1.EXPERIMENTAL STUDY FOR THE EFFECT OF TSPG ON THE EXPRESSION OF IL-3 IN HEMATOPOIETIC STROMAL CELLS

Objective Panax ginseng is a well-known Chinese traditional medicine functional in “invigorating qi”,and saponins are one of its main effective fractions.Our study investigates the effect of total saponins of panax ginseng(TSPG)on human hematopoiesis at early stage and its possible regulative mechanism to clarify the hematonic mechanism of Panax ginseng. Methods The techniques of culturing hematopoietic progenitor cells in vitro,bioassay of hematopoietic growth factor(HGF),immunocytochemistry and nucleic acid in situ hybridization were used to study the effect of TSPG on IL-3’s expression in hematopoietic stromal cell and its possible mechanism. Results TSPG directly added into culture system in vitro can markedly increase the colony forming yields of CFU-Mix;the different conditioned culture media prepared with TSPG can promote the proliferation and differentiation of CFU-Mix;the protein and mRNA expression of IL-3 in BMSC and cells of EcV304,THP induced by TSPG has been much intensified.Conclusion TSPG may regulate hematopoiesis at early stage by activating IL-3's gene expression in hematopoietic stromal cell,which is the most important element in hematopietic inductive microenvironment(HIM).

3.Analysis of rDNA ITS sequences of Radix et Rhizoma Salviae Miltiorrhizae and plants of Salvia L.

Objective To study the genetic diversity of ITS sequences of Radix et Rhizoma Salviae Miltiorrhizae (RRSM) and plants of Salvia L. and analyze the utility of ITS sequences in molecular authentication of RRSM and phylogenetic of plants of Salvia L. Methods The ITS gene fragment was amplified using a pair of primers. The PCR products were purified and sequenced by the methods of Sanger dideoxy. Results The DNA sequence of 222-224 bp ITS1, 220-223 bp ITS2 gene fragments and 5.8S rDNA were obtained from 13 samples of Salvia L. The interspecific substitution varied from 0% to 12.16% at ITS1 and 0.5% to 13.51% at ITS2. The intraspecific substitution of RRSM is 0% to 0.9% at ITS1 and 0% to 0.5% at ITS2. The substitution between group of Salvia L. and outgroup varied from 16.07% to 20.27% at ITS1 and 15.91% to 20.27% at ITS2. The phylogenetic tree based on ITS1 and ITS2 data was set up. Conclusion The ITS1 and ITS2 gene fragments are highly conservative at intraspecific level in RRSM, while they are less conservative at interspecific level in Salvia L. They are least conservative between group of Salvia L. and outgroup. Hence, the sequence of this fragment is a good molecular marker for authentication of RRSM and can be used for further study on the phylogenesis for plants of Salvia L.
4.CD44 MEDIATES EFFECTS OF HYALURONAN ON PHAGOCYTOSIS OF MACROPHAGES

Objective To investigate the effects of hyaluronan(HA) on phagocytosis of macrophages and to explore the mechanism.Methods CD44 of macrophages was labeled with immunochemical method.In phagokinetic track assay,the areas without gold particles around the cells were measured with an image analyzer,shape of the cells was viewed under a scanning electron microscope.In fluorescent bead phagocytosis assay,the phagocytosed beads were conunted,the distribution of intracellular F-actin was viewed.In HA-coated bead phagocytosis assay,the phagocytosed beads were counted,shape of the cells was viewed with scanning electron microscope,the level of the intracellular Ca~(2+) was analyzed with confocal laser scanning microscope when the beads were phagocytosed.After treatment with anti-CD44 antibody,the changes of phagocytosis of the cells were examined.Results CD44 expressed on macrophages.The phagocytosing cells were round,their lamellipodia and filopodia were short.The membrane of the cells formed cup-like invagination when they engulfed particles and beads.In HA group,the phagocytosed gold particles and fluorescent beads increased.There was rich F-actin in the area where the fluorescent bead was engulfed.In HA-coated group,number of the phagocytosed HA-coated beads was great,the level of the intracellular Ca~(2+) in the area where the bead was engulfed increased significantly.After treatment with anti-CD44 antibody,the phagocytosed gold particles,fluorescent beads and HA-coated beads decreased.Conclusion HA promotes phagocytosis of macrophages,CD44 mediates effects of HA on phagocytosis of the cells.
5.STUDY ON THE SIGNAL TRANSDUCTION AND REGULATION IN ERYTHROPOIESIS BY TOTAL SAPONINS OF PANAX GINSENG

Objective To investgate the signal transduction and regulation in erythropoiesis by total saponins of Panax ginseng(TSPG) to clarify the mechanism for Panax Ginseng promoting hematopoiesis.Methods The colony-forming ability of mononuclear cells(MNCs) from human umbilical cord blood was assayed in methylcellulose assay systems,various dilutions of TSPG(0,10-100mg/L) were then added to each methylcellulose culture mixture;MNC proliferation in response to erythropoietin(Epo) was measured by MTT;To investigate whether the preferential differentiation of MNCs into erythroid lineage cells was caused by an increase in the expression of erythropoietin receptor(EpoR) on the cells,immunoblotting analyses were performed;Immunoprecipitation of JAK_2/STAT_5 was used for the effect of TSPG on Epo/EpoR-induced tyrosine phosphorylation of JAK_2/STAT_5. Results 1.TSPG(10-75mg/L) can promote the colony formation of BFU-E,CFU-E,and the preferential differentiation into erythroid lineage cells was induced from 25mg/L of TSPG;2.Using the colorimetric MTT assay,MNCs exhibited proliferous responses to Epo(2?10~3-5?10~4IU/L) reaching maximum at 5?10~3IU/L Epo;3.The addition of TSPG did not increase the expression of EpoR;4.After MNCs were incubated in the presence of with or without TSPG for 24 h,the cells were stimulated with Epo for 0,2,5,30min,and tyrosine phosphorylation JAK_2/STAT_5 was measured.The pretreatment with TSPG enhanced Epo-induced tyrosine phosphorylation of JAK_2 and STAT_5(STAT_(5a) and STAT_(5b)).Conclusion In the present study,we have shown that the Epo/EpoR-mediated signals are enhanced,and JAK_2/STAT_5 signals plays an essential role in the effect of TSPG on erythropoiesis,whereas the expression level of EpoR remains unchanged.
6.Comparison of the quality of life before and after treatment of eczema in infants with or without food allergies

Objective To explore life quality of infants with eczema and their families before and after treatment. Methods The infants with eczema were divided into food allergic group (FA group,n=46) and non-food allergic group (N-FA group,n=47). The eczema was evaluated by the areas of eczema and severity index (EASI). After 2 months of standardized intervention, the quality of life before and after treatment were assessed by the Food Allergy Quality of Life Questionnaire-Parent Form (FAQLQ-PF) and the Infant’s Dermatitis Quality of Life Index (IDQOL).Results There was no difference between the two groups in age, gender, family history of allergy and feeding patterns (allP?>?0.05). In FA group, there were 34 cases (73.91%) of egg allergy, 20 cases (43.48%) of milk allergy and 2 cases (4.35%) of carrot allergy. After the intervention, the scores of EASI, FAQLQ, and IDQOL were decreased obviously in the two groups, and there were statistical difference before and after treatment (P all< 0.05). Before treatment, the total score and each part of FAQLQ-PF in FA group was similar with N-FA group (P all?>?0.05); After two months of intervention, the total score of FAQLQ in FA group was higher than that in N-FA group (Z =2.83,P=0.005); the subscales of emotional impact, anxiety about food, and social/dietary restrictions were also signiifcantly different between FA group and N-FA group (Z=?2.13-2.89, allP< 0.05). Conclusions Parents of infants with food allergies are more likely to worry about infant's emotional changes and social, dietary restrictions. FAQLQ-PF is more speciifc for assessing the quality of life in food allergies.
7.Effect of Naoxintong Capsule on the Myocardial Perfusion and Platelet Activation Function in Patients with Acute Myocardial Infarction after PCI

OBJECTIVE:To explore the effect of Buchang naoxintong capsule on the myocardial perfusion and platelet activation function in patients with acute myocardial infarction (AMI) after PCI. METHODS:100 AMI patients were randomly divided into control group and observation group by parallel random sampling,50 cases in each group. Control group was given conventional PCI, preoperatively chewing 300 mg aspirin+300 mg clopidogrel,postoperatively taking 100 mg/d aspirin+75 mg/d clopidogrel. Observation group was additionally treated 2-4 with Buchang naoxintong capsules(after bracket release),3 times a day,the treatment course for both groups was 7 d,observation period was 1 month. Infarct-related artery myocardial perfusion grade(TMPG)in 2 groups,platelet activation function indexes [CD62P,CD63 and monocyte platelet aggregation (MPA) positive expression rates] before and after treatment,the incidences of adverse cardiac events (MACE) and complications were observed and compared 15 min after PCI. Meanwhile,the incidence of adverse reactions was recorded. RESULTS:After treatment,the cases of TMPG perfusion grade 3 in observation group were significantly higher than control group,the incidence of MACE was significantly lower than control group, with statistical significance(P<0.05);there was no significant difference in the incidences of complications and adverse reactions in 2 groups(P>0.05). Before treatment,there was no obvious difference in CD62P,CD63,MPA positive expression rates in 2 groups;after treatment,CD62P,CD63,MPA positive expression rates in 2 group were significantly lower than before,and observation group was lower than control group,with significant differences(P<0.05). CONCLUSIONS:In PCI treatment of AMI patients,Buchang naoxintong capsule can rapidly improve platelet activation functions,TMPG perfusion,reduce the incidence of MACE,with good safety and no increase of complications.
8.Perspectives of Japanese Clinical Research Coordinator Industry Development and Experience Reference

OBJECTIVE:To provide reference of Clinical Research Coordinator(CRC)industry development in China. METH-ODS:By using the method of literature research combining with field survey research,the development status,training certifica-tion and working mode of CRC industry in Japan were summarized,and suggestions on the development of the CRC industry in China were proposed. RESULTS:Japanese CRC industry gradually matured,and staff tended to be stable,the standardized train-ing and certification system have been established,the services include the support on medical institutions,clinical trials,clinical researches and other multiple aspects. CRC industry in China should perfect the company system construction,carry out standard-ized training and certification examination, strengthen the hospitals' direct management of CRC, and expand the CRC services in clinical research. CONCLUSIONS:The experience of CRC industry development in Japan,including system construction,stan-dardized training and working mode,is a good example for China CRC industry to learn.
9.The role of cystine/glutamate antiporter on the pathogenesis of glioma and the relative applications of anesthetics

The incidence of glioma has been increasing in recent years,and threatened the human health seriously.Cystine/glutamate antiporter (system xc-) is one of the important glutamate transporters in the central nervous system,both the expression of system xc and the extracellular glutamate concentration increase in the course of brain tumor's development.Hence system xc-plays an essential role in the process of brain tumor genesis,it has become an important research field in brain tumor in the past decades.Our present review expound the important role of system xc-in the process of glioma genesis and development as well as provide theoretical foundation and new strategies for the clinical treatment and the application of drugs by connecting with anaesthetics and the effects of system xc on glioma.
