1.Study on Children′s Social Adaptive Capability and Their Relative Factors

Objective To study children′s social adaptive capability and their relative factors.Methods Five hundred and twenty-seven children in 4 different kindergartens were surveyed by using the social adaptive capability-measuring list from infant to junior school student which revised by ZUO Qi-hua and questionnaire of family situation which compiled by TAO Gong-min and Zhu Xiao-qin.Results Twenty-five point three percent of the surveyed children′s social adaptive capabilities were higher than normal level.Sixty-eight point nine percent of them were at normal level.Four point seven percent of them were at edge level.The rest were at a slightly lower level.The influencing factors included the educational level of parents,the educational manner of the parents and their marriage relationship.Conclusions It is necessary to do some investigation to find problem eariler on children′s social adaptive capability.Suitable educational methods and good family environment are important to improve children′s social adaptive capability.
2.Relation of Phlegm-dampness Constitution and Insulin Resistance in the First-degree Non-diabetic Relatives of Patients with type 2 Diabetes

Objective Observe the relation of phlegm-dampness constitution and insulin resistance in first-degree non-diabetic relatives of patients with type 2 diabetes.Methods Collected first-degree relatives without mabetes of patients with type 2 diabetic patients 87 cases.Among these 87 cases,39 cases were phlegm-dampness constitution and 48 were not.Fasting plasma glucose(FPG)and fasting insulin(FINS)were measured,and insulin resistance index(IR)was calculated.Results There were significant differences between the two groups in FINS and IR(P<0.05)..Conclusion The IR of the First-degree non-diabetic relatives with phlegm-dampness constitution of patients with type 2 diabetes have greater insulin resistance than the First-degree non-diabetic relatives with non-phlegm-dampness constitution of patients with type 2 diabetes.

DNA damage response (DDR) in different cell cycle status of human peripheral blood lymphocytes (PBLs) and the role of H2AX in DDR were investigated. The PBLs were stimulated into cell cycle with phytohemagglutinin (PHA). The apoptotic ratio and the phosphorylation H2AX (S139) were flow cytometrically measured in resting and proliferating PBLs after treatment with camptothecin (CPT) or X-ray. The expressions of γH2AX, Bcl-2, caspase-3 and caspase-9 were detected by Western blotting. DDR in 293T cells was detected after H2AX was silenced by RNAi method. Our results showed that DNA double strand breaks (DSBs) were both induced in quiescent and proliferating PBLs after CPT or X-ray treatment. The phosphorylation of H2AX and apoptosis were more sensitive in proliferating PBLs compared with quiescent lymphocytes (P<0.05). The expression levels of anti-apoptotic proteins Bcl-2 were reduced and cleaved caspase-3 and caspase-9 were increased. No significant changes were observed in CPT-induced apoptosis in 293T cells between H2AX knocking down group and controls. It was concluded that proliferating PBLs were more vulnerable to DNA damage compared to non-stimulated lymphocytes and had higher apoptosis rates. γH2AX may only serve as a marker of DNA damage but exert no effect on apoptosis regulation.

Objective To observe the effect of somatostatin (SST) on the expression of metalloproteinase-2(MMP-2) and tissue inhibitor matrix metalloproteinase-2(TIMP-2) of implanted tumor in nude mice after partial hepatectomy.Method Nude mice were divided into group A(n=10) implanted by human hepatocellular carcinoma(HCC),group B(n=10, partial hepatectomy) and C(n=10,HCC implantation after liver resection). SST(250 ?g/kg,b.i.d) was given intraperitoneally in group C. Mice were sacrificed on 35th d, tumor was measured, the expression of MVD.MMP-2 and TIMP-2 was detected by immunochemical staining and quantitative image computer-analysis. Objective To compare and evaluate the methods of building model of cervical spondylosis of vertebral artery type (CSA)rats by measuring the learning and memory ability,anxiety and depression,and degree of nervous tension of three kinds of CSA model rats.Methods We randomly divided 120 rats into mixed modeling group,bone graft compression modeling group,mechanical balance disorder modeling group,and blank control group with 30 rats in each.Morris water maze,elevated plus maze test and open field test were used to detect and compare the rats’abilities of learning and memory,anxiety and depression,and degree of nervous tension.We then evaluated the three CSA rat models.Results Compared with those in the blank group,the learning and memory abilities in the mixed modeling, bone graft compression and mechanical balance disorder modeling rats were significantly decreased,the anxiety and depression and degree of nervous tension were significantly increased (P<0.05).Compared with bone graft compression and mechanical balance disorder modeling groups,the mixed modeling group could restore the characteristics of CSA.Conclusion The three kinds of modeling methods can successfully reproduce the CSA animal model;the mixed modeling is superior and thus worthy of promotion.
8.Protective effect of Huoxue-Dingxuan capsule-medicated serum on mouse brain microvascular endothelial cell line bEnd.3 with hypoxic injury

AIM:To investigate the protective effect of Huoxue-Dingxuan capsule-medicated serum on mouse brain microvascular endothelial cell line bEnd .3 with hypoxic injury .METHODS:The Wistar rats were randomly divided into control group and Huoxue-Dingxuan capsule group .The serum was collected at the 7th day after drug administration . The bEnd.3 cells were divided into normal group , hypoxia serum model group and Huoxue-Dingxuan capsule serum group . After administration of the medicated serum , bEnd.3 cell were treated with hypoxia for 6 h.The cell morphology was ob-served under microscope , the apoptosis rate and cell cycle were detected by flow cytometry , and the activity of superoxide dismutase ( SOD) and the content of malondialdehyde ( MDA) were detected by ELISA .RESULTS:Compared with other groups, Huoxue-Dingxuan capsule-medicated serum significantly resisted the injury caused by hypoxia , obviously improved the morphology of bEnd .3 cells, significantly reduced the number of apoptotic cells induced by hypoxia , and effectively in-hibited the occurrence of hypoxia-induced G1/S phase arrest in the bEnd.3 cells.At the same time, the medicated serum inhibited MDA production , and increased SOD activity .CONCLUSION: Huoxue-Dingxuan capsule attenuates hypoxia-induced bEnd .3 cell damage by enhancing the antioxidant capacity of cells and inhibiting cell apoptosis .
9.Investigation and analysis of otolaryngology nurses' knowledge and attitudes toward pain management in Chongqing

Objective To explore otolaryngology nurses' knowledge of pain and attitude toward pain management and to provide reference for pain management.Methods A total of 102 otolaryngology nurses who participated in 2014 in Chongqing of Otolaryngology Head and Neck Surgery Symposium were surveyed by the nurses' general condition and current management situation and attitude survey regarding Chinese Knowledge and Attitude Survey Regarding Pain(KASRP).Results Average correct response rateon the KASRP from otolaryngology nurses was 45.9%,the interquartile range was 38.9%-51.0%,nurses' knowledge was relate to continuing education,P<0.05,while irrelevant to age,work experience,education,job title,school education,clinical application of pain assessment tools,etc,P>0.05.The proportion of nurses who accept the education of pain knowledge was 69.6% (71/102),there was 74.5% (76/102) nurses assess the patient by pain assessment tools in their daily work.Nurses first want to get the information of pain assessment,followed by pain medication,operational knowledge of analgesia pump and non-drug interventions for pain.Conclusions Nurses lack knowledge of pain,need to strengthen pain management training by continuing education and get knowledge of pain from academic journals,to provide reference for pain management.
10.Effects of the dominant negative form of protein phosphatase 2A catalytic subunit α driven by alpha-fetoprotein enhancer/phosphoglycerate kinase promoter on hepatoma cell xenografts

Objective To investigate the effects of AFP enhancer/pgk promoter driven expression of the dominant negative form of the PP2A catalytic subunit α (DN-PP2Acα) in vivo.Methods The previously constructed AFpg promoter-driven DN-PP2Acα was recombined into an adenovirus,and the expression of PP2Ac was tested using Western blot.Cell growth was tested using the MTT and flat plate clone formation assays.In vivo studies were performed in tumor xenograft models.Results AFpg promoter-driven expression of DN-PP2Acα exerted cytotoxic effects against the AFP-positive human hepatoma cell line HepG2,but did not affect AFP-negative human hepatoma cells (SKHEP-1) or normal human liver cells (L-02).Moreover,AFP enhancer/pgk promoter driven expression of DN-PP2Acα inhibited the growth of AFP-positive HepG2 tumors in nude mice bearing solid tumor xenografts,but did not affect AFP-negative SK-HEP-1 tumors.Conclusion The recombinant AFP enhancer/pgk promoter-driven DN-PP2Acα expression adenovirus presented selective cytotoxicity against AFP-positive hepatoma cells and provides a useful gene therapy strategy to selectively target hepatocellular carcinoma.
