1.Effects of orphanin FQ On simple and norepinephrine-activated cardiac function in isolated rat heart

Objective To investigate the effects of orphanin FQ (OFQ) on simple and norepinephrineactivated cardiac function in isolated rat heart.Methods Male SD rats aged 3-4 months weighing 250-280 g were anesthetized with intraperitoneal 25% urethane 1.4-1.8 ml.Their hearts were excised and passively perfused with K-H solutionin a Lanngendorff apparatus.A fluid-filled latex balloon was inserted into left ventricle for measurement of developed pressure.Left ventricular systolic pressure(LVSP),left ventricular end-diastofic pressure(LVEDP),left ventricular developed pressure(LVDP=LVSP-LVEDP),HR and±dp/dtmax were measured.Sixty isolated rat hearts were randomly divided into 10 groups(n=6 each):Ⅰ control group (group C);Ⅱ,Ⅲ,Ⅳ OFQ1,OFQ2,OFQ3 group--OFQ 10-6,10-7,10-8 mol/L were administered via perfusion system respectively;Ⅴ OFQ+UFP group--OFQ 10-6 mol/L+UFP-101 (all OFQ antagonist) 10-6 mol/L were administered;Ⅵ NE group--norepinephrine 10-5 mol/L;Ⅶ,Ⅷ,Ⅸ NE+OFQ groups--NE 10-5 mol/L+OFQ 10-6,10-7,10-8 mol/L were administered respectively;Ⅹ NE+OFQ+UFP group--NE 10-5 mol/L+OFQ 10-6mol/L+UFP-101 10-6mol/L.Results LVSP,LVDP and±dp/dtmax were significantly increased while HR was significantly decreased in the 3 OFQ groups as compared with control group.LVSP,LVDP±dp/dt max and HR were significantly increased in the 3 NE+OFQ groups as compared with NE group.Conclusion OFQ has positive inotropic action and negative chronotropic effect on the isolated rat heart.OFQ Can enhance the positive inotropic and chronotropic effect of NE on the heart.
2.Clinical study of CDFI in the diagnose of perforation of ventricular septum resulting from myocardial infarct

Objective To evaluate the clinical applicational value of color doppler flow image(CDFI)to perforation of ventricular septum resulting from myocardial infarct.Methods Before 7 cases confirmed perforation of ventricular septum though DSA,were examined by CDFI.The two results were compared to study the difference of bothes.Results All patients' results though CDFI were as the same as those though DSA,and accuracy rating was 100%.Conclusion CDFI may be applicated to early diagnose perforation of ventricular septum.
3.Effect of medicated serum of Radix Morindae Officinalis on skeletogenous cell bionomics

Objective:To explore the essence of Radix Morindae Officinalis in strengthening bones through observing the effect of medicated serum of Radix Morindae Officinalis on cultured osteoblast in vitro. Methods:The medicated serum of Radix Morindae Officinalis was added to the osteoblasts which were separated from the cranium of 24-hours newborn SD rat,in order to determining the effect of TGF-?1 and the proliferation and secreting of alkaline phosphatase and osteocalcin of the osteoblasts. Results:The proliferation of the osteoblasts were signifi cantly stimulated by the alcohol and the n-butanol extract from the Radix Morindae Officinalis; and the n-butanol extract from the Radix Morindae Officinalis could promote differentiating OB excreting the ALP and OC;in addition,the ability of the osteoblasts expressing TGF-?1 was promoted by the n-butanol extract from the Radix Morindae Officinalis. Conclusion:The n-butanol extract from the Radix Morindae Officinalis can promote the ability of the osteoblast in expressing TGF-?1 mRNA and excreting the ALP,and further promote the growth of bone.
4.Current Research Status and Analysis of Electroacupuncture Parameters for Post-operative Pain

Objective To find the optimal stimulation parameters of electroacupuncture for incision pain and visceral dragging pain after surgery.Method Databases including the China National Knowledge Infrastructure (CNKI), WanFang (WF), VIP and PubMed were retrieved by using computer. The eligible literatures related to the clinical and experimental studies on electroacupuncture for post-operative pain were summarized and the frequencies of the involved electroacupuncture parameters were counted.Result For incision pain, electroacupuncture was used after surgery, with continuous wave, usually a low frequency ranged 2~15 Hz and a current of 2~5 mA; for visceral dragging pain, electroacupuncture was used before surgery, with sparse-intense wave, alternate frequencies majorly at 2/100 Hz and a medium or low current intensity within patient’s endurance. Conclusion In clinical application, the elctroacupuncture parameters should be chosen carefully according to the type of post-operative pain.
5.The complications of intravitreal triamsinolone acetonide injection in vitrectomy for proliferative diabetic retinopathy

· AIM: To investigate the complications of intravitreal triamsinolone acetonide (TA) injection in vitrectomy for proliferative diabetic retinopathy (PDR).· METHODS: From February 2005 to January 2007, 18patients (18 eyes) with PDR who were injected with TA in vitrectomy were observed retrospectively after surgery.· RESULTS: During a postoperative follow-up period of 3 to 6 months (mean 4.6 months), some complications including deposit of TA granules on the macular region and surface of the retina (3 eyes), postoperative vitreous hemorrhage (3eyes), elevated intraocular pressure (2 eyes) and pseudohypopyon (1 eye)were observed.· CONCLUSION: The complications after surgery such as deposit of TA granules on the macular region and surface of the retina and pseudohypopyon could be cured without any special treatment. All eyes with elevated intraocular pressure after surgery were controlled by drug. Re-operation may be an effective method for patients with unabsorbable vitreous hemorrhage after vitrectomy.
6.Enlightenment of big-data era on clinical research of TCM and acupuncture.

The big-data era has arrived, which involves all professions and trades. Its impact on the medical field has gradually revealed. With the characteristics of big data "4V" (volume, velocity, variety, value) and its theory generality with TCM (holistic thinking, correlativity) as well as regularity of disease development, combined with present status of acupuncture clinical research, the influences of big-data era on trial design, data collection, analysis and sharing of acupuncture clinical research are discussed in this paper, aiming to provide references for solving the difficulties of study design and determining the future research direction of acupuncture clinical research.

8.Thinking on designation of sham acupuncture in clinical research.

Randomized controlled trials (RCT) is the source of the raw data of evidence-based medicine. Blind method is adopted in most of the high-quality RCT. Sham acupuncture is the main form of blinded in acupuncture clinical trial. In order to improve the quality of acupuncture clinical trail, based on the necessity of sham acupuncture in clinical research, the current situation as well as the existing problems of sham acupuncture, suggestions were put forward from the aspects of new way and new designation method which can be adopted as reference, and factors which have to be considered during the process of implementing. Various subjective and objective factors involving in the process of trial should be considered, and used of the current international standards, try to be quantification, and carry out strict quality monitoring.

10.Expressions of E-cadherin and N-cadherin in prostate cancer and their implications.

OBJECTIVETo investigate the differences in the expressions of E-cadherin and N-cadherin between high-risk prostate cancer and low- and medium-risk prostate cancer, and analyze their correlation with the age, serum PSA level, and Gleason score of the patients.
METHODSWe retrospectively analyzed the clinical data of 42 cases of prostate cancer, which were divided into a low- and medium-risk group (group A, n = 15) and a high-risk group (group B, n = 27). We measured the expressions of E-cadherin and N-cadherin by immunohistochemical staining, compared their differences between the two groups, and analyzed their correlation with the age, serum PSA level, and Gleason score of the patients.
RESULTSImmunohistochemical staining showed that the expression of E-cadherin was significantly higher in group A than in B (6.1 +/- 0.51 vs 4.2 +/- 0.37, P < 0.01), and so was its positive rate (73.3% vs 25.9%, P < 0.05). The positive rate of E-cadherin was also markedly higher in the patients with serum PSA < 20 microg/L than in those with serum PSA > or = 20 microg/L (66.7% vs 29.6%, P < 0.05), and so was it in the patients with the Gleason score 5-7 than in those with 8-10 (60.9% vs 21.1%, P < 0.05). On the contrary, the N-cadherin expression was significantly lower in group A than in B (3.7 +/- 0.32 vs 7.5 +/- 0.58, P < 0.01), and so was its positive rate (13.3% vs 59.3%, P < 0.01). The positive rate of N-cadherin was also remarkably lower in the patients with the Gleason score 5-7 than in those with 8-10 (26.1% vs 63.2%, P < 0.05). However, there were no statistically significant differences in the N-cadherin expression between the patients with serum PSA < 20 microg/L and those with serum PSA > or = 20 microg/L (P > 0.05), nor in the expressions of E-cadherin and N-cadherin between the patients aged > or = 70 years and those aged < 70 years (P > 0.05).
CONCLUSIONThe expressions of E-cadherin and N-cadherin are significantly different between high-risk prostate cancer and low- and medium-risk prostate cancer, which suggests that both may correlate with the invasion and metastasis of prostate cancer as well as with the serum PSA level and Gleason score of the patients.
