1.Characteristics of proliferation and myotube cell formation of rat skeletal muscle satellite cells cultured in vitro

Objective To establish a method of isolation and purification of rat skeletal muscle satellite cells,and observe the characteristics of proliferation and myotube cell formation of skeletal muscle satellite cells cultured in vitro. MethodsPurified skeletal muscle satellite cells were obtained by improved two-step enzymatic digestion method and pre-plating technique.Immunohistochemical staining was employed to identify the skeletal muscle satellite cells cultured in vitro.The growth of skeletal muscle satellite cells cultured in vitro was examined by MTT assay.The differentiation of skeletal muscle satellite cells was observed by inverted microscopy. Results The skeletal muscle satellite cells with higher purity were obtained and confirmed by the high expression of desmin.When cultured in vitro,the latent phase of skeletal muscle satellite cells was the first to the second day,and the platform phase was the fifth to the sixth day.Myotube cells gradually formed when cell confluence was more than 60% to 70% or differential medium with lower fetal bovine serum was used.Conclusion The combination of improved two-step enzymatic digestion method and pre-plating technique serve as an easy and practical way to obtain skeletal muscle satellite cells with higher purity.Skeletal muscle satellite cells can form myotube cells with contraction characteristics without any special induction.
2.Application of reduced glutathione in protecting liver function in HCC patients after interventional therapy: a clinical study

Objective To evaluate the clinical effect of reduced glutathione in protecting the liver function for patients with hepatocellular carcinoma (HCC) after receiving international therapy.Methods The clinical data of 110 HCC patients,who were admitted to authors' hospital during the period from January 2010 to June 2013,were retrospectively analyzed.According to the random number table method the patients were divided into the control group (n=50) and the treatment group (n=60),and transcatheter arterial chemoembolzation (TACE) and transcatheter arterial infusion (TAI) were carried out in all patients.After the treatment,intravenous infusion of 2.0 g vitamin C,0.2 g vitamin B6,2.0 g inosine,and 250 ml branched chain amino acid was employed per day for one week for the patients of both groups,and additional intravenous dripping of 1.8 g reduced glutathione (added in 250 ml of 5% glucose solution) was adopted for the patients of the treatment group.Various liver function indexes were tested before and one week after the treatment,the results were compared between the two groups.Results Before the treatment,no obvious differences in the hepatic functions and in the clinical condition existed between the two groups.One week after the treatment,in both groups the aminotransferase (ALT and AST) and total bilirubin (TBIL) were significantly increased when compared with the preoperative data (P<0.05),while the serum pre-albumin (PA) was remarkably decreased when compared with the preoperative data (P<0.05);these changes of various indexes in the treatment group were apparently better than those in the control group (P<0.05).Conclusion The use of reduced glutathione can effectively relieve the liver function damage caused by interventional therapy in HCC patients,therefore,reduced glutathione has a very good clinical application value.
3.Development of an embedded mobile terminal for real-time remote monitoring of out-of-hospital cardiac patients.

A kind of real-time remote monitoring embedded terminal which is combined with mobile communication technology and GPS localization technology, has been developed. The results of preliminary experiments show that the terminal can transmit ECG signals and localization information in real time and continuously, supply a real-time monitoring of out-of-hospital cardiac patients and trace the patients.

4.Extramammary Paget's disease of the scrotum: report of 79 cases.

OBJECTIVETo investigate the clinicopathologic feature and treatment result as well as prognosis of Paget's disease of the scrotum.
METHODSThe data of 79 patients with Paget's disease of the scrotum confirmed pathologically from 1993 to 2004 were retrospectively analyzed. The median age of these patients was 62.3 years. The median duration of the disease was 3 years and 7 months. Locally extensive excision and intra-operative frozen section analysis of resection margin were performed in 78 patients. Inguinal and iliac radical lymphadenectomy was done in 7 patients with inguinal lymph node metastasis.
RESULTSLocal recurrence developed in 9 patients, and 3 patients died of the disease during follow-up.
CONCLUSIONThe prognosis of Paget's disease of the scrotum after surgical resection is good. To highly watch out for the disease, to perform histological examination and surgical resection as early as possible are keys to control it successfully.

5.Association between self-assessed somatotypes and symptom depression among children and adolescents

Objective To examine the association between self-assessed somatotypes and depression related symptoms.Methods The study sample included 5555 students aged 9 to 18 years who had attended the Physical Fitness and Health Surveillance of Chinese School Students(2010)in Chizhou city,Anhui province.Association between self-assessed somatotypes,body mass index(BMI)and depression symptom were examined.Results There was a slight consistency between self-assessed somatotypes and BMI in both boys and girls(Kappa=0.217,P=0.000; Kappa=0.203,P=0.000).Significant difference in the prevalence of depression was found among weight misperception groups in both genders(x2=145.223,P=0.000).The prevalence of umderestimation of somatotypes was significantly higher in boys than in girls,while the result was reversive on the prevalence of overestimation.Additionally,the prevalence of depression was higher in girls than in boys(x2=5.199,P=0.023).Through logistic regression,data showed that self-assessed somatotypes and miscalculated groups were significantly associated with depressive symptoms.Compared to the group that self-assessment as being normal,those students when self-assessed as being slim,overweight or obesity were more likely to be depressive,with odds ratio(ORs)as 1.255(95%CI:1.066-1.478),1.538(95%CI:1.275-1.856)and 1.713(95% CI:1.035-2.834),respectively.Overestimated and underestimated somatotypes appeared to be risk factors causing symptoms of depression(OR=1.705,95%CI:1.382-2.105; OR=1.241,95%CI:1.059-1.454).Conclusion Slight consistency was found between self-assessed somatotypes and BMI,while the misjudged somatotypes were the risk factor related to depressive symptoms.It was suggested that life skills education should be carried out as preventive intervention approach,to improve the physical and mental health well-being of children and adolescents.

Objective To study the eco-geographic landscapes of natural plague foci,in China.Methods According to the surveillance records on plague epidemics and the eco-geographic landscapes of natural plague foci based on the county level,the criterion for classifying the ecological geographic zone of Chinese natural plague foci was established.Results 12 types and 19 subtypes of eco-geographic landscapes on Chinese natural plague foci were identified.Conclusion Scientific basis for Chinese natural plague foci classification was provided.
7.Psychopathological sub-healthy status as predictor on self-or unintentional injuries among Chinese adolescents: a school-based follow-up study

Objective To investigate the association between psychological sub-healthy status and risks on injuries and related predictors among adolescents in China,in order to develop reasonable intervention programs.Methods Adolescents were selected from middle schools and colleges in eight cities.Baseline status on sub-healthy that related to psycho-pathology was measured using the Multidimensional Sub-health Questionnaire of Adolescents (MSQA).Data on Self- or unintentional injuries were collected using a standardized injury surveillance questionnaire every 3 months,for three times (T0,T1,T2 and T3).Results A total of 12 113 students completed the study.Prevalence rates related to self-injury among boys and girls were 19.4% and 19.9%,respectively.The rates of having more than 4 types of self-injuries in boys and girls were 4.7% and 4.2%,respectively.Boys had a significantly higher unintentional injury rate than that of girls (62.2% vs.57.3% ).The differences in the rates of having more than 4 types of unintentional injury between boys (3.8%) and girls (3.4%) were also statistically significant.Rates on self- and unintentional-injuries increased in T1,T2 and T3 when the psychological symptoms had an increase.Data from Multiple regression analysis demonstrated that the baseline psychological sub-healthy status was a predictive factor for higher risk of self- and unintentional- injuries at each of the follow-up waves.Conclusion Sub-healthy status of psycho-pathology was associated with an increased risk on self-and unintentional injuries among adolescents.Integrative psycho-pathological and behavioralintervention programs might help in reducing the risk of injuries among the Chinese adolescents.
8.Registration and epidemiological characteristics of pulmonary tuberculosis among students in Hunan Province from 2012 to 2017

Objective To analyze the registration of pulmonary tuberculosis (PTB)among students in Hunan Province,understand the epidemiological characteristics,provide evidence for improving tuberculosis control strate-gy in schools in Hunan Province.Methods Registration rate and epidemiological characteristics of students with tu-berculosis in Hunan Province were analyzed through data registered in China Tuberculosis Information Management System between 2012 and 2017.Results From 2012 to 2017,7 940 students with PTB were found in Hunan Prov-ince,the registered incidence was 13.23/1 00 000,2 203 cases were smear positive for PTB,registered incidence was 3.67/1 00 000.Registered incidence of active PTB students in 2012-2017 was significantly different (χ2=80.079,P<0.001);registered incidence of smear positive PTB students in 2012-2017 was significantly different (χ2=112.213,P<0.001).The number of registered PTB students in the second quarter was the largest (32.2%), mainly male (60.8%)and students aged 15-19 years (61.8%).There was a significant difference in the registra-tion of PTB students in different cities from 2012 to 2017 (χ2=320.432,P<0.001).The top three regions of the total number of registrations were Changsha,Xiangxi and Hengyang.From 2012 to 2017,the registered PTB students were mainly referral (38.8%),99.8% of the patients received anti-tuberculosis treatment,diagnosis and treatment were mainly for smear-negative,non-severe,non-drug-resistant,and newly treated patients,accounting for 67.9%,95.2%,99.5%,and 99.3% respectively.Conclusion It is necessary to strengthen the prevention and control of tuberculosis in schools,screen tuberculosis among freshmen in high schools and universities,publicize tu-berculosis knowledge,and improve awareness of tuberculosis prevention and control in schools.
9.Relationship between Fractional Exhaled Nitric Oxide Level and Efficacy of Inhaled Corticosteroid in Asthma-COPD Overlap Syndrome Patients with Different Disease Severity.

This study explored the relationship between the fractional exhaled nitric oxide (FeNO) level and the efficacy of inhaled corticosteroid (ICS) in asthma-chronic obstructive pulmonary disease (COPD) overlap syndrome (ACOS) patients with different disease severity. A total of 127 ACOS patients with ACOS (case group) and 131 healthy people (control group) were enrolled in this study. Based on the severity of COPD, the ACOS patients were divided into: mild ACOS; moderate ACOS; severe ACOS; and extremely severe ACOS groups. We compared FeNO levels, pulmonary function parameters including percentage of forced expiratory volume in 1 second (FEV1) to predicted value (FEV1%pred), ratio of FEV1 to forced vital capacity (FEV1/FVC), inspiratory capacity to total lung capacity (IC/TLC) and residual volume to total lung capacity (RV/TLC), arterial blood gas parameters, including PH, arterial partial pressure of oxygen (PaO₂) and arterial partial pressure of carbon dioxide (PaCO₂), total serum immunoglobulin E (IgE), induced sputum eosinophil (EOS), plasma surfactant protein A (SP-A), plasma soluble receptor for advanced glycation end products (sRAGE), sputum myeloperoxidase (MPO), sputum neutrophil gelatinase-associated lipocalin (NGAL) and Asthma Control Test (ACT) scores, and COPD Assessment Test (CAT) scores. Compared with pre-treatment parameters, the FeNO levels, RV/TLC, PaCO₂, total serum IgE, induced sputum EOS, plasma SP-A, sputum MPO, sputum NGAL, and CAT scores were significantly decreased after 6 months of ICS treatment, while FEV1%pred, FEV1/FVC, IC/TLC, PH, PaO₂, plasma sRAGE, and ACT scores were significantly increased in ACOS patients with different disease severity after 6 months of ICS treatment. This finding suggests that the FeNO level may accurately predict the efficacy of ICS in the treatment of ACOS patients.

10.Prospective study on the use of nickel-titanium temperature-dependent memory-shape device (CAR27) for anastomosis after colorectal surgery.

OBJECTIVETo prospectively evaluate the safety and efficacy of nickel- titanium temperature-dependent memory-shape device(CAR27) for colorectal anastomosis.
METHODSSixty colorectal cancer patients were randomly divided into two groups and received colorectal anastomosis with CAR27 or traditional stapling device. Complications, bowel function return, and the extrusion of anastomosis ring were prospectively monitored.
RESULTSBoth CAR27 and stapler group had one case of anastomotic leakage. Other complications such as stricture or obstruction were not found. Time for anastomosis of the two groups were (10.1±1.2) minutes and (11.2±2.1) minutes respectively. Time to first flatus was(3.2±1.2) days and (3.5±1.4) days respectively. Time to food intake resumption was (4.0±1.4) days and (4.3±1.3) days respectively. The differences above between the two groups were not statistically significant(P>0.05). The ring was expelled with stool within 7-16 days. The two groups were similar in operative time and the return of bowel function.
CONCLUSIONCAR27 is safe and simple for colorectal anastomosis.
