2.Comparative study of optical coherence tomography angiography and ICGA in central serous chorioretinopathy

AIM:By analyzing optical coherence tomography angiography (OCTA) characteristics of central serous chorioretinopathy (CSC) and comparing the differences of CSC between OCTA and indocyanine green angiography(ICGA), to explore if OCTA can substitute ICGA for diagnosis of CSC patients, and guide the treatment of photodynamic therapy (PDT).METHODS: We reviewed 30 eyes of 30 patients with CSC, who were diagnosed by fluorescein angiography (FFA) and ICGA at Beijing Tongren Eye Center from November 2015 to March 2016.All patients underwent best-corrected visual acuity (BCVA) measurement, intraocular pressure, slit-lamp examination, indirect ophthalmoscope, color fundus photography, FFA, ICGA and OCTA.FFA and ICGA were captured by Spectralis HRA + OCT (Spectralis HRA + OCT;Heidelberg Engineering, Heidelberg, Germany).OCTA was performed by RTVue XR Avanti device (OptovueInc, Fremont, CA) with 6mm×6mm Angio Retina mode.The software (version 2017.100.0.1;OptovueInc) automatically segmented the tissue into four layers, the characteristics of choriocapillaris layer were analyzed.At the same time, the differences between OCTA and ICGA images were compared among CSC patients.The maximum diameters and areas of both choroidal hyperperfusion in ICGA and high flow signal in OCTA were measured.Then, the paired t test was used to analyze the differences between the maximum diameter and area of OCTA and ICGA measurement.RESULTS: Among 30 cases, high blood flow signals of OCTA were clearly visible in 27 cases, namely the coarse grain region;the inner low flow signals surrounded by high blood flow signals were seen in 21 cases;the outer low flow signals surrounding high blood flow signals were seen in 7 cases.High blood flow signals of OCTA were corresponded with the choroidal hyperperfusion of ICGA images;among these 30 cases, there were low reflection shadows in choroidal hyperperfusion with ICGA for 22 cases, for 21 cases out of these 22 cases, low flow signals inside of high flow signals of OCTA could be seen;9 out of 30 cases, there were low reflection halo outside of choroidal hyperperfusion of ICGA, and 7 out of these 9 cases, low flow signals outside of high flow signals of OCTA could be seen;still for those 30 cases, leakage point in late ICGA could be seen with 14 cases, however, special flow signals in OCTA could not be seen for them.For ICGA, the maximum diameter of choroidal hyperperfusion was 1.589±0.295mm, whose area was 0.705±0.131mm2;while for OCTA, the maximum diameter of high flow signal was 1.576±0.293mm, whose area was 0.745±0.138mm2.By using paired t test, there was no statistical difference between the maximum diameter of choroidal hyperperfusion in ICGA and the maximum diameter of high flow signal in OCTA, nor difference between the area of ICGA and OCTA.CONCLUSION: The high flow signals can be clearly visible in OCTA, which are corresponded with choroidal hyperperfusion in ICGA.OCTA can substitute ICGA for diagnosis of CSC patients, and guide the treatment of PDT.

4.Methodological evaluation of urinary mercury detection using Zeeman atomic absorption mercury analyzer

Objective To evaluate the method for detection of urinary mercury using a Zeeman atomic absorption mercury analyzer and to provide a reference for selecting a convenient method for mercury detection in experiments and clinical diagnoses. Methods Urinary mercury was detected by Zeeman atomic absorption spectroscopy (ZAAS) and hydride generation atomic absorption spectrometry (HG-AAS), and the detection limit, accuracy, precision and consistency of the two methods were compared. Results The Data collected by ZAAS and HG-AAS showed a good linear relationship in the range of 0 -1000 ng/mL (ZASS, R2 =1. 0000) and 0 -20 ng/mL (HG-AAS, R2 =0. 9990). The detection limits of ZAAS was 0. 156 ng/mL and that of HG-AAS was 1. 593 ng/mL, indicating that ZAAS is more sensitive. The recovery rate of standard addition of ZAAS was between 97. 5% and 103. 2%, and that of HG-AAS was between 95. 6% and 104. 5%. After measurement of 10 ng/mL and 100 ng/mL mercury standard solutions repeated for 10 times, the relative standard deviation (RSD) of ZAAS was 0. 30% and 0. 36% respectively, and the RSD of HG-AAS was 2. 82% and 1. 11%, respectively. The accuracy and precision of both the two method met the standards of GBZ/T 210. 5-2008, and the precision of ZAAS was better. A total of 30 urine samples were measured by these two methods. The results were compared with paired-samples t-test and showed a non-significant difference (P > 0. 05), indicating a high consistency of these two method (R2 =0. 9961). Conclusions ZAAS is a convenient and accurate method for the detection of urinary mercury, with a relatively low detection limit and better precision.
5.Establishment of the deep cervical lymph node metastasis model of tongue VX2 carcinoma and observation of its metastatic features

Objective To establish the deep cervical lymph node metastasis model of tongue VX2 carcinoma in rabbits and to observe its metastatical features. Methods The VX2 carcinomas were transplanted into the left ventral submucosas of tongues of 16 rabbits by injecting carcinoma mass suspension. According to the time the rabbits were killed after the transplantation of VX2 carcinoma, they were divided into four groups, or 7-, 14-,21- and 28-day groups, and 4 rabbits in each group. The specimens of transplanted tongue carcinomas and cervical lymph nodes in the rabbits were examined histopathologically after they were killed. Results All rabbits developed VX2 tongue carcinomas at the transplanted sites, which were confirmed with pathological examination, and the mean diameters of tongue carcinomas of 7-, 14-, 21- and 28-day groups were (0.74±0. 06)cm(x+s), (1.62±0.06)cm, (1.82±0.04) cm and (2. 52±0. 07)cm respectively. No cervical lymph node was palpable at bilateral necks in each rabbit of 7-, 14- and 21-day groups, and only 4 rabbits of 28-day group had palpable lymph nodes at ipsilateral necks. The histopathological examination revealed no lymph node metastasis in the rabbits of 7-day group. Ipsilateral deep cervical lymph node metastases were confirmed histopethologically in each rabbit of 14-, 21- and 28- day groups, which located beside larynx-trachea. Two of 4 rabbits in 28-day group occurred bilateral deep cervical lymph node metastases. The mean diameters of the ipsilateral deep cervical lymph nodes of 7-, 14-, 21- and 28-day groups were (0. 52±0. 03 ) cm, (0. 78±0. 04) cm, (0. 82±0. 03 ) cm and ( 1.42±0. 08) cm respectively. No metastasis from the transplanted tongue carcinomas was found in superficial cervical lymph nodes and submandibular lymph nodes in all rabbits. Conclusions The deep cervical lymph node metastasis develops at 2 weeks after the transplantation of tongue VX2 carcinoma and a deep cervical lymph node metastasis model of tongue VX2 carcinoma can be established.
6.Effect of Peimine on ERCC1 mRNA and LRP Expressions of A549/DDP Multidrug Resistance Cell Line.

OBJECTIVETo explore the effect of peimine on excision repair cross-complementation 1 (ERCC1) mRNA and lung resistant protein (LRP) expressions in A549/cisplatin (DDP) multidrug resistance (MDR) cell line.
METHODSLung cancer A549/DDP cells were cultured in vitro.Cells at logarithmic growth phase were divided into 4 groups, i.e., the blank control group, the DDP group, the ligustrazine group (DDP+ligustrazine), the peimine group (DDP + peimine). After 48-h drug action, ERCC1 mRNA expression was detected by RT-PCR and LRP expression detected by cell immunofluorescence.
RESULTSThere was no statistical difference in expression levels of ERCC1 mRNA and LRP between the DDP group and the blank control group (P > 0.05). Compared with the DDP group, expression levels of ERCC1 mRNA and LRP obviously decreased in the ligustrazine group and the peimine group (P < 0.05). They were obviously lower in the peimine group than in the ligustrazine group (P < 0.05).
CONCLUSIONSPeimine could reverse MDR of A549/DDP cell line. Its mechanism might be associated with down-regulating ERCC1 mRNA and LRP expression levels.

7.Application of confirmatory factor analysis in studying the achievements of index system in clinical sciences and technologies

To explore the role of confirmatory factor analysis in checking the construct validity of index system in clinical sciences and technologies and to determine the weighting of each index.Data were collected based on the achievements regarding the index system of sciences,technologies and analyzed by softwares SPSS and AMOS.Confirmatory factor analysis was performed to assess the construct validity and to identify the weighting.The P values for testing the two-order confirmatory factor models were bigger than 0.05,indicating that the actual data were in agreement with theory in designing the index system.Statistics on the goodness fit index(GFI)such as GFI were bigger than 0.90,indicating that they were satisfactory.Weightings for each index were identified based on factor loading of confirmatory factor analysis,showing that they were highly correlated with that from the Delphi method.Confirmatory factor analysis appeared to be an appropriate method in analyzing the associations among the index variables,and could be widely used to assess the construct validity of index system and identifing the weightings.

The sequences of ITS, matK, rbcL and psbA-trnH of 9 Gynostemma species or variety including 38 samples were compared and analyzed by molecular phylogeny method. Hemsleya macrosperma was designated as outgroup. The MP and NJ phylogenetic tree of Gynostemma was built based on ITS sequence, the results of PAUP phylogenetic analysis showed the following results: (1) The eight individuals of G. pentaphyllum var. pentaphyllum were not supported as monophyletic in the strict consensus trees and NJ trees. (2) It is suspected whether G. longipes and G. laxum should be classified as the independent species. (3)The classification of subgenus units of Gynostemma plants is supported.
