1.Research on the detection rate of couples at high risk of bearing babies with severe thalassemia among the people at reproductive age in foshan city,Guangdong province

Objective To investigate the detection rate of couples at high risk of bearing babies with severe thalassemia and the occurrence rate of birth defect caused by severe thalassemia among the people at reproductive age in Foshan city ,Guangdong prov‐ince .Methods Peripheral blood samples were collected from 17 522 preparing pregnant couples of child bearing age .Mean corpus‐cular volume (MCV) <80 fL was used as the positive indication for screening thalassemia .And 348 positive samples were detected by molecular biological technique for further analysis .Statistical analysis was conducted based on the couple′s genetic test results . Results Among the people at reproductive age ,the detection rates of couples bearing babies with Hb Bart′s Hydrops Fetalis ,He‐moglobin H disease andβ‐thalassemia were 0 .36% ,0 .47% and 0 .20% respectively ,the total was 1 .03% .The occurrence rates of birth defect caused by the above severe thalassemia were 9 .0/10 thousand ,11 .8/10 thousand and 5 .0/10 thousand ,the total was 25 .8/10 thousand .Conclusion This research illustrated the degree of damage of birth defect caused by severe thalassemia among the people at reproductive age ,which could provide valuable fundamental materials for the launch of eugenic work in this region .
2.A Progress in Detection of Interactions Between Macromolecules: Linked FRET Using Three Color Fluorophore

Fluorescence resonance energy transfer (FRET) is the energy transfer from an activated donor fluorophore to a receiving fluorophore. The efficiency of the energy transfer is the function of the distance between the two fluorophores, and is very sensitive to the distance. Its effective response distance is between 1~10 nm, thus it can be used to measure the distance between atoms or molecules. The feature of FRET is very useful in researches on conformational changes, interaction between macromolecules and signal transductions within live cells, and FRET has become a powerful tool in biomedical investigations. However, biological processes often involve interactions between more than two macromolecules, and FRET using two color fluorophores cannot meet the research demand. Recently, two research groups made breakthrough, establishing a novel FRET technique using three color fluorophores based on confocol microscopy and flow cytometry, respectively. The invention will significantly advance researches in biological and related fields.
3.Changes before and after docetaxel chemotherapy and significances of T lymphocyte subsets of patients with non-small cell lung cancer

Objective To investigate the changes before and after docetaxel chemotherapy and significances of T lymphocyte subsets of patients with non-small cell lung cancer.MethodsThe clinical data of 100 cases of patients with non-small cell lung cancer treated in our hospital from January 2013 December 2016 were retrospectively analyzed, these patients were regarded as lung cancer group, another 100 cases of healthy subjects in our hospital received over the same period were selected as healthy group, the changes of T lymphocyte subsets of the two groups were statistically analyzed.ResultsThe CD+3, CD+3CD+4, CD+4/CD+8, NK cells before and after chemotherapy of the lung cancer group were significantly lower than the healthy group (P<0.05), the CD+3, CD+3CD+4, CD+4/CD+8, NK cells before chemotherapy were significantly lower than after chemotherapy (P<0.05), the difference of CD+4/CD+8 between the two groups was not significant;The CD+3, CD+3CD+4, CD+4/CD+8, NK cells of the disease control patients in the lung cancer group were significantly lower than the disease progression patients, healthy group (P<0.05), the differences of CD+3, CD+3CD+4, CD+4/CD+8, NK cells between the disease control patients and healthy group were not significant, the difference of CD+4/CD+8 between the two groups was not significant.ConclusionThe changes of T lymphocyte subsets before and after docetaxel chemotherapy of patients with non-small cell lung cancer can provide effective basis for clinical evaluation of cellular immune function and efficacy of chemotherapy of patients with non-small cell lung cancer, so is worthy of the clinical's full attention.
4.Effect of basic fibroblast growth factor gene transfection on bone marrow mesenchymal stem cell transplantation for diabetes mellitus

BACKGROUND:Existing studies have shown that bone marrow mesenchymal stem cel s can significantly improve islet function in diabetic rats to decrease excessively high blood glucose level, which may be related to the enhancement of differentiation ability of autologou pancreatic stem cel s.
OBJECTIVE:To observe the therapeutic efficacy of basic fibroblast growth factor gene eukaryotic expression vector (PEGFP-C3-BFGF) transfection of bone marrow mesenchymal stem cel s in diabetic rats.
METHODS:Recombinant adenovirus (Ad.aFGF) mediated PEGFP-C3-BFGF was transfected into bone marrow mesenchymal stem cel s, and PEGFP-C3-BFGF expression was observed using fluorescence microscopy. Eighty Sprague-Dawley rats were randomly divided into normal control group, diabetes group, transplantation group, gene transfection group, with 20 rats in each group. After modeling, rats in different groups were given portal vein injection of normal saline, PBS, 1 mL of bone marrow mesenchymal stem cel suspension, and 1 mL of PEGFP-C3-BFGF-transfected bone marrow mesenchymal stem cel suspension. RT-PCR method was used to detect mRNA expression of matrix metal oproteinases in pancreatic tissue of rats in each group. Blood glucose levels of rats were detected at 24 hours, 3, 7, 14, 21 days after transplantation. ELISA method was used to detect plasma insulin levels in rats. Pathological changes of the pancreas were observed using hematoxylin-eosin staining.
RESULTS AND CONCLUSION:Under the fluorescence microscope, PEGFP-C3-BFGF transfected into cel s after 48 hours showed significant specific red fluorescence. Two weeks after transplantation, matrix metal oproteinases mRNA expression was significantly increased in the diabetes group compared with the control group (P<0.05), while it was decreased in the transplantation and gene transfection groups compared with the diabetes group (P<0.05). 5.Efficacy and safety of budesonide/formoterol single inhaler therapy versus budesonide inhaler plus formoterol inhaler in children with bronchial asthma

Objective To compare the efficacy between budesonide/formoterol single inhaler therapy and budesonide inhaler plus formoterol inhaler in children with bronchial asthma.Methods In randomized, open, parallel-group trial, 80 cases of children with bronchial asthma in children asthma center of Children's Hospital of Hebei Province were divided into B/F group ( budesonide/formoterol single inhaler therapy ) and B +F group (budesonide inhaler plus formoterol inhaler), 40 cases in each group.All patients in B/F group inspired budesonide formoterol sing inspirator [80μg (budesonide)/4.5μg(formoterol)/snuff], and all patients in B+F group inspired budesonide(100μg/snuff) and formoterol(4.5μg/snuff), 1 snuff/time, respectively, both groups were treated with 2 times/d,12 weeks.The visual analog scale(VAS), lung function as forced expiratory volume in one second(FEV1)%, peak expiratory flow(PEF) and clinical symptoms were investigated.Results Compared with B+F group in the same time period, PEF% and FEV1%on the 12th week were higher, VAS score on the 4th, 8th and 12th week were lower, symptoms scores on 8th and 12th were lower in B/F group, respectively(P<0.05).Conclusion The efficacy of budesonide/formoterol single inhaler therapy is better than that of budesonide inhaler plus formoterol inhaler in children with bronchial asthma, which is more convenience and has a better compliance.
6.Research advances in circulating miRNAs for diagnosis of hepatobiliary diseases

MicroRNAs (miRNAs)are involved in the pathological and physiological process of life activities,and some of them exist stably and abundantly in serum,plasma,and other body fluids.Previous studies have indicated that there are significant differences in the expres-sion patterns of circulating miRNAs between patients with viral hepatitis,liver fibrosis,liver cirrhosis,hepatocellular carcinoma,drug-in-duced hepatitis,and other hepatobiliary diseases at different disease stages.Among these circulating miRNAs,some can act as novel nonin-vasive biomarkers for the early diagnosis,disease progression,and prognostic evaluation of related diseases.The latest research advances in circulating miRNAs for the diagnosis of hepatobiliary diseases are reviewed.

Nitric oxide, an intracellular messenger molecule linked to activation of N Methyl D aspartate(NMDA) receptor, plays a role in morphine tolerance and dependence. Nitric Oxide Synthase (NOS) inhibitors attenuate or block morphine actions. NO/NOS system has an effect on morphine induced place preference. Actions of NO/NOS system are involved in the activation of opioid receptors. The effect of NO/NOS system on the expression of morphine withdrawal symptoms may be partially mediated by noradrenergic neuron system. Thus reviews on interactions between NO/NOS system and opioid system are very important towards applications of opioid analgesic as well as treatment against opioid tolerance and withdrawal.

Acidic environment of organelles of eukaryotic cells plays an important role in endocytosis, the secretion of lysoso-mal enzymes and other physiological activities.V-ATPase (vacuolar ATPases) and CLC (chloride channel) family are widely distribu-ted and present in virtually all eukaryotic cells in intracellular membranes and in the plasma membrane.They are responsible for the a-cidification of the vesicular interior.In this paper, the distribution, structure, mechanism of action and physiological and pathological significance of V-ATPase and CLC protein are discussed.
9.Research on the psychological intervention of peri-operation in the patients with endometrial carcinoma

Objective:To evaluate the effectiveness of psychological intervention on improving the survival quality of patients with endometrial cancer during the operation. Methods:Sixty-two patients included in the eligible subjects were randomly divided into experimental group (n=31) and control group (n=31). The control group was given routine nursing intervention and the experimental group was given psychological intervention. Admission, discharge and discharge after one year follow-up measurement of quality of life were recorded. Results:Two patients discharged from the SF-36 scale score difference were statistically significant (t=11.3152, P<0.05), statistically significant long-term effect one year after the score difference (t=9.2025, P<0.05), the test group was better than control group. Conclusion:Psychological intervention is effective to improve the quality of life of patients with endometrial cancer during operation period and long-term.
10.Inhibition of viral myocarditis by Astragaloside through IL-23/IL-17 signaling pathway

Objective To explore the role of interleukin-23 (IL-23)/interleukin-17 (IL-17) signaling pathway in viral myocarditis (VMC) and evaluate the intervention effect of Aastragaloside. Methods Seventy-five male BALB/c mice were randomly divided into 4 groups, control group (n=15), model group (n=20), low-dose intervention group (n=20) and high-dose intervention group (n=20). Mice in control group were inoculated with 0.1 ml virus cultivation solution intraperitoneally while mice in the other 3 groups were treated with 0.1ml virus cultivation solution containing 1×102 TCID50 coxsackievirus B3 (CVB3) to establish VMC model. On the day of inoculation, mice in low- and high-dose intervention groups were intra-gastrically administered with 0.1 ml of 1% and 9%Astragaloside solution respectively, whereas those in control and model groups were treated with 0.1 ml carboxymethycellulose solution. Astragaloside or carboxymethycellulose was given once a day and continued 15 days. The number of mice death and the performance of mice were recorded in experimental period. All mice were sacrificed on day 15. The heart and blood sample were obtained. Histological cross sections of heart were stained with hematoxylin-eosin and scored for myocardial histopathologic changes under optical microscope. Th17 cells were analyzed by flow cytometry. The mRNA and protein expression levels of myocardial IL-23 and IL-17 were detected by real-time quantitative PCR and Western blotting, respectively. Results The mortality was statistically significant differ-ences among the four groups (P= 0.013), which was the lowest in the control group. The myocardial histopathologic scores, the percen-tage of Th17 cells, as well as expression levels of myocardial IL-23 and IL-17 mRNA and protein were significantly lower in high-dose intervention group than those in model group and low-dose intervention group, but higher than those in control group (P < 0.05). The myocardial histopathologic scores, the percentage of Th17 cells, as well as ex-pression levels of myocardial IL-23 and IL-17 mRNA and protein were significantly higher in model group and low-dose in-tervention group (P < 0.05). There were no significant difference in the above mentioned indicators between low-dose inter-vention group and model group (P > 0.05). Conclusions Astragaloside may dose-dependently protect against VMC by in-hibiting IL-23/IL-17 signaling pathway.
