1.Predictors of response in patients with HBeAg-positive chronic hepatitis B with adefovir dipivoxil treatment

Objective To investigate the effective predictors of response in HBeAg-positive patients treated with adefovir dipivoxil(ADV)and to provide evidence for individualized treatment.Methods Patients administered with ADV for 48 weeks in a randomized,placebo-controlled,multicenter trial were studied.Statistical analyses,such as Backward stepwise logistic regression and 2?2 method were used for predictors analysis at week 48.Results The baseline serum ALT levels,HBV DNA levels,and undetectable serum HBV DNA by PCR at week 24 were predictors for HBV DNA negativity at week 48.The median of serum ALT levels and HBV DNA levels prior to treatment were 134.5 U/L and 6.57 lg copies/mL,respectively.Patients with baseline ALT levels higher than the median,HBV DNA levels lower than the median,and serum HBV DNA undectectable by PCR at week 24 had greater rate of HBV DNA negativity(93.3%),HBeAg loss(60%)and HBeAg seroconversion(40%)at week 48 than the others.47.8% of patients whose HBV DNA levels were positive at week 24 also achieved HBV DNA negativity at week 48,and 8.6% achieved HBeAg seroconversion.Conclusion Better response at week 48 has significantly higher serum ALT levels and lower HBV DNA levels prior to treatment and HBV DNA negativity at week 24 compared with non-response.Patients whose HBV DNA levels ware still positive at week 24 should continue therapy.

Objective To investigate the relationship between ?3 adrenergic receptor(?3AR) gene Trp~(64)Arg mutation and insulin resistance(IR) in Chinese.Methods The 120 patients with type 2 diabetes mellitus(group DM) and 80 nondiabetic subjects as normal controls(group N)were involved in the study.Polymerase chain reaction(PCR) and the restriction fragment length polymorphism analysis(PCR-RFLP) were used to determine the ?3AR genotype.The indexes such as body weight,height,waist circumstance,hip circumstance,blood pressure,lipids,fast plasma glucose and insulin were detected in all subjects.The IR was estimated by homeostasis model assessment(HOMA-IR).The patients in the diabetes group were divided into relative insulin sensitive group and relative insulin resistant group by the 75th percentile HOMA-IR.Results The BMI of the patients with the ?3AR Trp~(64)Arg mutation was significantly higher than that of those without the mutation(P

This investigation describes a new precise, sensitive and accurate stereoselective RP-HPLC method for determination of the enantiomers of a novel alpha- and beta-receptor blocking agent, 1-[4-(2-methoxyethyl) phenoxy]-3-[[2-(2- methoxyphenoxy) ethyl]amino]-2-propanol (TJ0711), in rat plasma. GITC was used for precolumn derivatization of TJ0711 enantiomers. Enantiomeric resolution was achieved on a Eurospher-100 C18 column (250 mmx4.6 mm ID, 5-mum particle size), with UV detection at 255 nm, and the mobile phase consisted of acetonitrile and water (58:42, v/v) containing 0.02% glacial acetic acid (v/v). Using the chromatographic conditions described, TJ0711 enantiomers were well resolved with mean retention time of 10.2 and 11.5 min, respectively. Linear response (r>0.999) was observed over the range of 0.125-12.5 mug/mL of TJ0711 hydrochloride enantiomers. The mean relative standard deviation (RSD%) of the results of within-day precision was [Symbol: see text] 10%. The proposed method was found to be suitable and accurate for the quantitative determination of TJ0711 enantiomers in rat plasma, and it can be used in pharmacokinetic studies.
5.Analysis of Drug Use Labeling for Pregnant and Lactating Women in 762 Drug Package Inserts

OBJECTIVE:To provide reference for drug use labeling for pregnant and lactating women. METHODS:The drug package inserts were collected from Linxi Hospital of Kailuan General Hospital during Jul. 2013-Dec. 2015. The information about drug use labeling for pregnant and lactating women was analyzed statistically. RESULTS:Among 762 drug package inserts,pack-age inserts which were not labeled with or labeled with indefinite drug use information for pregnant and lactating women accounted for 31.89% and 52.76% respectively. Among package inserts of 361 domestic chemical drugs and biological products,339 Chinese patent medicine and 62 imported drug,package inserts which were not labeled or labeled with indefinite drug use information for pregnant and lactating women accounted for 22.99% and 25.21%,44.54% and 88.50%,14.52% and 17.74%,respectively. CON-CLUSIONS:Except for poor drug use labeling for pregnant and lactating women in package inserts package,there still are other problems,such as items listed dispersedly,presentation content not consistent. Compared with imported drugs,the missing informa-tion for pregnant and lactating women are obvious in drug package inserts of domestic chemical drugs and biological products,and severe in those of Chinese patent medicine. It is recommended that drug manufacturers should strengthen drug tracing and monitor-ing after listed,and update and revise related content of package inserts timely;drug administration department should strengthen drug package inserts supervision,and unify and standardize labeled content management of drug use.
6.Expression of somatostatin receptors in nasopharyngeal carcinoma cell line CNE_2

Background and purpose:Somatostatin receptors have been found in a variety of tumors and are therefore amenable to treatment with somatostatin analogs, like octreotide. However, the study of SSTRs expression has been rarely studied in nasopharyngeal cancer.We investigated the expression of somatostatin receptors gene subtypes in human nasopharyngeal cancer cell line CNE_ 2 . Methods:We have harvested cultured human nasopharyngeal cancer cell line CNE_ 2 . Using both techniques, reverse transcriptase-polymerase chain reaction (RT-PCR) and immunohistochemical assay, we analysed mRNA of different subtypes of somatostatin receptors in human nasopharyngeal cancer cell line CNE_ 2 .Results:The positive rate of somatostatin receptors subtype SSTR_ 1 SSTR_ 2 SSTR_ 4 was manifested in the human nasopharyngeal carcinoma cell line CNE_ 2 by RT-PCR and immunohistochemical assay. According to immunohistochemical assay, SSTR_ 1 and SSTR_ 2A showed strongly positive expression and SSTR_ 3 and SSTR_ 5 negative expression,respectively.Conclusions:There are more than one SSTR subtypes expressed in the human nasopharyndeal carcinoma cell line CNE_ 2 . This study demonstrated the presence of SSTR_1,SSTR_ 2 in the human nasopharyndeal carcinoma cell line CNE_ 2 .
7.Effect of Underwater Treadmill Training on Patients with Incomplete Spinal Cord Injury

Objective To observe the effect of underwater treadmill training on patients with incomplete spinal cord injury (SCI).Methods 11 patients with incomplete SCI were treated with underwater treadmill training, besides other comprehensive rehabilitation. Motor and sense scores were assessed using ASIA2000 standard and recorded before and after the underwater treadmill training. Walking distances, maximal walking velocity when training and changes of activities of daily living (ADL) before and after training were also recorded.Results After underwater treadmill training, the sense score of ASIA increased ( P<0.05), motor score of ASIA, and scores of walking distance, maximal walking velocity and ADL increased significantly ( P<0.01).Conclusion The underwater treadmill training along with other routine rehabilitation treatment may be helpful to improve motor and sensory functions and ADL of patients with incomplete SCI.
8.Comparative analysis of subject novelty detection methods in medical literature

Objective To study the feasibility of novelty detection model in assessing the subject novelty of medical literature and comparatively analyze the advantages and disadvantages of words-overlap algorithm and co-words-based inverse file frequency quantitative algorithm. Methods Two novelty detection models were established for the 8 research subjects in PubMed-covered literature. The feasibility of two novelty detection models in assessing the subject novelty of medical literature was assessed according to the subject novelty of literature analyzed by experts, ROC curves and AUC values. Results Words-overlap algorithm showed that the fluctuating amplitude of subject novelty was rather high, which can thus reflect the difference between the contents in literature on the data. ROC curves and AUC values-based analysis revealed a high accuracy of words-overlap algorithm for judging the novelty of literature while co-words-based inverse file frequency quantitative algorithm displayed a low accuracy for judging the novelty of literature. Conclusion The novelty of literature detected with the two novelty detection methods is moderately related. The mean novelty value detected with the two novelty detection methods is of statistical signifi-cance. However, the novelty of literature detected with words-overlap algorithm is higher than that detected with co-words-based inverse file frequency quantitative algorithm.
9.Exogenous CCN1 promotes proliferation and migration of radiation-injured L929 cells

Objective To study the role of Cysteine-rich 61 ( Cyr61/CNN1) in repair of combined injury by observing whether exogenous CCN1 promotes the proliferation and migration of radiation-injured L929 cells. Methods A radiation model of L929 cells was induced by ? ray at a dose of 4 Gy. The irradiated L929 cells were cultured in a medium containing 2 ml adenovirus plasmids of CCN1 or RFP at 37 ℃ in an atmosphere containing 5% CO2,which served as a CCN1 group and a RFP group,respectively. Irradiated L929 cells cultured in a blank control medium served as a blank control group. Effects of CCN1 on proliferation and migration of radiation-injured L929 cells were detected by MTT assay,plate colony formation assay,cell cycle analysis,and scratch test of wound healing. 10.Differential expression of alpha-adrenoceptor subtypes in rat dorsal root ganglion after chronic constriction injury.

mRNAs of alpha-adrenoceptor (α-AR) subtypes are found in neurons in dorsal root ganglion (DRG) and change after peripheral nerve injury. In this study, the distribution of α-AR subtype proteins was studied in L5 DRG of normal rats and rats with chronic constriction injury of sciatic nerve (CCI). Using immunofluorescence technique, it was found that α1A-, α1B-, and α2A-AR proteins were expressed in large, medium, and small size neurons in normal DRG, and significantly increased in all size neurons 14 days after CCI. α1D- and α2C-AR was also expressed in all size neurons in normal DRG. However, α1D-AR was significantly increased and α2C-AR was decreased in small size neurons 14 days post CCI. α2B-AR neurons were not detectable in normal and CCI DRG. Co-expression of α1A- and α2A-AR in the same neuron was observed in normal DRG and increased post CCI. Collectively, these results indicated that there is distinct distribution of α-AR subtypes in DRG neurons, and the distribution and levels of expression of α-AR subtypes change differently after CCI. The up-regulation of α-AR subtypes in DRG neurons may play an important role in the process of generating and transmitting neuropathic pain.
