1.Angiotensin II-transient receptor potential channel C6 signaling pathway mediates podocyte injury

BACKGROUND:Transient receptor potential channel C6 (TRPC6) is a new and important slit diaphragm-associated protein in podocytes involved in regulating glomerular filter function. Glomerular TRPC6 expression is closely associated with proteinuria in diabetic kidney disease. OBJECTIVE: To investigate the expression of canonical TRPC6 in mouse podocytes induced by high glucose, and to explore the possible mechanism of diabetic kidney disease. METHODS:Mouse podocyte cels were cultured and divided into normal glucose group (5.6 mmol/L D-glucose), normal control group (5.6 mmol/L D-glucose+25 mmol/L mannitol) and experimental groups which were in the environment of high glucose (30 mmol/L). The experimental groups included high glucose group, valsartan treatment groups (10-5 mol/L) and U73122 control group (10μmol/L U73122). After 48 hours, the expressions of mRNA and proteins of TRPC6, nephrin and angiotensin II (AngII) were detected respectively by real-time quantitative PCR and western blot analysis. RESULTS AND CONCLUSION:Compared with the normal control group, the expressions of mRNA and proteins of TRPC6 and angiotensin II were markedly elevated in the high glucose group (P < 0.01), while the expressions of mRNA and proteins of nephrin were decreased (P < 0.01). The mRNA and proteins of TRPC6 and angiotensin II expressions were significantly down-regulated by valsartan (P < 0.05,P < 0.01), while the mRNA and protein expressions of nephrin were effectively up-regulated (P < 0.05). Compared with the high glucose group, the expressions of mRNA and proteins of TRPC6 and angiotensin II were ameliorated in the U73122 control group. The expressions of mRNA and proteins of TRPC6, nephrin and angiotensin II had no statistical significance between the normal control group and normal glucose group (P > 0.05). Angiotensin II-TRPC6 signaling pathway may mediate high glucose-induced podocyte injury, meanwhile it provides a new theoretical basis for the treatment of diabetic kidney disease, by which the angiotensin receptor blockers can protect podocytes in diabetic kidney disease.
2.Polymorphisms of the high-affinity receptor for IgE alpha-chain (FcεRIα) gene in patients with atopic dermatitis

Objective To assess the relationship between single nucleotide polymorphisms of FcεRIαgene and atopic dermatitis. Methods Genomic DNA samples were extracted from peripheral blood of 97 patients with atopic dermatitis and 283 normal human controls. The polymorphism at the distal promoter region of FcεRIα gene was determined by PCR-ligase detection reaction assay followed by gene sequencing. Results A G/T polymorphism was observed at position rs61828219 in the promoter region of FcεRIα gene, while all the tested individuals were homozygous for T/T at position rs12135235 and A/A at position rs36233780 in the promoter region of FcεRIα gene. The mutation frequency at position rs61828219 was 1.04% and 2.17% in patients with AD and normal human controls, respectively (both P > 0.05). Conclusions In the Chinese Han population, there is a G/T polymorphism at position rs61828219 in FcεRIα gene promoter region, which is unlikely related to the development of AD; however, no polymorphism is detected at position rs12135235 or rs36233780.
3.Correlation of apoptosis with androgen receptor in prostate cancer

Purpose To investigate relationship between androgen receptor and apoptotic index and to further understand the tumor biology of prostate cancer. Methods Fifty-six patients with histologically proven prostate cancer and 20 cases with benign prostatic hyperplasia(BPH) were collected. Androgen receptor(AR) were stained by 2H12 monoclonal antibody using immunohistochemical method. The apoptotic index (AI) was determined by the terminal deoxynucleotidyl transterase-mediated dUTP biotin nick end labeling (TUNEL) technique on serial sections of formalin fixed, paraffin embedded tissues. Results AR was not found significantly difference (P>0.05) between prostate cancer and BPH. The association of AR with AI according to Gleason score were not observed in prostate cancer. AI were significantly higher in prostate cancer compared to BPH (P<0.05).AI were also significantly higher in AR-positive prostate cancer than in AR-negative prostate cancer(P<0.05). Conclusion AR expression can induce prostate cancer cells to become apoptosis, and may be a useful predictor in functional classification and endocrine response of prostate cancer.
4.The relationship between interleukin-18 and podocyte injury in patients with lupus nephritis

Objective To investigate the relationship between interleukin (IL)-18 and podocyte injury of lupus nephritis (LN). Methods Sixty cases of biopsy proven LN patients were enrolled into the study. Thirty cases were selected as controls. The clinical and pathological data, blood and urine samples and renal tissues were collected. The Nephrin expression was detected by immunohistochemical method and IL-18 was measured by enzyme linked immunosorbent assay. The relationship between IL-18 and the Nephrin expression, clinical and pathological indicators of LN were analyzed. Results Thirty-eight cases were in active disease and 22 cases were in inactive disease in LN group according to SLE disease activity index (SLEDAI) 2000. One-way ANOVA showed that the level of plasma and urine IL-18 in the LN groups were higher than those in the control group [(200±38) ng/ml, (18±5) ng/ml] (F=110.84, 203.09, P<0.01). Plasma and urine IL-18 level in active LN group [(565 ±128) ng/ml, (200 ±47) ng/ml] was higher than that in the inactive group [(376 ±106) ng/ml, (67 ±22) ng/ml] (P<0.01), the level of IL-18 of type Ⅳ LN was significantly higher than that of typeⅢand type Ⅴ LN (P<0.01). The expression level of Nephrin in LN groups were lower than those in healthy control group (0.28±0.02)(F=136.39, P<0.01). The expression level of Nephrin in active LN group (0.13±0.03) was lower than that in the inactive group (0.18±0.02) (P<0.01), the level of typeⅣLN Nephrin was significantly 01). The Pearson correlation analysis showed that, compared with plasma IL-18, urine IL-18 level in the LN group was not only negatively correlated with the level of Nephrin and serum C3 (r=-0.780, -0.565, P<0.05), but positively correlated with 24 h UP, erythrocyte sedimentation rate (ESR), SLEDAI and GAI (r=0.546, 0.467, 0.599, 0.634, P<0.05). Serum IL-8 level was independent of albumin (ALB), C4, C reactive protein (CRP) and CI (P>0.05), and was negatively correlated with estimated glomerular filtration rate (eGFR) (r=-0.562, P<0.05). It was positively correlated with serum creatinin, blood urea nitrogen, AI, TLAI and inflammatory cell infiltration (r=0.529, 0.482, 0.665, 0.690, 0.671, P<0.05). Conclusion IL-18 has a very close relationship with podocyte injury in patients with LN, and the uIL-18 can be a potential non-invasive detection method to monitor podocyte injury in LN patients.
5.Analysis of distribution and antibacterial resistance of common pathogenic bacteria of clinical infectious diseases in hospital

Objective To analyse distribution and antibacterial resistance of common pathogenic bacteria of clinical infectious diseases and provide scientific basis for the treatment of infectious diseases ,rational use of antibacterial agents and nosocomial infec‐tion control .Methods The distribution and antibacterial resistance of common pathogens clinically isolated from inpatients and out‐patients from August 2012 to July 2013 were analyzed .Bacterial identification and drug susceptibility test were carried out by using MicroScan WorkAway 40 automated bacterial identification and drug‐susceptibility analyzer ,and the results of drug susceptibility test were analysed by using the WHONET5 .5 software .Results A total of 1 434 strains of pathogenic bacteria were isolated ,and gram‐negative bacteria ,gram‐positive bacteria and other pathogenic bacteria accounted for 53 .8% ,28 .1% and 18 .1% respectively . The detection rate of methicillin‐resistant Staphylococcus aureus and methicillin‐resistant coagulase negative Staphylococcus were 31 .6% and 57 .4% respectively .No strains of vancomycin‐resistant Staphylococcus were found ,while strains of vancomycin‐resist‐ant Enterococcus faecium accounted for 8 .8% .The detection rate of extended‐spectrum beta‐lactamases producing Escherichia coli and Klebsiella pneumoniae were 47 .2% and 12 .2% respectively .Isolates of Staphylococcus and Enterococcus faecium were sensi‐tive to vancomycin ,linezolid and moxifloxacin ,and the resistance rates were less than 10 .0% .While isolates of Staphylococcus were resistant to the penicillin ,macrolides ,quinolones ,tetracycline ,cephalosporin ,and the resistance rates were more than 30 .0% .Most isolates of gram‐negative bacteria were sensitive to ertapenem ,imipenem ,amikacin ,meropenem ,piperacillin/tazobactam ,ticarcillin/clavulanic acid and levofloxacin ,and the resistance rates were less than 10 .0% .Conclusion It is necessary to enhance monitoring of distribution and antibacterial resistance of pathogenic bacteria ,so as to provide references for guiding rational use of antibacterial a‐gents in different departments .
6.Correlation of Expressions of P170,TOPⅡ and Proliferation in Gliomas

Objective To investigate the correlation of the expressions of P170,topoisomerase Ⅱ(TOPⅡ)and the proliferation in gliomas.Methods Ki-67 was used as the marker of the proliferation in gliomas.The expressions of P170,TOPⅡ and Ki-67 were tested by S-P immunohistochemical technique using monoclonal antibody to their protein in 54 cases of low-grade gliomas(WHO Ⅰ~Ⅱ grade)and 54 cases of high-grade gliomas(WHO Ⅲ~Ⅳ grade),analysis of the clinical and follow-up were performed.Results The expressions of P170,TOPⅡ and Ki-67 had significant differences between low-grade and high-grade gliomas(P<0.01).Correlation analysis showed that the expression of P170 was negatively related with Ki-67(r=-0.276,P=0.019);the expression of TOPⅡ was positively related with Ki-67(r= 0.637,P<0.001);there was no correlation between the expressions of P170 and TOPⅡ.Conclusion The expressions of P170 and TOPⅡ are associated with the malignant prognosis in gliomas.
7.Relationship between white blood cell count and nonalcoholic fatty liver disease in an adult population

Objective To investigate the relationship between white blood cell count (WBC) and the prevalence of simple fatty liver disease (SFL) and nonalcoholic steatohepatitis (NASH). Methods We designed a large scale cross-sectional study in an adult population. Participants were selected from Tianjin Medical University's General Hospital-Health Management Centre. The diagnoses of simple fatty liver disease and nonalcoholic steatohepatitis were based on liver ultrasonography and serum alanine aminotransferase concentration. A total of 37507 subjects (8644 SFL and 2557 NASH) were included in this study. Multiple logistic regression analysis was used to assess whether the quartiles of WBC were associated with the prevalence of simple fatty liver disease and nonalcoholic steatohepatitis. Results After adjusting for potential confounders, the odds ratios (95% confidence interval) of simple fatty liver disease and nonalcoholic steatohepatitis for increasing quartiles of WBC were: simple fatty liver disease, 1.00 (reference), 1.37 (1.24, 1.50), 1.70 (1.55, 1.86) and 2.09 (1.90, 2.29) (P for trend<0.0001);nonalcoholic steatohepatitis, 1.00 (reference), 1.39 (1.16, 1.66), 1.69 (1.43, 1.99) and 2.13 (1.81, 2.50) (P for trend<0.0001). Conclusions This study proves the correlation between WBC and the prevalence of simple fatty liver disease and nonalcoholic steatohepatitis. Further study is needed to clarify whether WBC has a predictive value for the occurrence of nonalcoholic fatty liver disease.
8.Association of urinary kidney injury molecu le-1 with renal tubulointerstitial lesions and its clinical significance in lupus nephritis patients

Objective To explore the association of urinary level of kidney injury molecule-1 (KIM-1) with renal tubulointerstitial lesions in patients with lupus nephritis (LN). And to find a potential clinical biomarker, which could reflect the specific tubulointerstitial lesions of LN. Methods Sixty cases of biopsy proven new-onset LN patients were enrolled into the study. And 10 normal kidney tissues were collected, which came from 10 patients with kidney trauma or renal tumor nephrectomy. Sixty patients with LN were divided into mild disease group, moderate disease group, severe disease group and the extremely severe disease group according to the criteria of 2000 Hill's morphologic index for the evaluation of renal pathology. Urine and renal tissues specimen were collected. The clinical and pathological data were analyzed retrospectively. Urine level of KIM-1 was detected by enzyme linked immunosorbent assay (-). Immunohistochemical staining was used to observe the expression of KIM-1 in the renal tissue. T-test, One-Way analysis of variance(ANOVA) test or rank sum test and Pearson or Spearman correlation analysis were used for statistical analysis. Results ①With the aggravation of tubulointerstitial lesions, the urinary level of KIM-1 was increased gradually, which was shown in the control group [(0.32 ±0.03) ng/ml], mild group [(2.31 ±0.30) ng/ml], moderate group [(6.12 ±0.51) ng/ml], severe group [(12.51 ±1.83) ng/ml] and the extremely severe group [(15.30 ±2.11) ng/ml] (all P<0.05); ② With the aggravation of tubulointerstitial lesions, the expression of KIM-1 in the renal tissue was increased gradually which was shown in the control group [(2.13±0.12)%], mild group [(35.01±0.33)%], moderate group [(51.12± 2.11)%], severe group [(63.50 ±1.80)%] and the extremely severe group [(75.31 ±3.22)%] (all P<0.05);③ Urinary KIM-1 level in LN patients was positively correlated with the expression of KIM-1 in the renal tissue (r=0.870, P<0.01);④Urinary KIM-1 level in LN patients was positively correlated with 24 hUP, systemic lupus erythematosus disease activity index (SLEDAI), renol (R)-SLEDAI, glomerular activity index (GAI), tubulointerstitial ativity imdex (TLAI), chronicity (AI) (r=0.826, 0.741, 0.824, 0.743, 0.871, 0.741, P<0.05), and was negatively correlated with estimated glomerular filtration rate (eGFR) (r=-0.726, P<0.05), while was not correlated with CI (r=0.532, P>0.05). The expression of KIM-1 in the renal tissue was positively correlated with serum creatinine (SCr), quantity of 24 h UP, SLEDAI, R-SLEDAI, GAI, TLAI and AI (r=0.780, 0.780, 0.812, 0.727, 0.735, 0.910, 0.701, P<0.05), and was negatively correlated with eGFR (r=-0.727, P<0.05), while it was no correlated with CI (r=0.543, P>0.05). Conclusion Urinary KIM-1 enables to assess the tubulointerstitial lesion in LN patients, and it can be a sensitive biomarker to predict the tubulointerstitial damage and to evaluate the degree of tubulointerstitial lesions.
9.UPLC Fingerprint of the Root of Rosa Laevigata Michx from Different Regions

Objective:To establish the fingerprint analysis method for the root of Rosa laevigata Michx from different regions by UPLC. Methods:The column was ACQUITY UPLC? Phenyl(2.1 mm × 100 mm,1.7 μm). The mobile phase consisted of methanol-water with gradient elution. The flow rate was 0. 2 ml·min-1 , the detection wavelength was 210 nm, the column temperature was 30℃, and the injection volume was 3 μl. Results:The fingerprint consisted of 15 common peaks. The range of similarity for twelve bat-ches of the root of R. laevigata Michx was 0. 489-0. 942. And the reference fingerprint of the root of R. laevigata Michx was estab-lished by UPLC. Conclusion:The fingerprint method is simple and reproducible, which can provide basis for the quality control and the medicinal resources exploration.
10.Systematic Review and Sequential Analysis of Efficacy and Safety of Baogong Zhixue Granules for the Treatment of Abnormal Uterine Bleeding due to Ovulation Disorder

Objective To systematically evaluate and sequentially analyze the clinical efficacy and safety of Baogong Zhixue Granules for the treatment of abnormal uterine bleeding due to ovulation disorder.Methods The randomized controlled trials(RCTs)of Baogong Zhixue Granules for the treatment of abnormal uterine bleeding due to ovulation disorder were retrieved from the databases of CNKI,Wanfang Data,VIP,China Biology Medicine,PubMed,Embase,and Cochrane Library from the establishment of the databases to January 5th,2023.Literature screening and data extraction were conducted according to Cochrane Handbook for Systematic Reviews of Interventions,and Meta-analysis was performed by RevMan 5.4.Results Totally 15 articles were included,involving 1 425 patients.Meta-analysis results showed that Baogong Zhixue Granules could effectively improve the clinical efficacy of abnormal uterine bleeding due to ovulatory disorder(RR=1.17,95%CI[1.13,1.23],P<0.000 01),shorten hemostasis time[bleeding control time(MD=-6.35,95%CI[-8.89,-3.81],P<0.000 01),complete hemostasis time(MD=-12.56,95%CI[-16.22,-8.89],P<0.000 01)],reduce endometrial thickness(MD=-1.26,95%CI[-1.71,-0.80],P<0.000 01),reduce hemoglobin loss(MD=13.35,95%CI[11.41,15.29],P<0.000 01),improve the score of uterine bleeding(MD=-2.22,95%CI[-2.43,-2.01],P<0.000 01),and reduce the recurrence of disease(RR=0.37,95%CI[0.19,0.72],P=0.004).There was no statistical significance between the test group and the control group in terms of adverse reactions(P=0.82).The trial sequential analysis of the total efficiency was carried out,and the cumulative included research passed the traditional threshold and trail sequential analysis threshold.Conclusion Baogong Zhixue Granules have a certain clinical efficacy in the treatment of abnormal uterine bleeding due to ovulation disorder,but there may be publication bias in the study,and higher quality clinical RCTs need to be carried out.
