4.Detection of bence jones proteins of nonconcentrated urine by immunofixation electrophoresis

Objective To evaluate immunofixation electrophoresis (IFE) and to compare it with conventional manual heat test method for detection of Bence Jones (BJ) protein in nonconcentrated urine. Methods We performed IFE and heat test for urinary protein analysis in 116 urine samples and evaluated a new immunofixation electrophoresis system by urinary protein analysis in 20 patients with multiple myeloma. Results 20 patients with multiple myeloma were detected to have BJ proteins (8 ? and 12?) in urine by IFE, whereas no urine BJ proteins by heat test. Conclusion The heat test for BJ proteins should be replaced by IFE because of its insensitivity and unspecificity. The IFE method is higherly sensitive and specific for screening and identification of BJ proteins in urine.
5.Analyzing the risk factors and related psychological factors in patients with gastroesophageal reflux disease and surveying the curative effect

Objective To analyze, the risk factors and related psychological factors in patients with gastroesophageal reflux disease (GERD)and survey the curative effect. Methods All of 200 outpatients with GERD(GERD group) and 200 healthy persons (control group)from January 2006 to January 2008 were randomly selected, and the personal data were investigated. The GERD questionnaire (RDQ) and the Zung-self-rating depression scale (SDS) test were used. Results In GERD group, the proportions of high education, city living, marriage instability ,heavy working pressure, high body mass index (BMI),smoking and drinking habit, peptic disease in family, using nonsteroidal anti-inflammatory drugs (NSAIDs) were higher than those in control group (Pall < 0.01). After treatment, the patients in GERD group had lower scores in the RDQ and SDS test than those before treatment (P< 0.01). Conclusions There are many risk factors in patients with GERD, especially the psychological factors. The therapy of GERD should combine with psy-chotherapy and avoid high BMI, smoking, drinking and overusing the NSAID.

Gut microbes play an important role in human health and are believed to be closely related to obesity , diabetes and some other metabolic diseases .Recently, intestinal flora has become a research hotspot and new target for disease prevention.The problems facing intestinal flora research included variability between individuals ,huge amounts and varieties of gut microbes .Caenorhabditis elegans, with the advantages of living on E.coli, short growth periods , easy observation,applicability to high throughput screening of drugs , is an ideal model organism for research of microbe-host interactions .It has been used in probiotics screening and interaction mechanism research of multifold bacteria .This review summarizes some new researches and applications in this area .

Objective To establish a method for detecting intestinal bacteria in Caenorhabditis elegans.Methods The gut flora number of 1, 20 or, 50 C.elegans was quantified and compared using real-time quantitative PCR (qPCR). Results The gut flora of a single C.elegans could be detected using qPCR method , which could also reflect the difference in the number of gut bacteria between different samples .Conclusion The qPCR method can be uased to accurately quantify intestinal bacteria even in only one C.elegans and has the advantages of low-cost, high-sensitivity and good-specificity.
10.Effect of leptin on protecting rat islet ?-cells against lipotoxicity induced by free fatty acid

Objective To observe whether leptin protects rat islet ?-cells from lipotoxicity induced by free Taity acid ( FFA). Methods Pancreatic islets were isolated from Wistar rats.
