1.Clinical Observation of Zuo’s Yang-warming Acupuncture Therapy for Treatment of Allergic Rhinitis

Objective To observe the clinical efficacy of yang-warming acupuncture therapy established by Zuo Changbo for the treatment of allergic rhinitis. Methods Sixty patients with allergic rhinitis were randomly divided into observation group and control group, 30 cases in each group. The observation group was given Zuo’s yang-warming acupuncture therapy and the control group was given conventional acupuncture therapy. Acupuncture was performed once every other day, and 10 times constituted one course of treatment. After treatment, the therapeutic effect was evaluated, and the symptom scores of nasal itching, stuffy nose, sneezing and nasal discharge as well as their visual analog scale ( VAS) scores were observed. Results ( 1) Total effective rate of the observation group was 93.3%, and that of the control group was 63.3%, there was significant difference between the two groups ( P<0.01). ( 2) After treatment, the scores of the four symptoms were decreased obviously ( P<0.05 or P<0.01) , and the decrease in the observation group was superior to that in the control group ( P<0.01) . ( 3) After treatment, the VAS scores in both groups were decreased ( P<0.01) , and the decrease was more obvious in the observation group (P<0.01). Conclusion The curative effect of yang-warming acupuncture therapy on allergic rhinitis is better than that of the routine acupuncture group.
2.Effects of Living with Hope Program on family caregivers of patients with advanced lung cancer

Objective To explore the effects of Living with Hope Program(LWHP) on family caregivers of patients with advanced lung cancer.Methods Totally 60 family caregivers of patients with advanced lung cancer were randomly divided into the experimental group and the control group.The experimental group received LWHP intervention and routine care knowledge education,the control group only received routine care knowledge education.Hope,self-efficacy,anxiety and depression,and quality of life were evaluated at the first week,the second week,the first month,and the third month.Differences between two groups were compared using variance analysis of repeated measurements.Results The self-evaluation of the intervention was 76.6%,and the main effects of hope level,selfefficacy,anxiety and depression,and mental health were statistically significant (P<0.05);the time effect on hope level and self-efficacy were statistically significant (P<0.05);there were interaction effects between intervention and time on hope level,self-efficacy,anxiety and depression,and mental health (P<0.05).Conclusion LWHP can effectively improve hope level,self-efficacy and mental health status,and alleviate anxiety and depression of family caregivers of patients with advanced lung cancer.
3.Salmonella choleraesuis C500 delivering DNA immunization against classical swine fever virus.

Classical Swine Fever Virus (CSFV) E2 protein eukaryotic expression plasmid pVAXE2 was constructed. The plasmid pVAXE2 was transformed into Salmonella choleraesuis C500 (S. C500) attenuated vaccine strain by electroporation to generate Salmonella choleraesuis engineering strain S. C500/pVAXE2. The characterization of S. C500/pVAXE2 in morphology, growth, biochemistry and serology indicated that it retained the same properties as its original strain S. C500 with exception of kanamycin resistance originated from the plasmid pVAXE2. The plasmid stable in the bacteria after 15 passages. Kunming mice and rabbits were vaccinated three times at two weeks interval with S. C500/pVAXE2 in oral and intramuscular routes at the dosage of 1 x 10(8) CFU for mice and 2 x 10(9) CFU for rabbits each time. The specific antibody response against CSFV and Salmonella choleraesuis was detected by ELISA. Two weeks after the third boost the immunized rabbits were challenged with 20 ID50 of hog cholera lapinized virus (HCLV), followed by a virulent strain of Salmonella choleraesuis two week later than HCLV challenge. The results showed that all immunized mice and rabbits produced significant antibodies against CSFV and Salmonella choleraesuis, and the immunized rabbits demonstrated the effective protection against the challenge of HCLV and virulent Salmonella choleraesuis. These results indicated the potential of developing multiplex swine DNA vaccine by using this bacteria as the vector.

4.Expression of cathepsin B and its clinical importance in colorectal cancer.

OBJECTIVETo investigate the expression of cathepsin B (CatB) in colorectal cancer tissues and serum levels of CatB in patients with colorectal carcinoma and to study the association of CatB expression with lymph node and li ver metastasis.
METHODSImmunohistochemistry was used to detect CatB expression in tissues, and enzyme linked immunosorbent assay was applied to test CatB levels in peripheral vein blood in 83 patients with colorectal cancer.
RESULTSThe expression rates of CatB in primary lesions, normal colon mucosa, lymph node metastases and hepatic metastases were 56.6%, 31.3%, 88.4%, 85.0% respectively. The positive rates of CatB in primary lesions, hepatic and lymph node metastases were higher than that in normal mucosa (chi (2)=45.6124, P< 0.01). The CatB expression rates in lymph node and hepatic metastases were higher than that in primary lesions chi (2)=11.5982, 4.3747, P< 0.05). The positive rate of CatB was higher in Dukes C and D tumors than that in Dukes A and B tumors (chi (2)=18.8871, 25.1650, P< 0.01), higher in poorly differentiated and mucous adenocarcinomas than that in well-moderately differentiated adenocarcinomas chi (2)=14.2338, P< 0.05). The mean serum level of CatB in 83 patients with colorectal cancer was (5.9+/- 2.9) ng/ml, higher than (2.3+/- 1.1) ng/ml in the controls of 30 healthy volunteers (t=6.6975, P< 0.01). The serum level of CatB in the patients with Dukes C, D stages were higher than that with Dukes A, B stages.
CONCLUSIONSEnhanced expression of CatB in colorectal cancer tissues is associated with tumor infiltration and metastasis. Monitoring serum CatB level in patients with colorectal cancer is important in the prediction and diagnosis of lymph node and hepatic metastasis,and valuable for evaluation of the therapeutic effect.

5.The construction of yeast two-hybrid method in the protein-interactions and its significance in hepatic metastasis of colorectal carcinoma.

OBJECTIVETo construct the yeast two-hybrid system, and screen the proteins which interact with FasL, and investigate the relationship of FasL and hepatic metastasis of colorectal carcinoma.
METHODSWe have cloned the FasL gene into the pGBKT7 vector as the bait, then screened the fetal liver cDNA library, and have got a series of specific proteins that interact with FasL protein. Using the bioinformatics, we analyzed the interacting proteins in the mechanism of hepatic metastasis of colorectal carcinoma.
RESULTSWe have screened several proteins that interaction with FasL protein, including metallothionein 1K, 1G, 2A, cathepsin B, fatty acid synthase, interferon alpha-inducible protein 27, phospholipid scramblase, Ser/Thr-like kinase, anchor attachment protein, fibulin-5.
CONCLUSIONSWe have successfully constructed the yeast two-hybrid system, and preliminary identified that the interaction between FasL, metallothionein, cathepsin and anchor attachment protein is radically related to the hepatic metastasis of colorectal carcinoma.

6.Expression of long non-coding RNA associated with radiotherapy-resistance in colorectal cancer cell lines with different radiosensitivity.

OBJECTIVETo screen long non-coding RNA (lncRNA) associated with radiosensitivity in colorectal carcinoma cell lines.
METHODSColony formation assay was performed in colorectal cancer cell lines HT29, SW480, RKO, Lovo and HCT116 after irradiation with different radiation doses. Radiation sensitivity of these 5 cell lines was detected through survival fraction at 2 Gy (SF2 value). High-throughput lncRNA chip was used to screen lncRNA genes with expression differences more than 2 folds among SW480, RKO and Lovo. Further experiment on the expression differences of lncRNAs selected was conducted by realtime PCR.
RESULTSThe radiosensitivity order of these 5 cell lines from low to high (SF2 value from high to low) was HT29 (0.83 ± 0.03), SW480 (0.69 ± 0.02), RKO(0.53 ± 0.02), Lovo (0.47 ± 0.05), HCT116 (0.32 ± 0.03) (P < 0.01). Five lncRNAs associated with radiation sensitivity were screened. Among them, expression levels of R05532, NR_015441, and NR_033374 were positively correlated with radiation resistance(all P < 0.01), and expression levels of the other 2 lncRNAs, NR_073156 and AA745020, were not correlated with radiation resistance of colorectal cancer cells (both P>0.05).
CONCLUSIONSlncRNA R05532, NR_015441 and NR_033374 may be used as the predictive marker of radiosensitivity of colorectal cancer cells. Higher expression of these genes shows radiation resistance.

7.Analyzing the occupational hazard of 258 construction projects of Shunde in 2010 and 2011.

OBJECTIVETo understand the changing trend of occupational hazard of industries in Shunde area and to provide the scientific evidence for the prevention of occupational diseases.
METHODSThe pre-evaluation of occupational hazard was carried out for the construction projects. The data about employee number scale, industry species, occupational hazard factors, hazard levels and prevention measures for construction projects were collected and analyzed. The data of Shunde Occupational Health Survey in 2011 served as the control data.
RESULTSThere were 258 construction projects in 2010 and 2011, in which the proportions of medium and large scales increased to 8.1% and 2.4% respectively, the proportions of the furniture and chemical industries decreased from 25.6% or/and 5.1% to 2.7% or/and 1.8%, the proportions of the high-tech industries increased from 1.8% to 9.2%. The proportions of the projects with serious, medium and slight occupational hazard levels were 4.3%, 67.8% and 27.9%, respectively. The proportions of the projects with harmful chemicals, dusts and physical factors were 34.2%, 46.8%, 42.6%, respectively.
CONCLUSIONThe feature of occupational hazard in Shunde industries has changed at upgrading stage. We should pay attention to the prevention of occupational diseases in high-tech industries.

8.Expression of metallothionein and FasL in colorectal cancer and its clinical significance.

OBJECTIVETo study the expression of metallothionein (MT) and FasL in colorectal cancer and their relation to lymph node and liver metastasis.
METHODSImmunohistochemistry and quantitative RT-PCR were used to detect expression of MT and FasL in protein and mRNA levels in 93 cases of colorectal cancer.
RESULTSThe rates of MT expression in primary foci, non-cancerous colon mucosa, lymph node metastasis and liver metastasis were 58.1%, 32.3%, 81.1%, 64.3% respectively. And the rates of FasL expression were 41.9%, 19.4%, 62.3%, and 92.9% respectively. The positive rates of MT and FasL in primary foci, liver and lymph node metastasis were higher than that in non-cancerous mucosa (chi(2) = 35.2421, 57.5152, P < 0.01). MT expression rate in lymph node metastasis was higher than that in primary foci (chi(2) = 8.0565, P < 0.01). In liver metastasis, FasL expression rate was higher than in lymph node metastasis and primary foci (chi(2) = 8.6674, 22.4455, P < 0.01). The positive rates of MT and FasL in Dukes stage C and D were higher than that in Dukes stage A and B (chi(2) = 18.8871, 25.1650, P < 0.01). And higher rates of MT and FasL expression were observed in low differentiation adenocarcinoma and mucus adenocarcinoma than in middle-high differentiation adenocarcinoma (chi(2) = 11.1546, 9.2239, P < 0.05). High MT mRNA level was found in lymph node metastasis and high FasL mRNA level in liver metastasis.
CONCLUSIONSEnhanced expression of MT and FasL was associated significantly with lymph node and liver metastasis of colorectal cancer. Assay of MT and FasL expression has prognostic values for colorectal cancer patients.

9.Application of clinical pathway on rehabilitation exercise of postoperative shoulder arthroscopy

Objective To explore clinical pathway applied for shoulder joint operation under arthroscopy treated Bankart lesions of recurrent instability ( BLRI ) , and investigate the effect of postoperative rehabilitation exercise. Methods A total of 80 patients with BLRI under shoulder arthroscopy and their families were randomly divided into two groups on average: experimental group received clinical pathway for patients rehabilitation exercise and control group with routine health education. We recorded and statistically analyzed postoperative shoulder joint activity, postoperative pain score, nursing job satisfaction, activities of daily living and postoperative complications. Results The ASES and Constant-Murley scores of shoulder joint range of motion were (93. 86 ± 8. 67) and (95. 90 ± 4. 35) in the experimental group better than (63. 42 ± 10. 85) and (91.80±5.15)ofthecontrolgroupafter7daysandonemonthoperation(t=15.862,9.753;P<0.05). After 3 days operation, the VAS score was (4. 45 ± 2. 25) in the experimental group apparently lower (5. 50 ± 3. 75) in the control group (t=1. 328, P<0. 05), but the self-care ability (Barthel indexes) score was low, (85. 26 ± 12. 96) in the experimental group, and (81. 35 ± 14. 11) in the control group ( t=10. 236, P<0. 05). Comparing the postoperative complication of recurrent in stability, bone fibrillation, infection, the incidence of complication just was suffered one cases in the experimental group, but 6 cases in the control group (χ2 =3. 914, P<0. 05). Conclusions Using clinical pathway is an effective option for the patients to improve the quality of life and satisfaction rate of nurses, and it also can alleviate patients′ pain and reduce the postoperative complications.
10.Identification of 3-demethylchuangxinmycin from Actinoplanes tsinanensis CPCC 200056.

Chuangxinmycin (CM) from Actinoplanes tsinanensis was an antibiotic discovered by Chinese scientists about 40 years ago. It contains a new heterocyclic system of indole fused with dihydrothiopyran, whose biosynthetic mechanism remains unclear. CM is used as an oral medicine in the treatment of bacterial infections in China. The simple structure makes CM as an attractive candidate of structure modification for improvement of antibacterial activity. Recently, we analyzed the secondary metabolites of Actinoplanes tsinanensis CPCC 200056, a CM producing strain, as a natural CM analogue. We discovered the first natural CM analogue 3-demethylchuangxinmycin (DCM) as a new natural product. Compared to CM, DCM exhibited a much weaker activity in the inhibition of the bacterial strains tested. The finding provides valuable information for the structure-activity relationship in the biosynthesis of CM.
