6.Mechanism of genuineness of Glycyrrhiza uralensis based on SNP of β-Amyrin synthase gene.

β-Amyrin synthase (β-AS) genes of Glycyrrhiza uralensis from 6 different regions were analyzed by PCR-SSCP and sequenced, then the correlationship between β-AS SNP and regions of Glycyrrhiza uralensis were determined. According to the 1 coding single nucleotide polymorphism on the first exon of β-AS gene at 94 bp site, Glycyrrhiza uralensis could be divided into 3 genotypes. In these genotypes, the percentage of 94A type in genuine regions was much higher, and it had significant differences with the percentage in non-genuine regions (P < 0.001). The results of the experiment proved that different β-AS genotypes at 94 bp site from different regions may be one of the important reasons to result in the genuineness of Glycyrrhiza uralensis.

7.An analysis of the dental plaque microbiology of root surface caries in elderly patients

Objective To study the microbiology of root surface caries in elderly patients. Methods Seventy-five elderly people (aged 60~77 years) were divided into 2 groups:Control group of patients without root caries (n=30) and root caries group of patients with root caries without apicitis and pulpitis (n=45).Plaque samples were collected,cultured in selective and non-selective media.After the bacteria were isolated,the total count and the detection rates and bacterium numbers of porphyromonas,pervotella,streptococcus mutants group,actiomyces and lactobacillus were compared between the groups of control and root caries.Results The count of total bacteria, streptococcus mutants group,actinomyces,lactobacillus and of root caries group were significantly higher that those of the control group(4.73?0.75)lg(CFU/ml+1)vs(4.17?0.47)lg(CFU/ml+1), (3.89?0.89)lg(CFU/ml+1) vs (2.84?1.14) lg (CFU/ml+1),(3.24?1.89) lg (CFU/ml+1) vs (2.19?0.11)lg(CFU/ml+1),(3.24?1.11)lg(CFU/ml+1)vs(2.43?0.95)lg(CFU/ml+1), (2.67?0.70)lg(CFU/ml+1)vs (3.24?0.21)lg(CFU/ml+1),(P
8.Cloning, Expression and Property Analysis of Arabinosidase in Pichia pastoris

SMART-RACE was performed after isolating the total RNA of Armillariella tabescens to amplify the full-length cDNA of arabinosidase (GenBank Accession No. AJ620046). Bioinformatics analysis was used to analyze the code frame of arabinosidase, to predict its structure and function. Recombinant plasmid pPIC9-AF was constructed and then electroporated into methylotrophic yeast Pichia pastoris GS115. The secreted 6 ? His fusion protein was purified to analyze its enzymology property. This arabinosidase had high activity at 30-35℃ under acid condition, and was stable within wide range of pH and temperature. It maintained about 80% activity at the range of pH4. 0-8.0 and 20-40℃,wider than many other cloned arabinosidase. So it was worthy to go step further to study this enzyme, and recombinant expression provided a chance of highly expressing arabinosidase.
