2.Study on Extraction Process of Polysaccharides from Elaeagnus angustifolia L. by Enzymatic- assisted Ultrasonic Methods

Objective To optimize the technological conditions of polysaccharide extraction from Elaeagnus angustifolia L. by enzymatic-assisted ultrasonic. Methods The effects of different enzymes, enzymolysis temperature, enzyme dosage, enzymolysis time, material-water ratio, crushing degree, ultrasonic time and ultrasonic temperature on the extraction rate of Elaeagnus angustifolia L. polysaccharide was analyzed. Results The optimal technological condition was as follows:1.5% cellulase, 1.5% pectic enzyme, enzymolysis time 40 min, enzymolysis temperature 55 ℃, ultrasonic time 25 min, ultrasonic power 400 W, material- water ratio 1∶30, and temperature 60 ℃. In these conditions, the extraction efficiency of polysaccharide was 12.35%. Conclusion The process is simple and practicable, and can be used for the extraction of Elaeagnus angustifolia L. polysaccharide.
3.Laparosocopically assisted treatment of lower digestive tract hemorrhage in children

Objective To explore the value of laparoscopic diagnosis and treatment for Meckel’s diverticulum in children. Methods Laparoscopic exploration was performed in 11 children with suspected intestinal Meckel’s diverticulum. In other 2 children, the diverticulum was encountered during laparoscopic appendectomy, and then the umbilical incision was prolonged to take out the corresponding intestine for resection. Results No conversions to laparotomy were required in all the 13 children. The operation time lasted 30~90 min (mean, 75 min). The children were discharged at 5~6 postoperative days without complications. Conclusions Laparoscopic diagnosis and treatment for Meckel’s diverticulum in children is effective.
4.Application of methyl in drug design.

The methyl group plays an important role in the rational drug design. Introducing methyl into small molecules has become an important strategy of lead compound optimization. The application of methyl in drug design is reviewed in this paper. Methyl can modulate the physicochemical, pharmacodynamic, and pharmacokinetic properties by ortho effect, inductive effect, and conformational effect. It also improves the metabolic stability as a soft metabolic point. In addition, introducing methyl into drug molecules can also be applied as a strategy in new uses of old drugs and generate me-too drugs.
5.Investigation of KGF and KGFR expression in external auditory canal skin adjacent to tympanic membrane perforation

OBJECTIVE To investigate the continuous expression of keratinocyte growth factor(KGF)and keratinocyte growth factor receptor(KGFR)in external auditory canal skin adjacent to tympanic membrane perforation and analyze the role of KGF and KGFR in the different turnover of chronic otitis media.METHODS The external auditory canal skin adjacent to tympanic membrane perforation from 20 cases with cholesteatoma otitis media and the corresponding cholesteatoma tissue and normal external ear skin were examined by immunohistochemical S-P method and quantitative analysis.The positive rate was compared with 20 cases of non-cholesteatom otitis media's external auditory canal skin adjacent to tympanic membrane perforation.RESULTS The staining for KGF and KGFR in cholesteatoma otitis media's external auditory canal skin adjacent to tympanic membrane perforation was consistently stronger than that in non-cholesteatoma otitis media.The positive rates of the two tissue was(33.135?6.364)% and(19.965?10.570)%,(19.380?2.827)% and(13.145?7.935)% respectively,revealing a significant difference.CONCLUSION The activity of hyperproliferation of the external auditory canal skin adjacent to tympanic membrane perforation in cholesteatoma otitis media is stronger than that of non-cholesteatoma otitis media.KGF and KGFR may play a more important role for hyperproliferation of cholesteatoma.
6.Impact of transplant nephrectomy on panel reactive antibodies

Objective To observe the impact of transplant nephrectomy on panel reactive antibodies and a secondary renal transplantation.Methods Panel reactive antibodies in 15 patients with a failed renal transplant admitted in our hospital between 2004 to 2007 were measured before transplantation,before and 1 month,6 months,12 months after transplant nephrectomy,and the pathological changes were observed after transplant nephrectomy.Results Panel reactive antibodies was increasing after renal transplantation,and reached the highest level one month after transplant nephrectomy,then grandually got down.New HLA allosensitization sites were discovered after transplant nephrectomy.Large amount of C4d was stained in failed transplant.Conclusion Serum PRA increased after transplant nephrectomy.New HLA allosensitization sites were found,which may be useful in HLA matching.
7.Study on Extraction Method for Assaying Jinyou Tankeqing Tablets

Objective To optimize the extraction method for assaying Jinyou Tankeqing Tablets(JTT).Methods With the total flavonoids content as the index,Orthogonal Experimental Design was used to optimize the extraction condition for assaying JTT.Results The optimized ultrasonic extraction method was as follows: adding 0.2 mL?mg-1 powder to 95 % ethanol,extracting for 30 min;the optimized Sohlex extraction method was: adding 1.2 mL?mg-1 powder to 95 % ethanol,extracting for 2 h,the former method being better than the later.Conclusion The optimized extraction method for assaying JTT is adding 0.2 mL?mg-1 powder to 95 % ethanol and extracting for 30 min with ultrasonic wave.
8.Inhibitory effect of SIN-1 on ICAM-1 expression in rat kidney after ischemia/reperfusion injury

Objective To explore the therapeutic mechanisms of nitric oxide donor (SIN 1) in ischemic acute renal failure (IARF). Methods The effects of SIN 1 on intercellular adhesion molecule 1 (ICAM 1) expression and inflammatory cell infiltration in rat kidney after ischemia/reperfusion (I/R) injury were examined using ICAM 1, ?2 integrin polyclonal antibody, and immunohistochemistry. The renal functions were measured simultaneously. Results Progressive increased expression of ICAM 1 was found in kidneys, especially in vasa recta, after IR injury. ?2 integrin positive cells increased simultaneously in the outer medulla. The two indexes reached the peak values at 24 h after I/R injury. SIN 1 infusion at the beginning of reperfusion could remarkably inhibit the enhanced ICAM 1 expression, reduce the local infiltration of inflammatory cells, and ameliorate the renal functions. Conclusion NO donor can inhibit the renal expression of ICAM 1 and the infiltration of leukocytes after ischemia reperfusion injury, which may play an important role in the treatment of ischemic acute renal failure.
9.Adsorption and Desorption of Licorice Flavonoid with Macroporous Resins

Objective To study the adsorption and separation of licorice flavonoid with macroporous resins. Methods Eight types of macroporous resin were selected to compare their performances in absorbing and desorbing licorice flavonoid. The optimal type for licorice flavonoid was decided, meanwhile, its kinetic curve and dynamic absorbing behavior were studied. Results HPD300 resin possessed higher adsorption and desorption capacity. The appropriate adsorption and desorption conditions were as follows:concentration of sample was 2.0 mg/mL, velocity of sample solution was 1.5 BV/h, volume of sample solution was 2 BV (bed volume);velocity of 80%ethanol was taken as eluant 1.5 BV/h, and the volume was 3 BV. Flavonoid content was increased more than 2 times under above conditions. Conclusion HPD300 macroporous resin showed better comprehensive adsorption property. It can be used to purify and separate licorice flavonoid.
10.Effect of Proprioception Disturbance on Activities of Daily Living after Stroke

ObjectiveTo observe the effect of proprioception disturbance on the activities of daily living (ADL) after stroke.Methods67 stroke patients were divided into group A (32 cases with proprioception disturbance) and group B (35 cases without proprioception disturbance) according to if having proprioception disturbance. All patients of two groups were treated with routine medicine and rehabilitation treatment. Modified Barthel Index (MBI) was used to assess ADL when admission and 8 weeks later.ResultsIn all 67 stroke patients, the incidence of proprioception disturbance was 48.6%. After 8 weeks treatment, MBI scores of both groups improved significantly (P<0.05~0.01) and the average scores of group A was obviously lower than that of group B (P<0.05).ConclusionProprioception disturbance has bad affect on recovery of ADL after stroke obviously. Examination for proprioception disturbance should be performed in early stage after stroke and rehabilitation training should be carried on as soon as possible.
