Objective To investigate the influence of Helicobacter pylori(Hp) infection on gastric mucin MUC5AC and MUC6 expression in children.Methods Sixty-six cases with gastric biopsy specimens were obtained from 66 children undergone gastroscopy from Jan.2005 to Jun.2006 for episodic or continuous abdominal pain,nausea,vomiting,abdominal distension,retching and dyspepsia,and so on.Among these children,39 cases were male and 27 cases were female,owning a average age(8.8?3.0)years.These specimens were divided into 2 groups followed by the presence of Hp,which was detected by rapid urease tests and Hp-PCR.Gastric mucin MUC5AC and MUC6 mRNA were also measured by reverse transcription PCR(RT-PCR),and Hematoxylin and Eosin staining were used for pathology observation.Comparisons between every groups were performed using t test and ?2 test,and statistical significance was defined as P

Objective: To investigate the potential role of me latonin in the adrenal cortex of human embryo. Methods:Specifi c melatonin receptors was localized and characterized in the adrenal cortex of h u man embryo by means of immunohistochemistry. Results: mt1 (Me l1a)and MT2 (Mel1b)subtype of melatonin receptors was principally localize d to cytoplasm in zona glomerulosa, fasciculata and reticularis. Conclu sion: It is possible that mt1 and MT2 subtype of melatonin receptors co-exist in the adrenal cortex of human embryo.

Objective: To determine protein binding characteri stic and signal transmission pathway of melatonin(Mel) receptor(MR) in human e mbryonic peripheral organ tissues. Methods: MR was measured by radio ligand-binding assay and the effect of GTPγS on melatonin specific bindi ng was studied. Results: Mel specific binding sites were det ermined in 16 kinds of human embryonic tissue and this binding could be inhibit ed by GTPγS, supporting the theory that MR is coupled to inhibitory G-proteins system. Conclusion: MR is measured in human embryo tissue, the se results provide experimental data for elucidating the mechanism of the effect of Mel.
4.Detecting melatonin receptor in thyroid of human embryo with immunohistochemistr y and in situ hybridization

Objective: To make it clear whether there exists m elatonin receptor in the thyroid of human embryo. Methods: Thyr oid was collected and sliced up to be stained with methods of immunohistochemis try and in situ hybridization. Results: The thyroid tissue w as p ositively dyed, melatonin receptor mt1 and MT2 were with both immunohistoche mistry and in situ hybridization while brown granules dep osited in the membrane, plasma and nuclear of the thyroid cell were with the imm unohitochemistry. Conclusion: There exists melatonin rece ptors in human embryo thyroid, either mt1 or MT2, and they exist in the memb rane, plasma and nuclear.
5.Action of sulfonylurea on mRNA levels of components of ATP-sensitive potassium channel(SUR1,SUR2 and Kir6.2)in rat brain

The effects of streptozotocin-induced diabetes or long-term glyburide administration on mRNA levels of components of ATP-sensitive potassium channel(SUR1,SUR2,Kir6.2)in rat brain were observed. Streptozotocin-induced diabetes itself did not affect the mRNA levels of SUR1,SUR2,and Kir6.2 in the brain, and glyburide-treatment increased the Kir6.2 mRNA level in brain by 23% in non-diabetic rats than those in normal control but did not change SUR1 and SUR2 levels.The effects of glyburide on SUR1,SUR2 and Kit6.2 mRNA levels did not manifest in brain of diabetic rats.
6.Determination of orientin and vitexin in Trollius chinesis preparation by HPLC.

OBJECTIVETo establish a HPLC method for determining orientin and vitexin in Trollius chinesis preparation.
METHODThe separation was carried out on Hypersil ODS column with a mixture of acetonitrile-1% acetic acid solution (15:85) as the mobile phase, at a flow rate of 1.0 mL x min(-1). The detection wavelength was set 340 nm.
RESULTOrientin showed a good linear relationship in the range of 8.42-84.2 microg x mL(-1) (r = 0.9999) and the average recovery is 98.3%, RSD is 0.8%. Vitexin showed a good linear relationship in the range of 8.07-80.7 microg x mL(-1) (r = 0.9999) and the average recovery is 102.4%. RSD is 1.7%.
CONCLUSIONThe method is sensitive, specific, and accurate with a good repeatability and can be used for the quality control of Trollius chinesis preparation.

9.Changes of biological characteristics of colon carcinoma cell line - LoVo induced by Kai1/CD82 transfection.

OBJECTIVETo study the influence of Kai1/CD82 transfection on the growth, adherence, separation and invasion potential of LoVo colon carcinoma cell line.
METHODSKai1/CD82 cDNA was transfected into LoVo cells, and a stable expressing clone was established. In vitro methodology was used to obtain the growth curve and also to detect the adherence, separation and invasion potential of the transfected LoVo cells, in comparison with those of control cells without transfection.
RESULTSCompared with the control, no change was observed in the growth pattern of transfected LoVo cells. The numbers of adherent cells in the two groups were 0.08, 0.63, 0.83, 0.91 (x 10(5)) for the transfected cells and 0.04, 0.48, 0.71, 0.82 (x 10(5)) for the control cells respectively after 10, 20, 30, 40 minutes culture with shaking. The difference at 20, 30 and 40 minutes was statistically significant (P < 0.05). The separation rates of each group were 13%, 20%, 53% for the transfected cells and 11%, 28%, 60% for the control cells, respectively after 5, 10, 15 minutes culture with shaking. The difference at 10 and 15 minutes was statistically significant (P < 0.05). The aggregation rate of the transfected cells was higher than that of the control cells after culture with mild shaking for 5 hours (64.8% vs. 58.6%, P < 0.05). After co-incubation with endothelium cells ECV304, the number of invading cells decreased more in the transfected cells than that in the control cells (6.33/field and 17.67/field, P < 0.05).
CONCLUSIONTransfection expression of Kail/CD82 into LoVo cell line results in an increase of cell adherence and aggregation, but a diminished capability of separation and invasion, suggesting that the expression of Kai1/CD82 gene may inhibit the metastatic potential of colon carcinoma.
