative colitis (UC) ,157 patients had Crohn disease(CD) and 19 patients had undifferentiated IBD .The ratio of male to female was 1 .23 ∶ 1 .00 , and the average onset age of IBD was (47 .93 ± 16 .78)years old .The peak onset age of UC was from 40 to 60 years old ,while CD was more prevalent from 20 to 40 years old .Most of these patients had IBD less than 5 years .Cases from urban areas were more than from the country side .The majority of these hospitalised cases were moderate IBD patients and more UC cases than CD . 64 .4% of the UC cases were chronic and relapsing ;while in the CD cases ,51 .0% were inflammatory and 41 .2% were stenotic type .The main symptoms of IBD were diarrhea (91 .0% ) ,abdominal pain (84 .6% ) ,bloody stool (79 .4% ) and so on .About 6 .1%IBD patients had extraintestinal manifestations and 10 .6% of IBD patients had complications .The auxiliary diagnosis methods were colonoscopy (97 .7% ) and pathology (29 .2% ) .Colonoscopy revealed that UC lesions were in the rectum and sigmoid colon ,and CD lesions were in ileocecal and terminal ileum .Aminosaliccylic acid and steroids were the main medications and immunosupressant was much less used .The effective rate of medical treatment alone was 92 .4% with a mortality rate of 0 .4% .Conclusion The number of hospitalized IBD patients increased in recent years and most of them are young and middle aged .The lesions are mainly moderate . The majority UC cases are chronic and recurrent ;while CD cases belong to inflammatory type and stenotic type predominantly .Par-enteral lesions and complications are rare .Sulphasalazine (SASP)/5-ASA and steroids are still the mainstay agents used for the treatment of IBD .This disease need for long-term maintenance treatment .

4.DIAGNOSIS AND TREATMENT OF FRACTURE OF THE AXIS ARCH

The fracture of the axis arch is a rare uppercervical vertebrae fracture. It is easy to misdiag-nose. From 1971 to 1989, 7 cases of the fracturewere admitted to our hospital. Diagnoses were con-firmed by injury history, clinical features and X-ray films. The treatment results were as follows:six patients were in excellent condition and one pa-tient was in good one. The three following prob-lems were discussed: ①The mechanism of injury.②The misdiagnosis causes and its preventivemethods. ③Most patients can be treated conserva-tively and can be up. Only a few Type III fractureswill require operation after the failure of non-oper-ative treatment or at the time of conplicated injuryof the spinal cord occurring lately.
5.Clinical Application and Efficacy of Tazarotene Cream for Acne Vulgaris

OBJECTIVE:To provide optimal therapeutic scheme for patients with acne vulgaris. METHODS: By a single-blind randomized controlled method,a total of 122 patients with acne vulgaris were assigned to one of the following 3 groups: tazarotene cream group(n=42),Water chlorine tincture group(n=38) and combined treatment group(n=42). The curative efficacy was observed after treatment for 8 consecutive weeks. RESULTS: The number of skin lesions in all the 3 groups reduced as the course of treatment increased,and there were significant differences across the 3 groups after 8-week treatment as compared with before treatment (P0.05). CONCLUSION: Tazarotene cream is effective for non-inflammatory acne. The superior efficacy of water chlorine tincture as compared with tazarotene for patients with impetigo might be related to the antibacterial action of chloramphenicol. Tazarotene cream combined with local application of 0.5% water chlorine tincture is effective for inflammatory acne.
6.Determination of Diclofenac Sodium in Huamn Plasma by HPLC

OBJECTIVE:To establish a HPLC procedure to determine diclofenac sodium in human plasma.METHODS:The plasma sample was deproteinized by adding acetonitrile,A Hypersil-BDS(250?4.6mm,5?m)column with a mobile phase consisting of methanol-water-10%phosphate acid(60∶40∶1)was used,the detecting wavelength was276nm,column tem?perature was35℃.RESULTS:The linear range was20.50~2050.0ng/ml(r=0.9998,n=5),the detective limit was10ng/ml.The recovery was99.47%~105.4%with the within-day RSD from3.77%to5.81%(n=5),and the between-day RSD from4.93to8.96%(n=5)at three different concentrations.CONCLUSION:The method is simple,rapid and suitable for the pharmacokinetic study of diclofenac sodium.
7.Observation of effects of zinc on the injury induced by oxygen free radical in cultured heart cells

In present study, the injury induced by oxygen free radical in cultured cardiac cells was observed by adding xanthine and xanthine oxidase into the medium The effects of zinc on the injury of cells were also observed, taking the electro physiological changes and threshold concentration of BaCl_2 which induces cell stop beating as index. The results showed that the electric parameters of action potential (the amplitude, overshoot, maximum diastolic potential, threshold, maximum rate of depolarization and duration of action potential ) were decreased in XOD group, compared with the control. The threshold concentration of BaCl_2 was decreased too. However, there were no significant differences between Zn group and control group except the TP value. The electric parameters of action potential was increased notab(?)y in Zn group, compared with XOD group. Furthermore, the threshold concentration of BaCl_2 in Zn group was increased too. It is suggested that Zn possessed the protective effects on cultured cardiac cells from the injury induced by oxygen free radical.

9.Stem cell therapy for ischemic heart disease

BACKGROUND:Stem cel transplantation is a promising strategy for treatment of ischemic heart diseases, which has obtained great achievements in recent years. OBJECTIVE: To analyze the clinical trends of stem cel therapy for ischemic heart diseases reported from 2001 to 2015 in Medline, CNKI, Wanfang, ClinicalTrials.gov and Chinese Clinical Trial Register. METHODS:The relevant articles addressing stem cel therapy for ischemic heart diseases were retrieved using the keywords of “stem cel transplantation” and “ischemic heart disease” in Chinese and English in Medline, CNKI, Wanfang, ClinicalTrials.gov and Chinese Clinical Trial Register folowed by statistical analysis. The retrieval time was from 2001 to 2015, including 2001-2005, 2006-2010 and 2011-2015. RESULTS AND CONCLUSION: In Medline database, there were 219 clinical studies about stem cel therapy for ischemic heart disease published from 2001 to 2015, and the number of retrieved articles was most from 2006 to 2010, and decreased from 2011 to 2015. In CNKI and Wanfang databases, the number of relevant articles which had a similar trend with that in the Medline database decreased remarkably after 2011. In ClinicalTrials.gov, there were 63 clinical trials about stem cel therapy for ischemic heart disease, most of which were registered from 2006 to 2015 and came from Europe and North America. In Chinese Clinical Trial Register, there was no clinical trial about stem cel therapy for ischemic heart disease, which may result from the strict management in China.
10.Influence of cerebral hemorrhage stress on the proliferation and senescence of endothelial progenitor cells

Objective To explore the influence of cerebral hemorrhage stress on endothelial progenitor cells(EPCs) proliferation and senescence. Methods Mononuclear cells from peripheral blood of patients with cerebral hemorrhage (CH group) and patients with the same underling diseases (control group) as the front were isolated and induced to EPCs. Proliferation, numbers of colonies and cells, senescence of EPCs were checked and compared between the two groups. Results Compared with control group, proliferation (0.270 ± 0. 074 vs 0. 185 ±0.050, t=3.802) ,numbers of colonies( (5. l0 ±0.79) cell/HPF vs ( 1.90 ±0.41 ) cell/HPF, t= 14.407) and cells( (75.15 ± 10.48) cell/HPF vs(57.10 ± 9.28 ) cell/HPF, t = 5. 157 ) of EPCs in CH group on the stress of cerebral hemorrhage were increased significantly (P < 0.01 ), but senescence of EPCs ( (45.54 ± 5.74 ) cells/100 cells vs ( 46.86 ± 5.06 ) cells/100 cells, t = 0. 686 ) in two groups were not different (P > 0.05 ). Conclusion Cerebral hemorrhage stress can increase proliferation of EPCs,but can not change the senescence destiny of EPCs.
