Objective: To investigate the mechanism of combined growth hormone and fibrin glue on early healing of intestinal anastomosis in intra-abdominal sepsis rats. Methods: Male Wistar rats of abdominal sepsis were randomized into 3 groups.Control rats were given intestinal anastomosis after intestinal fistula operation.Glue group was given intestinal anastomosis and fibrin glue after intestinal fistula.GH group was given intestinal anastomosis and fibrin glue and growth hormone after intestinal fistula.FN,?-SMA,VEGF,and apoptosis in anastomoses were measured. Results: Expression of intestinal FN,?-SMA,and VEGF in GH group were higher than those in other groups and apoptosis were lower than others. Conclusion: It is indicated that combined growth hormone and fibrin glue ameliorates healing of intestinal anastomosis in intra-abdominal sepsis rats.

Objective: To investigate the mechanism of intestinal anastomosis in a rat model of intra-abdominal sepsis. Methods:Male Wistar rats were randomized into 2 groups.Control rats underwent intestinal anastomosis.Sepsis group underwent intestinal anastomosis after intestinal fistula operation.fibronectin(FN),?-smooth muscle actin(?-SMA),vascular endorthelial growth factor(VEGF) and apoqtosis in anastomoses were measured.Results:FN,?-SMA,VEGF in control group were higher than sepsis group and apoptosis were lower than the other. Conclusion:It is indicated that the repair of tissue was out of balance after infection, and the organization repair was retarded. The collagenoblast and the epithelial cell migration were slowed down, and the blood capillary regeneration was slowly. Not only the granulation organization was reduced, but also apoptosis was prematurely appeared.

Journal of Applied Clinical Pediatrics 2006;0(23):-
Objective To summarize the treatment results, operative complications and experiences of laparoscopic modified pull-through(Soave)precedure.Methods Twenty children with Hirschsprung disease(HD),16 males and 4 females, with age from 3.5 months to 4 years old, underwent laparoscopic modified pull-through(Soave)precedure under whole anesthesia and tracheal intubation, and the pathological colon was freed by ultrasonic surgical knife. Results All cases were operated on successfully. The average operating time was 132 min.The average resection colon was 35 cm and the average volume of bleeding was about 14 mL without blood transfusion. Two patients had postoperative enterocolitis;3 patients were no complications postoperation, such as constipation, soiling, intestinal obstruction, preianal intestine prolapse or cuff narrowing, and all patients had 1-2 bowel movements per day during a period of 6 months to 3 years follow-up.Conclusion Laparoscopic modified pull-through(Soave)precedure has smaller incision, less bleeding, quick recovery and less complications.

Danshen is one of the traditional Chinese herbal medicines and nas a long history or being used clinically in the treatment of cardiovascular and cerebrovascular conditions such as coronary heart disease and angina pectoris. Tanshinone IIA is a derivative of phenanthrene-quinone isolated from Danshen. It has been reported to be the major bioactive compound of Danshen and has diverse biological effects. Recent studies demonstrated that tanshinone IIA had neuroprotective effects on experimental ischemic stroke through its antiinflammatory, anti-oxidant, anti-apoptosis effects and its inhibitory effect on excitatory amino acid toxicity. In this review, we summarized all the recent progresses on the protective effect of tanshinone IIA on cerebral ischemic stroke. Hopefully, this article will throw some light on further study and application of tanshinone IIA.

Background Oxidative stress is a pathophysiological process of retina,so it is very important to explore a protective way against retinal oxidative stress.Studies determined that extract of ginkgo biloba (EGb) has antioxidant,anti-apoptosis,anti-thrombosis and anti-inflammatory effects, however, the effect of EGb on human Müller cells in oxidative stress is still below understood. Objective This study was to investigate the protection of EGb against oxidative stress of human retinal Müller cells induced by As2O3 in vitro. Methods Human retinal Müllercell line was cultured in DMEM/F12 with 10% fetal bovine serum (FBS),2 μmol/L glutamine and 1% antibiotics.As2O3 solution at the final concentration 5 mg/L was added in the medium for 24 hours to establish oxidative models, and then the EGb with the final concentrations of 5,10 and 20 mg/L was used to cell models for 24 hours, respectively.Cell viability was detected by MTT assay, and reactive oxygen species (ROS) levels in the cells were detected with CM-H2DCFDA fluorescent probe. The relative expression levels of caspase-3 mRNA in cytoplasm and cell nuclei were assayed by reverse-transcription PCR (RT-PCR),and the expressions of Nrf2 protein were quantitatively detected by Western blot. Results Müller cells adhered well 24 hours after cultured. At 6-7 days after culture,Müller cell body was large with abundant cytoplasm and mosaic-like arrangement. However, floating cells were seen after As2 O3 treatment.Cell viability (absorbance) was significantly different among the normal culture group,As2 O3-treated group,As2 O3 + 5 mg/L EGb group,As2 O3 + 10 mg/L EGb group and As2 O3 + 20 mg/L EGb group, with the strongest viability in the normal culture group and the weakest viability in the As2 O3-treated groups (F=163.57,P =0.00).The fluorescence intensity of ROS was the weakest in the normal culture group and the strongest in the As2 O3-treated group and was gradually weakened with the increase of EGb doses, showing a remarkable difference among the groups (F =4 013.61,P =0.00).The relative expression level of caspase-3 mRNA in the cells was gradually reduced with the increase of EGb doses, with a statistically significant difference among the groups (F =2 199.72,P =0.00).In addition,no considerable difference was seen in the expression level of Nrf2 protein (grey scale) in cytoplasm among the groups (F=15.42,P=0.40);while in the nuclei, the expression levels of Nrf2 protein were 100.01 ±0.04,46.59±0.63,54.51 ±0.62,59.93 ±0.17 and 67.60±0.24 in the normal culture group,As2 O3-treated group and As2O3+5 mg/L EGb group,As2O3+10 mg/L EGb group,As2O3+20 mg/L EGb group respectively, with a significant difference among them (F=7 271.72,P=0.00).Conclusions EGb can protect human retinal Müller cells against As2O3-induced damage in a dose-dependant manner by antioxidant and antiapoptotic effects in vitro, and the activities occur primarily in cell nucleus.
10.Recent progress in researching the reactive oxygen species-mediated myocardial remodeling

Reactive oxygen species(ROS) in the cardiac muscle are increased in cardiovascular diseases such as hypertension, coronary heart disease and heart failure.Oxidative stress mediates myocyte hypertrophy, myocyte apoptosis and interstitial fibrosis. And these changes are particularly pronounced in elderly patients or senescent animal disease models. The major sources of ROS in myocardium are mitochondria and the reduced nicotinamide adenine dinucleotide phosphate(NADPH) oxidase.Specific ROS such as hydrogen peroxide play an important role in myocardial remodeling in heart failure.Oxidative modifications of calcium handling proteins result in reduction of their activities, leading to decreased myocardial contractility.Reduction of oxidant levels and inhibition of oxidative modifications of specific proteins through inhibiting ROS sources may provide new strategies for the treatment of myocardial remodeling.
