1.Expression of interleukin-8 in interstitial fibroblast cells of breast cancer before and after application of docetaxel chemotherapy

Objective:To study and investigate the expression effects of interleukin-8 (IL-8)in interstitial fibroblast cells of breast cancer before and after the application of docetaxel chemotherapy.Methods:46 cases of breast cancer without chemotherapy surgical resection in patients with resected tissue specimens from August 2014 to February 2016 in our hospital were selected,all specimens were cultured to form stable breast cancer stromal fibroblast cells,each cell line after raising Daipei were divided into the observation group and control group,the control group cells without any treatment,the observation group of 24h cells treated with docetaxel(20ng/mL),two groups of cells were extracted from the mRNA and analyzed the gene expression and differential gene expression analysis,and then to PCR and protein to detected the expression level of electrophoresis detection of interleukin two interleukin-8 cells,the expression of interleukin-8 in two groups of breast cancer cells was observed and compared.Results:After docetaxel chemotherapy,the breast cancer stromal fibroblast gene expression changed,and compared to the control group,the observation group were given docetaxel chemotherapy after breast cancer between matter into fiber cell interleukin-6 serum IL-8 mRNA and protein expression levels were significantly higher than that of the control group,the difference had statistical significance (P<0.05).Conclusions:Interleukin IL-8 in the breast cancer matter into fiber cells were significantly up-regulated after docetaxel chemotherapy,indicating that the expression changes of breast cancer may be related with the effect of chemotherapy and chemotherapy drug resistance related.
2.Clinical study on the treatment of male immune infertility by Huzhangdanshenyin.

OBJECTIVETo investigate the efficacy of the Chinese medicine Huzhangdanshenyin on male immune infertility, and provide an effective therapy for this disease.
METHODSNinety men with immune infertility were selected as the research subjects and randomly divided into two groups, 60 in the treatment group, treated by Huzhangdanshenyin, and the other 30 in the control, treated by prednisone, both for 3 months. The improvement of clinical symptoms, immunologic indexes (antisperm antibodies in serum and seminal plasma) and sperm indexes (semen liquefied duration, motility, viability, density and abnormal morphology rate) were observed and the results analyzed.
RESULTSThe total antisperm antibody reversing ratio of the treatment group was higher than that of the control (P < 0.01), especially the serum antisperm antibody reversing ratio. There were significant differences in the clinical cure rate and total validity rate between the treatment group and the control (P < 0.01). After the treatment, the markers of the clinical symptoms (P < 0.01), especially the serum antisperm antibody reversing ratio. There were significant differences in the clinical cure rate and total validity rate between the treatment group and the control (P < 0.01). After the treatment, the markers of the clinical symptoms were lower (P < 0.01), and the improvement of the clinical symptoms was better in the treatment group than in the control (P < 0.01), especially the symptoms of pain in the back and knees, distending and bearing-down sensation of the perineum and testis, hypersexuality and topalgias. Compared with pre-treatment, sperm motility and viability of the treatment group significantly improved (P < 0.01), and so did sperm density (P < 0.05). However, there were no significant differences in sperm density, semen liquefied duration, abnormal morphology rate and pH (P > 0.05) before and after the treatment.
CONCLUSIONThe Chinese medicine Huzhangdanshenyin works more effectively than prednisone in the treatment of male immune infertility. It could improve the antisperm antibody reversing ratio, clinical symptoms and signs and ameliorate sperm indexes with no obvious advierse effects.

3. Protection mechanism of deacetylase inhibitor on spleen of rats with severe hemorrhagic shock

Objective To explore the protection and molecular mechanism of histone deacetylase inhibitors (HDACIs) on the spleen of rats with hemorrhagic shock. Methods A total of 60 SPF male SD rats were selected for the modeling of severe hemorrhagic shock using the method of arterial and venous cannulation with the time-divided bleeding. The measurement of mean arterial blood pressure and blood lactic acid was used to verify the modeling. The modeled rats were randomly divided into shock group, shock + suberoylanilide hydroxamic acid (SAHA) group, shock + autogenous transfusion group and shock + SAHA + autogenous transfusion group. Three hours after the treatment, the spleen of rats was collected and TUNEL method was employed to detect the apoptosis of spleen cells in each group. The statistical analysis was performed. Afterwards, real-time PCR and western blot were employed to detect the expression of BCL-2, BAX and caspass3 in the spleen of rats in each group. Results A total of 53 rats had successful modeling of severe hemorrhagic shock, with success rate of 88%. Cell apoptosis in the severe hemorrhagic model group was the most serious. After the intervention of HDACIs and the autogenous transfusion, the tissue injury was a bit recovered. Cell apoptosis was least in the shock + SAHA + autogenous transfusion group (P < 0.05). After the intervention of HDACIs and the autogenous transfusion, the relative expression of BCL-2 was significantly increased (P < 0.05), with highest relative expression of BCL-2 in shock + SAHA + autogenous transfusion group (P < 0.05). After the intervention of HDACIs and the autogenous transfusion, the relative expression of BAX was significantly decreased (P < 0.05), with lowest relative expression of BAX in the intervention group of single HDACIs. The change in the expression of caspass3 was similar to BAX, namely the relative expression of caspass3 was significantly decreased after the intervention of HDACIs and the autogenous transfusion (P < 0.05). Conclusions HDACIs and autogenous transfusion can all protect the spleen injury because of the severe hemorrhagic shock. Its molecular mechanism may be related to the regulation on the expression of BCL-2/BAX and caspass3, which may affect the apoptosis process of cells.
