1.The intervention effects of prevention and treatment modes of hypertension in people living in agricultural and pastoral areas

Objective To explore an effective mode for the prevention and control of hypertension in agricultural and pastoral areas in Xinjiang by comparing the effect of mode A with that of B in blood pressure-lowering treatment. Methods 1445 patients with hypertension in agricultural and pastoral areas were included in this study. They voluntarily received mode A or B blood pressure-lowering treatment. The changes in heart rate , blood pressure, lipid, and glucose, and the incidences of cardiovascular and cerebrovascular events and adverse events were noted;and the effect of modes A and B was compared. Results 87.07%of the patients chose mode A group, while 12.93%chose mode B. The rate of pressure control was 70.71%in mode A group and 68.75%in mode B group , with no significant statistical difference (P>0.05). In 12th month of treatment, the decreased level of systolic pressure was (19.09 ± 20.33)mmHg in the group with mode A and (14.14 ± 17.85) mmHg in the group with mode B, showing a significant statistical difference between the two groups (P=0.047); and the declined level of diastolic pressure was (11.17 ± 13.23)mmHg and (8.17 ± 11.17) mmHg, respectively, with no significant statistical difference. Conclusion Mode A blood pressure-lowering treatment can effectively control blood pressure in hypertensive patients living in agricultural and pastoral areas in Xinjiang.
2.Optimal computer aided measure for screw internal fixation in the cavitas glenoidalis through human coracoid process of scapula

Objective To investigate a special optimization technique for computer aided measure, and provide anatomical basis for screw internal fixation in the cavitas glenoidalis through the coracoid process of scapula. Methods Thirty accurate scapula models were reconstructed from CT data sets. First, special optimization objective function was designed for single screw internal fixation configuration, and the optimal placement of screw was found automatically under constraints. Then, the placements of double screws internal fixation configuration were searched taking advantage of principal component analysis. Finally, statistical measure data were provided according to new anatomical reference landmarks for clinical use. Results For single screw internal fixation configuration, the distance from the optimal screw entry point P to the acromion process point X was (39.15±2.28) mm, to the coracoid process point Y was (28.66±2.68) mm, to the angulus superior point Z was (61.13±6.57) mm;The angle was (81.27±7.15)° between PX and PY, and (133.27±6.84)° between PX and PZ. The mean inclination of the lag screw was (104.08±4.41)° for the angle with line PX, (101.29±3.51)° with line PY, and (76.23±5.03)° with line PZ. For double screws configuration, the distance from the original single screw entry point P to the screw entry point E was (5.12±1.37)mm,to the screw entry point F was (3.88±0.94)mm. The angle between the long axis of coracoid process and line EF was (27.41±3.51)°. Conclusion The automatic optimization technique for computer aided measure is very efficient and has many advantages over the conventional manual dissection methods, and is convenient to design new anatomical reference landmark system for clinical use.
3.CT perfusion of pulmonary carcinoma:the correlative study with fluoro-deoxyglucose positron emission tomography and tumor microvessel density

Objective To investigate the correlation between CT perfusion parameters of pulmonary carcinoma and standardized uptake values(SUV)derived from ~(18)F-fluoro-deoxyglucose positron emission tomography(~8F-FDG PET)and tumor microvessel density(MVD),and to determine the validity of CT perfusion in assessing tumor angiagenic activity of pulmonary carcinoma.Methods Fifty patients(mean age 57.5,17 females)with pulmonary carcinoma underwent CT perfusion using 16-slice helical CT.Blood flow(BF,ml?100g~(-1)?min~(-1)),blood volume(BV,ml?100g~(-1)),mean transmit time(MTF,s)and permeability surface area product(PS,ml?100g~(-1)?min~(-1))were analyzed.SUV of PET was calculated in 14 patients.The CD34 immunohistochemical staining was used for tumor microvessel counting.CT perfusion parameters of pulmonary carcinoma were correlatively studied with SUV and tumor MVD.Pearson's correlation analysis was performed to evaluate the association between CT perfusion parameters and SUV and MVD.Results The average values of BF,BV,MTT and PS were 97.30 ml?100g~(-1)?min~(-1), 8.86 ml?100g~(-1),6.75 s and 34.52 ml?100g~(-1)?min~(-1),respectively.The average value of MVD was 61.82/FOV.The mean value of SUV was 5.96.There was positive correlation between BF and SUV(r= 0.727,P
4.Two-Step MS-PCR Combined With ELISA Method for the Detection of Drug Resistance Mutations in HIV-1 RT Gene

Highly Active Anti-Retroviral Therapy (HAART) has effectively inhibited the prevalence of HIV-1 and reduced the death rate caused by AIDS. In recent years,the emergence of resistance-conferring RT gene mutations in HIV-1 strains has become the major reason for HAART failure. The detection of drug resistance is important for the HAART regimen choice and novel drug development. A novel assay for the detection of HIV-1 RT drug resistance mutations was developed. HIV-1 drug resistance and wild strains in B subtypes were investigated using Two-Step Mutagenically-Separated PCR (MS-PCR),and point mutations including M41L,K70R,K103N,Y181C,T215F were detected. A longer mutant type primer was designed,using microplates hybridization and ELISA technique to detect several point mutations within a mixed mutant-wild type population. The results indicate that the Two-Step MS-PCR is as sensitive and specific as that in the traditional MS-PCR and MS-PCR combined with ELISA can give a good P/N quotient with better sensitivity,low cost,relatively less time consumption and high-throughput screening. It will be used in clinic usage for the detection of HIV-1 drug resistance mutations as well as other point mutations.
5.Study on the Extracting Method and Producing Conditions of Phyllosticta commelimecola Toxin

The crude toxin was extracted from hypha and culture solution of Phyllosticta commelimecola through three different polarity solvent: benzinum, puncificatum ethyl acetate and chloroform. The result indicated that the toxin secreted by Phyllosticta commelimecola not only was in hypha but also in culture solution and the extracting effect of ethyl acetate was the best. The soybean median and PSK media can be respectively used as solid and liquid culture media to produce toxin and grow mycelium. The optimal cultural conditions for producing toxin were temperature 32℃,cultured period 14d, cultured ways shaking of 150r/min.
6.Prokaryotic Expression and Functional Study of HIV-1 Envelope Glycoprotein gp41 Helical Bundle

HIV-1 envelope glycoprotein gp41,which is a hopeful target for HIV-1 fusion inhibitors,plays a critical role in the fusion of viral and cellular membranes.In order to build up the screening assay of HIV-1 fusion inhibitors targeting gp41,HIV-1 gp41 5-helix and 6-helix were expressed in prokaryotic cells.Gp41 5-helix and 6-helix recombined plasmids were constructed by using PCR,enzyme digestion and ligation taking the clade B HIV-1 genome as a template.The plasmid was transferred into E.coli BL21(DE3)and then induced by IPTG.The expressed protein was purified by affinity chromatography after denaturation and renaturation.The SDS-PAGE analysis was used during expression and purification.Native-PAGE was used to identify the interaction between gp41 5-helix and T-20.The result will be helpful to build up the screening assay of HIV-1 fusion inhibitors targeting gp41.
7.Influence of periacetabular muscle contraction on acetabular fractures

Objective To study the influence of periacetabular muscle contraction on acetabular] fractures.Methods Twenty intact adult cadaveric pelvis(40 hip joints)treated antiseptically with bilat- eral 1/2 femoral shafts were selected and divided randomly into two groups(20 hip joints in each group). Then,the specimens were trimmed to fit the RMT-150B rock mechanics measuring system and the special in- creasing pressure system.After pressure of 3 500 N was pressed to the control group and pressure of 4500 N to the experimental group,acetabular fracture model was made by beatening with special striking machine,when striking force was recorded.Results We made forty aeetabular fracture models from 40 cadaveric hips. Pearson correlation coefficient of pressing force and striking force was -0.923(P<0.01).The striking force of the control group and the experimental group was 445-550 N and 290-400 N,respectively(t= 14.727,P<0.01).Conclusion The increase of contraction force of the periaeetabular muscles can decrease aeetabular stress and strain,influence the result of violent injury and play a vital role in inducing acetabular fractures.
8.DNA damages of liver cells and expressions of DNA damage repair genes in rats exposed to vinyl chloride monomer.

OBJECTIVETo study DNA damages of liver cells in rats exposed to vinyl chloride monomer (VCM), and the expressions of DNA damage repair enzymes including O(6)-methyl guanine-DNA methyl transferase (MGMT), X-ray repair cross-complementing group 1 (XRCC1) and X-ray repair cross-complementing group 3 (XRCC3); and to explore the repair mechanism of DNA damage induced by VCM.
METHODSRats were exposed to VCM by intraperitoneal injection. DNA damages were detected by single cell gel electrophoresis (comet assay). The expressions of DNA damage repair enzymes were measured by immunohistochemical methods.
RESULTSThe percentages of comet cells in low, moderate, and high dose groups (11.75%, 12.38%, and 17.63%, respectively) were greater than that of control (5.67%). The latter two groups were significantly different from that of control (P < 0.05, P < 0.01). The expressions of MGMT and XRCC1 decreased, and XRCC3 increased with the dose of VCM increased. DNA damage was correlated with the expression of XRCC3 (r = 0.438, P = 0.067).
CONCLUSIONVCM can cause DNA damage of liver cells with dose-response relationship. DNA damage repair enzymes take part in the repairing of DNA damage induced by VCM.

10.Clinical observation on tongue acupuncture for treatment of stroke.

OBJECTIVETo observe therapeutic effect of tongue acupuncture on stroke.
METHODSOne hundred and ninety cases were randomly divided into an observation group and a control group, 95 cases in each group. The patients in the observation group were treated by tongue acupuncture combined with body acupuncture, and the control group by simple body acupuncture.
RESULTSThe total effective rate was 95.8% in the observation group and 80.0% in the control group, the observation group being significantly better than the control group.
CONCLUSIONTongue acupuncture has a better therapeutic effect on stroke.
