Objective] Observing Cortex Lycii Radicis' effect(Cortex Lycii) on rat insulinoma cells(INS-1) proliferation and apoptosis. To explore the mechanism of Cortex Lycii on Pancreatic Beta Cell Proliferation and apoptosis.[ Methods] After primary culture, cells were randomly divided into blank control group:control,11.1mmol·L-1 glucose,HG,30mmol·L-1 glucose, HG+Cortex Lycii(1g·L-1), HG+Cortex Lycii(2g·L-1), HG+Cortex Lycii(4g·L-1), the survival rate of cells was observed by cell counting kit-8(CCK-8);the apoptosis rate was observed by Annexin V stammg. [Results] ①Compared with HG the better effect of cell proliferation groups of Cortex Lycii(P<0.01), the best is group of Cortex Lycii(2g·L-1)(P<0.05) ②Compared with HG group the higher survival rate is group of Cortex Lycii(P<0.01), the lower apoptosis rate of Cortex Lycii(2g·L-1) compared with Cortex Lycii(1g·L-1). [Conclusion] Cortex Lycii can promote the proliferation of pancreatic beta cell, inhibit the apoptosis to protect the pancreatic beta cell. The optimal concentration is 2g·L-1.

Angiogenesis is an important pathophysiological process of body response after cerebral ischemia.Angiogenesis is activated in a few hours after cerebral ischemia. It can promote neuronal remodeling and neurological function recovery.Studies have shown that the microvessel density is positively correlated with the long-term survival rate in patients with stroke after cerebral ischemia. This article reviews the regulatory mechanisms and imaging evaluation of angiogenesis after cerebral ischemia.

New epidemic broke out in recent year which was suspected to be caused by variant Muscovy duck parvovirus (MDPV). For this reason, new MDPV detection methods are needed for the new virus strains. In this study, a pair of primers were designed according to the full-length genome of MDPV strain SAAS-SHNH, which were identified in 2012, and were used to amplify the vp3 gene of MDPV by polymerase chain reaction. After being sequenced, the vp3 gene was subcloned into the prokaryotic expression vector PET28a. The recombinant plasmid was transformed into E. coli BL21 and induced with IPTG. SDS-PAGE and Western blotting analysis showed the MDPV vp3 gene was successfully expressed. After being purified by Ni2+ affinity chromatography system, the recombinant protein was used as antigen to immunize rabbits to obtain antiserum. Western blotting analysis showed that the acquired antiserum could react specifically with VP3 protein of J3D6 strain and MDPV vaccine strain. The antiserum could also be used for detection of cultured MDPV from primary duck embryo fibroblasts by immune fluorescence assay (IFA). It could be concluded that the VP3 protein and its antibody prepared in the research could be used for detection of VP3 antiserum and antigen respectively.

Objective To analyze the clinical characteristics of urinary tract infection after kidney transplantation and to analyze the distribution of pathogenic virulence factors. Methods From January 2014 to June 2016,a total of 255 cases were subjected to DCD renal transplantation in our center, and 45 cases of urinary tract infection occurred after operation.Among them, urinary tract E.coli infection occurred in 32 cases. The virulence factors gene aer,hly,irp2 and iucD in the 32 strains of E.coli isolated from urine were tested using PCR and multiple PCR.Results Thirty-two cases (71.1%) of urinary tract infection after renal transplantation was caused by E.coli, and divided into 9 cases of nosocomial infection group, and 23 cases of community-acquired group. The positive rate for aer,hly,irp2 and iucD in Nosocomial infection group was 66.7%,33.3%,88.9% and 77.8%,and that in community-acquired group was 30.4%,8.70%,39.1% and 34.7%,respectively.There were statistically significant differences among four kinds of virulence factors between two groups (P<0.05).The positive rate of virulence factors was significantly higher in nosocomial infection group than in community-acquired groups. The total resistance rate was 84.38%,and there was significarit difference in multi-drug resistance rate between two groups (P<0.01).Conclusion E.coli is the main pathogenic bacteria of urinary tract infection after renal transplantation, especially nosocomial infections. The detection rate of virulence factor is high, and the multiple drug resistance is outstanding,so the attention should be paid clinically.

Objective To evaluate the safety and efficacy of Zonisamide(ZNS) as adjunctive therapy in patients with refractory partial seizures receiving other antiepileptic drags (AEDs).Methods This was a randomized,double-blind,placebo-controlled study conducted at multi-centers.All 240 subjects were randomized to either the ZNS group or the placebo group in a 1:1 ratio. The double-blind treatment phase included a titration phase during which zonisamide dose inereased from 100 mg/day to 300 mg/day over 4 weeks and then a 12-week fixed-dose phase. The primary efficacy endpoint was,the median % reduction from baseline in all pattial seizure frequency(CPS+SPS+SGS)during the fixed-dose phase. The important secondaw endpoint wag the responder rate.Safety profiles and tolerance were also evaluated. Results The FAS analysis showed the median reduction from baseline in the ZNS group was greater than in the placebo group(48.4%vs 26.6%),the difference was significant for ZNS compared with placebo(F=4.904,P=0.028);The responder rates for all partial seizures(48.6%vs34.9%,X2=4.046,P=0.044)and for complex seizures(52.2% vs 33.3%,X2=5.607,P=0.018)were significantly higber in the ZNS group than in the placebo group in the FAS population. The overall adverse events(AEs)profile was comparable between the two groups. The most frequent AEs considered to be related to zonisamide by the investigator were headache, dizziness, somnolence, anorexia, nausea, etc. Conclusions ZNS is superior to placebo in reducing the frequency of partial seizures and well-tolerated.ZNS could be a choice of adjunctive therapy in patients with refractory partial seizures.

OBJECTIVETo observe the effect of volatile oil of amomum (VOA) on the expressions of mastocarcinoma-related peptide (PS2) and platelet activating factor (PAF) in helicobacter pyloriassociated gastritis (HPG) and to analyze its potential mechanism.
METHODSEighty patients with HPG were randomly assigned to two groups, 42 patients in the treated group treated with 0.5 mL VOA, thrice per day; and the 38 patients in the control group receiving Western tertiary medicinal treatment. Gastroscopic picture and helicobacter pylori (HP) infection (by quick urease and Warthin-Starry stain) of the gastro-membrane, expressions of PS2 and PAF (by immunohistochemical assay and Western blotting) as well as the contents of aminohexose and phospholipid (by Neuhaus method) in the gastric membrane of all patients were detected before treatment and 4 weeks after treatment. The clinical efficacy in the two groups was compared.
RESULTSThe total effective rate in the treated group was 88.1%, which was significantly higher than that in the control group (78.9%, P<0.05). After treatment, in the treated group, gastric membranous contents of aminohexose and phospholipid was increased, expression of PS2 elevated but that of PAF lowered, all showing significant difference as compared with those in the control group (P<0.01). In the control group, the expressions of PS2 and PAF changed insignificantly. The radical eliminating rate of HP in the treated group and the control group was insignificantly different between them (76.1% vs. 65.8%, P>0.05).
CONCLUSIONThe mechanism of VOA for anti-gastritis might be related with its action in increasing the expression of PS2 and decreasing the expression of PAF, and thus regulating the hydrophobicity of the gastric membrane.
