1.Influence of intensive atorvastatin therapy on CIN and inflammatory response in patients with ischemic cerebrovascular disease after interventional surgery

Objective:To explore influence of intensive atorvastatin therapy on contrast-induced nephropathy(CIN) and systemic inflammatory response in patients with ischemic cerebrovascular disease(ICD)after interventional sur-gery.Methods:A total of 118 ICD patients undergoing interventional surgery in our hospital were selected.Accord-ing to atorvastatin dose,they were divided into intensive group(n=59,received atorvastatin 80mg/d before surgery and 40mg/d after surgery)and routine control group(n=59,received atorvastatin 20mg/d 3d before surgery and af-ter surgery).Levels of renal function indexes and inflammatory indexes 24h before and 72h after surgery were com-pared between two groups.Results:Compared with before treatment,after treatment,there was significant rise in cystatin C(CysC)level in intensive group;significant rise in levels of serum CysC,creatinine(Scr),and β2-micro-globulin(β2-MG),and significant reduction in creatinine clearance rate(Ccr)in routine control group(P<0.05 or<0.01);compared with routine control group after treatment,there were significant reductions in levels of Scr [(93.97 ± 13.83)μmol/L vs.(78.44 ± 17.36)μmol/L],β2-MG[(2.88 ± 0.64)mg/L vs.(2.46 ± 0.61)mg/L] and CysC[(1.24 ± 0.07)mg/L vs.(0.88 ± 0.10)mg/L],and significant rise in Ccr[(68.92 ± 17.38)ml/min vs. (82.22 ± 15.94)ml/min]in intensive group,P=0.001 all.There were significant reductions in inflammatory index levels in both groups after surgery,compared with routine control group,there were significant reductions in levels of high sensitive C reactive protein[(13.53 ± 3.82)mg/L vs.(11.31 ± 3.72)mg/L],interleukin-6[(99.87 ± 24.76)ng/L vs.(85.73 ± 24.17)ng/L]and tumor necrosis factor α[(286.67 ± 78.38)ng/L vs.(252.61 ± 80.02) ng/L]in intensive group,P<0.05 or <0.01. Incidence rate of CIN in intensive group was significantly lower than that of routine control group(1.69% vs.11.90%,P=0.028).Conclusion:Intensive atorvastatin therapy can sig-nificantly lower incidence rate of CIN and inhibit systemic inflammatory response in ICD patients after intervention -al surgery,and it′s safe and reliable.

Objective To design a new method to verify the position of multileaf collimator(MLC)leaf using a two-dimensional ion chamber array(2D-array).Methods 2D-array of PTW T10018 Seven29~(TM) was used to calibrate the accuracy of MLC leaf position of Elekta Precise accelerator.The edge function of the leaf position of MLC was measured and used as the reference value.The precision of MLC leaf was then evaluated through comparing the measured and reference values.Results The accuracy of MLC leaf position was found within?0.1 mm.Conclusion This method of verifying the accuracy of multileaf collimator leaf position is easy,simple and reliable

This study is to investigate the effect of total flavonoids of Uygur medicine bugloss (BTF) on rats with myocardial ischemia/reperfusion injury, and to explore the mechanisms by which it acts. Left anterior descending (LAD) coronary artery in rats was occluded for 30 min followed by 4 h reperfusion. Meanwhile, BTF dissolved in saline was administered intraperitoneally at dosage of 10, 30 and 50 mg x kg(-1). Electrocardiograph, infarction index, serum myocardial enzymes and heart function were determined to evaluate the effect of BTF. Some other observations were carried out to explore whether inhibiting inflammation and apoptosis is involved in the mechanisms underlying BTF. Our results showed that in ischemia/reperfusion injured rats BTF could dose-dependently reduce myocardial infarction index and myocardial enzyme leakage, and enhance heart function, indicating that it possesses significant cardio protection. ELISA analysis showed that BTF could decrease the content of myocardial inflammatory cytokines such as IL-1beta, IL-6 and TNF-alpha. Western-blotting confirmed that BTF could increase the expression of anti-apoptotic protein Bcl-2 and reduce the expression of proapoptosis protein Bax. Further more, the phosphorylation level of PI3K and Akt was upregulated by BTF treatment. BTF can protect rat against myocardial ischemia/reperfusion injury. Anti-inflammation and inhibition of apoptosis through upregulating PI3K/Akt signal pathway may contribute to the protective effect of BTF.

4.Puerarin reduces oxidative damage to vascular endothelial cells by improving mitochondrial respiratory function

This study investigated the effect of puerarin on human umbilical vein endothelial cells (HUVEC) injured with hydrogen peroxide (H2O2). HUVEC were divided into three groups: a control group, a model group (H2O2 400 μmol·L-1) and a puerarin-treated group (3, 10, 30 and 100 μmol·L-1). HUVEC were cultured with varied concentration of puerarin for 2 h and treated with H2O2 for another 24 h. Cell proliferation was detected by a CCK-8 assay. 5.Effects of intensive insulin therapy on insulin resistance and serum proteins after radical gastrectomy.

OBJECTIVETo investigate the effects of intensive insulin therapy on insulin resistance(IR) and serum proteins after radical gastrectomy.
METHODSTwenty-two gastric cancer patients were randomly divided into the control (n=11) and intensive insulin therapy group (n=11), and underwent distal radical subtotal gastrectomy under epidural anesthesia. Fasting blood glucose (FBG), fasting insulin (FINS) and serum proteins were assayed preoperatively and at day 1, 3, 7 postoperatively. Insulin resistance index was calculated using homeostasis model assessment (HOMA). The length of hospital stay and postoperative complications were recorded respectively.
RESULTS(1)The levels of FBG, FINS, lnHOMA-IR (P<0.01,P<0.05) and the incidence of insulin resistance were remarkably decreased by intensive insulin therapy after the surgical procedure.(2)The levels of serum transferrin (TRF), prealbumin (PRE) and retinal binding protein (RBP) in the intensive insulin therapy group were significantly improved as compared to control group after operation(P<0.05). (3) The duration of fever, antibiotic use, passage of gas by anus, length of hospital stay and the occurrence of postoperative complications were also significantly lower than those in control group(P<0.01,P<0.05).
CONCLUSIONCompared to routine therapy, the intensive insulin therapy has more beneficial effects on the patients undergone distal radical subtotal gastrectomy in decreasing the insulin resistance, improving the status of nutrition and preventing postoperative complications.

This study aimed to evaluate the vasorelaxant effect and mechanisms of compound reserpine and triamterene tablets (CRTTs) and its component triamterene on isolated rat thoracic aorta rings. Isolated rat thoracic aorta rings pre-contracted by high potassium or norepinephrine (NE) were used to evaluate the vasodilatory effect of CRTTs and its component triamterene. The mechanisms concerning endothelium, potassium channels and calcium channels were studied through the interventions of several tool drugs. Animal welfare and experimental procedures followed the requirements of the Laboratory Animal Management and Animal Welfare Ethics Committee of the Institute of Materia Medica, Chinese Academy of Medical Sciences and Peking Union Medical College. 9.Intrasplenic tumor model of nude mice in the anti- metastasis roles of NGX6 gene against colon cancer.

OBJECTIVE:
To establish a liver metastasis model of nude mice in colon cancer so as to determine the function of NGX6.
METHODS:
The cells of Group HT-29, pcDNA3.1(+)/HT-29, and pcDNA3.1(+)/NGX6/HT-29 were implanted into the spleen of nude mice, respectively. Everyday we measured the weight of the nude mice and observed their ingestion, movement and mental status. The nude mice were killed after 45 days, and the effect of NGX6 on the malignant behavior of HT-29 was assessed by this experiment.
RESULTS:
In contrast to the other two groups, the metastasis in the liver and xenograft tumor in the spleen of pcDNA3.1(+)/NGX6/HT-29 group was significantly reduced (P<0.01).
CONCLUSION
The metastasis of HT-29 colon cancer cell line was significantly inhibited by NGX6 gene. This model of liver metastasis in the nude mice is a proper model to determine the anti-metastasis mechanism of NGX6 gene.

10.Total flavonoids of bugloss limits left ventricular remodeling after myocardial infarction in mice

OBJECTIVE To investigate the effects of total flavonoids of bugloss(TFB)on left ventricular (LV)remodeling after myocardial infarction(MI),LV size and function was compared in mice subjected to left anterior descending coronary artery ligation. METHODS 28 d after MI, the infarcted fraction of the LV and LV mass, systolic and diastolic function were measured. Capillary density and myocyte width in the nonischemic portion of the LV were also determined.RESULTS 28 d after MI,both groups had dilated LVs with decreased fractional shortening and lower ejection fractions. Although the infarcted size of the LV was similar in both groups,LV end-diastolic internal diameter,end-diastolic volume,and mass were lower, but fractional shortening, ejection fraction, and the maximum rate of developed LV pressure(dp/dtmax)were greater in TFB treated mice than in control mice.Impairment of diastolic func-tion, as measured by the time constant of isovolumic relaxation (t) and the maximum rate of LV pres-sure decay(dp/dtmin),was more marked in control mice than in TFB treated mice.Mortality after MI was greater in control mice than in TFB treated mice.In control mice,capillary density and myocyte width in the nonischemic portion of the LV did not differ before and 28 days after MI, whereas in TFB treated mice, capillary density increased and myocyte width declined after MI. CONCLUSION These results suggest that the presence of TFB limits LV dysfunction and remodeling in a murine model of MI in part by decreasing myocyte hypertrophy in the remote myocardium.
