1.Effects of purslane herb on stress ability of aging mice induced by D-galactose

OBJECTIVE: To investigate the effects of purslane herb aquenous extracts (PHAS) on the stress ability of aging mice induced by D-galactose. METHODS: We observed the survival time to hypoxia and heat survival rate of the mice treated with different doses of PHAS and vitamin E. The contents of lipofuscin and malondialdehyde (MDA), and the activity of superoxide dismutase (SOD) and catalase (CAT) in the brain and liver of the mice were tested. RESULTS: As compared with vitamin E, three doses of PHAS (1.6, 0.8 and 0.4 ml/d) prolonged the survival time to hypoxia and the pole climbing time and increased the heat survival rate, and the 0.8 ml/d PHAS had the best effect. In the group of 0.8 ml/d PHAS, the activity of SOD and CAT decreased less, and the contents of lipofuscin and MDA decreased significantly. The effect of vitamin E was not as good as the PHAS. CONCLUSION: PHAS can prevent the stress ability of the aging mice. One of its mechanisms may be increasing the activity of SOD and CAT, hence decreasing the damage of the oxidation products to the body.
2.Effect of PTTG on the growth and fluorouracil sensitivity of cholangiocarcinoma cell Line QBC939

Objective To investigate the effct of PTTG gene on the growth and sensitivity to 5-FU of cholangiocarcinoma cell.Methods The pcDNA3.1-PTTGas that contained full-length antisense PTTG,and pcDNA3.1(+) was transfected into the cholangiocarcinoma cell line QBC939.We successfully established and identified cell sublines tQBC939(PTTG-),tQBC939(pcDNA3.1).Cell growth curve was mapped out,cell proliferation rate was assessed by MTT assay and cell cycle percentage was counted by flow cytometry.After treatment with 5-FU,cell survival rate was assessed by MTT assay and IC50 was calculated.Flow cytometry and Hoechst staining were used to demonstrate discrepancy of cell apoptosis rates.Results Compared with other two groups,tQBC939(PTTG-) grew more rapidly and had a higher proportion of cells in S phase,whereas less cells in G2/M phase(P
3.Analysis of higher vocational nursing students participating in opening professional social practice

Objective To analyze the significance of higher vocational nursing students participating in opening professional social practice,and to explore the effective way of combining social practice with professional learning and clinical practice.Methods After completing the opening professional social practice in summer,the reflection diaries of 30 nursing students were analyzed and the concerns were refined.Questionnaire survey about self assessment of comprehensive effect of the practice of nursing students were conducted.Results Nursing operation and soft skills were mostly concerned by nursing students' in opening professional social practice.Vocational learning,occupation of responsibility,and the occupation ability were improved by practical activities.Conclusions Nursing students who participate in opening professional social practice before clinical work,can re-understand the professional learning,adapt and integrate themselves into clinical practice through the perception of clinical nursing work.

6.Clinical efficacy of postoperative hydrophallous after penile lengthening surgery

ObjectiveTo introduce a new procedure of penile lengthening which yields satisfactory outcomes and avoids persistent postoperative hydrophallus with the results of fresh body researches.MethodsIndirect intralymphatic injection and routine H-E staining were used to investigate the ramification of lymphatics of penis on fresh bodies.We found that the lymphatic return of penis was primarily mediated by dorsal and bilateral superficial lymph vessels,which converged dorsally at the root of penis and then ramified into the superior region of pubic symphysis and bilateral inguinal grooves.However,most previous incisions were done at the converging region of lymph vessels at the dorsal root of penis.It blocked most lymphatic return.Our modified method avoided interruption of lymph vessels in the converging region.Based on the results of the above investigations,the operative incision at the root of penis was shifted upward by 1.5 cm,and then the lengthened cavernous body of penis was covered by scrotal flap.ResultsEighty patients who accepted the new method achieved satisfactory outcomes.Postoperative hydrophallous decreased gradually from the 3rd day and vanished basically on the 6-7 day.ConclusionsOur modified procedure of penile lengthening reduces the damage of these vessels.Consequently,postoperative hydrophallus resolves fast and the new procedure yields high success rates,sound appearance and sensation of penis.Therefore,the new procedure is an ideal option for surgenons in clinical practice.
7.Character of drug targeting lymph nodes after regional injection of carboplatin-activated carbon in patients with breast cancer

Purpose: To examine the drug concentration in axillary nodes after regional injection of carboplatin-activated carbon suspension (CP-CH) in patients with breast cancer, and compared the result with what happened after regional injection of carboplatin solution ( CP-Sol). Methods: 32 patients with breast cancer were divided into 2 groups randomly with 16 patients in each group. One group received regional subcutaneous injection of CP-CH adjacent to the primary tumor. The contrast group received CP-Sol under the same conditions. Modified mammectomy was done 1, 12, 24, 72 hours later. The carboplatin concentration in axillary lymph nodes was examined by atom absorbing spectrometry. Results: At 1, 12, 24, 72 hours after injection of CP-CH, the carboplatin concentration in lymph nodes was 11.23?5. 66u,g/g, 26.40?11. 18u.g/g, 18.72?7. 14u.g/g, 15. 44?6. 92u,g/g, respectively. At 1, 12, 24 hours after injection of CP-Sol, the carboplatin concentration was 0. 24? 0. 06ug/g, 0. 13? 0. 08ug/g, 0. 12? 0. 04ug/g, respectively. Traditional Chinese medicine (TCM), an important component of complementary and alternative medicine, has evolved over thousands of years with its own unique system of theories, diagnostics and therapies. TCM has been increasingly used in the last decades and become well known for its significant role in preventing and treating cancer. We believe that TCM possesses advantages over Western medicine in specific aspects at a certain stage of cancer treatment. Here we summarize the advantages of TCM from three aspects: preventing tumorigenesis; attenuating toxicity and enhancing the treatment effect; and reducing tumor recurrence and metastasis.
9.Rhein on diabetic nephropathy rat podocytes nephrin expression

Objective To investigate rhein on podocyte nephrin gene expression and nephrin on glomerular in diabetic nephropa-thy (DN)rats .Methods DN rats mode were made by Intraperitoneal injection of streptozotocin (STZ) 60 mg/kg once .the DN model was divided into 2 groups:rhein treatment group (A) ,model group (B) ,and in addition ,set up a control group (C) .body weight ,kidney weight ,relative kidney weight ,24-hour urinary protein excretion ,serum creatinine ,blood urea nitrogen of rats were detected after 8 weeks of intervention .kidney pathological changes were observed by using light microscope ,electron microscope . immunohistochemical technique used to nephrin protein expression .Results Both the DN rats 24 -hour urinary protein excretion and podocyte nephrin gene expression of group A decreased significantly (P<0 .05) ,improvement in renal function .Conclusion Rhein on DN kidney has a protective effect ,which may be related to its podocyte nephrin gene expression.
10.MicroRNA profiling on oxidative stress induced apoptosis of human lens epithelium

AIM: To identify the changes in microRNA ( miRNA ) profile of human lens epithelium ( HLE) induced by H2 O2 and the role of miRNA in oxidative stress induced apoptosis.
METHODS: HLE cell line HLE - B3 was treated by 100μmol/L H2 O2 for 24h and the total RNA were isolated by Trizol reagent. miRNA profile was generated by miRCURYTM LNA microRNA Array. The target genes of differentially expressed miRNAs were predicted by bioinformatics software.
RESULTS:Twenty-eight miRNAs showed significantly differential expression after H2 O2 treatment, 18 miRNAs upregulated and 10 miRNAs downregulated. The differentially expressed miRNAs may involve in apoptosis of lens epithetium and development of cataract through targeting BCL2L2 and MIP.
CONCLUSION: H2 O2 can induce dramatically changes in miRNA profile of HLE, which may play a pivotal role in the pathogenesis and development of cataract.
