1.A method of on-column sample stacking in capillary electrophoretic analysis of micro-scale amino acids

Purpose The aim is to develop a method for the analysis of micro-scale amino acids in biological samples.Methods After having been extracted and dried,the residues were dissolved in a running buffer or a distilled water and acetonitrile.The samples were loaded into column for 5,25 or 50s at a hydrodynamic injection and were separated by capillary electrophoresis.The effect of different dissolvent on the sample stacking was observed.Results A 100-fold improvement in the amount of material that can be injected into a capillary column without loss of resolution was shown.Conclusion The marked effect of sample stacking was achieved by the using distilled water and acetonitrile(1∶1,v/v) as dissolvent.
2.Study of the Preparation Technique and Stability of Ketorolac Tromethamine Eye Drops

OBJECTIVE:To establish a reasonable formulation and preparation technique of Ketorolac Tromethamine eye drops and to study the stability.METHODS:The content,related substances and pH value were taken as the observing pa?rameters to design the formulation and preparation technic.The stability of3batches of sample was tested.RESULTS&CONCLUSION:The eye drops was rational in formulation and feasible in preparation technic.The preparation in commercial packing was stable after6-month acceleration test and9-month storage at room temperature.
3.Clinical research on effect of Shenfu Injection in treating vertigo of mild vertebrobasilar stenosis

Objective: To evaluate the effect of Shenfu Injection in treating vertigo of mild vertebrobasilar stenosis. Methods: From January 2005 to July 2008, 66 patients with the primary symptom of vertigo undergoing digital subtraction angiography (DSA) examination were diagnosed as mild vertebrobasilar stenosis (MVS) according to the North American symptomatic carotid endarterectomy trial (NASCET) criteria, and then randomly divided into Shenfu (SF) group (34 cases) and Danshen (DS) group (32 cases), the DS group acting as the control. Degrees of vertigo of the patients were evaluated by the screening version of the dizziness handicap inventory (DHI-S) and scoring system for the dizziness assessment rating scale (DARS) on the admitted day, the third and sixth day after treatment and discharged day respectively. The mean of blood ? ow velocity (Vm), resistance index (RI) of right and left vertebral arterys and basilar artery, hematocrit (Hmc) and fi brinogen (Fbg) level in serum were all examined on both the admitted and discharged days. Results: There was no signifi cant difference on the score of vertigo, the parameter of TCD and the index in serum between SF and DS groups at the admitted time. 4.Study of morphological and immunohistochemical characteristics of phyllodes tumors

Objective To investigate the morphological and immunophenotypic characteristics of phyllodes tumors (PT) and explore their significance in differential diagnosis and evaluation of prognosis.Methods 48 specimens with PT were observed under light microscope with HE and immunohistochemical staining (EnVision method).The antibodies including CD34,CD10,CD117,p53,and Ki-67 were used.10 cases of fibroadenoma were compared and the clinical data and follow-up results were analyzed retrospectively.Results All of 48 cases with PT presented as well-circumscribed masses with typically leaflike structures composed of double-layered epithelial component arranged in clefts and overgrowing hypercellular mesenchymal component.31 benign,12 borderline,and 5 malignant PT were diagnosed based on stromal overgrowth,cellular pleomorphism,mitosis,margins,and others.Focal necrosis was detected in 3 malignant cases and liposarcoma component existed in 1 case.Immunohistochemical staining showed that the positive rates of CD34 were 93.5 %,58.3 %,0 in benign,bordline,and malignant PT respectively,there were significant differences between each groups (P < 0.05).The expression of CD10 were 25.8 %,83.3 %,80.0 %,its expression in benign group was significantly different compared with bordline and malignant PT (P < 0.05).p53 was expressed in three groups with no significant difference (P > 0.05).CD117 and Ki-67 had high expression comparatively in malignant PT.All cases were treated surgically with a local recurrence rate of 22.9 %.Conclusions Reasonable surgery pattern based on accurate histopathological diagnosis is crucial to reduce the local recurrence rate of PT.Immunohistochemistry examination in PT is helpful to the differential diagnosis and evaluation of prognosis.
5.Hepatic and kidney gray-scale ratio: a new index for quantifying hepatic fibrosis degree

Objective To investigate hepatic parenchyma and kidney cortex's gray-scale ratio as a new index in evaluating hepatic fibrosis degree.Methods Sixty patients with hepatic fibrosis verified by liver biopsy and 40 normal individuals used as controls were scanned with Acuson Sequoia 512 with the same set-up of all the parameters.The hepatic-right kidney section was selected for offline analysis.The gray-scale value of near-area hepatic,mid-area hepatic,far-area hepatic parenchyma and right kidney cortex were measured by histogram of Photoshop software.The gray-scale ratio of hepatic parenchyma to kidney cortex was calculated and compared with the hepatic firbrosis degree.Results With development of hepatic fibrosis,the ratio of hepatic and kidney gray-scale value was increased.The results between control and experimental groups were significant(P<0.01).Conclusions Hepatic and kidney gray-scale ratio obtained from gray scale ultrasound was rarely affected by individual conditions of acoustic windows.It could be used as a new index for diagnosing of hePatic fibrosis and evaluating its therapeutic effects.
6.The application of high-resolution melting curve analysis in the detection of BRCA1/2 gene mutation in the population with high breast cancer risk

Objective To analyze the gene mutation and clinical characteristics of BRCA 1/2 by high resolution melting curve ( HRM ) in breast cancer patients and high risk groups , and to discuss the application value for BRCA 1/2 mutation detection by using HRM curve analysis in people at high risk of breast cancer.Method The clinical control analysis was used ,Peripheral blood samples of 52breast cancer patients,their first-degree relatives (52 cases consisting of high risk group ) and 40 healthy people without family history of breast cancer cases were collected from Anhui Province Tumor Hospital or Bozhou People ′s Hospital during March 2015 to June 2016.To establish an effective method for BRCA 1/2 mutation detection by using HRM curve, and the mutation positive results were validated by gene sequencing .To analyze the correlation between the of BRCA 1/2 gene mutation and the risk factors , a logistic method was used .Results 9 cases of BRCA1/2 gene mutations were found in 52cases of breast cancer patients and the mutation rate was 17.3%.5 cases of BRCA1/2 gene mutations were found in 52 cases of breast cancer high risk groups and the mutation rate was 9.6%.In 40 healthy people who without family history of breast cancer cases ,we found only 1 gene mutation case.All the mutation positive results detected by HRM curve analysis were matched with gene sequencing results .BRCA1/2 gene mutations and the risk factors such as primiparous age and chronic mammary gland disease have a certain correlation .In the 9 cases of BRCA1/2 gene mutations , we found 5 cases of BRCA1/2 gene mutations in their first-degree relatives, with the consistency of 44.4%(4/9).Conclusion Thebreast cancer patients′s first-degree relatives have a high mutation rates on BRCA1/2 gene and they can be the key screening objects .HRM curve analysis can be used in health screening and risk assessment at the breast cancerhigh risk groups .
7.Iodine nutritional status and thyroid function among pregnant women in different periods of pregnancy in Tianjin

Objective To evaluate the iodine nutritional status and thyroid function of pregnant women in different periods of pregnancy,analyze the relationship between iodine nutritional status and thyroid function and provide a basis for scientific supplementation of iodine to pregnant women.Methods In 2013 and 2014,using stratified random sampling method,six counties were selected in the city;around 30 early,middle and late pregnant women each in every county were selected;a total of 545 (173 early,203 middle and 169 late) pregnant women were investigated in the 6 counties;instant random urine samples of 25 ml were collected and urinary iodine was tested.A total of 151 (52 early,55 middle and 44 late) pregnant women were selected to collect 3 ml venous blood samples for determination of thyroid hormone and thyroid autoantibodies.Results Median urinary iodine levels of early,middle and late pregnant women were 132.2,128.9 and 113.5 μg/L,respectively,under the condition of iodine deficiency.Prevalence rates of low frce thyroxine FT4,subclinical hypothyroidism,hypothyroidism and hyperthyroidism in pregnant women were 0.66% (1/151),15.23% (23/151),0.66% (1/151) and 0.66% (1/151),respectively.The positive rate of TgAb and TPOAb was 15.23% (23/151) and 11.92% (18/151),respectively.There were significant differences in the levels of free three iodine thyroid (FT3) in different periods of pregnancy (F =7.723,P < 0.05);FT3 in late pregnancy was lower than that in early and middle pregnancy (P < 0.05).There were significant differences in the levels of FT4 in different periods of pregnancy (F =3.762,P < 0.05);FT4 in late pregnancy was lower than that in early and middle pregnancy (P < 0.05).There were significant differences in the levels of thyroid stimulating hormone (TSH) in different periods of pregnancy (F =13.199,P < 0.05);TSH in early pregnancy was higher than that in middle and late pregnancy (P < 0.05).Subclinical hypothyroidism prevalence rate in late pregnancy was higher than that in middle pregnancy (x2 =3.912,P < 0.05).The positive rate of TgAb in early and middle pregnancy was higher than that in late pregnancy (x2 =9.883,3.906,all P < 0.05).Urinary iodine ≥250 μg/L was a risk factor for subclinical hypothyroidism in pregnant woman [odds ratio (OR) =5.076,P < 0.05].Conclusions The iodine nutrition of pregnant women in Tianjin is insufficient.Excessive urinary iodine is an increased risk of subclinical hypothyroidism.We should monitor the urinary iodine and thyroid function in pregnant women.
8.Coronary disease characteristics of ACS patients ≥75 years and outcomes of one-year follow up after PCI

Objective:To explore the clinic and coronary disease lesion features in patients with acute coronary syn-drome (ACS) ≥ 75 years old and the outcomes of one-year follow up after percutaneous coronary intervention (PCI) .Methods :A total of 355 ACS patients ,who hospitalized in our hospital from Jan 2012 to Dec 2012 and re-ceived PCI ,were divided into ≥75 years group (n=205) and <75 years group (n=150) .Clinical and coronary dis-ease lesion characteristics and results of one-year follow up after PCI were compared between two groups .Results:Compared with < 75 years group , there were significant rise in incidence rates of hypertension (66.83% vs . 78.67% ) ,diabetes mellitus (28.29% vs .39.33% ) ,arrhythmia (17.07% vs .44.00% ) ,and percentage of type C lesions in coronary angiography (41.95% vs .56.00% ) in ≥75 years group , P<0.05~ <0.01 .There were no sig-nificant difference in incidence rates of adverse events during one-year follow up between two groups ( P>0.05 ) . Conclusion :Higher incidence rates of clinical complications and complex lesions are found in ≥75 years patients with acute coronary syndrome ;but no significant difference is found in one-year prognosis between the two groups .
9.Role of complement activation and CVF intervention in lung inflammatory injury induced by influenza virus

Objective To investigate the mechanism of pulmonary inflammation induced by influenza virus , and provide reference for the development of effective drugs for viral pneumonia .Methods An influenza PR8 infection mouse model was established .The levels of inflammatory cytokines and complement molecules were determined using RT -PCR and ELISA.The pathological changes were examined using biopsy .The complement inhibitor cobra venom factor ( CVF) was injected intraperitoneally at a dose of 50 μg/( kg· 24 h) , and then body mass .The survival rate and inflammatory factors were examined .Results Compared with the control group , the expressions of complement regulatory molecule Crry and CD59 were significantly decreased (P<0.01), while those of complement C9 and complement receptor C3aR and C5aR were significantly increased in the lungs of influenza model mice (P<0.01).Pro-inflammatory cytokines TNF-α, IL-6 and IFN-γwere highly expressed , but anti-inflammatory cytokine IL-2 was lowly expressed in serum .Treatment with CVF caused a sight body mass loss, a survival rate increase and a lung index decrease (P <0.05).Moreover, an IL-2 expression increase and a decrease of IL-6, TNF -αand INF-γexpression were observed in CVF treatment mice ( P< 0.05).Conclusion Inhibition of complement activation can increase the survival rate of mice with influenza pneumonia and decrease pulmonary indexes .thus delaying the pathogenesis of PR 8.
10.The role of indocyanine green fluorescence imaging in laparoscopic partial splenectomy

Objective:To study the role of indocyanine green(ICG)fluorescence imaging in laparoscopic partial splenectomy (LPS).Methods:The data of 4 patients who underwent ICG fluorescence imaging technology for LPS at Beijing Luhe Hospital Affiliated to Capital Medical University from May 2017 to May 2020 were retrospectively analyzed. There were 3 females and 1 male, aged 46, 41, 27 and 12 years respectively. The extents of spleen preservation were compared between ICG fluorescence imaging with ordinary white light during operation. The residual splenic remnants were tested with fluorescence imaging after splenectomy, which showed fluorescence fading indicating good vascular perfusion.Results:ICG fluorescence imaging was performed on 4 patients. The operation time ranged from 180.0 to 250.0 min, and the intraoperative blood loss ranged from 40.0 to 200.0 ml. The postoperative hospital stay ranged from 4 to 14 days. There were no serious complications. Postoperative histopathology showed: splenic cyst ( n=1), splenic hemangioma ( n=2), and splenic laceration ( n=1). Conclusions:ICG fluorescence imaging technology had a significant role to play in partial splenectomy. This study showed this technique to improve safety of laparoscopic partial splenectomy.
