1.Advances on the study of the pharmacological effects of artemisinin and its derivatives

Artemisinin is a sesquiterpene lactone compounds,their derivatives include mainly dihydroartemisinin,artesunate,artemether and Artemisia ether. These drugs are now mainly used for clinical treatment of malaria. With the pharmacological action of artemisinin and its derivatives have been further studied,except antimalarial effects,researchers have found their anti-inflammatory,anti-sepsis,antitumor,radiosensitization,antimicrobial sensitivity,anti-fibrosis effects in recent years. Here,we collect and review the most newly pharmacological effects of artemisinin and its derivatives all over the world in past years.
2.Intraosseous pressure and blood flow of the distal femoral shiaft after blocking the proximal and middle femoral medullary cavity

3.Experimental study of the effect on the the distal femoral joint by the high intraosseous pressure induced with blocking of the proximal and middle femoral medullary cavity with bone cement

[Objective]To explore the effects on the distal femoral joint by high intraosseous pressure after blocking of proximal and middle femoral medullary cavity with bone cement.[Method]The rabbit model was established whose proximal and middle femoral medullary cavity of left side was infused with PMMA while the right side served as the control.Twenty-six rabbits were cavided randomly into 4 groups,8 for normal control group,the remained modeled rabbits were grouped according to the observing time-point into following groups:4th(T4),8th(T8) and 16th week(T16).The rabbits was killed to obtain the samples of distal femoral articular synovium,cartilage and the bone under cartilage.Then the structural changes of these tissues were observed by HE staining,toluidine blue staining,immunohistochemistry and transmission electron microscope.[Result](1)HE staining:the damage to articular cartilage,synovial tissue and the bone under cartilage aggravated with time.At the 16th week,it showed the articular cartilage was destructed,the bone structure was severely damaged with the synovial tissue proliferation and swelling.(2)Toluidine blue staining(16th week):all the layers of articular cartilage appeared loss of stain while the superficial layers were slightly loss of stain in control group.(3)Immunohistochemistry(16th week):①CollagenⅡ:the chondrocytes were positively stainned.②TGF-?1:chondrocytes stain was positive;the staining of articular synoviocytes appeared positive.(4)Transmission electron microscope:The lesions to articular chondrocytes and bone cells aggravated with time,part of which presented with necrosis and collapse at the 16th week.[Conclusion]The distal femoral intraosseous and intramedullary blood circulation is severely damaged after blocking of the proximal and middle femoral medullary cavity with bone cement,leading to continuous high intraosseous pressure.It makes the bone metabolism disordered in distal femur,which results degeneration and necrosis in distal femur,cartilage and synovial tissue.
4.Determination of the Concentration of 5-Fluorouracil in Human Plasma With HPLC

OBJECTIVE:To establish a HPLC method for the determination of 5-fluorouracil in human plasma METHODS:The plasma samples were extracted with ethyl acetate-isopropanol(85∶15,V/V)following precipitation of plasma proteins with ammonium sulfate The solvent was evaporated to dryness under a stream of nitrogen The dry extract was dissolved in mobile phase and then injected into the Diamonsil C18 column(150mm?4 6mm,5?m)and 0 01mol/L KH2 PO4(pH5 5) was taken as mobile phase The flow rate was 1 5ml/min,UV detection wavelength was 267nm The internal standard was 5-bromouracil RESULTS:The minimal detectable drug concentration in plasma was 0 025?g/ml The calibration curve was linear over a range from 0 3?g/ml to 10?g/ml The intra-day RSD≤5 0%(n=5) and the inter-day RSD≤11 5%(n=5) CONCLUSION:The method can be used for studying the pharmacokinetics and bioavailability of 5-fluorouracil
5.Analysis on Learning Curve of Laparoscopic Pancreaticojejunostomy in Pigs

Objective To evaluate the surgical effects of laparoscopic pancreaticojejunostomy in pigs and investigate the learning curve. Methods A total of 40 porcine cases of laparoscopic pancreaticojejunostomy were performed by a single surgical team from September 2012 to February 2013.According to the operation sequence, pigs were divided into 4 groups with 10 in each group.For analyzing the efficacy of surgery in different stages, the operation time and efficacy of anastomoses were compared between these groups. Results The operation was finished successfully in all the 40 cases.There were statistical significances in operation time among the four groups.The group Ⅳ had significant shorter time (112.7 ±8.3) min than those in the group Ⅰ [(145.1 ± 16.6) min, q=9.154, P<0.05], the groupⅡ[(133.0 ±8.7) min, q=5.735, P<0.05], and the group Ⅲ[(137.2 ±9.0) min, q=6.922, P<0.05], respectively.There was no difference between the group Ⅱ and the group Ⅰ in operation time (q=3.419, P>0.05), so as to other groups.The results of anastomoses among the four groups were not significantly different (χ2 =0.586,P=0.900). Conclusions Laparoscopic pancreaticojejunostomy in pigs is feasible.The learning curve of the operation is about 30 cases.
6.Application of enhanced CT-guided percutaneously transthoracic lung biopsy

Objective To investigate the clinic value of enhanced CT-guided percutaneously transthoracic lung biopsy. Methods A total of 423 patients who underwent CT-guided percutaneously transthoracic lung biopsy were retrospectively analyzed. The percentage of specimen are satisfactory,the rate of sample with pathological findings,the number of cases with complications,the average goes of unsuccessful biopsy and the times of changing puncture point were compared between the enhanced CT group and the plain CT group. Results Compare parameters in patients whose procedure were guided with the enhanced CT group and with plain CT group, the average goes of unsuccessful biopsy, the times of changing puncture point in patients, the percentage of specimen that were satisfactory, the rate of sample with positive pathological findings and the rate that complicated with pneumothorax or hemorrhage show statistical significance.(3.08±1.05 vs 3.75±1.34, 8.52%vs 18.54%,90.13%vs 73.60%, 76.62%vs 63.36%, 15.51%vs 27.53%, 7.76%vs 14.04%, P<0.05). Conclusion In patient whose biopsys were guided by enhanced CT, the percentage of samples that are satisfactory is higher, the incidence of complication is fewer and the sample with pathological finding is higher compared to those in patients whose biopsys were guided by plain CT. Therefore, enhanced CT-guided percutaneous transthoracic biopsy is more valuable than plane CT.
7.The adhesive characters of Lactobacillus to enterocyte-like HT-29 cells

Objective To study the adhesive characters of Lactobacillus to enterocyte-like HT-29 cells.Methods A HT-29 cell culture was employed to investigate the adherent ability of Lactobacillus.Bacterial suspension and HT-29 cells were co-incubated at 37℃ for 1h.The cells with adherent bacteria were Gram-stained,and then examined microscopically under oil immersion.The pH of the medium,bacterial growth phase,co-incubation time,bacterial concentration,D-mannose and type of host cells,which were involved in the process of adherence of Lactobacillus to enterocyte-like HT-29 cells,were also investigated.Results The adherent level increased substantially for Lactobacillus reuteri JCM1081 as the pH decreased from 6.0 to 4.0.Maximum adherence occurred within 90min and remained stable until the end of incubation.The bacterial concentration exerted a certain influence on the adherence of Lactobacillus reuteri JCM1081 to HT-29 cells,and it seemed to be concentration dependent.Maximum adherence occurred in the concentration of 109cfu/ml and remained stable until the end of incubation.Lactobacillus strain that grew into stationary phase showed higher ability to adhere to HT-29 cells than that in logarithmic phase.The addition of D-mannose dramatically decreased the level of Lactobacillus adherence.The Lactobacillus strain adhered in higher levels to the enterocyte-like cells or gastric epithelium cells than other cell origins,and the adhesion of Lactobacillus showed host cells specificity.Conclusion The adhesive characters observed with Lactobacillus suggested that adhesive properties are species and host specific.The pH,bacterial growth phase,co-incubation time,bacterial concentration and D-mannose may influence the adhesion of Lactobacillus to enterocyte-like HT-29 cells.
8.Study on the short -term effect of entecavir in the treatment of 50 patients with decompensated liver cirrhosis

Objective To observe the short -term effect and safety of entecavir in the treatment of 50 patients with decompensated liver cirrhosis.Methods 92 patients with decompensated liver cirrhosis met the inclusion criteria in our hospital were selected as study subjects.The patients were randomly divided into the observation group (46 cases)and the control group(46 cases).The control group received routine treatment,including the liver,liver, diuretic,jaundice,symptomatic and supportive treatment and prevention of complications.The observation group was given entecavir for 48 weeks on the basis of the control group,0.5 mg /times,qd.The clinical effects were assessed after treatment.Results The HBV -DNA,ALT,TBIL,Child -Pugh score of the observation group and the control group after treatment were (2.05 ±0.32)log10 IU /mL and (3.11 ±0.36)log10 IU /mL,(32.1 ±8.5)U /L and (90.3 ±12.6)U /L,(27.8 ±7.1 )μmol/L and (35.3 ±8.2)μmol/L,(6.42 ±1.24)points and (9.20 ± 1.46)points,which were significantly lower than before treatment (t =6.563 and 5.341,11.266 and 7.044,7.346 and 6.013,6.252 and 4.423,all P <0.05).The ALB levels were (38.5 ±2.9)g/L and (34.4 ±3.0)g/L,which were significantly higher than before treatment(t =4.812 and 3.701,all P <0.05).The HBV -DNA,ALT,TBIL, Child -Pugh score of the observation group after treatment were significantly lower than the control group (t =3.335, 3.431,3.411,3.204,all P <0.05).The ALB of the observation group after treatment was significantly higher than the control group (t =3.034,P <0.05).There were two cases of deaths(4.45%)in the observation group,8 cases of deaths(17.39%) in the control group because of progressive deterioration,there was significant difference in treatment outcome between the two groups(χ2 =4.039,P <0.05).Conclusion Entecavir can delay progression, effectively reducing HBV -DNA,improve liver function and Child -Pugh score in the treatment of patients with decompensated liver cirrhosis,it can be used as first -line drug use.
9.Influence of Insoine on Expressions of Vascular Endothelial Growth Factor and Cyclooxygenase-2 in Newborn Rat with Hypoxic-Ischemic Encephalopathy

Objective To observe the expressions of vascular endothelial growth factor(VEGF) and cyclooxygenase-2(COX-2) in the cerebral tissue following hypoxic-ischemic encephalopathy(HIE) in newborn rats and explore the machanism of inosine in protecting against hypoxic-ischemic brain damage(HIBD).Methods Sixty-six newborn rats aged 7 days were divided into sham,control and experimental groups.HIE models were made by clamping the right cervical artery and making hypoxia for 2 hours.The rats in experimental group began injecting inosine,at 1 day before experiment,and the rats in the sham and control groups saline solution with same dose.The samples were made at the given time,and expressions of VEGF and COX-2 were investigated by immunohistochemical technique.Results The cerebral tissue had no expression of VEGF and COX-2 in sham group.From 2 hours on cortex and striatum after HIE in control and experimental groups,expressions of VEGF and COX-2 increased rapidly,peaking at 12-24 hours,and then decreased gradually.Expressions of VEGF and COX-2 were higher in experimental group(All P
