AIM: To identify the changes in microRNA ( miRNA ) profile of human lens epithelium ( HLE) induced by H2 O2 and the role of miRNA in oxidative stress induced apoptosis.
METHODS: HLE cell line HLE - B3 was treated by 100μmol/L H2 O2 for 24h and the total RNA were isolated by Trizol reagent. miRNA profile was generated by miRCURYTM LNA microRNA Array. The target genes of differentially expressed miRNAs were predicted by bioinformatics software.
RESULTS:Twenty-eight miRNAs showed significantly differential expression after H2 O2 treatment, 18 miRNAs upregulated and 10 miRNAs downregulated. The differentially expressed miRNAs may involve in apoptosis of lens epithetium and development of cataract through targeting BCL2L2 and MIP.
CONCLUSION: H2 O2 can induce dramatically changes in miRNA profile of HLE, which may play a pivotal role in the pathogenesis and development of cataract.

It is an important measure in improving the practical teaching quality at Chinese westem medical colleges to strengthen the training practice of part-time assistant teachers in clinical teaching bases. To establish a good training model,we should take into consideration its operability and practicability, as well as the positivity of the teachers.The key to making the training succeful is to ascertain the direction and contents, to set up an instructors' team, and to make a specific training plan.

BACKGROUND:Alterations in secondary hemodynamics around the stent edge can be caused by stent planted. Is there different effect of shear stress on platelet growth factors-A, B (PDGF-A, B) and basic fibroblast growth factor (bFGF) of endothelial cells around stent edge after stent planted?OBJECTIVE: To explore the effect of shear stress on growth factors of endothelial cells around stent edge.DESIGN: Observational comparative study.SETTING: Department of Cardiology, Yijishan Hospital, Wannan Medical College; Laboratory of Biomechanics (National Laboratory), Medical College of Shanghai Jiao Tong University.MATERIALS: The experiment was carried out in the Laboratory of Biomechanics, Medical College of Shanghai Jiaotong University from April to October 2006. The main reagents were detailed as follows: trypsin (Hyclon Company); M-199 medium (Gibco BRL Company); fetal bovine serum (Hangzhou Sijiqing Bioengineering Material Institute); aFGF, Heparin and Hepes (Sigma Company); thymide, Ⅷ factor monoclonal antibody (Rabbit anti-human),biotinylated horse anti-rabbit IgG and propidium lodide (Sigma Company); neonate belly band (Delivery Room of Shanghai the Fifth People's Hospital); trizol (Invitrogene Company); reverse transcription polymerase chain reaction (RT-PCR) kit (Fermentas Company); penicillin/streptomycin (AB/M), objective gene primer (Shanghai Bioengineering Company).METHODS: Traditional parallel-plate streaming cavity was modified into rectangular and gradient shear stress models;while static group was established at the same time. Two shear stress groups were subjected to 11.37 dyne/cm2 and 5.66-14.38 dyne/cm2 shear stress, respectively for 3 hours, 6 hours and 12 hours.MAIN OUTCOME MEASURES: The expressions of PDGF-A, B and bFGF mRNA of human umbilical endothelial cells (HUVECs) were measured by RT-PCR at different time points.RESULTS: Compared with static group, the maximal expression of PDGF-A, B and bFGF mRNA was reached peak at 3 hours in gradient shear stress group and this level of PDGF-A mRNA expression remained till 12 hours. in rectangle shear stress group, the peak value of mRNA expression of these growth factors was all reached at 6 hours, and then gradually decreased.CONCLUSION: Over-expressions of PDGF-A, B and bFGF induced by gradientshear stress after stent planted may play an important role in stent restenosis through intimal hyperplasia.

Object To study the effect of melittin on growth and proliferation of hepatocarcinoma cells. Methods Hepatocarcinoma cell line SMMC7721 was treated by melittin. The growth and proliferation was observed by cell counting and MTT assay; necrosis was estimated by Trypan blue staining. The expression of proliferating cell nuclear antigen (PCNA) and cycle phase were detected by flow cytometer. Results Melittin could significantly inhibit the growth of SMMC7721 and block up cells progress through G 2/M. The percentage of PCNA positive cell was decreased gradually when concentration of melittin was increased. Conclusion Melittin could inhibit the proliferative activity of hepatocarcinoma cell line SMMC7721 and reduce of PCNA positive cell expression and arrest cells in S phase.

0.05),that there was negative expression of PP2Ac protein in the cytoplasm of the human lung squamous carcer and small cell lung cancer while positive expression of PP2Ac protein in intercellular substance,and that there were negtive expression of PP2A in human lung adencarcinoma and overexpression of p-AKT protein in human lung adencarcinoma.Western blot technique analysis showed that there was nomal expression of PP2A protein in normal human lung tissue,and lower expression in YTLMC-90 cell than in nomal human lung tissue.While sharp low expression GLC-82 cell.Adding okadaic acid(OA),an inhibitor of PP2A,the expressiong of p-AKT in GLC-82 cells was sharply higher than in normal lung tissue at 6 h,12 h and 24 h,indicating that inverse correlation between PP2A and p-AKT protein expression.Conclusions PP2A is involved in human lung carcinomar,its expression level is related to histological type of human lung carcer.Activition of AKT(p-AKT)is related tightly to human lung carcer.

Objective To systematically assess the efficiency of colonoscopy screening for the asymptomatic elderly.Methods A search was performed to identify studies related to colonoscopy screening for the aged in Pubmed and Embase.The key words used for the search included colorectal neoplasm, adenoma, screening, elderly, and colonoscopy.The detection rate of colorectal adenomas, advanced adenomas,colorectal cancer in the elderly and non-elderly were extracted.The studies were evaluated and analyzed by software RevMan 5.3.Results Twelve studies involving 312 111 cases were included(25 107 elderly and 287 004 non-elderly).The total AADR of the elderly was 9.97％ ,ADR was 11.33％ ,while CRC detection rate Was 1.79％.The meta-analysis showed that there were significant differences in the AADR(RR =1.57,95％ CI: 1.51-1.64), ADR(RR =1.22, 95％ CI: 1.19-1.25) and CRC detection rate(RR =2.6,95％ CI:2.34-2.90) between the elderly and the younger patients (P ＜ 0.000 01).The detection rate of AADR,ADR, CRC in the elderly were significantly higher than the non-elderly.Conclusion The elderly have higher AADR and CRC detection rate than the younger.But further studies are needed because life expectancy and complications are factors that need to be considered for screening.
