1.A preliminary study on the resin casting of normal male canine prostatic duct and acinus system

Objective To investigate the three-dimensional structure of normal male canine prostat- ic duct and acinus system,and to study the mechanism of intraprostatic urinary reflux(IPUR)resulting from high pressure of the posterior urethra by experiment.Methods Using scanning electron microscopy (SEM)and corrosion casting methods,high pressure of the prostatic urethra was produced based upon IPUR in normal male canines.Acrylonitrile-butadiene-styrene(ABS)casting solution was injected into 8 Beagle canines'normal prostates to induce intraprostatie resin solution reflux.Results Six casting molds of nor- real canine prostate specimens were obtained,and the casting failed in 2.By corrosion and casting treatment, the prostate appeared to consist of several glandular lobes.The columnar apophyses of glandular surface were observed.The most glandular ducts pointed outward from urethral peristome.Each glandular duct system was made up of many bifurcated branches from the bough as a duct tree.More than 90% of the bifurcations were Y-shaped,and 6 bifurcated branches were rarely seen.The tip of the duct was vesicular.The diameter and length of the bough were(0.37?0.14)mm and(1.14?0.04)mm,respectively.The intersectional angle of urethra and peripheral glandular duct was the biggest.Peripheral glandular duct was perpendicular or con- verse to urinary flow.Peripheral glandular aeinus appeared elliptic and velvet.The mesh-like and crater-like depressions were shown on the surface of central glandular aeinus.Ejaculatory ducts independently and branchlessly entered the urethra.Conclusions The resin corrosion casting methods combined with SEM can clearly show the three-dimensional structure of normal male canine prostatic duct and acinus system. IPUR may easily occur in the peripheral glands from the morphological view.It is suggested that prostatitis occurs more commonly in peripheral glands,confirming a fact that high pressure of the posterior urethra can induce IPUR.
2.Dynamic changes of estradiol and pregnenolone levels in serum and hippocampus of female rats with epilepsy

Objective To observe the dynamic changes of estradiol(E2)and pregnenolone (PREG)levels in serum and hippocampus of female rats with epilepsy and explore the relationship between E2 level in the hippocampus and severity of seizure. Methods The seizure model was established by injecting kainite(KA)into the hippocampus of amygdala-kindled rats.The behaviors at the moment of epileptic seizure were observed and serum and hippocampus samples were collected at 0,1,2,3,4,5,6,12 and 24 h after seizure.Rats in the control group was injected the same volume of saline at the sanle time points.The levels of E2 and PREG in the Seizure and control groups were measured by radioimmunoassay and enzyme linked immunosorbent assay,respectively.These results were statistically analyzed. Results Epileptic Seizure was showed after KA injection 5 to 10 min with the strongest seizure appearing at 3 h.The level of E2 was not significantly different in serum of the rats with epileptic seizure.While the level of E2 significantly changed in the hippocampal tissue after seizure:it elevated 1h after epileptic seizure,peaked at 4 h and returned to normal at 12 h;statistically significant differences of E2 level in the hippocampai tissue was showed among different time points(P<0.05).Besides,the E2 level in the hippocampus was increased gradually with the aggravation of the seizure(R~2=0.646,P<0.05).No significant differences were found in the PREG level of the hippocampal tissues between the rats treated with KA and the rats in the control group. Conclusion Epileptic seizure can induce local E2 synthesis in the rat hippocampus,thus,the E2 level in the hippocampus and severity of seizure is positively correlative.
3.Pharmacokinetics of tramadol hydrochloride in the extracellular fluid of mouse frontal cortex studied by in vivo microdialysis.

The paper aims to explore the studying method for the pharmacokinetics of drugs in target organs, the pharmacokinetic process of tramadol hydrochloride in the extracellular fluid of frontal cortex (FrCx) of mice was investigated. Six male mice (Kunming strain) were anaesthetized (urethane, 1.8 g x kg(-1), ip) and secured on a stereotaxic frame. A microdialysis probe was implanted into the FrCx and perfused with artificial cerebrospinal fluid at a flow rate of 2 microL x min(-1). One hour later, mice were administrated (ip) with tramadol hydrochloride (50 mg x kg(-1)) and dialysates were collected continuously at 12-min intervals (24 microL each) for 6 h. The tramadol concentration in dialysates was determined by HPLC-Ultraviolet detection method, and the concentration-time curve and pharmacokinetic parameters of tramadol were calculated with DAS software. The results showed that the pharmacokinetic process of tramadol in the FrCx extracellular fluid of mice was fitted to a two-compartment open model, and the main pharmacokinetic parameters t1/2alpha, t1/2beta, t(max), C(max) and AUC(0-infinity) were (0.27 +/- 0.05) h, (2.72 +/- 0.24) h, (0.50 +/- 0.10) h, (2 110.37 +/- 291.22) microg x L(-1) and (4 474.51 +/- 441.79) microg x L(-1) x h, respectively. In conclusion, a studying method for pharmacokinetics of drugs in the target organ is established, which is simple and feasible. Tramadol hydrochloride shows a two-compartment model in the extracellular fluid of the mouse FrCx, and the distribution- and elimination half-life are 0.5 h and 2.7 h, respectively.

4.Analysis of harmful heavy metals, sulfur and main trace elements from Paeoniae Radix Alba before and after sulfur-fumigated process by ICP-AES method.

OBJECTIVETo analyze the contents of harmful heavy metals, sulfur and trace elements from Paeoniae Radix Alba before and after sulfur-fumigated process.
METHODSamples were pretreated by wet digestion and then subjected to elements analysis by inductively coupled plasma-atomic emission spectrometry (ICP-AES).
RESULTThe contents of Cu and S elements were increased, while the content of Zn element was decreased in Paeoniae Radix Alba after sulfur-fumigated process compared with the samples dried in the sun. Moreover, heavy metallic elements, such as As and Hg, were detected in Paeoniae Radix Alba after sulfur-fumigated process.
CONCLUSIONSulfur fumigated process may cause residual of heavy metals and sulfur which are harmful to human body and change the contents of some trace elements.

Adolescent idiopathic scoliosis(AIS)is a complex three-dimensional spinal deformity,characterized by lateral curvature and vertebral rotation.The medical imaging techniques are essential for determination of severity of scoliotic spine, prediction of progression and assistance in the decision-making process of treatment for scoliosis, including radiograph, stereoradiography, computed tomography, magnetic resonance imaging and ultrasound.This paper reviewed their application from the view of measure parameters,reliability and va-lidity,as well as merits and demerits.It is possible to assess the three-dimensional nature of scoliosis in the future.
6.Analysis on establishment of ISO9001 quality management system in project management institutions

7.Development of SYBR Green I real-time RT-PCR for the detection of Ebola virus.

In order to establish a rapid and accurate method for the detection of Ebola virus (EBOV), the primers used in SYBR Green I real-time RT-PCR were designed based on the EBOV NP gene sequences published in GenBank. The SYBR Green I real-time RT-PCR was established and optimized for the detection of EBOV. The EBOV RNA that was transcribed in vitro was used as a template. The sensitivity of this method was found to reach 1.0 x 10(2) copies/microL and the detection range was 10(2) - 10(10). No cross reaction with RNA samples from Marburg virus, Dengue virus, Xinjiang hemorrhagic fever virus, Japanese encephalitis virus, Influenza virus (H1N1 and H3N2) and Porcine reproductive and respiratory syndrome virus E genomic RNA was found. The method would be useful for the detection and monitoring of EBOV in China.

OBJECTIVETo identify zebrafish mutants with myelopoiesis defects by ENU mutagenesis and large-scale forward genetic screening.
METHODSMale zebrafish were mutagenized with N-ethyl N-nitrosourea to induce mutations in the spermatogonial cells to generate the founders, which were outcrossed with AB to raise F1 fish. The F1 fish from different founders were mated to generate the F2 families. The F3 embryos from F2 sibling crosses were screened by Sudan black B staining and neutral red staining.
RESULTSA total of 350 F2 families from F1 sibling crosses were screened, and 1424 F2 crosses were analyzed. Six mutations were identified resulting in abnormal Sudan black B staining and neutral red staining, indicating the involvement of neutrophil deficiency or macrophage abnormalities.
CONCLUSIONIt is simple and cheap to induce and screen myelopoiesis deficiency in zebrafish by ENU chemical mutagenesis and Sudan black B staining and neutral red staining. These mutants shed light on the identification of the genes important to myelopoiesis in zebrafish.

OBJECTIVETo screen and identify zebrafish mutants with erythropoiesis defects by N-ethyl-N-nitrosourea (ENU) mutagenesis and large-scale forward genetic screening using beta e 1 as the marker.
METHODSThe chemical mutagen ENU was used to treat healthy wild-type male fish (AB strain, F0). The surviving ENU-treated fish were mated with wild-type female fish to generate F1, and further F2 family was generated by F1 family intercross. The adult F2 fish were intercrossed within each F2 family and the resulting F3 embryos from each crossing were subjected to whole mount in situ hybridization (WISH) with the beta e 1 probe. Mutagenesis was performed by treating the male zebrafish with ENU to induce mutations in pre-meiotic germ cells to generate the founders, which were outcrossed to obtained the F1 fish. The F1 fish from different founders were mated to generate the F2 families. F3 embryos from the sibling cross in the F2 family were examined by whole mount in situ hybridization using beta e 1-globin probe. The putative mutants were then characterized with different hematopoiesis markers.
RESULTS AND CONCLUSIONWe identified 4 beta e 1-deficient mutants with erythropoiesis defects, including two with specific erythiod lineage defects and two with concurrent lymphopoiesis defects.

10.Prophylaxis of Bortezomib-Induced Peripheral Neuropathy in Patients with Multiple Myeloma by High-Dose Intravenous Mecobalamin.

OBJECTIVETo evaluate the efficacy and safety of high-dose intravenous mecobalamin (HDIME) for the treatment of bortezomib-induced peripheral neuropathy(BIPN) in the patients with multiple myeloma (MM).
METHODSA total of 65 newly diagonsed patients with multiple myeloma receiving bortezomib in Tianjin Medical University General Hospital were enrolled in this single-centre randomized clinical trial from July 2012 to May 2016. Out of 65 patients 38 in control group received bortezomib-based chemotherapy and 27 patients in HDIME group received the additional high-dose intravenous mecobalamin.
RESULTSThe incidence of BIPN in HDIME group was lower than that in control group(29.63% vs 55.26%, χ=4.197,P<0.05). Whether the BIPN rate of Grade 2 or 3 and above in HDIME group significantly decreased as compared with control group(18.52% vs 47.37%,χ=5.746,P<0.05) (3.71% vs 21.05%, χ=3.983,P<0.05). The BIPN rate of less than 5 cycles of bortezomib was not significantly different between HDIME and control groups(χ=2.714,P>0.05). Overall effective rate of HDIME group and control group was 77.78% and 73.68%(P>0.05) respectively. Neither PFS nor OS was significantly different between HDIME group and control group(P>0.05). Treatment-related toxicity was only mild rash in 1 case. No other side-effects including nausea, abdominal pain, and hypotension occurred.
CONCLUSIONHDIME has a good efficacy for the prophylaxis BIPN and and without serious side effects.
