1.Expression of caveolin-1 in osteosarcoma and its significance for clinicopathological features

Objective By detecting the expression of caveolin-1 in osteosarcoma tissues,to explore its possible roles in the occurrence,progress and metastasis of osteosarcoma and its relationship with clinicopathological features.Methods The expression level of caveolin-1 protein in 48 osteosarcoma tissues and 24 normal bone tissues were detected by immunohistochemistry of SP method.Its relationship with clinical and pathological features were also analyzed.Results among the 48 osteosarcomas examined,38 were stained weak or negative for caveolin-1,whereas the strong caveolin-1 staining was observed in all the 24 normal bone tissues.The absorbent optical density value of caveolin-1 protein in osteosarcoma tissues and normal bone tissues were(0.2292 ± 0.0329) and(0.6428 ± 0.0028),respectively.The expression of caveolin-1 in osteosarcoma tissues was significantly lower than that in normal bone tissues (t =21.48,P < 0.01 ).Caveolin-1 protein expression was significant correlated with the pulmonary metastasis of tumors ( t =11.84,P < 0.05 ),but had no correlation with nge,sex,tumor size and pathologic grades.Conclusion The low expression of caveolin-1 may participate in osteosarcoma invasion and metastasis,and may be a new prognostic molecular marker for osteosarcoma and a new therapeutic target for tumor.

3.STUDIES ON ELEMENTS IN ANOPHELES SINENSIS INFECTED WITH BRUGIA MALAYI MICROFILARIAE

Amounts of Certain elements in Anopheles sinensis, both normal ones (emerged mosquitoes, d0, d5, d8, d12 and d18 after taking blood meal) and those infected with Brugla malayi microfilaria (d5, d 8, d12 and d18 after infection), were determined using IL-551 and PE-603 atomic absorption spectrometer(AAS). The results showed that 9 kinds of trace elements including Fe, Zn, Cu, Cd, Al, Pb, Mn. Ni and Cr and 4 kinds of macro elements as K, Na, Ca, Mg were present in both the noninfected and infected mosquitoes. Comparing the contents of the elements between the noninfected and infected mosquitoes, the amounts of many kinds of elements reduced obviously in mosquitoes infected with microfilaria. The quantity and kinds of elements reduced in mosquitoes with the days of infection, for instance, 10 kinds of elements on the 5th day, 11 on the 8th day, 9 on the 12th day and 7 on the 18th day after infection(Tables 1, 2).
4.Advances in the pharmacological study of Morus alba L.

Morus alba L. (mulberry) is a well-known deciduous tree, belonging to the genus of Morus of Moraceae famlily. Its leaves, twigs, roots (bark) and fruits are widely used in the traditional Chinese medicine. The active constituents of mulberry contained flavonoids, alkaloids, steroids, coumarins, with the significant hypoglycemic, hypolipidemic, antihypertension, anti-oxidation, anti-inflammatory, anti-bacterial, anti-tumor and immunomodulatory activities. This review summarized the research progress of the major pharmacological activity, pharmacokinetics and drug-drug interaction based on CYPs and transporters of mulberry and its active constituents.

5.Screening and identification of serum biomarker of anovulatory dysfunctional uterine bleeding and its expression in the menses

Objective To screen and identify the serum biomarker of anovulatory dysfunctional uterine bleeding (ADUB) , to determine the expression of biomarker protein in menses of ADUB pa-tients, and to investigate the relation between ADUB and the biomarker proteins. Methods Subjects included 128 ADUB patients and 93 age-matched controls( normal women ). Their serum and super-natant of mense were collected and stored for use at -80℃. The differential proteins in the serum of the 2 groups were detected by CM 10 and analyzed by Biomarker WizardTM3.2 software. Then, the differential proteins were identified by Trieine-SDS-PAGE gel separation, spectrometry identifica-tion, and immunoprecipitation. The expression of the protein identified above in the menses was test-ed by ELISA, RT-PCR, and Western blotting. SPSS 14.1 was applied for statistical analysis and chart drawing. Results Five differential protein peaks were screened and their peak values were 11.80, 13.59, 13.79, 13.85, and 14.20 km/z, respectively. The intensity of protein peak ( 11.80 km/z ) which was identified as serum amyploid protein A ( SAA ) of ADUB was significantly higher than that of the controls (P<0.05). While the intensity of protein peak (13.59 km/z) which was identified as vascular endothelial growth factor (VEGF) of ADUB was obviously lower than that of the controls (P<0.05). The intensity of protein peak 13.08, 13.85, and 14.20 was not different between the cases and controls. SAA expressed highly in the menses of ADUB but low in that of the controls. Conversely, VEGF expressed highly in the menses of the control but low in that of the ADUB. Conclusion Two biomarkers which might be related with ADUB have been correctly screened and identified as SAA and VEGF. It needs further study whether the increased expression of SAA and reduced expression of VEGF are the cause or result of ADUB.
6.Bone marrow mesenchymal stem cell transplantation effect on expression of HCN2 and HCN4 gene following myocardial infarction

BACKGROUND: Studies have demonstrated that hyperpolarization-activated andcyclic nucleotide-gated cation channels (HCN) 2 and HCN4 are abnormal expressed in ventricular muscle following heart disease, which is closely correlate to ventricular arrhythmia. Bone marrow stem cells (BMSCs) transplantation can improve damaged cardiac muscle; however, its effect on remodeling of ion channel is unclearly. OBJECTIVE: To detect the changes of HCN2 and HCN4 expression in left ventricle following BMSCs transplantation.METHODS: Five Sprague-Dawley (SD) rats, aged 3 weeks, were prepared for BMSCs by Percoll method. Additional 30 health, male, SD rats, were randomly divided into DMEM, cell transplantation, sham operation and control groups. At 4 weeks after model preparation, DMEM culture medium was injected into the infarcted area and surroundings with 5 points in the DMEM group. The 3rd generation of cultured BMSCs (200 μL, with 5×10~6 cells) were injected into rats in the cell transplantation group with the same methods. The mRNA and protein expression of HCN2 and HCN4 gene was detected by RT-PCR and Western blot respectively.RESULTS AND CONCLUSION: In non-infarcted region, the mRNA and protein expression of HCN2 and HCN4 among each groups had no significant difference (P > 0.05). Compared with the control and sham operation groups, mRNA and protein expression of HCN2 and HCN4 in surrounding of the infracted region was elevated in the DMEM group (P < 0.05), which was greater than that of the cell transplantation group (P < 0.05). The mRNA and protein expression of HCN2 and HCN4 was smaller in the center area of infracted region in the DMEM group than that of the control and sham operation groups (P < 0.05), which was similar to the cell transplantation group (P > 0.05). Acute myocardial infarction can increase mRNA and protein expression of HCN2 and HCN4 in infracted region surrounding. BMSCs transplantation may lower the fatality rate of ventricular arrhythmia by reducing HCN2 and HCN4 expression.
7.Association between Expression of HDAC1 Protein and HPV16/18 Infection in Cervical Intraepithelial Neoplasia and Cervical Squamous Cell Carcinoma

Objective To explore the anomalous expression of HDAC1 in cervical intraepithelial neoplasia(CIN)and cervical carcinoma and its relationship with the infection of HPV16/18. Methods The expressions of HDAC1 protein and HDAC1 infection were detected by in situ hybridiza?tion(ISH)and immunohistochemistry staining(SP)in tissues from patient with chronic cervicitis(the chronic cervicitis group),CIN(the CINⅠgroup,the CINⅡgroup and the CINⅢgroup)and cervical squamous carcinoma(the cervical carcinoma group). Results The infection rate of HPV16/18 was 8.33%,34.78%,48.00%,65.21%,78.00%in patients with chronic cervicitis,CINⅠ,CINⅡ,CINⅢand cervical carcinoma re?spectively. There was significant difference between groups(P<0.05). The positive expression rate of HDAC1 was 0,13.04%,32.00%,47.82%and 76.00%in chronic cervicitis,CINⅠ,CINⅡ,CINⅢand cervical carcinoma,respectively. The differences were statistically significant(P<0.05). The positive expression of HDAC1 in the HPV16/18 infected patients was significantly higher than those uninfected with HPV16/18(P<0.05). The HDAC1 positive expression had no relationship with clinical stages of cervical carcinoma,however,it was related to the tissue differentia?tion,stromal invasion and pelvic lymph node metastasis(all P<0.05). Conclusion The infection of HPV16/18 may play an important role in the carcinongenesis of SCC through increasing the expression of HDAC1.
8.Differential expression of nucleus-cytoplasm transport receptor protein importin13 in limbal neoplasms

Background The study on normal stem cell markers provides a new way of thinking of that pathogenesis of cancer research and looking for specific markers of cancer stem cells.Importin13 (IPO13) is a novel nucleus-cytoplasm transport receptor protein of importin β family,the study on the biological behavior of IPO13 in ocular tissue and limbal neoplasms is lacking.Objective This study was to investigate the differential expression of IPO13 and p63 in human benign and malignant conjunctiva-cornea neoplasms.Methods The specimens of normal donor limbal and conjunctival tissues (6),conjunciva-cornea papilloma (CCP) (6) and conjunctiva-cornea intraepithelial neoplasia(CCIN) (6)were collected from Xiangya Hospital of Center South University.The expressions of IPO13 and p63 in the corresponding tissue were qualitatively and quantitatively detected using immunochemistry.The A values of IPO13 and p63 positive response were calculated and compared among the 3 types specimens.Results The immunohistostaining on frozen sections showed that IPO13 was expressed in nuclei of basal cells of limbal and conjunctival epithelium and the cellular nuclei of basal cells and suprabasal cells of CCP and CCIN epithelium.The A values of IPO13 positive expression were 1687± 1014,3546± 1375 and 7635 ±2854 in the normal limbal specimen,CCP and CCIN,respectively,showing a significant difference among them (F =7.23,P<0.05),and the A value was higher in the CCP and CCIN than that in the normal limbal tissue (q =4.02,5.13,P<0.05),and that in the CCIN was significantly elevated in comparison with CCP(q =3.45,P<0.05).p63 was expressed in nuclei of basal cells and suprabasal of limbal,conjunctival and CCP epithelium,and was expressed in nuclei of the entire CCIN epithelium.The expressions of p63 in the normal conjunctiva-cornea tissue,CCP and CCIN were 2110± 1229,3966± 2129 and 6650± 2136 respectively,with significant difference among the three different specimens (F =6.17,P< 0.05),and the A value of p63 positive expression was significantly elevated in the CCP and CCIN compared with normal limbal specimen (q =4.33,5.01,P<0.05),and that in the CCIN was significantly elevated in comparison with CCP(q=3.83,P<0.05).Conclusions IPO13 is more specifical in marking the poorly differentiated cells in limbal epithelial proliferative lesions than p63.Compared with the normal limbal specimen,the expressions of IPO13 and p63 in CCP and CCIN specimens gradually upregulat,which suggests that IPO13 and p63 may play a positive regulatory role in conjunctiva-cornea proliferative lesions.The differential expression of IPO13 and p63 is predominant between benign and malignant limbal epithelial proliferative lesions,indicating that IPO13 and p63 may play an important role in regulating the abnormal proliferation and differentiation of limbal epithelial proliferative lesions.
9.Influence of Maca on Resist-fatigue and T-SOD, Liver Glycogen of the Mice After Exhaustive Sports

Objective To establish the resist-fatigue animal test through using Maca, in order to provide reference for the relevant departments to exploit new sports nutrition . Methods Thirty male KM mice were randomly divided into Quiet control group (A),Exhaustive group (B,high altitude),Maca exhaustive group (C), there were 10 mice in each group. Drinking water containing Maca (Maca =500 mg/L) was supplied in group C. Additional exhaustive group (D,low altitude) was set.Thirty days later, all mice were killed after one exhaustive test, then we tested the exhaustive time, the level of T-SOD and liver glycogen. Results The exhaustive time of group C was significantly longer than group B (P<0.05), the T-SOD level of group C was significantly higher than group A and group B (P<0.05), and the liver glycogen content of group C was significantly higher than group B (P<0.05) . Conclusion Maca can raise T-SOD and liver glycogen level of the mice after exhaustive swimming, and has function of resist-fatigue.
10.Chemical constituents from Pleione yunnanensis.

This study was to investigate the chemical constituents from pseudobulbs of Pleione yunnanensis, one of the source of traditional Chinese medicine "Shancigu". The chemical constituents were isolated by various chromatography methods, including silica gel, ODS, Sephadex LH-20, and semi-preparative HPLC. Fourteen compounds were isolated and identified from the EtOAc fraction of 90% ethanol extract, including five dihydrophenanthrenes, four bibenzyls, two triterpenoids, and three phenylacrylic acids. Their structures were identified on the basis of the spectral data as 4, 7-dihydroxy-2-methoxy-9,10-dihydrophenanthrene (1), 4, 7-dihydroxy-1-(p-hydroxybenzyl)-2-methoxy-9,10-dihydrophenanthrene (2), (2,3-trans)-2-(4-hydroxy-3-methoxyphenyl) -3-hydroxymethyl-10-methoxy-2,3,4,5-tetrahydro-phenanthro[2,1-b]furan-7-ol (3), pleionesin B (4), blestriarene A (5), batatasin III (6), 3, 3'-dihydroxy-2-(p-hydroxybenzyl) -5-methoxybibenzyl (7), 3', 5-dihydroxy-2-(p-hydroxybenzyl) -3-methoxybibenzyl (8), 3,3'-dihydroxy-2,6-bis(4-hydroxybenzyl) -5-methoxybibenzyl (9), triphyllol (10), pholidotin (11), (E) -p-hydroxycinnamic acid (12), (E)-ferulic acid (13), and (E)-ferulic acid hexacosyl ester (14). Compounds 5,10-14 were separated from this plant for the first time.
