Background Proliferation of the human Tenon capsule fibroblasts(HTFs) is a main cause of failure of filtering surgery.To search the drug of inhibiting the growth of the HTFs is essential for the improvement of successful rate of filtering surgery.Objective The aim of this study was to investigate the inhibitory effect of apigenin on HTFs and its mechanism.Methods Human Tenon capsular tissue was obtained during the strabismus correction surgery.HTFs was primarily cultured using explant method and identified using vimentin by immunochemistry.The 3-5 generation of cells were incubated to 96-well plate.Apigenin of 0,20,40,80,160 μmol/L was added into the medium,respectively,for 24,48,72 hours,and the proliferation of HTFs was detected by sulfonyl chloride (SRB) at the wavelength of 560 nm (A560).Bromodeoxyuridine (BrdU) of 10 μg/L was added to culture the cells for 48 hours to calculate the labeling rate of BrdU.The morphology of the cells was observed using Hoechst 33258 staining,and apoptosis and cells cycle were evaluated by flow cytometry.Results Cultured cells grew well with the positive response for vimentin,showing the green fluorescence in cytoplasm.SRB assay showed that the A560 value was gradually declined with the increase of the dosage of apigenin and prolong of time (Fgroup =480.306,P =0.000 ; Ftime =555.144,P =0.000).The labeling rate after 0,40,80 μmol/L apigenin acted for 48 hours was (87.860 ±0.632)%,(61.520±4.306)% and (23.480±4.472)%,showing a significant difference among the three groups (F =299.347,P =0.000).The labeling rate of HTFs for BrdU was significantly decreased in the 40 and 80 μmol/L apigenin groups compared with the 0 μmol/L apigenin group (P<0.05).Hoechse 33258 staining found that the number of the HTFs was gradually decreased and the cell number of karyopyknosis and nuclear deformation was increased with the increase of apigenin dosage.Percentage of cells in G0/G1 phase were raised and that in S and G2/M phase were declined in the higher dosage apigenin group,with a significant difference among the different groups (FG0/G1 =58.621,P=0.000;Fs =32.357,P=0.001 ;FG2/M =83.998,P=0.000).In the 72nd hour after acted by 0,40,80,160 μmol/L apigenin,the apoptosis rate of HTFs was (4.77±0.21) %,(13.24±1.35)%,(18.33±1.86) %,(31.58 ± 2.77) %,respectively,with a statistically significant difference among the four groups (F =204.791,P<0.05).Conclusions Apigenin restrains the growth of HTFs by evoking G0/G1 cell cycle arrest and inducing apoptosis in a dosage-and time-dependent manner.
3.Effects of immunosuppressor against kidney disease in treatment of 21 patients with cytomegalovirus pneumonia

Objective To study the clinical feature,treatment,and prognosis of the cytomegalovirus (CMV)pneumonia patients treated with immunosuppressor against kidney disease.Mlethod The patients received immunosuppressor against kidney disease in The First Affiliated Hospital of Sun Yat-sen University from June 1999 to December 2006.CMV antigen of leucocyte in the peripheral blood and/or bronchoalveolar lavage fluid of these patients were detected with immunocytochemical methods,and 21 patients were found suffering from CMV pneumonia.The 21 patients were introvenously injected with ganciclovir 5~10 mg/(kg?d),and the immunosuppressive agent treatment suspended.Their clinical feature and prognosis were retrospectively analyzed. Results The 21 patients received corticosteroids before CMV pneumonia contracted,of them,13 patients had been intensively treated with Methyllprednisolone with mean total dose(3.2?0.6)g.Of them,15 had been treated with cyclophosphamide with mean total dose(3.8?1.3)g.The median time from the beginning of using immunosuppressor to the onset of CMV pneumonia was 25(13~92)days.All patients had fever,cough, shortness of breath and X-ray showed interstitial pneumonia,of them,19 patients developed hypoxemia,and 11 patients' CMV antigen was positive in the leucocyte from bronchial lavage fluid.The result showed 9 patients survived and 12 died.The average duration of treatment with ganciclovir was(26.2?6.3)days. CMV pneumonia is a serious complication in patients who were treated with immunosuppressor against kidney disease.The mortality is high.Ganciclovir is a medicine of choice to treat CMV pneumonia.
4.Changes of physical functions among Chinese minority students from 1985 to 2005

Objective To study the changes of physical functions among minority students in China from 1985 to 2005, to provide evidence for health promotion to the minority students in the country. Methods Using data from the Chinese national survey on students physical fitness and health from 1985 to 2005, we analyzed the physical functions of 15 minority groups from primary and secondary schools, including Mongolian, Hui, Uygur, Zhuang, Korean, Tibetan, Yao, Li, Qiang, Buyi, Dong, Miao, Tu, Salar, Kirgiz, aged 7-18 years old. Results Some of the minority students had the higher levels of physiological functions and athletic ability, but some indicators decreased significantly. In terms of items related to 'speed' , most of the minority boys and girls improved, but items related to 'endurance' or 'vital capacity', were significantly declining. Part of the minority students bad better 'endurance' than students of Hart ethnicity in the cities. All the indicators of athletic ability declined in schoolboys and schoolgirls of Korean ethnicity. Kirgiz students were outstanding in vital capacity and speed quality while Uighur students had better physical development and vital capacity, but were poorer in running and jumping, reflecting the weakness of explosive power of lower limbs. On the contrary, Li and Zhuang students had shown a very good quality in running, jumping, male shoulder muscular strength and endurance items, etc. Conclusion The physical functions and the athletic ability of the minority students in China had the advantages and characteristics, which might be related to their living environment and habits.
5.Chemical study on fruiting bodies of Boletus vioaceo-fuscus.

OBJECTIVETo investigate the chemical constituents of Boletus vioaceo-fuscus.
METHODThe compounds were isolated with column chromatography. The structures were determined by spectroscopic techniques.
RESULTSix compounds were isolated from the fruiting bodies of Boletus vioaceo-fiuscus. They were identified as ergosta-5, 7, 22-triene-3beta-ol (1), dihydrofuran-2, 5-dione (2), (22E, 24R)-5alpha, 6alpha-epoxyergosta-8, 22-diene-3beta, 7alpha-diol (3), (22E, 24R)-5alpha, 6alpha-epoxyergosta-8 (14), 22-diene-3beta, 7alphadiol (4), cerebroside B (5) and adenosine (6), respectively.
CONCLUSIONAll the Compounds were obtained from the fruiting bodies of Boletus vioaceo-fiscus for the first time.

6.Differential protein analysis in rat renal proximal tubule epithelial cells in response to acetazolamide and its relation with the inhibition of AQP1.

AIMTo study the endogenous mechanism for the inhibition of aquaporin-1 expression in rat renal proximal tubule epithelial cells in response to acetazolamide.
METHODSPrimary cultured rat renal proximal tubule epithelia cells were divided into two groups: one was subjected to 1 x 10(-5) mol.L-1 acetazolamide, the other served as normal control. When grown to sub-confluency, the cells were disintegrated to perform isoelectrofocusing electrophoresis in order to find the differential proteins induced by the acetazolamide treatment. The differential proteins were defined by peptide mass fingerprinting technology.
RESULTSTwo differential proteins were found in the cell disintegrant. The pI 3.8 protein was reduced after treatment, which showed 21.4% similarity with the brush border membrane myosin from rat brain and testis, and 27% with glycogen phosphorylase; The pI 5.5 protein was increased on the contrary, with 20% similarity to phosphatidylinositol transfer protein alpha isoform.
CONCLUSIONAcetazolamide inhibited AQP1 expression probably by affecting the expression of pI 3.8 and pI 5.5 proteins.

7.Cross-sectional study on growth and development of Chinese Han students.

OBJECTIVETo analyze the current situation of the growth and development on Chinese Han children and adolescents.
METHODSUsing the 2004 National Survey on Students' Physical Fitness and Health, the body height, weight and chest circumference of 161,477 subjects aged 7 to 18 enrolled in this study were analyzed.
RESULTSIt showed that the levels of the growth and development in urban students were better than those of rural students (P < 0.01). The average level on stature of 12-aged students were 152.78 cm and 148.66 cm respectively for urban and rural boys, and the deference was 4.12 cm; the mean heights of urban and rural girls were 152.68 cm and 149.24 cm respectively, and the deference was 3.44 cm. The average level on body weight of 12-aged students were 44.75 kg and 39.27 kg respectively for urban and rural boys, and the deference was 5.48 kg; the mean weights of urban and rural girls were 42.91 kg and 39.17 kg respectively, and the deference was 3.74 kg. The average level on chest circumference of 12-aged urban students was beyond the rural students 3.68 cm and 2.56 cm respectively by gender. The average level on body height and weight of 7 to 14 aged students of east inshore areas were more than those of southwest, northwest and middle areas (P < 0.01). The mean body height and weight of students of southwest area were the lowest in whole country (P < 0.01).
CONCLUSIONIt reveals that attaching further importance to improve the work of students' physical fitness and health and nutrition in inland areas, especially in southwest areas and rural areas, would be contributory to the growth and development of the children and adolescents.

8.Analysis of motor performance status in students of Han nationality in fifteen provinces in China.

OBJECTIVETo analyze the motor performance status in students of Han nationality in fifteen provinces in china.
METHODSTotally, 161 804 students of Han nationality aged from 7 to 18 years old were involved in the Chinese Surveillance on Students' Physical Fitness and Health in 2004. Motor abilities were accessed with the aid of gripping power, 50 m dash, standing long jump, and 1-min sit-ups. Based on general statistical description, principal component analysis and linear regression, the development characters of students' motor performance were explored.
RESULTSThis research showed some characters similar to those of last ones: motor capability was improved in correlation with age; boys did better than girls, the difference between 18 year-old rural boys and rural girls was 15.3 kg, -2.0 s, 58.6 cm, 8.7/min; the urban students performed better than the rural ones, the difference between 15 year-old urban boys and rural boys was 0.9 kg, -0.2 s, 3.5 cm, 3.5/min. The first principal component might represent the 4 tests greatly. Regression analysis on principal component quantitatively interpreted the influence of factors such as age, sex and area.
CONCLUSIONSThe general principles of exercise quality development of students are still in work. Principal component analysis should be adequate and convenient in motor performance analysis.

9.Step-test on surveillance of students' physical fitness and health.

OBJECTIVETo analyze the role of step-test on large sample surveillance in students' physical fitness and health.
METHODSTotally, 36 237 students aged 12-, 14- and 17-year-old from fifteen Chinese provinces taking part in the 2004th National Surveillance on Students' Physical Fitness and Health were enrolled in this study. All of them passed through the step-test, of which the indices were calculated in combining with the performance time and pulse rates in resuming period. The means of step-test index were analyzed among various youth groups, and compared with the obesity, overweight and the normal-weight subjects.
RESULTSAs compared with the general level of physical growth, the means of step-test index were found higher in rural groups than in urban ones, and higher in southern groups than in northern ones. The highest means of index was found in the rural groups in having the lowest socioeconomic status, and also the lowest means found in metropolis groups having the highest socioeconomic status. These gaps were due to the change of life style, absence of physical exercise and relatively to the high body weight of the high socioeconomic-level groups. Among the obesity, overweight and normal-weight youths, the means of step-test index was shown in a gradient decrease tendency, and those significant gaps might be found between the obesity and the overweight, and the overweight and the normal-weight groups, showing that there should be harmful influences of overweight and obesity on the youth's cardiovascular function.
CONCLUSIONAs a safe and easily-taken measure for cardiovascular function, the step-test should be useful in regularly measuring the national students' physical fitness and health status.

10.Chemical constituents from the fruiting bodies of Abortiporus biennis

AIM To study the chemical constituents from the fruiting bodies of Abortiporus biennis (Bull.) Singer.METHODS The ethyl acetate fraction of methanol extract from A.biennis fruiting bodies was isolated and purified by silica,Sephadex LH-20 and RP-18 column,then the structures of obtained compounds were identified by spectral data.RESULTS Six compounds were isolated and identified as (22E,24R)-ergosta-5,7,22-trien-3ββ-ol (1),(22E,24R)-5α,8α-epoxyergosta-6,22-dien-3β-ol (2),cerevisterol (3),(22E,24R)-ergosta-4,6,8 (14),22-tetraen-3-one (4),1,3-dioleoyl-2-1inoleoylglycerol (5),cereboside B (6).CONCLUSION All the compounds are isolated from this fungus for the first time.
