1.STUDY ON CONDITIONS OF SWINE BLOOD MEAL FERMENTATION WITH COMPOUND STRAINS

Two high protease producing strains, whic h are desi gnated Aspergillus oryzae AS100 and AS102, were screened and applied as main fermentation strains of swine blood meal. A Saccharomyces cerevisiae Y113 and a Bacillus Asp007 were also used as assistant fermentation strains. The enzyme production abilities we re studied,and a series of parameters of the fermentation technology were deter mined. Through swine blood meal fermentation, a high-protein fermented feed wa s produced, which is strong soy flavor and was rich in protein(69%), free amino acid, vitamin such as VitD 3 and niacin and organic elements such as Fe. It is a kind of high-protein feed for animals and it could be used as feedstuff addi tive.
2.Differentiation of Human Amniotic Mesenchymal Cells into Cardiomyocyte-like Cells in Vitro

To explore the plasticity of human amniotic mesenchymal cells(hAMCs) into cardiomyocyte-like cells,hAMCs were isolated from human amnion with collagenase digestion.Phenotype of the isolated cells was analyzed by flow cytometry(FCM).hAMCs were treated with 5-azacytidine and basic fibroblast growth factor(bFGF) to investigate their ability of differentiation into cardiomyocytes.The induced differentiated cells were evaluated by immunofluorescence for desmin and ?-actin expression and by RT-PCR for Nkx2.5,GATA-4 and alpha-myosin heavy chain(?-MHC) mRNA expression.The results showed that,after primary culture,hAMCs could reach a confluence of 80% with swirl like growth at 6 days.The cells proliferated rapidly after passages with a 100% confluence at 3~4 d.hAMCs were positive expression of CD44 and vimentin,but negative for CK19.After induced differentiation at 8~10d,the differentiated cells have close-up arranged with long spindle-shape.At 2 weeks and 4 weeks,induced cells expressed ?-actinin and cardiac-specific transcription factor Nkx2.5.Expressions of GATA-4 and desmin can be detected but ?-MHC can not in the hAMCs both before and after the induction.In conclusion,hAMCs have the ability of differentiation into cardiomyocyte-like cells,which means that hAMCs can be regarded as candidate cells for cellular cardiomyoplasty(CCM).
3.Midterm curative efficacy of domestic drug eluting coronary stent in treatment of left main lesion

Objective To study midterm curative effect of domestic drug eluting coronary stent in treatment of left main (LM) lesion.Method From April 2004 to September 2005,totally 32 patients with LM lesion received percutaneous coronary intervention (PCI) without protection.Thirty-nine coronary stents (Firebird, Microport,Shanghai) were implanted.One stent was used when the lesion only involved the opening or the middle portion of the LM branch.If the bifurcation was involved,the stenosis was treated considering the following factors: the diameter of left main,left anterior descending (LAD) and the left circumflex branch (LCX);whether there was stenosis of the opening of circumflex branch.Methods included"T"stent,stent pass though the opening of circumflex branch and kissing stents.All patients were followed at 1,6 and 12 months after discharge.Coronary angiography of patients was performd at 5 months after discharge.Central necrosis,restenosis,target vessel reconstructive rate (TVR) and target lesion reconstructive rate (TLR) were analyzed.Results Most LM lesion involved bifurcation (53.1%).Treatment for bifurcation included"T"stent in 47% patients,stent passing through the opening of LCX in 35.3 %.No restenosis occurred in lesion involving the opening or middle portion of the LM during follow-up.The 62.3% patients received coronary angiography.Angiography showed 5% of restenosis rate,5% TLR,10% TVR and 6.3% central necrosis rate.Conclusions Drug eluting stent implantation in low risk unprotective left main lesion may achieve satisfactory clinical results.
4.The Observation on the Effect of Q-Switching Nd:YAG Treatment in Pigmented Dermatoses and Influence for Melanin Metabolism

Objective To observe the effect of Q-Switching Nd:YAG treatment in pigmented dermatoses and its influence for melanin metabolism. Methods 114 patients with pigmented dermatoses in our hospital from October 2015 to August 2016 were selected as the study object,and were randomly divided into the control group (57 cases) and the observation group (57 cases).The control group were treated with intense pulsed light therapy while the observation group were treated with Q-Switching Nd:YAG treatment on the treatment of control group. Then the total effective rates,recurrence rates,adverse reaction rates,skin lesion scores and serum melanin metabolism related indexes before and after the treatment of two groups were analyzed and compared. Results Clinically, the total effective rates of observation group were higher than those of control group ( <0.05)and the recurrence rate of observation group was lower than that of control group ( <0.05) .The adverse reaction rates of two groups were compared and the differences were not statistically significant ( >0.05) .The skin lesion scores and serum melanin metabolism related indexes of two groups before the treatment were compared and the differences were not statistically significant (>0.05) .The serum melanin metabolism related indexes of two groups after the treatment were all better than those of control group ( <0.05) . Conclusion The clinical effect of Q-Switching Nd:YAG treatment in the patients with pigmented dermatoses is better, and it has the active influence for the melanin metabolism,so it has higher clinical value in the treatment of pigmented dermatoses.
5.Preliminary experiences in minimally invasive transforaminal lumbar interbody fusion.

OBJECTIVETo explore clinical application of minimally invasive transforaminal lumbar interbody fusion (TLIF) for the management of lumbar disorders and discuss its indications, surgical techniques and clinical effectiveness.
METHODSFrom Jan 2005 to Dec 2006, 31 selected patients (22 males and 9 females, aged from 41 to 63 years) with degenerative lumbar diseases were treated with minimally invasive TLIF assisted by METRx X-Tube micro-endoscopy system. The index diagnosis was lumbar disc herniation with Lumbar spinal stenosis in 7, lumbar disc herniation with segmental instability in 16, grade 1 to 2 of lumbar spondylolisthesis in 8. The surgical methods was performed with bilateral or unilateral pedicle screws insertion and a single rectangle cage posterolateral placement. The operating time, blood loss, blood transfusion, drainage, visual analogue scale (VAS), preoperative and postoperative JOA scores were observed as well as radiographic evaluation. The results were compared with standard TLIF group respectively.
RESULTSA total of 116 pedicle screws and 31 cages were implanted of which 4 patients were treated with unilateral pedicle screws fixation. Four pedicle screws were found misplaced in CT scans after surgery. The average operating time was 199 min, blood loss 359 ml, volume of transfusion 32 ml drainage 81 ml, and VAS was 2.37 about 72 hours after surgery, which had statistic difference compared with control group. There was no statistic difference on postoperative improvement rate and JOA scores in two groups.
CONCLUSIONSMinimally invasive TLIF minimizes paraspinal muscle trauma and blood loss, shortens the operating and recovery time. A good long-term outcome can be gained compared with standard procedures.

8.Case-clinical studies on three different intervertebral fusion with bone graft and internal fixation in the treatment of cervical spondylotic myelopathy.

OBJECTIVETo evaluate the therapeutic effects of three different intervertebral fusion with bone graft and internal fixation in the treatment of cervical spondylotic myelopathy.
METHODSIn the retrospective study, 83 patients (38 males and 45 females, with an average age of 69 years) with cervical spondylotic myelopathy were divided into three groups. The patients in three groups were treated by autogenous iliac bone grafting (Robingson grafting) with anterior cervical plate-screw fixation, Pyramesh with anterior cervical plate-screw fixation and BAK fixation respectively. JOA scores and the bony fusion time were recorded and the results were evaluated by numeric computer system.
RESULTSThe JOA scores of all the patients were higher than those before surgery (t = 1.55, P < 0.05). There were no statistical difference of JOA scores in the three groups (F = 2.51, P > 0.05).
CONCLUSIONIt is concluded that internal fixation of the three types is promising to the treatment of cervical spondlotic myelopathy. Robingson grafting with anterior cervical plate-screw fixation provides good stability and bony fusion. BAK is better technically required but there was height loss in clinic. Pyramesh with anterior cervical plate-screw fixation combines the merits of both two techniques above but the cost is higher.

9.Ex vivo expansion of megakaryocyte progenitor cells for CD133(+) cells derived from human umbilical cord blood.

To study the expansion potentiality of megakaryocyte progenitor cells (MPCs) derived from human umbilical cord blood CD133(+) (UCB-CD133(+)) cells and determine the optimal harvest time. UCB-CD133(+) cells were purified from mononuclear cells (MNCs) by magnetic activated cell sorting (MACS) and seeded in serum-free liquid culture medium supplemented with thrombopoietin (TPO), interleukin-3 (IL-3), and stem cell factor (SCF) to expand MPCs. At day 0, 6, 10 and 14 of culture, the total cell number was counted and the dynamic changes of CD133, CD34, and CD41 antigen expression during ex vivo expansion were analyzed by flow cytometry (FCM). At different expansion times, the CD133(+) cells were collected and cultured in collagen semisolid medium to carry out CFU-MK colony culture. The incidence of CFU-MK was calculated and the morphology of MPCs and CFU-MK were detected by immunohistochemistry and Wright-Giemsa staining. The results showed that UCB-CD133(+) cells optimally expanded at day 7 with expansion multiple of 8.2 +/- 2.2 in serum-free liquid culture systems and the total cell number was expanded by 116-fold at day 14. At 10 days, each UCB-CD133(+) cell can form 2.5 +/- 1.0, 2.6 +/- 0.5 and 20.3 +/- 5.9 cells of CD133(+)CD41(+), CD34(+)CD41(+) and CD41(+) respectively, from which the number of CD133(+)CD41(+) and CD34(+)CD41(+) cells reach the highest. UCB-CD133(+) cells both before and after expansion could form CFU-MK, the total number of CFU-MK reached the peak from cells of 10 days expansion of UCB-CD133(+) cells and the expansion multiple of CFU-MK was 59.5 +/- 11.8. Immunohistochemical results indicated that the expanded megakaryocytic cells were immature and no sign of platelet formation. It is concluded that the human UCB-CD133(+) cells have a high ability of MPC expansion, 10 days of culture can be result in optimal expansion effect.
