1.Clinical significance of detection of GDF3,PSA combined with MUC1 for diagnosis of breast cancer

Objective To investigate the clinical significance of detection of growth differentiation factor 3(GDF3), prostate specific antigen(PSA) combined with mucin 1(MUC1)for diagnosis of breast cancer.Methods50 cases of breast cancer were selected as the breast cancer group, 50 cases of benign breast disease were selected as the benign breast disease group, and 50 healthy people were selected as the normal group.GDF3, PSA, MUC1 and tumor markets carbohydrate antigen(CA)15-3,CA125,carcinoembryonic antigen(CEA) were detected, and sensitivity, specificity and accuracy were analyzed.ResultsCompared with benign breast disease group and normal group, GDF3, PSA and MUC1 in breast cancer group were higher(P<0.05), CA15-3, CA125 and CEA were higher(P<0.05).The sensitivity, specificity and accuracy of PSA+MUC1 were higher than other group(P<0.05),the sensitivity, specificity and accuracy of combined detection of GDF3, PSA and MUC1 were higher(P<0.05).ConclusionDetection of GDF3 combined with MUC1 and PSA for early diagnosis of breast cancer has more diagnostic significance.

OBJECTIVE: To provide reference about the way to consummate hospital drug quality management system and ensure drug quality and patients' medication safety.METHODS: Efficient drug maintenance measures were explored by analyzing and summarizing the specific work and conservation procedures of drug maintenance in our hospital which included the humidity of storehouse,monitoring of storage condition,color-code management,expiration date management etc.RESULTS & CONCLUSIONS: Drug maintenance should be aimed at guaranteeing the drug quality;moreover,great importance should be attached to regular quality examination and maintenance of drugs and take timely effective measures aimed at the problems presented so as to ensure the drug quality,safety and effectiveness.
3.Mirena IUD on menstrual patterns in patients with adenomyosis and sexual quality of life in clinical research

Objective To investigate the effect on the menstrual patterns and sexual quality of life of patients with adenomyosis after treated with Mirena IUD.Methods56 patients with adenomyosis who were treated with Mirena IUD were recruited from August 2009 to December 2009.We used Menstrual blood loss chart method,Dysmenorrhea score method and Brief Index of Sexual Function for Women (BISF-W) to evaluate the menstrual pattern,the volume of uterus and sexual quality after1,3,6,9,12 months.{Results53 patients were followed up,the rate was 94.6%.The condition of 56 patients became better after treated with Mirena for 3,6,9,12 months,dysmenorrhea[Placed before:(3.15±0.91)points;After placing:(1.41±0.51)points,(0.22±0.09)points,(0.07±0.01)points,0 points]and menstrual quantity[Placed before:(12.0±57.5)points;After placing:(71±23.1)points,(29±19.4)points,(18±10.5)points,(10±4.7)points]were both reduced(P<0.05),the symptoms were not significantly improved after 1 month(P>0.05).D2 indicators was significantly improved after 3 months of treatment (P<0.05),sexual function was also improved after 6 months(P<0.05~0.01).ConclusionsMirena IUD has a good effect on the menstrual pattern and sexual function for patients with adenomyosis.
5.Prenatal genetic diagnosis for phenylketonuria families by combination of linkage analysis and mutation screening

Objective To explore the prenatal genetic diagnosis for classic phenylketonuria (PKU) families.Methods Probands and their family members from three classic PKU families were analyzed by combining linkage analysis through short tandem repeats (STR) polymorphism and PCR-sequencing for the exons within mutation hot spot of phenylalanine hydroxylase gene.Results Linkage analysis found uninformative for Family 1,while 100 % confirmative information was obtained from Family 2 and 3.Sequencing showed compound heterozygous mutations of phenylalanine hydroxylase gene for all of the three probands.Five mutations were detected,namely Y166X,R243Q,R413P,EX6-96A > G and IVS11-1G> C,and IVS11-1G > C was a novel identified muntation.Information from linkage analysis and mutation screening showed clearly that the fetus of Family 1 and 2 were affected,while normal for Family 3.Conclusions For those PKU families,reliable service of prenatal genetic diagnosis could be provided by combining linkage analysis with mutation screening of phenylalanine hydroxylase gene.
6.Discussion of the optimal time to remove urinary catheter in patients undergoing laparoscopic adrenalectomy

Objective To investigate the optimal time to remove the urinary catheter in patients undergoing laparoscopic adrenalectomy. MethodsPatients undergoing laparoscopic adrenalectomy were assigned to group A (6 h), group B ( 12 h), group C ( normal control group) with 20 cases in each. Subject feeling, adverse reactions, postoperative bed time and days of absolute diet were compared. Results The incidence of bacteriuria in group A (35%) and B (30%) were significantly lower than that of group C (85%)(F=14. 80, P =0. 000). The incidence of hematouria in group A (25%) and B (10%) were also significantly lower than that of group C ( 80% ) ( F =22. 98, P =0. 000). Conclusions 6 - 12 hours after laparoscopic adrenalectomy are the optimal time to remove the catheter. In this manner, it can reduce the incidence of bacteriuria and hematouria, promote the postoperative recovery, thus to reduce the cost of hospitalization and improve the nursing work efficiency.
7.A novel mutation of GJB6 in Chinese sporadic non-syndromic hearing impairment

OBJECTIVE To investigate the contribution of the GJB6 gene [encoding connexin 30 (C?30)] mutations in Chinese population with sporadic non-syndromic hearing impairment. METHODS PCR reactions were performed with two pair of primers for the coding sequence of GJB6 gene and for the deletion of GJB6. PCR products bidirectional sequencing was subsequently applied in 214 patients with hearing loss and 86 normal controls. RESULTS A novel heterozygous mutation-233(C→A) was found, which results in amino acid change, A78D. This mutation wasn't detected in the control subjects. The altered valine residue lies within the second conserved transmembrane domain. The large deletion△(GJB6/ D13S1830)] of GJB6 was not found in this group. CONCLUSION The large deletion of GJB6 was not found in the Chinese deafness population. A novel heterozygous mutation of GJB6 was found. These results indicated GJB6 mutations are not a major cause of hearing loss in the Chinese population.
8.Detection of osteopontin in sera,urine and renal tissue in patients with systemic lupus erythematosus and its significance

Objective To investigate the expression of osteopontin (OPN)in serum,urine and renal tissue of patients with systemic lupus erythematosus (SLE)and its relevance with organ damage in and activity of this disease.Methods Enzyme-linked immunoabsorbent assay(ELISA)was applied to detect the concentration of OPN in the sera of 100 patients with SLE and 30 sex-and age-matched normal human controls as well as in the urine of 57 patients with SLE and 15 normal human controls.Renal tissue was obtained from 3 patients with lupus nephritis and subjected to immunohistochemistry for the observation of OPN.Results The level of OPN was significantly higher in the sera and urine of patients with SLE than in those of normal human controls (64.03 ±72.87 μg/L vs 29.88±1 1.28μg/L,454.87±231.63 μg/L vs 122.67±39.47μg/L,both P<0.05).Increased level of OPN in sera and urine was also observed in patients with active SLE com-pared with those with inactive SLE (80.92±87.49μg/L vs 36.43±23.48μg/L,584.36±207.15 μs/L vs 28 1.08±1 3 1.92μg/L,both P<0.05).A positive correlation was noted between the serum level of OPN and SLE disease activity index(SLEDAI)(r=0.462,P<0.01),and the level of urine OPN was positively correlated with both SLEDAI and the concentration of urine immunoglobulin G.urine mierualbumin.urine α1-microglo-bulin,urine β2-microglobulin(r=0.901,0.458,0.359,0.342,0.409,respectively,all P<0.05).OPN was found in renal tubule epithelia of the three patients with lupus nephritis.Conclusion OPN may be involved in the pathogenesis of SLE and associated with the renal damage in patients with lupus nephritis.
9.Expression of cyclooxygenase-1 in ovarian cancer and its significance

Objective To explore the expression of cyclooxygenase-1 in ovarian cancer and its significance. Methods Expression of COX-1 and CA125 was detected by immunohistochemistry in 37 cases of ovarian cancer and 31 cases of ovarian cyst. The serum level of COX-1 and CA125 was tested in 40 cases of ovarian cancer and 31 cases of ovarian cyst and 60 healthy volunteers by ELISA. Results The positive expression of COX-1 and CA125 was 78% and 57% in ovarian cancer and 3% and 7% in ovarian cyst,respectively. The expression of COX-1 and CA125 was 65% and 93% at the serum level. Conclusion Expression of COX-1 could act as an auxiliary diagnostic criterion in ovarian carcinoma.
10.Comparison of development of gastric insufflation related to different peak inspiratory pressures during facemask ventilation in pediatric patients: ultrasonographic measurement

Objective To compare the development of gastric insufflation related to different peak inspiratory pressures (PIPs) during facemask ventilation in the pediatric patients.Methods Ninety male pediatric patients,aged 2-4 yr,of American Society of Anesthesiologists physical status Ⅰ,scheduled for elective surgery under general anesthesia,were randomly divided into 5 groups (n =18 each) using a random number table:PIP 8 cmH2O group (group P8),PIP 10 cmH2O group (group P10),PIP 12 cm H2O group (groupP12),PIP 14cmH2O group (group P14) and PIP 16 cmH2O group (group P16).Anesthesia was induced with fentanil,propofol and rocuronium in sequence.After loss of eyelash reflex,positive pressure facemask ventilation was performed for a 120 s period in pressure-controlled mode.Gastric insufflation was detected by real-time ultrasonography of the antrum,and cross-sectional antral area was measured using ultrasonography before facemask ventilation and at 120 s of facemask ventilation.The pulse oximetry (SpO2),tidal volume (VT),end-tidal pressure of carbon dioxide (PETCO2) and end-tidal oxygen concentration (ETO2) were recorded at 30,60,90,and 120 s of facemask ventilation.The development of gastric insufflation and hypoventilation was recorded.Results Compared with group P8,the incidence of gastric insufflation was significantly increased in group P16 (P<0.01),and no significant change was found in the incidence of gastric insufflation in the other groups (P>0.05),the incidence of hypoventilation was significantly decreased,VT and ETO2 were increased,and PET CO2 was decreased in P12,P14 and P16 groups,and PETCO2 was significantly decreased at 120 s of facemask ventilation (P< 0.05 or 0.01),and no significant change was found in the other parameters in group P10 (P>0.05).Compared with P12 and P14 groups,VT was significantly increased,PEHTCO2 was decreased at 120 s of facemask ventilation (P<0.05),and no significant change was found in the incidence of hypoventilation and ETO2 in group P16 (P>0.05).There was no significant difference between group P12 and group P14 in the incidence of hypoventilation,VT,PETCO2 and ETO2 (P>0.05).The pediatric patients showed a certain CO2 accumulation [PETCO2 (40.6±4.0) mmHg] at 120 s of facemask ventilation in group P8,and the pediatric patients showed excessive ventilation [PETCO2 (23.6± 1.4) mmHg],and cross-sectional antral area was not measured using ultrasonography in three cases because of excessive gastric insufflation in group P16.Conclusion PIP at 12-14 mmHg in pressure-controlled ventilation mode can not only ensure adequate preoxygenation and but also avoid excessive gastric insufflation during facemask ventilation in the pediatric patients.
