Objective:To study e ects of Saikosaponin a(SSa) on tumour necrosis factor-alpha(TNF-?) release and its receptor expression in cultured hippocampal astrocytes induced by pentylenetetrazol(PTZ).Methods:The in vitro cultured primary hippocampal astrocytes were randomly divided into control group(group A),PTZ-induced group(group B)PTZ10mmol/L+SSa groups(group C and group D,the SSa concentrations were 1.25mg/L and 0.625mg/L respectively).The extracellular uid TNF-? level and the expression of tumour necrosis factor receptor type 1(TNFR1) in hippocampal astrocytes were respectively detected by ELISA and Western-blot after PTZ-induced 2 hours.Results:the TNF-? level and TNFR1 expression of group B were signi cantly higher than that of group A,group C and group D(P

Evaluation of the outcomes of completed projects funded by National Natural Science Foundation of China (NSFC) is a key process of funding management which is of great importance for management improvement and promoting transformation of scientific or technological achievements.Therefore,we evaluated all the projects completed at the end of 2016 funded by NSFC in the field of preventive medicine.The research achievements of selected projects were evaluated,as well as the problems.We hope our work could provide reference and datum for making discipline development strategy in the future.

Objective To analyze projects completed at the end of 2017 and funded by National Natural Science Foundation of China (NSFC) in the field of skin and its appendages,in order to provide references for preparing concluding reports and making discipline development strategies in the future.Methods The concluding reports of projects completed at the end of 2017 and funded by National Natural Science Foundation of China in the field of skin and its appendages were collected from the Internet-based Science Information System.The achievements were analyzed,including journal articles,patents,talent training and monographs.Results A total of 91 projects were completed at the end of 2017 in the field of skin and its appendages,and the total cost of funds was 43.51 million RMB Yuan.A total of 452 journal articles were published,including 324 articles in the Science Citation Index (SCI)-indexed journals and 42 in the A Guide to the Core Journal of China-indexed journals.Additionally,0.74 SCI journal article and 0.10 Chinese core journal article could be produced per 100 000 RMB Yuan.The average impact factor of journals publishing these SCI articles was 3.972,and in average,100 000 RMB Yuan yielded 1 article in the journal with an impact factor of 2.92.Of the 324 published articles,46 and 102 were published in journals ranked within the first (Q1) and second (Q2) quartile,respectively,by Chinese Academy of Sciences.In all the SCI articles,14 were published in journals with impact factor of ＞ 10.There were 4 institutions with more than 5 completed projects,and a total of 129 (39.84％) SCI articles were published.Published SCI articles in the field of immune skin diseases accounted for the highest proportion (148/324,45.68％).A total of 152 talents were trained.Conclusions The support of NSFC in basic researches in the field of skin and its appendages has led to substantial achievements.More attention should be paid to the writing of concluding reports.Innovation consciousness should be raised,and project leaders should make more original and leading achievements in the future.

Objective To understand the incidence and mortality of malignancies as well as tumor burden of Shouyang County, Shanxi Province in 2012. Methods According to the review methods and the standards from the National Cancer Registry, the data of the incidence and mortality of malignant tumors in Shouyang County in 2012 were collected, collated and statistically analyzed. Results There were 389 new malignancies cases of Shouyang County in 2012, including 210 males and 179 females. The incidence rate was 187.61/100000 (190.00/100000 in males and 184.88/100000 in females). There were 263 death cases , including 164 males and 99 females. The mortality rate was 126.84/100000 (148.38/100000 in males and 102.25/100000 in females). The top 10 incidence of malignancies of the whole county was followed by lung cancer, cervical cancer, gastric cancer, liver cancer, colorectal anal cancer, esophageal cancer, breast cancer, bladder cancer, gallbladder cancer and uterus cancer, accounting for 86.12 ％ of the overall malignant cancers. The top 10 death malignancies of the whole county was followed by lung cancer, liver cancer, gastric cancer, esophageal cancer, Hodgkin disease, leukemia, colorectal cancer, bone cancer, brain tumor and pancreatic cancer, accounting for 87.45 ％ of the overall malignant cancers. Conclusion Lung cancer ranks first in the incidence and mortality of malignant tumors of Shouyang County, Shanxi Province, and the prevention and control of major tumors should be strengthened.

We analyzed the project results of preventive medicine from the National Natural Science Foundation of China (NSFC) finished in 2017 based on the project-ending reports and data on science fund sharing service network. A total of 406 projects in this field were completed in 2017. A total of 3 122 published articles supported by these projects, including 1 789 articles in science citation index (SCI) journals and 525 articles in Chinese core journals. In addition, there were 224 patent application/software copyright and 589 trained postgraduates. The top three sub-disciplines of project were non-communicable disease epidemiology, human nutrition and hygienic toxicology, accounting for 45.32% of the total number of completed projects. There were 12 institutions which had more than 10 finished projects, accounting for 41.87%. During the recent 5 years, the number of SCI articles and patents/software copyrights per project showed a general uptrend. It should be noted that the number of articles in Chinese core journals and postgraduates decreased in recent two years. Our analyses demonstrated that the project results should be guided by the new era policy of science fund to promote sustainable development of scientific research.

OBJECTIVE： To investigate the protective effects of anemarsaponin B on hypoxia/reoxygenation injury astrocytes and its possible mechanism. METHODS： The primary astrocytes of neonatal SD rats were cultured and identified， and then randomly divided into normal group， model group， positive control group （nimodipine， 10 μmol/L）， anemarsaponin B low-dose， medium-dose and high-dose groups （1， 10， 100 μmol/L）， respectively. Normal group and model group were given complete medium 1 000 μL. Administration group was given complete medium with relevant medicine 1 000 μL. Except for normal group， hypoxia/reoxygenation injury model was established by oxygen-glucose deprivation/reperfusion in other groups. After reoxygenation， relative release rate of lactate dehydrogenase （LDH） in cell was detected by colorimetry. MTT assay was used to detect the relative viability of the cells. The contents of aquaporin 4 （AQP-4）， IL-6， IL-1β and TNF-α in cell were measured by ELISA. RESULTS： Compared with normal group， relative release rate of LDH， the contents of AQP-4， IL-6， IL-1β and TNF-α in cell were increased significantly in model group， while relative viability of the cells were decreased significantly （P＜0.01）. Compared with model group， relative release rate of LDH， the contents of AQP-4， IL-6， IL-1β and TNF-α in cell were decreased significantly in administration groups， while relative viability of the cells were increased significantly （P＜0.05 or P＜0.01）. CONCLUSIONS： Anemarsaponin B can significantly decrease cell injury degree， strengthen cell viability and protect hypoxia/reoxygenation injury astrocytes to certain extent. The effect may be related to the down-regulation of the secretion of AQP-4， IL-6， IL-1β and TNF-α.

AIM: To investigate the guiding role of individualized medication adjustment based on CYP2C19 metabolic typing in the treatment of ischemic stroke with clopidogrel, and to provide reference for clinical individualized medication. METHODS: The total of 80 patients with ischemic stroke were divided into the individualized drug instruction group with gene detection (n=40) and the control group without gene detection (n=40) according to whether they received CYP2C19 gene detection. According to the metabolism of CYP2C19, the individualized medication instruction group was divided into slow metabolic type, intermediate metabolic type, fast metabolic type and ultra-fast metabolic type. Patients with fast and ultra-fast metabolites were given clopidogrel dose of 75 mg once a day. Patients with intermediate metabolic type were given double clopidogrel dose of 150 mg once a day. Patients with slow metabolism were given tigrillo dose of 90 mg twice a day or aspirin dose of 100 mg once a day. The control group received 75 mg clopidogrel once a day. All patients enrolled in the groups were followed up for 3 months by outpatients or telephone. The incidence of vascular events and mRS scale scores were compared between the two groups. RESULTS: The incidence of vascular events in the individualized drug instruction group was significantly lower than that in the control group, and the incidence of mRS score(0-1) was significantly higher than that in the control group, with statistically significant differences (P<0.05). CONCLUSION: The individualized medication for patients with ischemic stroke by CYP2C19 gene detection can significantly reduce the incidence of adverse vascular events and improve the prognosis and living ability of patients.
