Objective To investigate the expression and prognostic significance of p27kip1 in patients with stage I and Ⅱ colorectal carcinoma ( CRC). Methods We retrospectively analyzed the expression of p27kip1 by immunohistochemistry in paraffin-embedded specimens from 114 consecutive patients operated for stage I and Ⅱ CRC. Statistical analysis was done with SPSS10. 0 package. Results The rate of p27kip1 over-expression in stage I and ⅡCRC was 85. 1 % (97/114). The expression of p27kip1 was statistically associated with TNM stage and histologic grade, while there was no significant relationship between p27kip1 expression and patient's age, tumor location. The 5-year disease free survival rate of the patients with p27kip1 over-expression was significantly higher than those with p27kip1 low-expression (88. 4% vs. 52. 9% , P

Objective To study the relationship between pain symptoms and the clinico-pathological features of pelvic endometriosis (EM).Methods One hundred thirty patients with laparoseopic diagnosis of EM were studied retrospectively and the relationship between pain symptoms including dysmenorrhea, chronic pelvic pain(CPP),dyspareunia and dysehezia and the anatomical features of pelvic endometriosis were evaluated. Results One hundred (76.9%)patients with pain symptoms and 30 (23.1%)without were included in this study. The number of patients with mild, moderate and severe dysmenorrhoea was 27 (20.8%),41(31.5%),and 32 (24.6%),respectively.Patients with dyspareunia,CPP and dyschezia were 46(35.4%),45(34.6%) and 67(51.5%),respectively.Compared with patients without dysmenorrhea, the proportion of deep utero-sacral nodules (45.0% vs 13.3%,P=0.00),recto-vaginal nodules (16.0% vs 0,P=0.01),complete obliteration of eul-de sac (41.0% vs 10.0%,P=0.00),and lesions of DIE (51.0% vs 16.7%,P=0.00) was significantly increased in patients with dysmenorrhea. The severity of dysmenorrhea was positively correlated with nodules in uterosacral ligaments (P=0.005,r= 0.302),and invasive depth of uterosacral ligaments (P=0.016,OR=5.085).Among patients with endometrioma, significantly more moderate to severe adhesions were found in patients with dysmenorrhea , compared with those patients without dysmenorrhea(29.1% vs 8.3%,P=0.029).Patients with CPP had more nodules in the utero-sacral ligaments(51.1% vs 30.6%,P=0.018)and DIE lesions(57.8% vs 35.3%,P=0.011),compared with those without.More nodules in the utero-saeral ligaments(46,3% vs 28.6%,P=0.028),recto-vaginal nodules(19.4% vs 4.8%,P=0.01),complete obliteration of cul-de sac(44.8% vs 22.2%,P=0.005)and DIE lesions(53.7% vs 31.7%,P=0.01)were found in patients with dyschezia, compared with those without.Nodules in the recto-vaginal pouch were an independent risk factor of dyspareunia. Conclusion Pain symptoms including dysmenorrhea, dyspareunia, chronic pelvic pain, and dysehezia are remarkedly related to endometriotic nodules at the posterior part of the pelvis or those with deep invasions.

Objective To determine the expression of G-protein coupled receptor 34 (GPR34) in hepatocellular carcinoma tissues. Methods Immunohistochemistry and tissue-array were used to determine GPR34 expression in human hepatocellular carcinoma tissues and matched adjacent tissues.A statistical analysis was performed to establish the potential correlation between GPR34 expression and the patients'clinicopathological characteristics, tumor progression, and prognosis. Results GPR34 is up-regulated in primary hepatocellular carcinoma tissues compared with matched adjacent tissues (P =0.003),and it was shown that GPR34 expression is significantly correlated with tumor size (P =0.024),small hepatocellular carcinoma(0.030) and infiltration(P =0.012),but not with sex, age, grade, chnical stage (all P >0.05) ; it was also shown that GPR34 expression had a significant influence on prognosis (X2 =5.617,P=0.018).Multivariate analyses showed that high GPR34 expression is an independent poor prognostic factor for overall survival (P =0.037).Conclusions The up-regulation of GPR34 acts as an potential prooncogene in the development and progression of hepatocellular carcinoma.GPR34 may be a useful diagnostic or prognostic molecular biomarker, and a potential target for therapeutic intervention.

OBEJECTIVE Gecko has been clinically used in China for many years. It has been proved that the gecko polypeptide mixture(GPM)extracted from gecko could inhibit the growth of multiple types of tumor cells. In order to investigate the possible anti-tumor molecular mechanisms of GPM,we used RNA-seq technology to identify the differentially expressed genes of human hepatocellular carci-noma(HCC)HepG2 cells treated with or without GPM.METHODS The HepG2 cells were treated with different concentration of GPM(0,0.1,0.2,0.3,0.4 mg·mL-1)for 6 h,12 h and 24 h,respectively.MTT assay was used to detect the viability of HepG2 cells. DAPI fluorescence staining was performed to observe nucleus morphological changes of HepG2 cells.Western blot analysis was applied to observe the expres-sion of apoptosis- related proteins and endoplasmic reticulum stress (ERs)-related proteins in HepG2 cells.Flow cytometry was also applied to detect reactive oxygen species(ROS)generation. In this report, we showed that GPM could induce HepG2 cells apoptosis and influence HepG2 cells proliferation in a dose-dependent manner. We applied many analysis methods, including differentially expressed genes analysis,Gene Ontology(GO)enrichment analysis,KEGG pathway enrichment analysis,protein-protein interaction network analysis to screen out possible molecular mechanisms.RESULTS ER-nucleus signaling pathway, cellular response to stress and apoptotic processes were identified the potential anti-cancer molecular biological process of GPM.GPM may also induce apoptosis in HepG2 cells via endoplasmic reticulum stress pathway. The GPM could induce ROS generation and up-regulate ERs-related proteins. CONCLUSION The present study revealed the potential anti-tumor mechanism of GPM.
