1.Efficacy and security of combinations of tirofiban with low-dose heparin in the treatment of progressing stroke of the elderly

Objective To investigate the efficacy and security of tirofiban with low-dose heparin for treating progressing stroke with intracranial vessel stenosis,to analyze the clinical value of tirofiban in the treatment of ischemic stroke.Methods Clinical data of 26 patients treated with tirofiban and low-dose heparin for progressing stroke were retrospectively collected with NIHSS increment ≥3 within 6 days of stroke as the diagnosis standard.The degree of intracranial vessel stenosis was checked by CT angiography (CTA).The efficacy of tirofiban for treating progressing stroke,and the correlation between the degree of intracranial vessel stenosis and the prognosis of progressing stroke were observed.Results The percentage of progressing stroke patients with intracranial vessel stenosis/lesion was higher than without intracranial vessel stenosis/lesion (88.5 %vs.11.5%,x2=14.786,P 0.000).There was a significant difference in NIHSS scores between progressing stroke patients with and without intracranial vessel stenosis/lesion before treatment (t 2.17,P=0.046).NIHSS scores were significantly decreased (t=3.59,P=0.021) in both two groups after treatment.MRS was lower in progressing stroke patients without intracranial vessel stenosis/lesion than in progressing stroke patients with intracranial vessel stenosis/lesion after treatment (t=6.79,P=0.001).The efficacy of tirofiban was higher in progressing stroke patients without intracranial vessel stenosis/lesion than in progressing stroke patients with intracranial vessel stenosis/lesion (60.2% vs.30.8%,x2=4.35,P<0.05).Conclusions Combination of tirofiban with low-dose heparin is safe and effective in the treatment of progressing stroke.The degree of intracranial vessel stenosis is correalted with progressing stroke prognosis.

Red cell isoenzymes EsD and PGM and serum Hp are of genetic polymorphism which is inheritedsccording to the Law of Meqdelian Segregation. The distribution of their gene frequencies varies withraces. In this paper,the phenotype frequencies of the EsD,PGM and Hp have been investigated by theemthod of mixed starch/agarose gel electrophoresis and polyacrylamide gel electrophoresis among fivenationalities in Inner Mongolia, namely Creqen, Ewenki, Dawuor, Northeast Mongolian and Buryat.The gene Oroqen,Ewenki,Dawuor,Northeast Mongolian and Buryat. The gene frequencies were alsocalculated. The phenotype frequencies of EsD,PGM and Hp are compared not only among the five nationalities but with those reported by other countries.
3.Effects of RNA-binding protein La on migration and invasion of cervical cancer cells

Objective To investigate the role of RNA-binding protein La protein in the invasion and migration of cervical cancer cells.Methods RNAi technology was used to silence the La protein in HeLa cell,a cell line of cervical cancer,and then screened by G418.Finally the stably expressed HeLa-shLa cell line was constructed and then wound healing,Transwell,Western blot and gelatin zymography assay were performed. Results After La protein HeLa was silenced by RNAi,the invasion and migration capabilities of HeLa cells were decreased significantly compared with those of the controls.Meanwhile,SiRNA-mediated depletion of La reduced the expression of MMP-2 and increased the expression of TIMP-2.Meanwhile the activity of MMP-2 was reduced too.Conclusion RNA-binding protein La promotes the invasion and migration of cervical cancer cells,which may be related to regulating its matrix metalloproteinases and inhibitors.
4.Real-time Fluorescence Quantitative PCR for Detecting Extended Spectrum-β-Lactamases Genotype

Objective:To establish a rapid method to detect drug-resistance genotypes of extended spectrum-β-Lactamases (ESBLs) produced by gram negative bacillus using the real -time fluorescence quantitative PCR. Methods: According to clinical common genotypes of ESBLs, SHV, TEM.CTX-M.OXA and their homology, 9 pairs of specific primers were designed including SHV, TEM, CTX-M-1, CTX-M-2, CTX-M-8, CTX-M-9, OXA-1, OXA-2 and OXA-10. To extract DNA template by boiling assay, and then establish and grade up SYBR GREEN I real-time fluorescence quantitative PCR reaction system, finally definite real-time fluorescence quantitative PCR method. Its precision and range of linearity were tested. With established assay 51 multi- drug resistant ESBLs- E. coli K. pneumoniae were detected and compared with improved three dimensional extract tests. Results: Except OXA-2, 8 genotypes SHV, TEM, CTX-M-1, CTX-M-2, CTX-M-8, CTX-M-9, OXA-1 and OXA-10 were amplified by quantitative PCR from 39 ESBLs+ and 51 multi-drug resistant ESBLs-E. coli K. pneumoniae and confirmed by sequence testing. The range of linearity was 3×10~3-3×10~8 copies/mL, r =-0.994 7. Repetitive experiments showed that the average coefficient of variation between -runs was 9.6%. Comparing with three dimensional extract test, there was no significant difference (χ2 = 1.125,P> 0.05). Conclusion: Testing drug-resistance genotypes of ESBLs with SYBR GREEN I real-time fluorescence quantitative PCR is a rapid,specific and sensitive method, which is capable of inspecting genotypes of ESBLs from clinical strains.
5.Development of multifunctional anorectal therapeutic apparatus

This article introduces the structure, principle and characteristics of a new anorectal therapeutic apparatus. The adoption of new technologies, new components and new materials lays the foundation for the compatibility for electric energy, magnetic energy and thermal energy as well as the safety and comfortability of the intracavitary electrode. The intracavitary electrode possesses the functions of electrotherapy, thermotherapy, magnetotherapy and massage, and thus can be applied to the physical therapies of colitis, irritable bowel syndrome and prostatitis. The apparatus has such advantages as reliable curative effects, no toxic and side effects, simple manipulations, low cost, etc, and thus should be popularized.
6.Mechanism of nerve root stretch injury

BACKGROUND:Under physiological conditions, nerve roots can move along with the movement of limbs and spine. However, the mechanisms of nerve root stretch injury under physiological conditions and the neurological dysfunction after injury are unclear.
OBJECTIVE:To review the reason of nerve root stretch injury, and to analyze the mechanism of nerve function from biomechanics, pathology and neurophysiology.
METHODS:A computer-based online search of PubMed database was undertaken by the first author to identify the articles related to the research of nerve root injury and nerve stretch injury between 1990 and 2012, with the key words of“nerve root, nerve, stretch injury”. A total of 391 articles were screened out. The articles on the anatomy and biomechanics research of nerve root were included, as wel as the pathology and neurophysiology research after nerve root stretch injury. Final y, 44 articles were included for review.
RESULTS AND CONCLUSION:Nerve root can be stretched along with limbs and spine movement. However, diseases or trauma may cause pathological nerve root stretch, and thus leading neurological dysfunction. The nerve root is often influenced stretch because of its anatomical and tissue structure. Mechanical injury mechanism of stretch composes of peripheral (peripheral nerve conduction stress) and central mechanism (displacement of spinal cord content). Pathological studies found that the local fibrosis is caused by the extracted serum protein that difficult to remove after injury, and this is because of the shortage of lymphatic system in nerve roots. Fibrosis can cause nerve root ischemia, affect the nerve function, and change the biomechanical properties of nerve root. Nerve root injury can cause primary and secondary injury of internal axons, and this is the main reason for neurological dysfunction after injury.
7.Effects of infrasound on the auditory function of guinea pigs

Objective To observe the effects of exposure to infrasound on auditory function of guinea pigs as reflected by the brain stem response (ABR) and distortion product otoaeoustic emission(DPOAE).Methods Seventy-two Guinea pigs were used in this study,of them,12 served as controls and 60 were divided into 2 experi- mental subgroups.ABR and DPOAE were detected after exposure to infrasound stimulation at 16 Hz,90 dB or 16 Hz,130 dB for 2 hours a day for 1 ,7,14,21 and 28 days.Results The threshold of ABR after exposed to infrasound at 16 Hz,90 dB in 1 day and 28 days was higher than the controls (P0.05),but significant difference was observed with 16 Hz 130 dB infrasound exposure on the 21st and,28th days when compared with the controls (P

9.The influence of all-trans retinoic acid on the craniomaxillofacial development of mice

Objective:To study the influence of all-trans retinoic acid (atRA)on craniomaxillofacial development of C57 mice. Methods:Pregnant C57BL mice were divided into 4 groups(n =5)at gestation day (GD)1 0.Mice in three atRA-induction groups were given atRA of 60,80 and 1 00 mg/kg,respectively.The mice in control group were given the equivalent volume of corn oil.All pregnant mice were sacrificed at GD1 9 and the embryos were collected.Stereo microscope was used to observe the craniomaxillofacial morphology.Standardized radiographs were taken and cephalometric analysis was performed.Results:The embryonic body length and body mass of control group surpassed those of 80 and 1 00 mg/kg atRA groups(P <0.05,P <0.01 ).atRA induced craniomaxillofacial malformations and maldevelopment.The mice induced by atRA exhibited a shorter mandibular body and more retrusive position of max-illary and mandibular(∠NAK and ∠NBD)when compared with their norm(P <0.01 ).Significant decrease in craniofacial length (Op-Rh)was observed in all atRA-induced groups(P <0.01 ).Decreases in cranial vault height(Fp-Os)and cranial vault length(Pa-Na)dimensions were observed in 80 and 1 00 mg/kg atRA groups(P <0.05,P <0.01 ).Conclusion:Exogenous atRA dose-depend-ently induces retardation of craniomaxillofacial morphology in embryo of C57BL mice by inhibition of the sagital and vertical dimension development of the bone.
10.A preliminary study on the role of the E2 subunit of pyruvate dehydrogenase modified by xenobiotics in the pathogenesis of primary biliary cholangitis

Objective To explore preliminarily the role of the E2 subunit of pymvate dehydrogenase (PDC-E2) modified by xenobiotics (e.g.2-octynic acid,2-OA) in the pathogenesis of primary biliary cirrhosis (PBC).Methods Patients of PBC (102 cases),primary sclerosing cholangitis (PSC,34 cases) and healthy controls (HC,50 cases) were selected.The anti-PDC-E2,anti-2-OA and anti-lipoic acid (LA) antibody in the peripheral blood of the 3 groups were tested by enzyme linked immunosorbent assay (ELISA).By inhibitive ELISA (iELISA),30 of the 102 PBC patients with anti-PDC-E2 antibody but without anti-2-OA antibody were selected to detect whether there was any new epitope on the PEC-E2 conjugated with 2-OA.The chi-square test and Fisher exact test were taken to analyze the enumeration data.The two-tailed unpaired t test with Welch's correction was used to compare the measurement data.Spearman rank correlation analysis was also employed for proper test.Results The positive rate of anti-PDC-E2,anti-LA and anti-2-OA antibody in PBC patients was 94.1%(96/102),73.5%(73/102) and 53.9%(55/102) respectively,all of which were statistically significantly higher than those in healthy controls group but were of no significant difference between PSC and healthy controls group.There was no significant relevance between the levels of Anti-LA and anti-2-OA antibody in the PBC group (r=-0.065,P=0.520).The iELISA results showed that the antibody,which only identified the epitopes on 2-OA-PDC-E2 induced by the 2-OA conjugation with PDC-E2,existed in 40%(12/30) of the PBC patients,and more interestingly,this antibody was predominantly appeared in PBC patients at their early clinical stage.Conclusion There are anti-LA antibody and anti-2-OA antibody in PBC patients,which have shown no significant association with each other.It is very likely that new antigenic conformational epitopes on PDC-E2 modified by 2-OA would emerge,which might led to the immune response in the individuals who are susceptible to PBC,and thus contribute for the breaking of PDC-E2 immune tolerance,and PBC occurrence finally.
