1.CYTOGENETIC STUDIES ON HPV-18-IMMORTALIZED CELLSFROM ESOPHAGEAL EPITHELIA OF HUMAN EMBRYO

Objective To study the cytogenetic changes in HPV-immortalized cells(SHEE)for further investigating the esophageal carcinogenesis. Methods Giemsa staining was used to show the modal number of chromosomes.G-banding was done to observe the chromosomal aberrations.Fluorescence in situ hybridization(FISH)of the interphase nuclei with the centromere probes of chromosome 1,7 and 8 were performed to determine the numerical abnormalities. Results The modal numbers at passage 10 and 20 were 58-63 and 57-64, respectively.Passage 61 had a bimodal distribution 58-61 and 63-65.The structural aberrations could almost be found in all of karyotypes.The following structural aberrations were identified:1.del(1)(q12);2.del(1)(p32);3.der(4),t(4;?)(q31;?);4.der(5),t(5:?)(q31;?).FISH results showed that trisomy 1 and tetrasomy 7 markedly increased with the increase of passages and disomy 8 kept in faint change in all passages.Conclusion The modal number moved right slowly in the graph of the modal distribution and the increases of chromosomal numbers were in imbalance with increases of passages of SHEE.The structural aberrations of chromosome were found in most of karyotypes.Our results suggest that the cell immortalization is a multistep process involved in the multiple chromosomes.;
2.Adenovirus-mediated T-bet gene transfers into lymphocyte to induce type-1 T-helper differentiation

AIM: To construct recombinant adenovirus containing transcription factor T-bet (T-box expressed in T cells),and induce type-1 T-helper differentiation of lymphocytes. METHODS: T-bet gene was cloned from total RNA of lymphocyte stimulated with IFN-? with RT-PCR methods,then subcloned into transfer vector pAdtrack-CMV in BgIII/SaII sites. The new transfer vector pAdtrack-CMV. T-bet was digested with Pme I,subsequently cotransformed into BJ5183 cells with adenoviral backbone plasmid pAdEasy-1. The resultant plasmid pAd. T-bet was linearized by Pac I and transfected into 293 cells with liposome LIPOFECTAMINE 2000 for producing Ad.T-bet. The recombined adenovirus Ad.T-bet was identified through RT-PCR and Western blotting methods. Lymphocytes purified from patients suffering from liver cancer was infected with liposome and Ad.T-bet with multiplicity of infection (m.o.i) 5000,and the concentration of IFN-? in culture media was evaluated with ELISA methods. RESULTS: T-bet gene was successfully cloned from lymphocytes and incorporated into recombinant adenovirus Ad.T-bet. Lymphocytes infected with Ad. T-bet constantly and strongly secreted Th1 cytokine IFN-?. CONCLUSION: Recombinant adenovirus Ad.T-bet effectively induces type-1 T-helper differentiation,which is a promising method for restoration of patients' immune reaction against cancer.
3.Introduction to revision of Technical Specification for Occupational Health Surveillance

To revise GBZ 188 Technical Specification for Occupational Health Surveillance based on national laws, regulations, standards, specifications and legal documents of occupational disease, and combination with the actual situation in China. The main modifications are as follows: the occupational health surveillance for workers exposed to toluene (xylene may implement by reference), bromopropane, methyl iodide, ethylene oxide, chloroacetic acid, indium and its compounds, coal tar, coal tarasphalt, asphalt, β-naphthylamine, dust of metal and its compounds(tin, iron, antimony, barium and its compounds), hard metal dust, erionite dust, low temperature, laser, tick-borne encephalitis virus, Borrelia burgdorferi, and human immunodeficiency virus, for scraper or grind operators, and underground workers using squatting or kneeling position, crawling position, side-lying position, or shoulder position for a long period of time are included. The emergency health screening for workers exposed to arsenic, fluorine and its inorganic compounds, and acrylamide are included. The occupational medical examination (OME) for workers exposed to amino and nitro compounds of benzene, phosgene, monomethylamine, organic fluorine and dimethyl sulfate has been adjusted and made mandatory, with corresponding assessments required upon leaving the job. The special occupational health surveillance for workers exposed to mycobacterium tuberculosis and hepatitis virus is removed. The OME conclusion of reexamination is removed, and standardize recheck/additional inspection requirements. The optional items in OME performed before, during and after leaving post are removed, but the optional items in emergency medical examination are retained. Additional OME items are added. The Guideline for OME Summary Reports is added as informative appendix, and so on. The revised GBZ 188 Technical Specification for Occupational Health Surveillance is more scientific and practical.
