Objective: To explicit whether miR-200b can suppress the proliferation and invasion by targeting CD133, and to explore a molecular mechanism that miR-200b plays a role in tumor suppression in glioma. Methods: The CD133 3'-untranslated region (3'-UTR) mRNA-luciferase reporter vector was constructed and the dual-luciferase reporter gene assay was employed to examine the effect of miR-200b on activity of luciferase. U251 cells were transfected with miR-200b mimics and CD133-small interfering RNA (siRNA) (as a positive control) by LipofectAMINE 2000, and the expressions level of CD133 protein was detected by Western blotting. The inhibition effects of CD133 on cell proliferation and invasion were observed after CD133 siRNA were transfected into U251 cells. U251-SC cell mammosphere assay was performed after cotransfection with miR-200b mimics and CD133 siRNA. Results: miR-200b could bind to the 3'-UTR of CD133 and inhibit the activity of luciferase. CD133 protein expression was significantly down-regulated when miR-200b was overexpressed in U251 cells. Overexpression of miR-200b inhibited the invasion of U251 cells. Overexpression of miR-200b antagonized a role of proliferation and invasion in U251 cells. Overexpression of miR-200b reduced mammosphere number and the size of glioma stem cells. Conclusion: miR-200b can suppress cell proliferation and invasion by targeting CD133 in glioma. Copyright © 2014 by TUMOR.
3.MRI tracking of transplanted neural stem cells labeled with superparamagnetic iron oxide in ischemic model

Objective To explore the migration of transplanted neural stem cells labeled with SPIO with 4.7T MR system and study cell differentiation by immunohistochemistry in ischemic rats. Methods Rat neural stem cells(NSCs)co-labeled with SPIO mediated by poly-L-lysine and bromode- oxyuridine(BrdU)were transplanted into the middle cerebral artery occlusion(MCAO)rats after 14 days when MCAO model was made successfully.4.7T MR scanner was used to monitor the migration of transplanted cells after 1.2,3,4,5 and 6 weeks post-transplantation.After MRI examination at 6th week,the rats were killed and Prussian blue staining and immunohistochemistric staining were per- formed to study migration and functional differentiation of NSCs.Results Three weeks after trans- plantation,linear hypointensity area derived from migration of labeled NSCs was observed in the corpus callosum adjacent to the injection site.Six weeks after transplantation,linear hypointensity area was moved toward the midline along the corpus callosurn.MRI findings were confirmed by Prussian blue staining and immunohistochemical straining.Co-labeled immunohistochemical methods demonstrated transplanted NSCs could differentiated into astrocytes and neurons.Conclusion MRI is very useful in demonstrating the migration path of labeled NSCs.Transplanted NSCs into ischemic rats can differen- tiated into astrocytes and neurons during the process of migration.

Objective: To study the effects of long chain PUFA, AA and/or DHA on the fatty acid composition, growth and development of the newborn rats hippocampal neurons in culture. Methods: The primary culture of the hippocampal neurons was carried out in vitro by using serum free medium and supplementing 4 ? mol/L AA, DHA, or AA and DHA, of which the total concentration were 4 ?mol/L, the ratios of AA∶DHA from 1∶2 to 16∶1, in medium. The hippocampal neuronal fatty acid composition was analyzed by gas chromatogram, and the neuronal size and length of hippocampal neurite were measured by image analysis. Results: There were significant positive correlations between the ratios AA∶DHA in medium and the AA percentage and the ratios AA∶DHA in hippocampal neurons. The soma area? body maximum? minimum diameter and process length of hippocampal neurons in the group, which the total concentration of AA and DHA was 4 ?mol/L, and the ratio AA∶DHA in medium was 2∶1 or 4∶1,were higher significantly than other groups and groups with other ratios of AA∶DHA. Conclusion: AA and DHA could promote the growth and development of hippocampal neurons in culture..
5.Intelligence Characteristics and Correlation with Motor Development of Children with Spastic Diplegia

ObjectiveTo investigate the intelligence characteristics and relativity with motor development of children with spastic diplegia.MethodsIntelligence and motor development of 46 children with spastic diplegia were tested by Wechsler Intelligence Scale and Motor Development Milestone (MDM), to find out difference of intelligence between the children with diplegia and the normal control group.ResultsIntelligence quotient (IQ) of children with spastic diplegia was lower significantly than the normal control, specially performance IQ (PIQ). PIQ was related with motor development. The more delayed motor development was, the more lower PIQ was. Subtest scores of animal egg, puzzle, block design and picture generalization were lower in spastic diplegic children with normal IQ than that of the normal control group, while vocabulary IQ (VIQ) was not different.ConclusionThe intelligence structure of children with spastic diplegia is imbalanced and PIQ of those children is significantly lower than the normal children, which is related to hands-eyes correspond, sight-motor correspond and speed, and space-sight ability.

The spores suspension of Streptomyces roseosporus D-38 irritated with 20mW He-Ne laser for 20 min were incubated on G1 medium plates containing 1. 9?g/mL of streptomycin. Ten percent of mutants increased the potency of daptomycin by streptomycin-resistance method, including the mutant LC-54, which could produce daptomycin 81. 2 mg/L, which was 39% higher than that of the beginning strain by flask fermentation.

Objective To investigate the values of three-dimensional visualization technology in the diagnosis and surgical treatment of hepatic hemangioma. Methods Thirty two patients with hepatic hemangioma who had been hospitalized during the period from July 2010 to January 2014 in our hospital were scanned by 64-slice spiral computerized tomography (CT) before surgical treatment. Three-dimensional (3D) reconstruction based on the CT data was carried out to achieve dignosis and surgical planning. Assisted with the 3D model , we chose the best surgical procedure for liver resection, protecting the major blood vessels near hemangioma and retaining normal liver tissue as far as possible. Results The location, size and shape of hepatic hemangioma, vascular variation, and spatial relationship with intrahepatic vessel were shown factually by three-dimensional reconstruction. All the hemangiomas were preoperatively assessed to be resectable. The compliance rate for preoperative surgical planning to actual surgery was 100%. Under assistance of the 3D model during surgery , 14 patients received laparoscopic hepatectomy and 18 underwent hepatectomy. Pringle′s maneuver was applied in 18 patients , with blocking time of (15.32 ± 7.12) min and blood loss of (188.63 ± 66.37) mL. The postoperative complications included pleural effusion in 5 patients and incision infection in one patients. Conclusions Three-dimensional visualization technology for the individualized diagnosis and treatment of hepatic hemangioma helps reduce surgical trauma and incidence of postoperative complications.
