With the fast development of biopharmacy,transgenic chicken oviduct bioreactor is going to be the focus of people’s attention.For its excellent advantages,transgenic chicken will be one of rising industries in the field of biopharmacy and hot spot of scientific study.Now,the most successful method for producing transgenic chicken is mediated by retrovirus transferring gene into it, and transgenic chicken have been produced by this method.The advantages and methods of producing transgenic chicken by retrovirus,and the progress in transgenic chicken research were summarized.The significance and existent problems of transgenic chicken are also discussed.
2.Effect of Qizhi Weitong Granules combined with rabeprazole on gastroesophageal dynamics and inflammatory factors in reflux esophagitis

Objective To observe the effect ofQizhi Weitong Granules combined with rabeprazole on gastroesophageal dynamics and inflammatory factors in reflux esophagitis (RE) and explore its clinical significance.Methods Nighty-six RE patients from Changping Chinese Medicine Hospital were selected in this study,which randomly divided into control group (n =48) and observation group (n =48).The control group was given rabeprazole 10 mg/time (2 times/d) for 4 weeks.The observation group was given rabeprazole 10 mg/time (2 times/d) and Qizhi Weitong Granule 5 g/time (3 times/d) for 4 weeks.The gastrin and motilin expression was detected by radioimmunity analysis.The IL-17 and IL-23 was detected by ELISA analysis.The rate of clinic effects and adverse reaction was compared.Results There was no significant difference ofgastrin and motilin between the two groups before treatment.After treatment,gastrin and motilin was increased in the two groups (P < 0.05,0.01).However,gastrin and motilin was higher in observation group than that in control group after treatment (P < 0.05).There was no significant difference ofIL-17 and IL-23 between two groups before treatment.After treatment,IL-17 and IL-23 was decreased in the two groups (P < 0.05,0.01).However,IL-17 and IL-23 was lower in observation group than that in control group after treatment (P < 0.05).The rate of clinic effects was 93.8% in observation group,which was better than 81.3% in control group (P < 0.05).The rate of adverse reaction was 10.4% in control group and 8.3% in observation group.There was no significant difference between two groups.Conclusion Qizhi Weitong Granules combined with rabeprazole can improve gastroesophageal dynamics and inhibit inflammatory response in RE patients,with significantly clinical effect and safety.
3.Technology research on the preparation of fosfomycin trometamol from fosfomycin sodium

Objective to investigate a new synthesis for the preparation of fosfomycin tromethamine and to seek for the opti‐mized process and crystallization conditions .Methods An inorganic method ,utilizing in situ generated tromethamine bisulfate to re‐act with fosfomycin sodium was used to gain fosfomycin tromethamine .Results After the optimization of reaction and crystalliza‐tion condition ,it can produce fosfomycin tromethamine .Conclusion Inorganic acid salt synthesis technology is feasible with high yield .
4.Lentivirus-mediated shRNA interference targeting ADAM17 down-regulates MMP-9 expression in airway epithelial cells via TNF-α/NF-κB signaling

Aim To study the role of TNF-α/NF-κB signaling in matrix metalloproteinase ( MMP)-9 expres-sion induced by lipopolysaccharide ( LPS ) in airway epithelial cells, and to investigate the effects of lenti-virus mediated RNAi targeting a disintegrin and metal-loproteinase 17 ( ADAM17 ) gene on MMP-9 expression induced by LPS. Methods The ADAM17 siRNA ex-pression vector was constructed, and packaged to re-combinant lentivirus in 293T cells. The HBE4-E6/E7 cells were pretreated for 30 min by NF-κB inhibitor ( PDTC) and a recombinant human TNFR p75-Fc fu-sion protein ( Etanercept) , or infected by the recombi-nant lentivirus for 72 h, and then stimulated for 24 h by LPS or TNF-α. The release of TNF-α was detected by ELISA. The mRNA and protein levels of MMP-9 were analyzed respectively by RT-PCR and Western blot. NF-κB activity was detected by electrophoretic mobility shift assay. Results LPS and TNF-α signifi-cantly increased MMP-9 mRNA and protein expressions and the activation of NF-κB in HBE4-E6/E7 cells ( P<0. 05 ) . Etanercept and PDTC significantly inhibited MMP-9 expression and the activation of NF-κB induced by LPS ( P<0. 05 ) . Lentivirus mediated RNAi targe-ting ADAM17 significantly decreased TNF-α produc-tion, inhibited MMP-9 mRNA and protein expressions and the activation of NF-κB induced by LPS in HBE4-E6/E7 cells ( P <0. 05 ) . Lentivirus mediated RNAi targeting ADAM17 did not inhibit MMP-9 mRNA and protein expressions and the activation of NF-κB in-duced by TNF-α ( P>0. 05 ) . And PDTC significantly inhibited MMP-9 mRNA and protein expressions and the activation of NF-κB induced by TNF-α ( P <0. 05 ) . Conclusions TNF-α/NF-κB signaling partic-ipates in the regulation of MMP-9 expression induced by LPS in airway epithelial cells, and lentivirus-media-ted RNAi targeting ADAM17 plays an important role in that signaling pathway upstream by regulating TNF-αrelease.
5.β-lactamase genotyping of multi-drug resistant Acinetobacter baumannii in a neonatal intensive care unit

Objective To investigate the correlation between drug resistance and β-actamase genes of multi-drug resistance Acinetobacter baumannii (MDR-AB) in neonatal intensive care unit to provide evidence for rational antibiotics administration and nosocomial infection control.Methods Twenty-six MDR-AB strains were separated and collected from clinical specimens.The minimum inhibitory concentrations of 13 antimicrobial agents were determined by agar dilution method.Genotypes of β-lactamase were detected by polymerase chain reaction.Results The resistant rates of the 26 strains to Ceftazidime,Cefoxitin,Piperacillin-tazobactam and Ciprofloxacin were 100.0%.About 80.8% to 96.2% of these strains were resistant to the other antimicrobial drugs.Among the 26 MDR-AB strains,100% (26/26) strains possessed oxa-51,77% (20/26) possessed oxa-23 gene,54% (14/26) carried arnpC gene,both oxa-23 and ampC were identified in 42% (11/26) strains,while oxa-24,oxa-58,imp-1,imp-4 and vim-2 gene were not identified.Conclusions The drug resistance of Acinetobacter baumannii is serious,oxa-23 and ampC are the major plactamase genes carried by MDR-AB in neonatal intensive care unit.
6.Autogenous iliac bone implantation after core decompression for the treatment of ischemic necrosis of the femoral head in dogs

BACKGROUND: The pathological change of femoral head was observed after treating necrosis with core decompression and autogenous iliac bone implantation, expected to obtain a better understanding of the prognosis of autogenous iliac bone implantation method.OBJECTIVE: To explore the curative effect of the ischemic necrosis of femoral head filling the hole of the core decompression with autogenous iliac bone implantation in animal models. DESIGN, TIME AND SETTING: Animal trial observation. The experiment was performed at the Central Laboratory of the Guilin Medical University from April 2006 to September 2007.MATERIALS: Nine hybrid adult dogs were prepared for necrosis of femoral head models.METHODS: The ischemic necrosis of femur head was made with the liquid nitrogen in dogs. The rod-shape autogenous cancellous bone that comes from the posterior superior iliac spine was driven into the full depth of the core r decompression tunnel to the subchondral bone. The dogs were divided into 3 groups, 3 dogs in each groups were sacrificed respectively at 3, 6, 12 weeks after operation. Radiograph and histological examination were used to evaluate the results.MAIN OUTCOME MEASURES: Histological changes of the femoral head necrosis and bone graft at the weeks 3, 6 and 12.RESULTS: No collapse of femoral head was observed at the 3 weeks after operation. The graft as well as perivascular tissues showed necrosis with empty lacune. Fiber granulation tissues could be seen ingrowth of capillaries and perivascular tissues into the bone graft. At 6 weeks after operation, basophilia blue area and new born osteoblast, cartilage, as well as chondroblasts zone could be found at the surface of bone trabecula. After 12 weeks of operation, new bone formation, representing typical process of "creeping substitution". And the cancellous bone of necrotic femoral head was rebuilt gradually. CONCLUSION: Core decompression combined with implantation of autogenous iliac bone implantation can accelerate femoral head repairing, delay or prevent femoral head from necrosis.
7.Treatment of 7 patients with medial meniscal cyst under arthroscopy.

OBJECTIVETo investigate clinical effects of arthroscopy in the treatment of medial meniscal cyst.
METHODSFrom June 2011 to January 2013, 7 patients with medial meniscal cyst were treated with arthroscopy. There were 3 males and 4 females,ranging in age from 27 to 63 years old,with a mean age of (43.93±2.10) years old. The cysts have been discovered for 3 to 30 months,with a mean time of (10.6±1.3) months. All the patients complained of knee pain,especially in the medial joint gap. The Pisani sign, Caklin sign and medial McMurray sign were all positive. Preoperative MRI examination confirmed the diagnosis. Lysholm score changes and clinical efficacy were observed through a six-month follow-up.
RESULTSThe postoperative Lysholm scores were all significantly higher than the preoperative scores. According to Sarimo standard, 6 patients got an excellent result, and 1 good.
CONCLUSIONArthroscopic treatment of medial meniscal cyst has replaced the traditional method, which could retain the normal meniscus as much as possible and repair the meniscus injury simultaneously, as well as get a good curative effect and a good recovery of knee function. This method is worthy of clinical application.

9.Drug susceptibility of wild-type and mutant H7N9 neuraminidase to zanamivir and oseltamivir.

This study aimed to investigate the drug susceptibility of wild-type and mutant avian influenza A (H7N9) virus neuraminidase (NA) to oseltamivir and zanamivir. Codon optimized DNA of H7N9 (A/ Hangzhou/1/2013) NA was synthesized and constructed into the pcDNA3.1/His vector (NA(H7N9-WT)). Mutant NA(H7N9-H274Y) and NA(H7N9-R292K) plasmids were constructed by directed mutagenesis PCR using NA(H7N9-WT) plasmid as the template followed by sequencing. NA plasmids were transfected into 293T cells and cell lysates containing NAs were collected 48 h post-transfection. Wild-type and mutant NAs were analyzed by Western blotting and their activities were tested by the 4-MUNANA-based assay. All three NAs were expressed and enzymatic activities were confirmed. The effects of oseltamivir and zanamivir on all three NAs were then tested. It showed that the half maximal inhibitory concentrations (IC50s) of oseltamivir carboxylate on NA(H7N9-WT), NA(H7N9-H274Y) and NA(H7N9-R292K) were 1.6 nM, 15.1 nM, and > 1 000 nM with fold changes of 9 and > 625, respectively. The IC50 values of zanamivir on NA(H7N9-WT), NA(H7N9-H274Y), and NA(H7N9-R292K) were 1.1 nM, 1.4 nM, and 38.0 nM with fold changes of 1.3 and 34, respectively. These results indicated that oseltamivir and zanamivir could significantly inhibit NA(H7N9-WT). NA(H7N9-R292K) showed high-level resistance to both drugs (34-fold and 625-fold) and NA(H7N9-H274Y) was sensitive to both (1.3-fold and 9-fold). These results indicated that both oseltamivir and zanamivir could be used for patients infected with the H7N9 virus. However, when patients carried the H7N9 virus with a NA R292K mutation, other medications would be preferred over oseltamivir or zanamivir.

10.Impact of mother's cognitive behavior intervention on premature infant-mother interaction

Objective To examine the impact of cognitive behavior intervention (CBI) on mother-infant interaction in the neonatal department. Methods Low-risk appropriate for gestational age premature infants with gestational age 28-36 weeks, who were admitted to the neonatal intensive care unit in our hospital between August 2007 and May 2008, and their mothers were assigned to intervention group and control group respectively. Intervention group mothers (n = 19)participated in CBI. Control group mothers (n= 19) were asked to read educational materials. Mothers in the intervention group, all received 4 times interventions and 2 h for each time. The CBI process include assessment, plan, implementation and evaluation. Mothers were educated for knowledge about appearance and behavioral characteristics of preterm infants and were guided about how to take care of their babies and participate in nursing care process. Mothers completed measures on mother-infant interaction (Nursing Child Assessment Feeding Scale, NCAFS) in the first follow-up after infant discharge. Results The results showed that the scores of NCAFS and total scores of the mothers in the intervention mother-infant dyads were 46.4 ± 8.5 and 35.8±6.3,higher than in the control group (42.7±5.9 and 33.3 ± 5.6), respectively, but there were no statistic difference between the two groups (t = 1.55 and 1.28, P> 0. 05). The scores of "mother's response to distress" were significantly higher in the intervention mother-infant group (12. 2± 1.7) than in the control group (10.7±2.4)(t=2.10,P<0.05). Conclusions The levels of premature infant-mother interactions are low. CBI can improve mother's response to premature infant's distress. Further study is needed to investigate the effects of CBI in improving mother-infant interaction.
