Chinese Journal of Biologicals 2023;36(12):1471-1475
Objective To express and purify the human renin(REN)and mature renin(REN-340)in prokaryotic cells, and detect the concentration. Methods According to the human REN gene sequence(5972)registered in NCBI,REN and REN-340 gene fragments were artificially synthesized and cloned into vector p ET-28a(+)after E.coli codon optimization, and the recombinant expression plasmids 6 × His-REN-p ET and 6 × His-REN-340-p ET were constructed. The recombinant plasmids were transformed into competent E.coli BL21(DE3)and induced at different induction temperatures(20,25,30and 37 ℃)and different final concentrations(0. 1,0. 2,0. 4 and 0. 5 mmol/L)of IPTG. Recombinant REN and REN-340were induced under the optimal induction conditions, and the concentrations were detected by ELISA after nickel ion affinity chromatography with Ni-NTA. Results The recombinant expression plasmids 6 × His-REN-p ET and 6 × His-REN-340-p ET were constructed correctly as identified by restriction analysis and sequencing. The optimum induction temperature of recombinant REN and REN-340 was 37 ℃ and 20 ℃,and the best final concentration of IPTG was 0. 1 and 0. 2 mmol/L,respectively. The concentrations of purified recombinant REN and REN-340 were 9 148. 21 and 15 115. 31 m IU/m L,respectively. Conclusion The prepared recombinant REN and REN-340 proteins have high concentration with short production cycle, which are expected to be used as candidate quality control products and standard substances for external quality assessment(EQA).

4.Investigation and analysis of data of abdominal injuries in Deyang after the Wenchuan earthquake
Chinese Journal of Digestive Surgery 2008;7(4):258-259
A magnitude of 8.0 earthquake struck on Wenchuan on May 12, 2008. Until July 1, 1393 injured persons had been admitted to Deyang People's Hospital. Of all injured persons, 32 were diagnosed with abdominal injury, including 18 men and 14 women. All the abdominal injuries were closed injury, and multiple abdominal viscera were involved in the abdominal injuries after the earthquake. Careful examination is crucial in preventing missed diagnosis. The incidences of the liver and spleen injuries were significantly higher than that of the intestine, and the reason may be that the liver and spleen are the parenchymal viscera. Diagnostic abdominocentesis can timely diagnose the parenchymal viscera with severe blood loss. Diagnostic peritoneal lavage combined with selective CT scan can timely diagnose the abdominal injuries with comparatively low cost.
5.Relationship between GRIM-19 and cancers
International Journal of Surgery 2010;37(11):759-762
Gene associated with retinoid-IFN-induced mortality-19(GRIM-19)is originally isolated as a growth suppressive gene using a genetic screen.GRIM-19, initially found in nucleus and mitochondria, is essential for the assembly and functioning of mitochondrial complex I.GRIM-19 involves the regulation of cell proliferation and apoptosis, and low expression or mutation of GRIM-19 contributes to abnormal proliferation.During viral oncogenesis, GRIM-19 may be a general target protein similar to other cellular tumor suppressors.GRIM-19 plays an important role in tumor formation and apoptosis inhibition, and may be as a new tumor marker to early cancer screening.
6.Effects of transcription factor HOXB7 silencing on viability, invasion and migration of SGC-7901 cells
Chinese Journal of Pathophysiology 2016;32(10):1824-1829
[ ABSTRACT] AIM:To explore the effects of transcription factor HOXB7 silencing on the viability , invasion and migration of SGC-7901 cells.METHODS:The siRNA was designed to specifically interfere the expression of HOXB7.To assess the silence efficiency of the siRNA , the expression of HOXB7 at mRNA and protein levels was detected by real-time PCR and Western blot in the SGC-7901 cells transfected with siRNA .The cell viability was measured by MTT assay .Cell cycle-related proteins such as cyclin D 1 and CDK4 were determined by Western blot .Transwell assay was performed to an-alyze the migration ability of the SGC-7901 cells. The mRNA and protein levels of the tumor invasion-and metastasis-relat-ed molecules, such as VEGF, PTEN and p-AKT, were also detected by real-time PCR and Western blot .RESULTS:The expression of HOXB7 at mRNA and protein levels was higher in the gastric carcinoma tissues than that in the normal gastric tissues. The expression of HOXB7 at mRNA and protein levels was knockdown by siRNA .After silencing of HOXB7 gene expression, the viability of the SGC-7901 cells decreased, the expression of cyclin D1, CDK4 and VEGF was down-regula-ted, and the phosphorylation level of AKT was also obviously decreased .CONCLUSION:HOXB7 effectively promotes the viability, invasion and migration of gastric carcinoma cells by up-regulating the expression of cyclinD 1, CDK4 and VEGF, and increasing the phosphorylation level of AKT to inhibit PTEN function .
7.Expression of KLF4 in diabetic mouse myocardium and the interventional effect of Tongxinluo capsule on diabetic cardiomyopathy
Chinese Pharmacological Bulletin 2015;(6):876-881
Aim To observe the expressions of KLF4 in the myocardium of diabetic mice and their changes under Tongxinluo capsule intervention. Methods For-ty KK/Upj-Ay mice were randomly divided into diabet-ic model group(n=10)and diabetic model with Tongx-inluo(TXL low, middle, high) groups(n =10,respec-tively). C57BL/6 mice were selected as control group ( n=10 ) . At the end of the 3 th month the mice were sacrificed and were weighed. The fasting blood-glucose (FBG),glycosylated hemoglobin(HbA1c),triglyceride ( TG) ,total cholesterol ( TC ) and insulin ( FINS ) were measured to calculate HOMA-IR. Radioimmunoassay was used to measure serum TNF-αand IL-6 . The path-ological changes in the myocardium of mice were ob-served by HE staining. KLF4 mRNA was examined by Real-time PCR, while KLF4 and NF-κB protein were measured by Western blot. Results Compared to the control group,FBG,HbA1c,TG,TC,FINS,HOMA-IR,
TNF-α and IL-6 in model group were markedly in-creased;the expressions of myocardial KLF4 were markedly decreased and the expression of nuclear NF-κB protein were markedly increased ( P < 0. 01 ) . Tongxinluo capsule could significantly reduce myocar-dial pathological damage, FINS, TG, TC, TNF-α, IL-6 level and the expression of nuclear NF-κB protein and up-regulate the expression of KLF4 ( P <0. 05 ) , but had no effect on FBG and HbA1c(P>0. 05). Conclu-sions KLF4 may be involved in the development of myocardial injury during diabetes. Tongxinluo capsule can ameliorate the myocardial damage and improve the function of diabetic myocardium by up-regulating the expression of KLF4 and inhibiting the activation of NF-κB inflammatory signal pathway.
8.Therapeutic Observation of Thread-embedding at Scalp Points for Insomnia
Shanghai Journal of Acupuncture and Moxibustion 2015;(5):418-420
Objective To observe the clinical efficacy of thread-embedding at scalp points in treating insomnia.Method Sixty-four insomnia patients were randomized into a treatment group of 33 cases and a control group of 31 cases. The treatment group was intervened by thread-embedding at scalp points, while the control group was by orally taking Estazolam. Four weeks were taken as a treatment course, and the clinical efficacies and changes of indexes were measured after a treatment course.Result The total effective rate was 90.0% in the treatment group versus 90.3% in the control group, and the difference was statistically insignificant (P>0.05). The item scores of Pittsburgh Sleep Quality Index (PSQI) were changed significantly after treatment in both groups (P<0.01). The changes of sleep latency and daily function disturbance in the treatment group were significantly different from that in the control group (P<0.05).Conclusion Thread-embedding at scalp points and oral administration of Estazolam both can improve insomnia symptoms. Thread-embedding at scalp points is inferior to Estazolam in improving sleep latency but is superior to Estazolam in improving daily function.
9.Ultrasonic Detection of Mesenteric Lymph Nodes in Children with Intermittent Abdominal Pain
Acta Medicinae Universitatis Scientiae et Technologiae Huazhong 2015;(5):575-577
Objective To examine the clinical application of ultrasonography to detection of mesenteric lymph nodes in chil‐dren with intermittent abdominal pain. Methods A total of 196 children who underwent abdominal ultrasonography for differ‐ent reasons were divided into the intermittent abdominal pain group and non‐abdominal pain group. The location ,size and num‐ber of mesenteric lymph nodes were recorded. Results Statistical difference in the long‐axis diameter(P=0.005)and ratio of short‐to‐long‐axis diameter was found among patients with different ages in non‐abdominal pain group(P= 0.015) ,while no significant difference was seen in short‐axis diameter(P=0.773).No significant difference was observed in the diameter of each axis between different genders in non‐abdominal pain group. There was a statistical difference between abdominal pain group and non‐abdominal pain group in the incidence of lymph nodes with short‐axis diameter of 6 mm and larger(P=0.002)and long‐axis diameter of 14 mm and larger(P=0.007).Conclusion Mesenteric lymph node with short‐axis diameter larger than 6 mm should be considered enlarged in children ,but should not be diagnosed with mesenteric lymphadenitis. It’s common to find en‐larged mesenteric lymph nodes in children without abdominal pain.Further investigations with a larger number of patients are required to confirm these findings .
10.Sensitivity of gastric cancer stem cells to 5-fluorouracil
Chinese Journal of Tissue Engineering Research 2015;(41):6606-6610
BACKGROUND:5-Fluorouracil is a common chemotherapy drug for gastric cancer, but it is more likely to develop drug resistance in clinical treatment. Studies have shown that tumor stem cels are lowly sensitive to chemotherapeutic drugs, which may be an important cause of chemotherapy resistance. OBJECTIVE:To analyze the sensitivity of gastric cancer stem cels to 5-fluorouracil in vitro, and to understand the mechanism of drug resistance associated with gastric cancer. METHODS:Based on the sorting strategy, human gastric cancer cel clones were isolated from AGS cel lines. CD44 and thymidylate synthetase expression in different clones was detected using immunocytochemistry analysis. Clone formation assay was used to evaluate self-renewal capacity of different clones. Cel counting kit-8 was used to determine the clonal growth inhibition rate of AGS under treatment with different concentrations of 5-fluorouracil. RESULTS AND CONCLUSION:The AGS cels that were inoculated with low density and cultured could differentiate into 32 forms, including the ful clone (16%, 5/32), the second clone (66%, 21/32) and the accessory clone (19%, 6/32). Among them, the ful clones highly expressed CD44 and thymidylate synthetase, and could generate a great amount of passage 2 clones after inoculation; the second clones showed a weak expression of CD44 and thymidylate synthetase, and could generate a smal number of passage 2 clones after inoculation; the accessory clones showed a weak or no expression of CD44 and thymidylate synthetase, and there was no passage 2 clone after inoculation. Under the effect of different concentrations of 5-fluorouracil, the growth inhibition rates of the secondary clones and AGS cels were both higher than that of the ful clones (bothP < 0.05). These findings indicate that gastric cancer stem cels have a relatively lower sensitivity to 5-fluorouracilin vitro, which is speculated to be an important mechanism of drug resistance.
