1.ANTIMICROBIAL ACTIVILIES OF SEVERAL SEAWEEDS IN QINGDAO COAST

Duplicate plates methods and two purposive extraction methods (extracting phenols with 80% ethanol for 3 times after extracting lipids with other for 13 hours in soxhlot extractor)were used to research antimicrobial ability of nine common seaweeds in Qingdao coast. All extracts of nine seaweeds showed considerable activity against Bacillus subtilis, however,only three extracts,belonging to three species respectively,were effective against Escherichia coli. Several seaweeds not showing antimicrobial activity in previous tests showed considerabal effects. Gas chromatography was applied to analyse the contents of fatty acids in six kinds of lipids. C12-C22 saturated and unsaturated fatty acids were detected but no positive relation between the contents of unsaturated fatty acids and inhibition abilities were found.
2.Studies on antioxidative activity of high molecular weight polyphenols from two kinds of brown algae

Object To evaluate the antioxidative activity of the high molecular weight polyphenols from Sargassum thunbeergii Kuntze and Sargassum kjellmanianum Yendo. Methods Three kinds of evaluation systems for antioxidative activity were adopted to test their efficiency to scavenge radicals (including hydroxyl free radical ?OH, superoxide redical O? 2 and DPPH? free radical). Results The high molecular weight polyphenols from the two kinds of brown algae had strong activity to scavenge the radicals above. Furthermore, they acted almost as the same level. Conclusion The high molecular weight polyphenols from the two kinds of brown algae had strong antioxidative activity, thus they are the potential, natural and marine antioxidants.
3.Application of Y chromosome specific probe to the detection of expression of endothelial progenitor cells grafted in the injured lung tissues of rats

Objective To investigate the efficient methods used for tracing endothelial progenitor cells(EPCs)after transplantation in injured lung tissue.Methods Thirty clean grade healthy syngeneic SD female rats were divided into three experimental groups(n =10):(1)sham group,rats treated with intravenous phosphate-buffered saline(PBS)instead of lipopolysaccharide(LPS)followed by EPCs graft;(2)PBS-treated group,rats treated with intravenous PBS after intravenous injection of LPS to produce acute lung injury;(3)EPC-treated group,rats treated with EPCs after intravenous injection of LPS to produce acute lung injury.The transplanted EPCs were got from the same genetic species of the SD male rats.Rats of each group were sacrificed 7 days after EPCs transplantation.Their whole lung tissues were harvested to detect the expression of Y-chromosome by using hybridization in situ and RT-PCR assay.Statistic package of SPSS16.0 was used for the data analysis and significant differences between means were evaluated by ANOVA analysis.Results Compared with the other two groups,positive signals of sex-determining region y were found in lung endothelium from the EPC-treated group.Conclusions Y chromosme specific probe can be one of efficient methods for tracing stem cells after transplantation.
4.Application of microwave technique to extraction of free anthraquinones in Rheum emodi

5.Optimum extraction of polysaccharides from Ulva pertusa established by ortho gonal test

AIM: To optimize and establish the extraction techn ology of water-soluble polysaccharides from Ulva pertusa. TLC was employed to determine the mainly neutral sugars in the Ulvan. METHODS: The orthogonal test was employed to test the effects of th e four factors including volume of water, temperature of extraction, time of ext raction and pH of water on the yield and total carbohydrate contents of water-s oluble polysaccharides from Ulva pertusa and optimize the extraction technol ogy. Five groups of parallel tests were carried out by the optimum parameters es tablished and the sugar constituents of products were analyzed by TLC. RESULTS: Temperature of extraction and time of extraction have sign ificant effects on the extraction yield of water-soluble polysaccharides from Ulva pertu sa, but, for total carbohydrate content, only temperature of extraction was a significant factor. The mainly neutral sugars were composed of rhamnose, xylose and glucose. CONCLUSION: The optimum extraction technology was A 1B 3C 3D 3 when the yield and total carbohydrate content were considered as preferential pa rameters.
6.Effects of Degraded and Natural Polysaccharides from Porphyra Haitanensis on Proliferation of Lymphocytes

Objective To analyze the property of natural polysaccharides(P) from Porphyra haitanensis T. J. Chang et B. F. Zheng and its degraded polysaccharides(P3),and to investigate their effects on proliferation of lymphocytes. Methods P3 and P were identified by IR spectra. Their effects on immunological function were studied by cell culture technique. Results P3 and P displayed significant effects on promoting the proliferation of T-lymphocytes,and had inhibitory effects on the proliferation of B-lymphocytes in a dose-dependent manner. The effects of P3 were more powerful than P. Conclusion The degraded and natural polysaccharides from Porphyra haitanensis(P3 and P) can regulate the proliferation of lymphocytes,which may be related to their molecular weights.
7.Effects of a strong pulsed magnetic field on the growth-related gene expression of human bladder cancer BIU-87 cells

Objective To estimate any influence of strong pulsed magnetic fields on the expression of growth-related genes in human bladder cancer BIU-87 cells. Methods Human BIU-87 cells were cultured in vitro and randomly divided into a magnetic field group and a control group. Each group was further divided into 24 h, 48 h and 72 h sub-groups. The magnetic field group cells were exposed to an 8 T magnetic field pulsed at 15 Hz for 2 h every day. The control group cells also placed on the same environment, but not exposed to any strong, pulsed magnetic field. The expression of B cell lymphoma/leukemia gene-2 (Bcl-2) mRNA, Bax mRNA and caspase-3 mRNA was measured with RT-PCR, and flow cytometry was used to evaluate the expression of the Bcl-2, Bax and caspase-3 genes of the tumor cells in vitro. Results The expression of Bax mRNA and protein was significantly higher in the cells exposed to the magnetic field than in the control groups. The expression of Bcl-2 mRNA and protein was significantly less. The expression of caspase-3 mRNA and protein in the two groups showed no significant differences.Conclusions A strong, pulsed magnetic field can inhibit the growth of bladder tumor BIU-87 cells and promote their apoptosis. The mechanism is probably related with the magnetic field promoting Bax mRNA and protein expression and inhibiting Bcl-2 mRNA and protein expression.
8.Clinical study on treatment of lung cancer hydrothorax with Yiqi Yangjing decoction combined with traditional Chinese medicine sticking therapy

Objective:To observe the clinical effect of Yiqi Yangjing recipe combined with sticking therapy on lung cancer pleural effusion.Methods:From December 2017 to August 2019, 68 patients with advanced lung cancer and pleural effusion in Pudong New Area Hospital of Traditional Chinese Medicine were selected and randomly divided into two groups by random number table method, with 34 cases in each group.The control group was given furosemide orally on the basis of routine nutritional support treatment, and the treatment group was given Yiqi Yangjing prescription orally and traditional Chinese medicine sticking therapy on the basis of the control group.Both two groups were treated for 6 weeks.The changes of pleural fluid volume and quality of life scores were observed and compared between the two groups.Results:The total effective rate in the treatment group was 70.5%(24/34), which in the control group was 47.1%(16/34), the difference was statistically significant between the two groups(χ 2=3.886, P<0.05). The total effective rate of TCM syndrome integral was 64.7%(22/34) in the treatment group, and 35.3%(12/34) in the control group, the difference was statistically significant between the two groups(χ 2=5.800, P<0.05). After treatment, the overall quality of life score of the treatment group(55.74±5.15)points, which was higher than that of the control group[(51.91±5.20)points]( t=56.130, P<0.05). Conclusion:Yiqi Yangjing recipe combined with traditional Chinese medicine sticking therapy can effectively improve the clinical efficacy of lung cancer hydrothorax, improve the quality of life of patients.
9.Association between negative emotion and interpersonal sensitivity among patients in methadone maintenance treatment.

OBJECTIVETo analyze interpersonal sensitivity level and influencing factors among patients in methadone maintenance treatment.
METHODSFrom 2010 June to 2011 June, the research was performed in Center of Methadone Maintenance Treatment of Nanjing Baixia District Hospital. A total of 272 methadone maintenance treatment was studied using a self-designed questionnaire, interpersonal sensitivity scale in symptom check list (SCL-90), self-rating depression scale (SDS), self-rating anxiety scale (SAS). If interpersonal sensitivity level ≥ 2 points, it was positive. In this study, through independent sample t test, analysis of the social demographic characteristics, depression and anxiety, interpersonal sensitivity level. Non conditional logistic regression analysis was used to explore a variety of factors, and the relationship of these factors with interpersonal sensitivity.
RESULTSThe depression score, the anxiety score and the level of interpersonal relationship sensitivity in 272 methadone maintenance treatment (1.65 ± 0.51, 47.66 ± 8.58, 42.38 ± 9.40) was higher than the national norm (1.65 ± 0.51, 41.88 ± 10.57, 29.78 ± 0.46) (t values were 8.23, 17.63 and 22.27, respectively; all P values < 0.001). In 272 methadone maintenance treatment, the percentage of men was 75.0% (204/272) and the percentage of women was 25.0% (68/272). SDS score of male and female was (48.38 ± 8.30) and (45.90 ± 9.34) points (t = 5.67, P = 0.037); Aged 20-29, 30-39, 40-49 and ≥ 50 years old group, SDS scores were (48.26 ± 7.59), (47.73 ± 8.38), (47.79 ± 9.22) and (48.09 ± 6.33) points (F = 3.90, P = 0.046); According to the drug's time, divided into 1-3, 4-6 and ≥ 7 years group, SDS scores were (45.92 ± 7.47), (47.41 ± 7.57) and (48.07 ± 8.57) points (F = 4.49, = 0.039). SAS score of male and female was (41.21 ± 9.53) and (42.91 ± 9.42) points (t = 2.23, P = 0.043). The positive rate of interpersonal sensitivity was 47.1% (128/272). Multivariate analysis showed that, with SDS and SAS scores increased by 1, respectively, interpersonal sensitivity increased 23.23 (8.62-96.68) and 11.97 (6.42-19.27) points.
CONCLUSIONInterpersonal sensitivity level in methadone maintenance treatment was higher than normal and depression; anxiety and depression both were the risk factors of interpersonal sensitivity level.
