1.Creep characterizes of rabbit tibia under torsion load following exercise

BACKGROUND:The creep performance of bone tissues during exercise is achieved through the microstrain,and the holosteous torsion can completely retiect the biomechanicel charactedstics of bone tissues.OBJECTIVE:To determine the creep characteristics of rabbit tibia under the torsion load following exercise.DESIGN,TIME AND SETTING:A parallel controlled animal observational experiment was performed at the Laboratory of Biomechanics in Orthopedic Hospital of Sichuan Province between May 2005 and July 2007.MATERIALS:A total of 32 Japanese white rabbits,irrespective of genders,aged 6 months,were divided into exercise group(n=24)and control group(n=8).METHODS:Exercise group animals were placed in cage to receive electrical stimulus.0.1-0.2seconds once every 20 seconds.Following the stimulation,the animals ran and jumped for several paces along with the cage and made a tum,15 minutes once,three times per day,totally 45 minutes,at the interval of 15 minutes.The exercise was carried out five days per week for 4successive weeks.The control group was fed as exercise group.but received no exercise.MAIN OUTCOME MEASURES:After 4 week exercise.the tibias were griped by the torsion clamping apparatus,the creep characterizes of anterior,posterior,medial and lateral sides of tibia under 0.52 N·m torsion load within 500 seconds were determined by strain gauge technique.RESULTS:Under torsion load,the creeps were present obviously in the rabbit tibia,reached the creep curve inflection point at 2seconds,and then gradually appeared.After 4 weeks,the creeps of tibias at anterior and posterior side in the exercise group were less than control group;in the control group,the anterior side of tibia was remarkably greater than the medial side.After exercise.the creep was the smallest in anterior side,which was less than the lateral side(P<0.05).CONCLUSlON:The creep characteristics of tibia are anisotropic,the deformation at the anterior and posterior sides is greater than that at the medial and lateral sides.Exercisecan alter the creep charactedstice of rabbit tibia and reduces the creep at the anterior and posterior sides.
2.Application feasibility of xenogeneic deproteinized cancellous bone scaffold in the treatment of spinal intertransverse fusion

BACKGROUND:Nowadays, most of the studies regarding tissue engineering bone have mostly focused on critical-size bone defects of the backbone; however, there are less studies and reports on its spinal fusion. OBJECTIVE:To explore the feasibility of xenogeneic deproteinized cancelous bone as bone tissue engineering scaffold in the treatment of spinal intertransverse fusion. METHODS:The cancelous part in the distal femur of adult pigs was obtained to prepare xenogeneic deproteinized cancelous bone. After combined with the recombinant human bone morphogenetic protein, the xenogeneic deproteinized cancelous bone was combined with bone marrow mesenchymal stem cels to prepare tissue engineering bone. Twenty-four goats were obtained to prepare intertransverse bone bed, and randomly divided into two groups: observation and control groups. In the observation group, the tissue engineered bone was implanted into the left side, and the xenogeneic deproteinized cancelous bone of recombinant human bone morphogenetic protein was implanted into the right side. In the control group, the autologous iliac bone was implanted into the left side, and xenogenic deproteinization cancelous bone was implanted into the right side. At the 4th, 8th and 12th weeks after implantation, the fusion segment was obtained for gross observation, X-ray observation, histological observation and biomechanical testing. RESULTS AND CONCLUSION:X-ray films showed that the implant materials from these two groups were fixed wel and reliably. At different time points after implantation, the implant materials from each group were al in good position. There were no purulent and necrotic tissues around the material. Soft tissue ingrow and wraping were present. There were no effusions and necrosis surrounding the implant materials. The imaging and histological performance in the tissue engineering bone group outperformed that in the recombinant human bone morphogenetic protein xenogenic deproteinized cancelous bone group and xenogenic deproteinized cancelous bone group, which was the most close to the autogenous bone. At the 12th week after implantation, the maximum bending load in the tissue engineering bone group was the most close to the autogenous iliac bone group. There was no significant difference between these two groups. These results demonstrate that as bone tissue engineering scaffold, xenogenic deproteinized cancelous bone has a certain application feasibility in the treatment of spinal intertransverse fusion.
3.The expression of VEGF,COX2 and mPGES mRNA in colon cancer

Objective To study the expression of VEGF,Cox2 and mPGES in colon cancer.Methods VEGF,Cox2 and mPGES mRNA expression in 32 paired samples(tumor and adjacent normal tissue)were de- termined by using real time RT-PCR.Results VEGF was overexpressed in 19 of 32(59.3 %)tumor tissues compared with that in 6 of 32(18.7 %)adjacent normal tissue;COX2 was overexpressed in 20 of 32(62.5 %) tumor tissues compared with that in 5 of 32(15.6 %)adjacent normal tissue;mPGES was overexpressed in 24 of 32(75 %)tumor tissues compared with that in 9 of 32(28.12 %)adjacent normal tissue.Conclusion Our result suggested that VEGF165,mPGES and COX2 overexpressed in colon cancer.
5.Cutaneous metastasis of cholangiocarcinoma in six cases

Objective To analyze the clinical characteristics and therapeutic effects of cutaneous metastasis from cholangiocarcinoma. Methods From 2006 to 2009, clinical data of 6 patients found with cutaneous metastasis from cholangiocarcinoma were reported. Perineural and vascular invasion were all found in the primary cholangiocarcinoma in these 6 cases with cutaneous metastasis and results were compared with 60 cases of cholangiocarcinoma without cutaneous metastasis. Follow-up to the 6 patients with cutaneous metastases was conducted at an interval of three months. Results One patient with mixed hepatocellular and cholangiocellular carcinoma developed cutaneous metastatic tumor and the pathological diagnosis was cholangiocellular carcinoma. Three patients with hilar cholangiocarcinoma and two patients with distal cholangiocarcinoma presented cutaneous metastases. All six cases of the primary cancers and cutaneous metastases were diagnosed as moderately- poorly differentiated adenocarcinoma. And there was perineural and vascular invasion in the primary tumors. Perineural and vascular invasion of primary cancer was found in 17of 60 patients with cholangiocarcinoma and without cutaneous metastasis. The cutaneous metastasis was positively correlated to the perineural and vascular invasion in the primary tumor (χ2 = 12. 7288, P <0. 001 ). Two of 6 cases presenting solitary nodular cutaneous metastasis received skin tumor excision. These two patients survived 8 and 10 months respectively. Among four patients with multiple cutaneous metastases,two cases received chemotherapy and survived 3 and 6 months respectively, the other two cases refusing treatment survived 3 and 4 months respectively. Conclusions Cutaneous metastasis usually occurred at cholangiocarcinoma with lower differentation and perineural and vascular invasion in the primary tumor.Solitary nodular cutaneous metastasis could be excised, multiple cutaneous metastases had poor prognosis.

9.Gray Matter Heterotopia:CT Diagnosis

Objective To analyze the CT appearances and the classification of gray matter heterotopia.Methods The clinical and CT data of 26 cases with gray matter heterotopia were retrospectively reviewed,11 were males and 15 were females,ranged in age from 2 days to 9 years with a mean of 2.6 years.Results The classification of heterotopia included:(1)Subependymal heterotopia in 14,5 cases with encephaloceles,1 case with Dandy-Walker malformation and 1 case with arachnoid cyst of cisterna megna.(2)Subcortical heterotopia in 7,4 cases with callosal agenesis were accompanied.(3)Band heterotopia in 5.Conclusion CT scan can not only reveal the appearances of the subependymal,subcortical and band types heterotopia,but also show other associated abnormalities.
10.Lung injury in heatstroke rats in xerothermic environment of desert

Objective The extremely xerothermic environment of desert easily causes heatstroke and consequently multiple organ damage.Few reports are seen at home and abroad on heatstroke-induced lung injury associated with dry heat of desert.This stud-y explored the changes of the injured lungs in heatstroke rats in xerothermic desert. Methods Forty-eight male SD rats were ran-domly divided into six groups of equal number: mild, moderate and severe heatstroke groups and their corresponding controls.The rats of the three heatstroke groups were exposed to a temperature of 41 ℃and humidity of 10%, while those in the three corresponding control groups to a temperature of 25 ℃ and humidity of 35% for 70, 110 and 145 minutes, respectively.Then all the rats were sacrificed and the bronchoalveolar lavage fluid ( BALF) and lung tissues were collected for observation of pathological changes, obtainment of lung in-jury scores, calculation of the wet/dry weight ratio of the lungs, measurement of the total protein level in the BALF, and determination of the malondialdehyde ( MDA) content in the lung tissue. Results The mild, moderate and severe heatstroke groups showed re-markable increases compared with the three corresponding control groups in lung injury scores (3.25 ±0.50, 6.50 ±0.58 and 8 ± 0.82 vs 1.25 ±0.50, 1.00 ±0.00 and 1.25 ±0.50), the wet/dry weight ratio of the lungs (3.50 ±0.57, 4.85 ±0.19 and 4.10 ± 0.07 vs 1.78 ±0.66, 1.67 ±0.68 and 1.67 ±0.25), the total protein level in the BALF ([93.51 ±5.54], [141.24 ±6.07] and [160.76 ±6.74] μg/mL vs [42.6 ±8.67], [45.25 ±6.78] and [41.68 ±3.62] μg/mL), and the content of MDA in the lung tis-sue ([1.79 ±0.08], [2.79 ±0.11] and [3.52 ±0.36] nmol/mg prot vs [1.21 ±0.12], [1.20 ±0.12] and [1.28 ±0.19] nmol/mg prot) .The lung injury scores, the total protein level in the BALF, and the content of MDA in the lung tissue were increased with the prolonging of the time of exposure to dry heat, while the wet/dry weight ratio of the lungs was the highest in the moderate heat-stroke group.The lung injury scores were positively correlated with the wet/dry weight ratio of the lungs, the total protein level in the BALF, and the content of MDA in the lung tissue (P<0.01). Conclusion Heatstroke in the xerothermic environment of desert can cause lung injury in rats, which deteriorates with the prolonging of the time of exposure to dry heat.As for the management of such heatstroke, more attention should be paid to the protection against lung injury.
