1.Study on application effects of new type glove constraint in intensive care unit

Objective To investigate the application effects of new-type glove constraint in intensive care unit. Methods A total of 113 eligible patients were randomly divided into two groups by random digits table method, of which 59 patients used the traditional restraint for body constraint in the control group while 54 patients applied the new-type glove constraint in the treatment group. The skin conditions of restraint position, tube coming off and the satisfaction of patients and relatives were compared between two groups. Results The incidence of skin abnormality at restraint position was 0 in the treatment group and 13.6%(8/59) in the control group. The difference had statistical significance (χ2=7.880, P<0.01). The satisfaction of patients and relatives was 33.3%(18/54) in the treatment group and 5.1%(3/59) in the control group, and the difference had statistical significance (χ2=16.619, P<0.01). Conclusions The use of new-type glove constraint may give a maximum activity space to patients;reduce patient′s psychological adverse impacts;and decrease the skin and body damages. It is worthy of clinical promotion and application.
2.Neuronal apoptosis and expression of caspase-12 mRNA and protein following focal cerebral ischemia-reperfusion in rats

Aim To study the expression of caspase-12 mRNA and protein following focal cerebral ischemia-reperfusion in rats,and explore the effect of endoplasmic reticulum pathway on neuronal apoptosis.Methods 60 male Wistar rats were randomly divided into sham-operated group and ischemic group.The middle cerebral artery occlusion(MCAO)models were established by using the intraluminal suture occlusion method,neuronal apoptosis was detected by TUNEL staining,the expression of caspase-12 protein was detected by immunohistochemical staining,the expression of caspase-12 mRNA was detected by RT-PCR method.Results In ischemic group,the number of apoptotic cells and the expression of caspase-12 mRNA and protein were gradually increased following prolonged cerebral reperfusion,reached the peak at 24 h.The number of apoptotic cells and the expression of caspase-12 mRNA and protein in ischemic group were significantly less than those of sham-operated group at all times(P
3.Assessment of right ventricular free wall longitudinal myocardial deformation using speckle tracking imaging in normal subjects.

To assess right ventricular free wall longitudinal myocardium deformation and examine the changes with normal age by speckle tracking imaging (STI), myocardial systolic peak strain (epsilon), systolic peak strain rate (SRs), early diastolic peak strain rate (SRe), late diastolic peak strain rate (SRa), the ratio of SRe/SRa were measured in the basal, middle and apical segments of right ventricular free wall in 75 healthy volunteers (age range: 21-71 y) by STI from the apical 4-chamber view. RV longitudinal strain and strain rate were highest in the basal segment of the free wall. Older subjects had lower early diastolic strain rate (SRe) than younger subjects, but they had higher late diastolic strain rate (SRa). A negative correlation between age and the ratio of SRe/SRa was found in all RV free wall segments (r=-0.466 - -0.614, P<0.01). It is concluded that RV diastolic strain rate changes with age and STI can be used for the study of RV myocardial deformation.

5.The effects of Newcastle disease virus on human leukemia in vitro

Objective To study the effects of Newcastle disease virus (NDV) on human leukemia in vitro. Methods The density and morphologic change of cells infected by NDV was observed. MTT assay was used to investigate the effect of NDV on the proliferation of the leukemia cells. Results The cells of the control were all regular and coarctate by inverted microscope. The effect of NDV were conspicuous in that the cells were irregular, sparsate, aggregate and fused in the experimental groups. MTT assay showed that the proliferation of SHI-1 cells were significantly inhibited by NDV with different concentration (P <0.01). The results also demonstrated that the difference within the concentration and the time were both statistically significant (P<0.01). The inhibition of NDV was further confirmed in a time-concentration-dependent manner.Conclusion Newcastle disease virus can inhibit human acute monocytic leukemic cell proliferation in vitro.The safety of NDV is reliable.

Isolation and purification of chemical constituents from solid culture of endophyte Chaetomium globosum in Imperata cylindrical was performed through silica gel column chromatography, gel filtration over Sephadex LH-20 and preparative HPLC. Nine compounds were obtained and their structures were determined as chaetoglobosin F(1), chaetoglobosin Fex(2), chaetoglobosin E(3) cytoglobosin A(4), penochalasin C(S), isochaetoglobosin D (6), N-benzoylphenylalaninyl-N-benzoyphenylalaninate(7), uracil(8) and 5-methyluracil(9), respectively, based on HR-MS and NMR data and comparison with literatures. Compound 7 was isolated from Chaeeomium sp. for the first time. In vitro cytotoxicity of compounds was evaluated using MTT mothed and 1,3,4 and 5 showed inhibition activity to the human cervical carcinoma cell HeLa with IC50 values of 99.43, 23.77, 97.92, 86.25 micromol x L(-1), while positive cotolocisnin Ad apno1ch alse IC50 24.33 micromol x L(-1).

9.Influence of simulated microgravity on rat islets morphology in culture

Objective To observe the influence of simulated microgravity on rat islet. Methods Isolated islet were assigned to flask-culture or bioreactor-culture. Gross structure and ultrastructure of islet were observed by transmission electron microscopy (TEM) and scanning electron microscopy (SEM). Results Islets ultrastructure on 7th day in bioreactor closely resembled fresh islets,with well-formed secretory granules and abundant mitochondria. SEM showed under microgravity islets communicating each other with cavity-like areas. Conclusions The ultrastructure of islets cultured under microgravity closely resembled fresh islets.

Objective To investigate the protective effect of losartan on acute ischemia-reperfusion(I/R)(injury) of pancreas in rats.Methods Seventy-two Wister rats were randomly divided into 3 groups:(1)(Control group);(2)Ischemia-reperfusion group:the anterior mesenteric artery and the celiac artery were(occluded) for 15 min,30min and 60min followed by 6 hours reperfusion;(3)Losartan group:losartan(40mg/kg)were administered by gavage at 12h and 1h before arterial occlusion.The pathologic changes of pancreatic tissue were observed under light microscopy;TUNEL was used to detect apoptosic of pancreatic cells;Bcl-2 expression in the pancreatic cells of rats was analyzed by immunohistochemistry technique.(Results) Losartan treatment reversed the histological abnormalities including infiltration of inflammatory cells and atrophy of acinar cells.Compared with losartan group,pancreatic tissue of I/R group exhibited increased MDA[((20.1?1.2))nmol/g and((34.9?2.6)) vs(17.9?2.1)nmol/g and(25.2?3.3)nmol/g,P
