1.Expression and location of IL-2/IFN-? fusion protein in colorectal tumor cell lines

Objective If IL-2 and IFN-? are united,perhaps we may see a more significant antitumor effect.A fusion gene of IL-2 and IFN-? was designed and constructed by DNA engineering.Methods CEA promotor、IL-2cDNA and IFN-?cDNA were obtained by PCR and RT-PCR.SDS-PAGE was used to examine the results;pcDNA-IL-2、pcDNA-IFN-?、pCEA-IL-2/ IFN-?、pEGFP-IL-2/ IFN-? were transfected to CEA high expression cell line(Lovo)by use of liposome.The fusion protein expression was examined by fluorescence microscope and Western blotting.Results IL-2/ IFN-? fusion gene can be expressed in tumor cells.Its expression is more obvious in CEA high expressed cells(Lovo),and can be located in cell plasma.Conclusion The fusion gene can be expressed in plasma of CEA high expressed cell lines.It may have a new anti-tumor effects.
2.Update on the research of diabetic dyslipidemia

Dyslipidemia is one of the major risk factors for cardiovascular disease in diabetes mellitus,but clinical practices in the management of diabetic dyslipidemia are still not satisfactory in China.The cornerstone of treatment for diabetic dyslipidemia consists of therapeutic lifestyle change and statins.Recent guidelines emphasize aggressive low density lipoprotein-cholesterol control with statins.However,intensive statin therapy may increase the risk of myopathy and diabetes.Therefore,it is important to consider its risk benefit ratio in individual patients.The additional benefit of statins in combination with other hypolipidemic drugs on cardiovascular risk needs verifying by further investigation.
3.Expression regulation of Bnip3 and its relationship with tumors

Bnip3,a kind of mitochondrion apoptosis gene,belongs to the Bcl-2 gene family and BH3-only subfamily.It is one of the downstream response factors of hypoxia inducible factor (HIF).Bnip3 can induce cell apoptosis and autophagy under the oxygen deficit condition.Many studies show that Bnip3 is closely related to the occurrence,development and treatment of tumors.Molecular targeting treatment aimed at Bnip3 combined with chemoradiotherapy has preferable application foreground in tumor therapy.
4.Effects of different target concentrations of remifentanil when combined with propofol on BIS in patients undergoing neurosurgery

Objective To investigate the effects of different target concentrations of remifentanil when combined with propofol on BIS in patients undergoing neurosurgery. Methods Fifteen ASA Ⅰ or Ⅱ patients, aged 18-64 yr, weighing 50-85 kg, scheduled for intracranial surgery, were involved in this study. The patients received propofol by target controlled infusion (TCI) at a target effect site concentration (Ce) of 3μg/ml. TCI of remifentanil was started after TCI of propofol reached the preset Ce and Ce of remifentanil was increased step by step from 2 ng/ml to 3, 4, 5, 6, 7 and 8 ng/ml. Invasive BP, MAP, HR and BIS were continuously monitored and recorded when remifentanil reached the each preset Ce. Vecuronium 0.1 mg/kg was injected iv when Ce of remifentanil reached 5 ng/ml, and then the patients were intubated and mechanically ventilated 3 min later. The changes in BP, MAP, HR and BIS were recorded. When HR < 50 bpm and/or MAP < 60 mm Hg, TCI of remifentanil was stopped and iv atropine or ephedrine was given. Results BIS was significantly decreased after TCI of remifentanil when propofol reached 3 μg/ ml compared with the baseline value ( P < 0.05 or 0.01). BIS was significantly decreased when Ce of remifentanil ≥6 ng/ml compared with that when propofol reached 3 μg/ml ( P < 0.05 or 0.01) .Conclusion Low-concentration of remifentanil when combined with propofol has no effect on BIS, but when Ce of remifentanil≥ 6 ng/ml, BIS is obviously decreased.
5.Therapeutic Observation of Cluster Needling at Anterior Oblique Line of Vertex-temporal plus Speech Training for Motor Aphasia after Cerebral Infarction

Objective To observe the clinical efficacy of cluster needling at the anterior oblique line of vertex-temporal plus speech training in treating motor aphasia after cerebral infarction.Method Forty patients with motor aphasia after cerebral infarction were randomized into a treatment group and a control group, 20 in each group. Both groups were given basic neurological treatment. In addition, the treatment group was intervened by cluster needling at the anterior oblique line of vertex-temporal plus speech training, while the control group was by speech training alone. After 4-week treatments, the speech function and communication ability were observed and the clinical efficacies were compared.Result The total effective rate was 90.0% in the treatment group versus 80.0% in the control group, and the difference was statistically significant (P<0.05). After treatment, the speech function score and Chinese Functional Communication Profile (CFCP) score were changed significantly in both groups (P<0.01). There were significant differences in comparing the speech function and CFCP scores between the two groups (P<0.01,P<0.05).Conclusion Cluster needling at the anterior oblique line of vertex-temporal plus speech training is an effective approach in treating motor aphasia after cerebral infarction.
6.Bone marrow mesenchymal stem cells combined with peptide hydrogel and chondrogenic factors for repair of articular cartilage defects in rabbits

BACKGROUND:Peptide hydrogel has good plasticity, and it can fil the injured site very wel; therefore, to use this material as a scaffold is a feasible exploration in bone and cartilage tissue engineering. OBJECTIVE:To test the effect of bone marrow mesenchymal stem cels combined with injectable peptide hydrogel and chondrogenic factors for repair of articular cartilage defects in rabbits. METHODS:Bone marrow mesenchymal stem cels from rabbits were isolated and cultured. A ful-thickness bone-cartilage defect model, 5 mm in diameter and 3 mm in depth, was made on the left knee joint of rabbits, and the right knee joint of rabbits with no treatment was used as control after modeling. There were three experimental groups: self-assembling peptide hydrogel group, peptide hydrogel+chondrogenic factors group, and peptide hydrogel+chondrogenic factors+bone marrow mesenchymal stem cels group. Transforming growth factor β1, dexamethasone and insulin-like growth factor 1 were mixed as chondrogenic factors and added into self-assembling peptide hydrogel or bone marrow mesenchymal stem cels. Twelve weeks after treatment, animals were sacrificed for gross and histological observation, X-ray radiography, and histological evaluation using immunohistochemistry method. RESULTS AND CONCLUSION:At 12 weeks after treatment, the self-assembling peptide hydrogel group showed excelent results in the cartilage repair, and better achievements in safranin-O staining, colagen II immunostaining, and histological scores than the other groups (P < 0.05); the peptide hydrogel+chondrogenic factors group had better repair effects similar to the self-assembling peptide hydrogel group, but the expression of proteoglycans was higher than that in the control group (P < 0.01); there were poorer repair effects and more osteophytes in the peptide hydrogel+chondrogenic factors+bone marrow mesenchymal stem cels group than the the peptide hydrogel+ chondrogenic factors group. Experimental findings indicate that the self-assembling peptide hydrogel can repair cartilage defectsin situ and improve cartilage repair, which is expected to improve current repairing effects on cartilage defects.

In the past decade, liquid chromatography-mass spectrometry has undergone rapid development in clinical laboratory medicine.Benefiting from the high sensitivity and specificity, as well as the capability of multi-component analysis in the single run, such technology is regarded as an excellent alternative to conventional immunoassays towards newborn screening for the genetic diseases, therapeutic drug monitoring and endocrine hormones detection.With the continuing progress in expertise-training, quality assurance and methodology standardization, it could be expected that liquid chromatography-mass spectrometry would become a dynamic and crucial part for routine clinical applications in the near future.

10.The value analysis on serum homocysteine and cystatin C in patients with cerebral infarction

Objective To analyze clinical Value of serum homocysteine(Hcy)and cystatin C(Cys C) in patients with cerebral in‐farction .Methods From August 2010 to December 2014 ,our hospital 118 patients with cerebral infarction and 62 healthy subjects for the study were divided into the control group and the observation group .The observation group was further divided into light , medium and heavy groups .Test results for the two groups were related to the study of serum homocysteine and Cys C were ana‐lyzed separately and compared between two groups of Hcy levels Cys C contact with cerebral infarction .Results Serum Hcy ,Cys C levels significantly higher in the observation group than that of control group ,differences were statistically significant(P<0 .05);The serum Hcy ,Cys C of patients with severe cerebral infarction group were significantly higher than that of light and medium cer‐ebral infarction group;Serum Hcy ,Cys C levels of patients with medium cerebral infarction group were also significantly higher than the light cerebral infarction group ,differences were statistically significant(P<0 .05) .Conclusion The serum Hcy and Cys C has important clinical value of detecting the disease assessment and treatment of patients with cerebral infarction .
