Object To investigate the effects on the active components in Polygonum cuspidatum Sieb. et Zucc. and its different processed products. Methods The qualitative analysis of piceid and emodin are taken by TLC, and content of emodin was determined by TLCS and HPLC. Results The contents of emodin significantly increased in samples fried with rice wine, vinegar and salt compared with the crude. Conclusion Different methods and ajuvants made a certain influence on the contents of emotion in P. cuspidatum.
2.Necessity and Methods for Validation of Equipments of Preparations in Medical Institutions

OBJECTIVE: To introduce validation to the equipments of self-made preparations in medical institutions.METHODS: The necessity and difficulties for the validation of the equipments of self-made preparations in medical institutions were analyzed from aspects of the specificity of self-made preparations,the status quo of the preparation equipment as well as the requirements on the equipments etc.RESULTS: The scope and the procedures of the validation on the equipments of preparations had been presented.CONCLUSIONS: Medical institutions should adopt suitable methods,approach and validation procedures to develop corresponding validation work for equipments of preparations based on their own condition.
3.Safety of Simvastatin in the Treatment of Patients with Dyslipidemia

OBJECTIVE: To explore the safety of different dosages of simvastatin in patients with dyslipidemia. METHODS: 269 patients with essential dyslipidemia were randomly assigned to receive simvastatin (5, 10 or 40 mg?d-1) for 8 weeks. The curative efficacy and the adverse reactions were compared among groups. RESULTS: There were significant differences across the 3 groups (5, 10 or 40 mg?d-1) with regard to the total effective rates on regulating total cholesterol, triglyceride, high-density lipoprotein cholesterol(P0.05). The total incidence of ADRs was 14.87%, which appeared mainly as gastrointestinal reaction and myosalgia, and the differences across the 3 groups were significant (P=0.03), but no significant difference was noted with regard to sex and age (P=0.46,P=0.07). CONCLUSION: Simvastatin may induce ADRs, but in most cases the ADRs were mild,and the incidence of the ADRs increased as the dosage of simvastatin increased.Tailoring the dosage of simvastatin to patients’ clinical characteristics is conducive to patients’ medication safety.
4.Relationship Between Gene Frequency of HLA-DRB1*03 Genes and Expression of IL-18 in Chronic Hepatitis B Patients

OBJECTIVE To study the relationship between HLA-DRB1*03 genes and expression of cytokine IL-18 in chronic hepatitis B patients.METHODS A total of 153 chronic hepatitis B patients and 138 normal healthy people were detected.HLA-DRB1*03 alleles were conducted in all the patients with polymerase chain reaction-sequence specific primer(PCR-SSP),and IL-18 was detected by ELISA.RESULTS Thirty-four individuals with chronic hepatitis B patients were HLA-DRB1*03 genes positive,the frequency was 22.22%,which was higher than the frequency of HLA-DRB1*03 in heathy individuals.The average level of IL-18 in chronic hepatitis B patients with HLA-DRB1*03 negatives was significantly higher than in HLA-DRB1*03 positives.CONCLUSIONS HLA-DRB1*03 closely relates with susceptibility to chronic hepatitis B.In chronic hepatitis B patients,the average level of Th1 cytokines in HLA-DRB1*03 negatives is higher than that in HLA-DRB1*03 positives.

6.A Preliminary Study on Anti-inflammatory Effects of Rhamnocitrin from Oxytropis falcata Bunge

Objective To observe the anti-inflammatory effects of rhamnocitrin in Oxytropis falcata Bunge;To provide experimental evidence for searching the anti-inflammatory active component of Oxytropis falcata Bunge. Methods Kunming mice or Wistar rats were randomly divided into model group, naproxen group and rhamnocitrin high-, medium-, low-dose groups, and corresponding medicines were administered by gavage for 7 days. Animal inflammatory models were established respectively by the mice ear swelling induced by dimethylbenzene, the mice abdominal capillary permeability induced by acetic acid, the rat paw edema created by injection of albumen and the rat granuloma induced by cotton pellet wool. The effects of rhamnocitrin on acute and chronic inflammatory animal models were observed. Results Different doses of rhamnocitrin groups could reduce the animal ear swelling, abdominal capillary permeability, paw edema and granuloma. Compared with the model group, there were obvious differences in rhamnocitrin groups (P<0.05). High dose rhamnocitrin group and naproxen group showed relatively strong inhibition effects on swelling, with statistical significance (P<0.05). Conclusion Rhamnocitrin has anti-inflammatory effects and is an important biological component of Oxytropis falcata Bunge.
7.Apoptosis induced by simvastatin in rat vascular smooth muscle cells through calpain and caspase-3-dependent pathways

AIM: Hydroxymethylglutaryl CoA (HMG-CoA) reductase inhibitors, such as simvastatin, have been shown to reduce atherosclerotic cardiovascular morbidity and mortality by mechanisms unrelated to its lipid-lowering effect. Several studies have shown that simvastatin induces apoptosis in a varieties of cell lines including vascular smooth muscle cells (VSMC). The aim of this study was to investigate the signal pathways involved in apoptosis induced by simvastatin. METHODS: Cultured VSMC were treated with simvastatin. Calpain activity was determined by measuring Ca 2+ ionophore-specific calpain substrate (suc-LLVY-AMC), caspase-3 activation was detected by Western blot, and apoptotic changes were distinguished by annexin Ⅴ binding and DNA laddering. 8.Application of ITS2 Barcoding in Identification of Oxytropis Falcate Bunge

Objective To identify Oxytropis falcate Bunge from its adulterants;To secure the quality and clinical efficacy of Oxytropis falcate Bunge. Methods Genomic DNA was extracted. ITS2 fragment of nuclear gene was amplified and sequenced. The homology of the sequences was analyzed, and the related data were analyzed by software MEGA. The NJ (neighbor-joining) tree was constructed based on ITS2 sequence. Results The intraspecific genetic distance of Oxytropis falcate Bunge (K2P distance) was 0, and the interspecific genetic distance (K2P distance) between Oxytropis falcate Bunge and its adulterants ranged among 0.068-0.084. The constructed NJ tree showed that ITS2 could identify Oxytropis falcate Bunge from its adulterants correctly. Conclusion ITS2 barcode can be used to identify Oxytropis falcate Bunge from its adulterants effectively, and this study provides important molecular evidence for the identification of Oxytropis falcate Bunge and safety in its clinical use.
9.Progress in Noninvasive Assessment of Liver Fibrosis in Patients with Chronic Hepatitis B

Chronic hepatitis B(CHB)is one of the most commom cause of liver fibrosis. Accurate assessment of liver fibrosis is essential for the strategy of treatment and judgement of prognosis . Liver biopsy is the gold standard for staging fibrosis,but it is invasive with high cost,low reproducibility and poor acceptance by patients. Therefore,it is urgent to explore a noninvasive modality for the assessment of liver fibrosis. Recent evidence highlights that elastographic techniques, biochemical markers and the diagnostic model consisted of several serum markers have the potential for the diagnosis of liver fibrosis. This article reviewed the progress in noninvasive assessment of liver fibrosis in patients with CHB.

AIM To examine the effect of metabotropic glutamate receptor ligand (s) 4 carboxy 3 hydroxyphenyl glycine 〔(s) 4C3HPG〕 on kainic acid induced epileptic seizures and brain damage in rats. METHODS Rats were given (s) 4C3HPG (500 nmol/rat) by icv infusion prior to kainic acid (KA) injection ip(10 mg?kg -1 ). The behavoir changes, electrographic seizure duration and histological alterations between different group of rats were compared.
