1.A novel chromene with anti-tumor activities from fungus Phomopsis sp.

A new chromene (1) and six known compounds identified as 6-hydroxymellein (2), 6-hydroxy-5-methylmellein (3) nectriapyrone (4), chermesinone A(5), chermesinone B(6), and pomopxanthone A(7), were isolated in our investigation of the cytotoxic constituents from the fermented rice substrate of endophytic fungus Phomopsis sp. HCCB03519. The structures of these com pounds were elucidated through spectroscopic data analysis. All compounds exhibited inhibitory activities against cancer cell lines. Compound 7 showed stronger inhibition against cancer cells than the positive control 5-Fu.

2.Changes and Clinical Significance of Glucose,Insulin and Insulin-Like Growth Factor-1 in Serum of Newborn Infants with Hypixic-Ischemic Encephalopathy

Objective To observe the changes and relationships of glucose (Glu),insulin(Ins) ,insulin-like growth factor-1 (IGF-1) in newborn infant with hypixic-ischemic encephalopathy (HIE) and the protective role which Ins,IGF-1 worked on neuron.Methods HIE 50 cases,the quantity of IGF-1 in serum was detected by radioimmunoassay,the quantity of Ins in serum was detected by chemiluminescent immunoassay,the quantity of glucose in serum was detected by the instrument with type of Rokangquan TM accutive.The umbilical vein blood 2 mL were drawn from 20 cases of controls after birth.The indexes were detected by the same methods.Results 1.In acute period,the level of serum IGF-1 reduced obviously in newborn infants with HIE,and Glu increased obviously.There was a negative correlation between IGF-1 and Glu.The changes related with neonate complications.In rehabilited pe riod,the Glu get to normal,the IGF-1 was a little higher than that in acute period.but it still was lower than that of control group.There was negative correlation between IGF-1 and Glu.2.The level of Ins in serum of newborn infants with HIE had no obvious changes compairing with control group in acute period and rehabilited period.It had no relations with Glu,Ins.Conclusions It approves that IGF-1 and Glu in serum of newborn infants with HIE have obviously changed.This result will provide an academic base on treatment of HIE.
3.Clinical efficacy of intravitreal conbercept injection and macular grid pattern photocoagulation in treating macular edema secondary to non-ischemic branch retinal vein oclussion

Objective To study and compare the clinical efficacy between intravitreal conbercept injection and (or) macular grid pattern photocoagulation in treating macular edema secondary to non-ischemic branch retinal vein occlusion (BRVO).Methods Ninety eyes of 90 patients diagnosed as macular edema secondary to non-ischemic BRVO were enrolled in this study.Forty-eight patients (48 eyes) were male and 42 patients (42 eyes) were female.The average age was (51.25 ± 12.24) years and the course was 5-17 days.All patients were given best corrected visual acuity (BCVA),intraocular pressure,slit lamp with preset lens,fluorescence fundus angiography (FFA) and optic coherent tomography (OCT) examination.The patients were divided into conbercept and laser group (group Ⅰ),laser group (group Ⅱ) and conbercept group (group Ⅲ),with 30 eyes in each group.The BCVA and central macular thickness (CMT) in the three groups at baseline were statistically no difference (F=0.072,0.286;P=0.930,0.752).Patients in group Ⅰ received intravitreal injection of 0.05 ml of 10.00 mg/ml conbercept solution (conbercept 0.5 mg),and macular grid pattern photocoagulation 3 days later.Group Ⅱ patients were given macular grid pattern photocoagulation.Times of injection between group Ⅰ and Ⅲ,laser energy between group Ⅰ and Ⅱ,changes of BCVA and CMT among 3 groups at 1 week,1 month,3 months and 6 months after treatment were compared.Results Patients in group Ⅰ and Ⅲ had received conbercept injections (1.20 ± 0.41) and (2.23 ± 1.04) times respectively,and 6 eyes (group Ⅰ) and 22 eyes (group Ⅲ) received 2-4 times re-injections.The difference of injection times between two groups was significant (P<0.001).Patients in group Ⅱ had received photocoagulation (1.43 ±0.63) times,9 eyes had received twice photocoagulation and 2 eyes had received 3 times of photocoagulation.The average laser energy was (96.05 ±2.34) μV in group Ⅰ and (117.41 ±6.85) μV in group Ⅱ,the difference was statistical significant (P=0.003).BCVA improved in all three groups at last follow-up.However,the final visual acuity in group Ⅰ and group Ⅲ were better than in group Ⅱ (t=4.607,-4.603;P<0.001) and there is no statistical significant difference between group Ⅲ and group Ⅰ (t=-0.802,P=0.429).The mean CMT reduced in all three groups after treating for 1 week and 1 month,comparing that before treatment (t=-11.855,-10.620,-10.254;P<0.001).There was no statistical difference of CMT between group Ⅰ and Ⅲ at each follow up (t=0.404,1.723,-1.819,-1.755;P=0.689,0.096,0.079,0.900).CMT reduction in group Ⅰ was more than that in group Ⅱ at 1 week and 1 month after treatments (t=-4.621,-3.230;P<0.001,0.003).The CMT in group Ⅲ at 3 month after treatment had increased slightly comparing that at 1 month,but the difference was not statistically significant (t=1.995,P=0.056).All patients had no treatment-related complications,such as endophthalmitis,rubeosis iridis and retinal detachment.Conclusions Intravitreal conbercept injection combined with macular grid pattern photocoagulation is better than macular grid pattern photocoagulation alone in treating macular edema secondary to non-ischemic BRVO.Combined therapy also reduced injection times comparing to treatment using conbercept injection without laser photocoagulation.

5.Application of flow cytometric score in the diagnosis of myelodysplastic syndromes

Objective To identify myelodysplastic syndromes (MDS) from non-MDS (mainly non-clonal cytopenias) by flow cytometric (FCM) score.Methods Four parameters (CD34+ myeloblast-related and B-progenitor-related cluster size,myeloblast CD45 expression and granulocyte side scatter value) were rated on 50 patients with MDS and 20 patients with non-clonal cytopenias.A diagnosis of MDS was formulated in the presence of FCM score value ≥ 2 (every abnormal parameter was scored 1).Results When compared with non-clonal cytopenias,patients with MDS had increased myeloblast-related cluster size and decreased B-progenitor-related cluster size,and had a lower granulocyte side scatter (P < 0.05),but no significant difference was found when myeloblast CD45 expression was compared (P =0.06).The FCM score value was significant higher in MDS patients (P < 0.001).45 cases were correctly classified (sensitivity 90 %).There was 1 false-positive case among 20 controls (specificity 95 %).Conclusion The FCM score may help to establish the diagnosis of MDS,and it is simple and feasible.
7.Simultaneous determination of 11 mycotoxins in malt by isotope internal standard-UPLC-MS/MS.

A suitable ultra-high performance liquid chromatography coupled with tandem mass spectrometry (UPLC-MS/MS) method was developed for the determination of 11 mycotoxins with isotope internal standard in malt. The mycotoxins in malt were extracted and purified by one-step ultrasonic extraction procedure using acetonitrile/water/acetic acid (80 : 19 : 1), and then detected and confirmed by UPLC-MS/MS, and quantified by isotope labeled AFB1 ([13C17]-AFB1) and ZEN ([13C18]-ZEN) internal standards. Rapid separation of the 11 mycotoxins was successfully achieved on a Phenomenex Kinetex C18 column (100 mm x 2.1 mm, 2.6 μm) with gradient elution using the mobile phase of methanol containing 0.1% formic acid and 2 mmol x L(-1) ammonium acetate in water. Simultaneous acquisition was performed in multiple reaction monitoring (MRM) mode with electrospray ionization (ESI) source operated in both positive and negative ionization modes. The established method provided a good linearity for the 11 mycotoxins within their respective linear ranges with correlation coefficients all higher than 0.999 1. The average recoveries ranged from 75.0% to 117.0% with relative standard deviations (RSDs) below 5.1%. The limits of detection (LODs) and quantitation (LOQs) ranged from 0.05 to 30 μg x kg(-1) and 0.15 to 87.5 μg x kg(-1), respectively, which were below the maximum residue levels (MRLs) set by the European Union. Twenty malt samples were analyzed and nine samples were detected with mycotoxins, which were confirmed according to the same fragment ions found in positive samples and the standards at the same retention time. This study has demonstrated that the one-step extraction procedure of mycotoxins from complex matrices coupled to UPLC-MS/MS method is simple, quick, accurate and sensitive for quantitative and qualitative analysis of multiple mycotoxins in malt.

8.A Meta analysis of Shensongyangxin capsule combined metoprolol in treatment of ventricular premature beats

Objective:To compare the therapeutic effects between Shensongyangxin capsule combined metoprolol and metoprolol monotherapy in treating ventricular premature beats.Methods:The related literature of Shensongyangxin capsule combined metoprolol in treatment of ventricular premature beats published from Aug 2005 to Aug 2013 were searched by computer from Cochrane Library,MEDLINE,EMbase,Wangfang Data,China National Knowledge Infrastructure (CNKI)and Chinese biomedical literature database (CBM),and were searched manually.RevMan 5.0 software was used to perform Meta analysis on data.Results:Symptomatic relief in treatment group was signifi-cantly better than that of control group [(Z=8.28,P <0.00001;OR=6.17,95%CI (4.01,9.48)];dynamic ECG improvement in treatment group was significantly better than that of control group [Z=9.37,P <0.00001;OR=3.41,95%CI (2.64,4.40)];there was no significant difference in incidence rate of adverse drug reaction be-tween two groups [Z=1.47,P =0.14;OR=0.70,95%CI (0.43,1.13)].All funnel schemas were symmetry in general,so there was small possibility of bias.Conclusion:Shensongyangxin capsule combined metoprolol possesses better therapeutic effect than metoprolol monotherapy in treating ventricular premature beats,and it doesn′t in-crease adverse drug reactions.
9.Effect of Ganglioside- 1 and Rehabilitation on Recovery of Neurologic Behavior in Rats with Hypoxic- ischemic Brain Damage

Objective To investigate the effects and the possible mechanism of Ganglioside-1 (GM1) and rehabilitation on neurologic behaviors of rats with hypoxic-ischemic brain damage (HIBD). Methods 48 immature rats developed as model of HIBD were divided into 5 groups: control group, GM1 group, rehabilitation training group (R group), GM1+R group, and sham group. They were evaluated with Climb, Slope and Water Maze Tests. The expressions of activated caspase-3 in neurons were detected with immunohistochemistry. Results The achievement of Climb and Slope Test were significantly different among groups (P<0.001), which ranked as the sham group> GM1+R group> R group> GM1 group> control group (P<0.05). The latency of Water Maze Test was significantly different among groups (P<0.01),which was longer in the control and GM1 groups than in the sham group (P<0.05). The activated caspase-3 expressed in hippocampus and frontal cortex was less in the GM1 and R groups than in the sham group (P<0.05), more in the GM1 group, R group and GM1+R group than in the control group in hippocampus (P<0.05), more in the R group and GM1+R group than in the control group in frontal cortex (P<0.05).The expression of activated caspase-3 in both hippocampus and frontal cortex correlated with the achievement of Climb and Slope Test (P<0.05), but not with the latency of Water Maze Test (P>0.05). Conclusion Rehabilitation training and GM1 are effective on recovery of neural function in rats with HIBD with synergia, which may associated with the activated caspase-3 in terminal synapse.
10.Association of MRP2 expression level and arsenic methylation in rats' liver treated with sodium arsenite and sodium arsenate

Objective To explore the effect of sodium arsenite (iAs3+) and sodium hydrogen arseuate (iAs5+) exposure on rat multidrug resistance protein 2 (MRP2) expression,and the correlation of liver arsenic and its methylation products,and to provide a basis for further elucidating the mechanism of arsenic toxicity.Methods Seventy wistar rats were randomly divided into seven groups,10 rats in each group.Control group was administrated with deionized water.Sodiun arsenite high-dose group,middle-dose group,low-dose group were administrated with different concentrations of sodium arsenite:20.0,6.7 and 2.2 mg/kg BW every day,respectively.Animals were sacrificed 90 days later by cervical dislocation to collect liver,the expression of MRP2 in the membrane of hepatocyte was determined by Western blotting.HPLC-HGAFS was employed to determine the content or level of arsenic and its methylation products.Correlation between expression of MRP2 and arsenic methylation products was analyzed using Pearson's linear correlation method.Results The MRP2 protein levels of control group,iAs3+ and iAs5+ high-,middle-,low-dose groups were 1.21 ± 0.13,1.85 + 0.09,1.65 + 0.19,1.61 + 0.18,1.69 + 0.04,1.42 + 0.19,1.27 ± 0.10.Compared to control group,MRP2 protein levels of iAs3+ high-,middle-,low-dose group and iAs5+ high-dose group were increased (P < 0.05); Compared with the low-dose group,MRP2 protein levels of iAs3+ and iAs5+ high dose groups were increased(P < 0.05); Comparing the matched doses group of iAs5+ and iAs3+,MRP2 protein levels of iAs3+ high-,middle-,low-dose group were higher than iAs5+(P < 0.05).Meanwhile,there was a significant positive relationship between the expression of MRP2 and the content of iAs3+,MMA and DMA in iAs3+ exposed group(r =0.575,0.678,0.395,all P < 0.05).There was also a significant correlation between the expression of MRP2 and MMA and IMA in iAs5+ exposed group(r =0.593,0.643,all P < 0.05).There was no significant relationship between the expression of MRP2 and the content of iAs5+in iAs5+ exposed group.Conclusions The expression of MRP2 is increased with increasing dose of arsenic exposure,thus resulting in compensatory changes on MRP2 expression in the liver cell membrane.MRP2 efflux may play an important role in the metabolic pathways of iAs3+.The level or load of arsenic methylation in liver is closely related with MRP2 expression.
