2.Establishment of a hepatic stem cell line stably expressing human interleukin-2

Objective:Establishing the hepatic stem cell(WB-F344)line that expresses human interleukin-2 stably and effectively.Methods: We constructed the retroviral vector Plpcx-il-2 containing human interleukin-2 gene, and transfected it into PT67 packaging cells.The infectious viruses were used to infect the hepatic stem cell WB-F344,then the puromycin resistant clones were acquired after puromycin selection.The expression of IL-2 was detected using RT-PCR,immunocytochemistry, Western blot and indirect immunofluorescence, respectively.Results: The retroviral vector containing human IL-2 gene was constructed.The titer of virus in the supernatant reached 10~5 CFU/ml after transfecting PT67 packeting cells.The puromycin resistant clones were acquired after infection of WB-F344 and the cell line WB-F344/Plpcx-IL-2 expressing human interleukin-2 stably was established.Conclusion: By retroviral-mediated transfecting human interleukin-2 gene, the hepatic stem cell stably and effectively expressing interleukin-2 are obtained.This provides a good basis for further research on immuno-gene therapy of liver cancer.
3.CT diagnosis and differential diagnosis of renal and retroperitoneal tumors in children

Objective To investigate the diagnosis and differential diagnosis of renal and retroperitoneal tumors in children with CT.Methods CT findings in 189 cases of pathologically confirmed renal and retroperitoneal tumors,including nephroblastoma in 71 cases,neurogenic tumor in 89 cases(neuroblastoma in 70 cases,ganglioneuroblastoma in 2 cases and ganglioneuroma in 17 cases),teratoma in 18 cases and lymphoma in 11 cases,were retrospectively analyzed.Results CT showed that nephroblastoma was a round or oval encapsuled mass usually resulting in renal destruction,the residual kidney was a crescent-shaped encapsuled mass which is easy to lead to thrombosis of renal vein and inferior vena cava due to tumor.The manifestations of neuroblastoma were similar to those of ganglioneuroblastoma,usually with calcification and embed vessels,and could invade the kidneys and often metastasize to lymph nodes,cranium and brain,bones and liver.The ganglioneuroma was manifested as a small oval-shaped low dense mass in adrenal gland area or in paravertebral, with no exacerbation or slight exacerbation in its early stage.Lymphoma was located around the median line and presented as an irregular mass fused by multiple lymph nodes embedding the vessels loosely.Teratoma,usually occurred on one side of the retroperitonium,was characterized by calcification/ossification and fat tissue.Conclusion CT is of a higher value for the diagnosis and differential diagnosis of renal and retroperitoneal tumors in children.

Objective To investigate the effects of DNaseⅠ-treated streptococcal antigens on peripheral blood mononuclear cells(PBMC)proliferation in psoriasis patients.Methods PBMC from 15 patients with psoriasis and 10 healthy controls were stimulated with three concentrations(2,10,25?g/mL) of streptococcal antigens(SA), nd DNaseⅠ-treated streptococcal antigens(DNaseⅠ-SA).After 72 h of culture,cellular proliferation was determined by ~3H thymidine incorporation.Results The DNaseⅠ-SA,at all the concentrations tested,was less potent than the corresponding concentrations of SA(P0.05),however,the lower concentrations(2,10?g/mL) of DNaseⅠ-SA were less potent than those of SA(P0.05).Also,the extent of proliferation response to SA and DNaseⅠ-SA was higher in the patients than in the controls(P

Objective To observe the clinical effect of herbal decoction Sishen Jian on acute osteoarthritis of knee joint.Methods The 60 osteoarthritis cases in the attack stage were randomized into treatment group(30 cases)and control group(30 cases).The treatment group was given the decoction Sishen Jian and Diclofenac Sodium Sustained Release Capsules and the control group was given the capsules only.Two weeks made one course of treatment.Before and after treatment,the symptoms such as pain and morning stiff- ness,the signs including joint swelling and tenderness,and the erythrocyte sedimentation rate(ESR)and C reactive protein were ob- served for comparison.Results The total effective rate of the treatment group and control group was 96.66% and 53.26% respec- tively.The difference of two groups was significant(P
7. In vitro percutaneous permeability and skin irritation of gastrodigenin

Objective: To investigate percutaneous permeability and skin irritation of gastrodigenin so as to provide the basis on the development of transdermal drug delivery formulations. Methods: The modified Franz diffusion cell method and the excised rat skin were used to evaluate the percutaneous permeability of gastrodigenin. The effects of various common-used solvents, transdermal enhancers, and particle carriers on the permeability of gastrodigenin were studied. The rabbit skin irritation test was used to evaluate the safety of gastrodigenin solutions and their gels. Results: Gastrodigenin was easy to permeate rat abdominal skin. The penetration rate constant (Js) was 134.1 μg/(cm2·h), and transit dose accumulated in 24 h was 3 mg/cm2. The solubility and permeability of gastrodigenin could be enhanced by 30% EtOH, so it was an ideal solvent for gastrodigenin. Improvement effects of the transdermal enhancers (Azone, turpentine, and borneol) and the transdermal particle carriers (microemulsion and ethosomes) were not significant. The skin irritation test results indicated that neither gastrodigenin soultions nor their gels exerted irritative effects to the rabbit skin. Conclusion Gastrodigenin has good percutaneous permeability without skin irritation, so it is suitable for the transdermal drug delivery in the treatment of central nervous system diseases.
8.Analysis of Narcotic Drug Use from 2002 to 2005 in Our Hospital

OBJECTIVE: To evaluate the current situation and trend of narcotic drug use in our hospital where the authors work in order to provide the reference for scientific management and rational use of these drugs. METHODS: The yearly amount of narcotic drugs administered in the whole hospital, the yearly amount in the separate departments, as well as drug expenditures and ratios between January 2002 and November 2005 were added up and analyzed statistically. RESULTS: The amount of bucinnazine use dominated in the first. The amount of morphine for oral use was increasing year by year. The amount of fentangl transdernal patch use was also bigger and increasing rapidly. However, the amount of pethidine and morphine for injection use was decreasing. CONCLUSION:Analgetics for oral and transdermal use will be the main categories of analgetics in the future.
9.Application of parenteral nutrition in critically ill patients undergoing cardiac surgery: a retrospective study

Objective To summarize the clinical application of parenteral nutrition (PN) in Fuwai Hospital. Methods The clinical applications of PN in 132 critically ill patients undergoing cardiac surgery in our hospital were retrospectively investigated and analyzed. Results The application of PN in Fuwai hospital was basically appropriate and standardized. Energy supply in PN formulation was reasonably low, but still met the energy needs of patients in the fasting period. The nutritional composition and energy/nitrogen ratio in the formulation were basically rational. However, problems including the lack of double energy in partial formulation, low proportion of glucose-supplied energy due to low glucose-lipid ratio, and insufficient supply of insulin still existed. One (0.76%)patient was discharged according to his own decision; 22 ( 16.67% ) patients died, which were not directly associated with PN; and the other 109 (82.58%) patients were cured and discharged. Conclusion Application of PN for critically ill patients undergoing cardiac surgery must strictly follow the operation indications and rationally select nutritional components and proportions to ensure its safety and effectiveness.
10.Comparison of the effects of sevoflurane and disoprofol to inflammatory factors in patients received abdominal surgeries

Objective To explore and compare the effect of sevoflurane and disoprofol to inflammatory factors in patients received abdominal surgeries.Methods Patients with sevoflurane and disoprofol respectively were anaesthetized and detected IL-6,IL-8,IL-10 and TNF-α in their serum at T1 (when anesthesia induction),T2 (after anesthesia induction),T3 (30min after beginning of operations) and T4 (after operations).Results Compared with T1,the serum level of IL-6,IL-8,IL-10 and TNF-α increased significantly in all patients(all P < 0.05) ; Compared with disoprofol group,the serum level of IL-6,IL-8 and TNF-α increased significantly (all P < 0.05) ; Compared with sevoflurane group,the serum level of IL-10 increased significantly(all P < 0.05).Conclusion Intravenous anesthesia with disoprofol in patients receiving abdominal operations is proper for maintenance of inflammatory balance.
