1.Expression and clinical significance of selenium binding protein 1 in gastric cancer

Objective To detect the expression of selenium binding protein 1 ( SBP1 ) in gastric cancer cell line SGC7901, BGC823, normal gastric epithelial cell line GES-1, gastric cancer tissues and normal gastric tissues, explore the relationship between SBP1 and pathologic features, and discuss the feasibility of SBP1 as an diagnostic marker of gastric cancer. Methods The clinical data of 135 patients with gastric cancer who were admitted to Zhongnan Hospital of Wuhan University from 2006 to 2007 were retrospectively analyzed. The expression of SBP1 in the gastric cancer tissues and 16 cases of normal gastric tissues were detected by immunohistochemistry. The mRNA and protein expressions of SBP1 of SGC7901, BGC823 and GES-1 were determined by Western blot and RT-PCR. All data were analyzed by using chi-square test and one-way analysis of variance.Results The mRNA expressions of SBP1 in BGC823 and SGC7091 were 0. 120 ± 0. 020 and 0. 133 ± 0. 015,respectively, which were significantly lower than 0. 907 ± 0. 015 in GES-1 ( F = 2106. 462, P < 0.05 ). The protein expression of SBP1 in BGC823 and SGC7901 were 0.253 ±0.015 and 0.273 ±0.015 ,respectively, which were significantly lower than 0.877 ±0.025 in GES-1 ( F = 1026. 758, P <0.05 ). A strong positive reaction of SBP1 was observed in 3 cases of gastric cancer tissues and 16 cases of normal gastric tissues. The decrease of the protein expression of SBP1 was correlated with clinical stages of the patients ( x2 = 12. 629, P < 0.05 ), rather than the sexes, ages, tumor histological types, tumor differentiation, infiltration depths and lymph node metastasis (x2 =2. 142, 0.860, 1.838, 5.001,4.858, 1.994, P>0. 05). Conclusions The decrease of SBP1 expression could be used as a marker in diagnosing gastric cancer. Down-regulation of SBP1 expression may play an important role in the genesis and prognosis of gastric cancer.
2.Isolation and culture methods of bone marrow stromal stem cells from the skull

BACKGROUND:PA6 cells are bone marrow stromal stem cells from the mouse skul , and scientists have found that PA6 cells co-cultured with some kinds of stem cells have shown neural differentiation, based on which, PA6 cells can be used for repair of nerve injury. Therefore, researchers give more and more attentions to PA6, but few studies have addressed isolation and culture methods of bone marrow stromal stem cells from the skul . OBJECTIVE:To isolate and culture bone marrow stromal stem cells from the skul of Sprague-Dawley rats and to observe cellular morpholopy in vitro and perform immunofluorescence identification. METHODS:Under sterile conditions, newborn Sprague-Dawley rat’s skul was cut into pieces. Smal skul pieces were washed using Dulbecco’s modified Eagle’s medium containing 10%fetal bovine serum. Single cellsuspension was made, and placed in culture flask. The culture medium was changed many times for cellpurification. The second passage of cells was obtained for morphology observation under an inverted microscope. cellsurface markers were detected by using immunofluorescence staining. RESULTS AND CONCLUSION:After the primary culture for 24 hours, cells exhibited adherent growth;after 3 days, cells were increased in number, and presented with irregular shapes, such as polygon, triangle, spindle and flat shape. After passage, cells were uniform in morphology, arranged in radial and bunch patterns, and exhibited strong adherent ability. Some cells grew in cluster, and proliferated faster than primary cells. Immunofluorescence staining showed that cells were positive for CD105, CD73, CD44, CD90, but negative for CD45, D34,CD14, HLA-DR. Results indicate that this method can obtain bone marrow stromal stem cells.
3. Bioinformatics analysis of hematopoietic injury and potential therapeutic drugs associated with SARS-CoV-2 infection

Objective To analyze and predict hematopoietic injury caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and potential therapeutic drugs, and to provide theoretical basis for clinical treatment of the hematopoietic injury. Methods The gene expression omnibus (GEO) database was used to screen the whole genome expression data related to SARS-CoV-2 infection. The R language package was used for differential expression analysis and the Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) enrichment analysis. The core genes were screened by protein-protein interaction (PPI) network analysis using STRING online analysis website. Then the self-developed apparent precision therapy prediction platform (EpiMed) was used to analyze diseases, drugs and related target genes. Results A total of 222 differential genes were screened, including 172 up-regulated and 50 down-regulated. GO enrichment analysis suggested that gene is mainly related to type I interferon response, cell cycle regulation, inflammatory cell migration, innate immune response, secretion of blood particles and vesicles, chemokines and their receptors. KEGG enrichment analysis suggested that gene is mainly related to viral infection, myocardial injury, complement and coagulation cascade, cell chemotaxis, platelet activation, acute inflammation, immune response, cellular signal transduction and so on. Ten core genes such as STAT1, IL-6, IRF7, TNF, MX1, ISG15, IFIH1, IRF9, DDX58 and GBP1were screened by PPI network analysis. EpiMed screened 10 drugs with potential intervention effects, including Rabdosia rubescens, sirolimus, glucocorticoid, Houttuynia cordata, Polygonum multiflorum, Red peony, tretinoin, Glycyrrhiza, cyclosporine A, fluvastatin and so on. Conclusions SARS-CoV-2 infection can damage the hematopoietic system by changing the expression of a series of genes. The potential intervention drugs screened from this have certain reference significance for the basic and clinical research of coronavirus disease 2019 (COVID-19).
4.Laparoscopic Radical Prostatectomy:Report of 11 Cases

Objective To evaluate the clinical efficacy of laparoscopic radical prostatectomy.Methods A total of 11 cases of laparoscopic radical prostatectomy were enrolled in this study.Among the cases,transperitoneal approach was used in 4 patients,and extraperitoneal approach was employed in 7.The prostate,seminal vesicles,and ampulla ductus deferens were resected,and then the bladder was anastomosed with the urethra.Results The operation was successfully performed in all the patients with a mean operation time of 304 min(180-390 min),and a mean blood loss of 520 ml(300-1200 ml).The postoperative catheterization time and hospital stay were 12-28 d(mean,19.8 d)and 12-24 d(mean,20 d)respectively.No injuries to the rectum,urinary incontinence,or urethral stenosis occurred in this series.The patients were followed up for 1-36 mon(mean 11 mon),during this period,no local recurrence or distant metastasis were found.The postoperative PSA level was 0-0.05 ?g/L.Conclusion Laparoscopic radical prostatectomy is safe and effective for the treatment of prostate cancer.
5.Sympathectomy for Raynaud disease

Objective To study the effect of thoracoscopic sympathectomy and/or chemical lumbar sympa- thectomy performed on patients with Raynaud disease. Methods Five patients, 1 male,4 females, aged from 30 to 65(mean 45. 3 ). Raynaud phenomenon appeared only on both hands in one patient, only on feet with a toe gangrene in another one , the other 3 cases on all limbs. Thoracoscopic symathectomy was performed for those whose hands were af- fected and chemical lumbar sympathectomy(12-3 ) was pererformed for those whose feet were affected. Results All patients were followed up from 12 to 48 months(mean 24 months). All experinced improvement with hands warm and satisfactory results after thoracoscopic sympathectomy. However, the original symptoms reccurred in two patients after postoperatire six months. All 4 patients performed chemical lumbar sympathectomy experienced improved symptoms with feet warm and satisfactory results and the symptoms did not recur up to now. The sympathectomy showed different results for hands and for feet. Conclusions Thoracoscopic sympathectomy for hands affected by Raynaud disease has efficiency temporarily, but is not satisfactory. The chemical lumbar sympathectomy for the feet affected by Raynaud disease has dramatically disappearence of symptoms and the results are very good
6.Changes and Relationship of Serum Insulin-Like Growth Factor-1 and Leptin in Children with Growth Hormone Deficiency

Objective To explore the changes and relationship of serum insulin-like growth factor-1(IGF-1) and leptin in children with growth hormone deficiency(GHD).Method Serum IGF-1 and leptin levels were measured by immunoradiometric assay in 20 prepuberal normal children and 23 children with GHD.Results Serum IGF-1 level in GHD group(51.158?29.988)?g/L was significantly lower than that in controls(112.680?41.540)?g/L.Serum leptin levels in GHD group(6.002?2.204)?g/L was significantly higher than that in controls(4.523?2.204)?g/L.There was no significant correlation between serum IGF-1 and leptin.Conclusion IGF-1 and leptin regulate growth in children with GHD in their individual way.
7.A clinical study on laparoscopic choledocholithotomy and primary suture in treatment of choledocholithiasis

Objective To explore the indication,technology and clinical significance of laparoscopic choledocholithotomy and primary suture in treatment of choledocholithiasis.Methods 78 patients with choledocholithiasis were divided into two groups receiving respectively laparoscopic choledocholithotomy and T-tube drainage treatment,and laparoscopic choledocholithotomy and primary suture after common bile duct exploration.The time of operation,postoperative hospital stay and complications were analyzed and compared.Results Bile leakage occurred in 2 cases in the primary suture group and 3 cases in the T-tube drainage group;No residual stones or biliary stricture was found in either groups.The time of operation,postoperative complications were not statistically different (P > 0.05).The difference in postoperative hospital stay and GI function recovery time between the two groups was statistically significant (P < 0.05).Conclusion In well selected cases,the primary suture of common bile duct after laparoscopic choledocholithotomy is feasible and safe.
8.Preparation, Characteristics and in vitro Drug Release of Silybin Lipid Emulsions

Objective:To prepare silybin ( SLB) lipid emulsions and evaluate the drug release behavior in vitro. Methods:Silybin lipid emulsions were prepared by high-pressure homogenization method. Under the guidance of single-factor test, the best formula of SLB lipid emulsions was obtained by orthogonal design. Results:The encapsulation efficiency of SLB lipid emulsions prepared by the optimal formula was (91. 45 ± 0. 005 2) % with the particle size of (74. 42 ± 14)nm and PDI of 0. 106, Zeta potential of ( -30. 2 ± 2. 13)mV and pH of (6. 48 ± 0. 04). The drug release of SLB lipid emulsions was up to 90% within 12 hours. Conclusion:The opti-mized preparation of SLB with sustained-release property is obtained successfully.

Objective To assess the efficacy and safety of argon plasma coagulation(APC) in treating patients with Barrett's esophagus.Methods Two reviewers independently searched the Cochrane Library(Issue 2,2008),MEDLINE(January 1948 to November 2008),and CNKI(January 1999 to November 2008),respectively.The quality of the included studies was assessed according to the guidance in the Cochrane Handbook of systematic reviews of interventions.Results Six randomized controlled trials involving 253 patients with Barrett's esophagus met the inclusion criteria and were included.One trial reported that the ablation rate of patients in the APC group was significantly higher than that in the endoscopic surveillance group.Followed up one year,the ablation rate in APC group was 63% compared with 15% in the control group(P
