Objective To observe the expressions of vascular endothelial growth factor(VEGF) and cyclooxygenase-2(COX-2) in the cerebral tissue following hypoxic-ischemic encephalopathy(HIE) in newborn rats and explore the machanism of inosine in protecting against hypoxic-ischemic brain damage(HIBD).Methods Sixty-six newborn rats aged 7 days were divided into sham,control and experimental groups.HIE models were made by clamping the right cervical artery and making hypoxia for 2 hours.The rats in experimental group began injecting inosine,at 1 day before experiment,and the rats in the sham and control groups saline solution with same dose.The samples were made at the given time,and expressions of VEGF and COX-2 were investigated by immunohistochemical technique.Results The cerebral tissue had no expression of VEGF and COX-2 in sham group.From 2 hours on cortex and striatum after HIE in control and experimental groups,expressions of VEGF and COX-2 increased rapidly,peaking at 12-24 hours,and then decreased gradually.Expressions of VEGF and COX-2 were higher in experimental group(All P
2.Clinical study of combination of acupuncture and medicine combined with auricular points treated with tourette syndrome in children

Objective To evaluate the clinical effect of combination of acupuncture and medicine combined with auricular pointsfor the children of MT.Methods A total of 70 children with MT who met the inclusion criteria, were divided into the treatment group and control group according to random number table method (n=35); The treatment group was given acupuncture treatment and prescription ofZishen-Tiaogan and auricular points; and the control group was given Haloperidol tablets. Both groups were treated for 2 months. The severity of the children's convulsing symptoms and the clinical efficacy were assessed by the Yale Global Tie Severityb Scale (YGTSS).Results After 1 course of treatment, the total effective rate of the treatment group was 82.9% (29/35), while the control group was 57.1% (20/35), and the difference was statistically significant (Z=-2.276,P=0.023); after 2 courses of treatment, the total effective rate of the treatment group was 88.6% (31/35) and the control group was71.4% (25/35).The difference was statistically significant (Z=-2.346, P=0.019). After 1 course of treatment, YGTSS score was (14.6 ± 6.9vs. 17.3 ± 7.5,t=4.604), and after 2 course of treatment,YGTSS score was (8.0 ± 5.3vs. 10.3 ± 4.9,t=5.521) in the treatment group wwere significantly lower than the control group (P<0.05).Conclusions Combination of acupuncture and medicine combined with auricular points can alleviate the clinical symptoms of children with MT and improve the clinical effect.
3.Effect of serum of patients with hepatopulmonary syndrome on myogenic differentiation, proliferation and migration of human lung fibroblasts

Objective To investigate the effect of the serum of patients with hepatopulmonary syndrome on the myogenic differentiation,proliferation and migration of human lung fibroblasts.Methods The human lung fibroblasts were seeded in plates or flasks and randomly divided into 2 groups (n =31each) using a random number table:serum of patients with hepato-pulmonary syndrome group and serum of healthy volunteer group.The human lung fibroblasts were incubated in the DMEM culture medium containing 10% serum of patients with hepatopulmonary syndrome or in the DMEM culture medium containing 10% serum of healthy volunteers.At 24,48 and 72 h of incubation (T1-T3),the expression of smooth muscle-α-actin (SM-α-actin) and smooth muscle myosin heavy chain (SM-MHC) in human lung fibroblasts was determined by Western blot,the proliferation of the human lung fibroblasts was determined using 3H-TDR incorporation assay,and the migration of the human lung fibroblasts was determined by Transwell chamber assay.Results Compared with serum of healthy volunteer group,the expression of SM-α-actin and SM-MHC in human lung fibroblasts was significantly up-regulated at each time point,and the proliferation and migration of the cells were significantly enhanced at T2,3 in serum of patients with hepatopulmonary syndrome group (P<0.05).Compared with the value at T1,the expression of SM-α-actin and SM-MHC in human lung fibroblasts was significantly up-regulated,and the proliferation and migration of the cells were significantly enhanced at T2,3in serum of patients with hepatopulmonary syndrome group (P<0.05).Compared with the value at T2,the expression of SM-α-actin and SM-MHC in human lung fibroblasts was significantly up-regulated,and the proliferation and migration of the cells were significantly enhanced at T3 in serum of patients with hepatopulmonary syndrome group (P<0.05).Conclusion The serum of patients with hepatopulmonary syndrome can promote the myogenic differentiation,proliferation and migration of human lung fibroblasts.

Xiamen Cardiovascular Hospital set up the first chest-pain center in Fujian province,and launched the first citywide regional chest pain network in China.Their experiences prove that the first-aid network can effectively integrate ambulances,primary hospitals,general hospitals,and special hospitals for seamless connection between EMS and in-house rescue.These efforts can minimize the mortality and disabilities of myocardial infarction,effectively elevating the myocardial infarction treatment efficiency and capabilities of the region.
5.Research and practice on basic skill training and technique improving of acupuncture and tuina major.

In order to summarize and perfect the basic skill practicing method in acupuncture teaching and form a characteristic teaching, the research on skill training and technique improving of acupuncture and tuina major was carried out. The undergraduate students in acupuncture and tuina major from College of Acupuncture-Moxibustion and Tuina were randomly divided into an experimental group and a control group. Students from both groups received the basic courses of acupuncture-moxibustion and tuina as requested in the syllabus and the special teaching was added to the experimental group, namely "three practice" (strength practice, coordination practice, targeted practice of acupuncture basic skills) and "three enlightenment" (technique enlightenment, mind and qi enlightenment, efficacy enlightenment). During and after the courses, the teaching effectiveness was evaluated on the basis of needling manipulation. The peacetime score in the experimental group was superior to that in the control group (38.03 +/- 1.14 vs 33.25 +/- 1.31, P < 0.05), the total score in the experimental group was superior to that in the control group (84.03 +/- 6.30 vs 78.05 +/- 6.55, P < 0.05). The special teaching method of "three practice" and "three enlightenment" can improve the effect of skill training teaching method. It highlights the unique feature of central plains by integrating the essence of Shaolin internal qigong and Chen-style Taiji, it can also provide new ideas for the practical teaching of TCM professional courses and the construction of characteristic professionals.

6.Expression of metadherin in retinoblastoma and its relationship with histopathological high-risk factors

Background Metadherin is a newly discovered oncogene.It is highly expressed in some solid tumors and plays a significant role in invasion and metastasis of neoplasm as an important biological marker of aggressive cancers.However,there is little data available for the relationship between metadherin expression and clinicopathologic features of retinoblastoma(RB).Objective This study was to investigate the expression of the metadherin in RB cell lines and specimens as well as its clinical significance.Methods The expression of metadherin mRNA in different metastatic potential of RB cell lines(Y79,WERI-RB1 and SO-RB50)was detected by real-time PCR,and the protein expression of metadherin (MDTH)in paraffin-embedded RB tissue was detected by immunohistochemistry.The relevance of metadherin expression to clinical histopathological features was statistically analyzed.Results The relative expression value of metadherin mRNA was 6.11±0.17,6.21±0.21 and 3.97±0.17 in Y79,SORB50,WERI-RB1,respectively,with a significant difference among the three types of cell lines(F =142.643,P<0.05).The relative expression value of metadherin mRNA was significantly higher in Y79 or SO-RB50 than that in W ERI-RB1 (P=0.000).The expression of MTDH protein mostly located in the cellular membrane and cytoplasm.Among the 54 RB sections,35 (64.81%)showed a high intensity in expression of metadherin protein.The total score of metadherin protein expression was higher in the E stage of RB than that of D stage(P=0.035),in the patients with high-risk factor than those without high-risk factor(P=0.002)and the cases with optic nerve invasion compared to non-invasion (P=0.017).However,no significant differences were found in the expression of metadherin protein between different ages (P =0.579),different genders (P =0.513),bilateral eyes (P =0.305),different disease courses (P=0.860),various types of differentiation (P =0.537),different degree of the ehoroid invasion (P =0.238),and with or without invasion of the anterior ocular segment (P =0.579).Conclusions Metadherin appears to be highly expressed in RB cells.The activation of the MTDH gene is associated with invasion and metastasis of RB.These results imply that the dynamic change of metadherin expression probably is a prognostic indicator of RB.
7.Study on mechanism of apoptosis of HL-60 cells induced by cytarabine.

To study the mechanism of apoptosis of HL-60 cells induced by cytarabine (Ara-C), morphological changes of the HL-60 cells stained with Giemsa were observed by microscopy; the DNA fragments were detected by agarose gel electrophoresis; the expressions of Bcl-2 and bax gene-related apoptosis were investigated by immunohistochemistry. The results showed that HL-60 cells in experimental groups had changed in morphology since they were cultured for 8 hours. HL-60 cells stained with Giemsa showed that their nuclear membranes were splitted. The purplish red chromatins were concentrated into pieces and apoptosis bodies were formed. The apoptosis rate increased in the experimental groups compared with the control group. The expression of Bcl-2 protein was lower and the expression of Bax protein was higher and the ratio of Bcl-2/Bax proteins in the experimental groups was lower than those in the control group. It is concluded that Ara-c can effectively induce apoptosis of HL-60 cells and simultaneously decrease the level of expression of Bcl-2 protein and elevate the level of expression of Bax protein. Decrease of expression ratio of Bcl-2/Bax proteins may be one of the main mechanisms in HL-60 apoptosis induced by Ara-C.

8.Experimental study on anti-tumor effect of xihuang pill and its immune clearance function.

OBJECTIVETo discuss the anti-tumor effect of Xihuang pill on tumor-bearing rats and its effect on the immune clearance function of tumor-bearing organisms.
METHODWalker256 tumor cells were adopted to establish the tumor-bearing rat model. The rats were randomly divided into five groups: the normal control group, the model control group, the lentinan group and Xihuang pill low dose, middle dose and high dose groups, with 10 rats in each group, and continuously treated and given drugs for 14 d after modeling. Blood and tumors were collected from abdominal aorta to calculate the tumor inhibition rate. The content of CD3+, CD4+, CD8+ T cells and adhesion molecule B7-1 (CD80) in peripheral blood were detected by flow cytometry (FCM). The expressions of IL-2 and IFN-gamma in were determined by ELISA.
RESULTThe tumor inhibition rate of the Xihuang pill high dose group was 33. 1 percent. Compared with the model group, the Xihuang pill large dose group showed significantly low IL-2, IFN-gamma, CD3+, CD4+, B7-1 in peripheral blood, with statistical significance in their differences (P < 0.05).
CONCLUSIONXihuang pill could show its anti-tumor effect by enhancing the immune clearance function and increasing IL-2, IFN-gamma, CD3+ T, CD4+ T, B7-1 in peripheral blood.

9.Effect of Acupuncture and Speech Rehabilitation on Motor Aphasia after Stroke

ObjectiveTo compare the effect of acupuncture and speech rehabilitation on motor aphasia after stroke with speech rehabilitation alone. Methods49 patients with aphasia were divided into treatment group (27 cases) and control group (22 cases) randomly. The cases in treatment group were treated with acupuncture and speech rehabilitation. The cases in control group were treated with speech rehabilitation alone. They were assessed before and 1 month, 3 months after treatment. ResultsThere was no significant difference between treatment group and control group 1 month after treatment (P>0.05), but was significant in auditory comprehension, speaking, reading, dictation and description (P<0.05) 3 months after treatment. ConclusionThe acupuncture can facilitate the recovery of motor aphas after stroke.
10.Comparative proteomic analysis in left ventricular remodeling following myocardial infarction in rats.

OBJECTIVELeft ventricular remodeling (LVR) following myocardial infarction (MI) is a key pathophysiological process in which MI develops into heart failure. The exact mechanism of LVR remains unclear. We performed differential proteomic analysis on the myocardia of rats with LVR after MI, to explore the mechanism of ventricular remodeling after MI.
METHODSIn the LVR group (n = 12), after the anterior descending coronary artery was ligated, the rats were fed for four weeks before the LVR models were established. Rats in the sham-operated group (n = 11) underwent thread-drawing without ligation. The hemodynamic parameters, pathological findings, and proteomics were compared between the two groups.
RESULTSIn the LVR group, the left ventricular end-diastolic pressure increased, the maximal left ventricular pressure increase/decrease ratio decreased significantly, and the left ventricular systolic pressure decreased. H-E staining and Masson staining of cardiac muscle tissues of the LVR group showed myocytolysis, disarray, and collagen proliferation. Twenty-one differentially expressed proteins were detected by proteomic analysis. We validated two proteins using western blot analysis. The differentially expressed proteins could be divided into six categories: energy metabolism-related proteins, cytoskeletal proteins, protein synthesis-related proteins, channel proteins, anti-oxidation- related proteins, and immune-related proteins.
CONCLUSIONThese differentially expressed proteins might play key roles in LVR following MI.
