2.IL-24 expression at maternal-fetal interface and its roles in trophoblast invasion.

In this study, the expression of IL-24 at maternal-fetal interface and the roles in extravillous trophoblast (the TEV-1 cell line) invasion were examined. Immunohistochemistry was used to detect the expression of IL-24 in villi and decidual tissue. The proliferation of TEV-1 cells under the effect of IL-24 was measured by MTT assay. The invasiveness of TEV-1 cells under the effect of recombinant IL-24 (rhIL-24) was examined by transwell system. Immunohistochemical detection showed that IL-24 was expressed in the villi and decidual tissue, and distributed in villous column, trophoblasts, stroma and blood vessels. The proliferation of TEV-1 cells was not inhibited by rhIL-24 of various concentrations. The examination of invasion in vitro showed that rhIL-24 could inhibit the invasion of TEV-1 cells in a concentration-dependent manner. The results suggested IL-24 could inhibit the invasion of TEV-1 cells. Therefore, IL-24 produced by maternal-fetal interface in human first trimester pregnancy may influence the invasion of trophoblasts and is involved in normal pregnancy.

4.The research progress of speech feature and functional image in Parkinson′s disease

Speech disorders are the earliest sign of motor impairment in Parkinson′s disease (PD), even before limb dyskinesia. Patient′s quality of life can be affected significantly by the presence and severity of speech pathology. The early diagnosis is particularly necessary for PD with speech disorders. The abnormal phenomena in acoustic analysis have occurred in the early stage of PD. The voice of PD patients manifests characteristic changes on acoustic analyses, which is helpful for the diagnosis of PD, while the neural correlation of these features are not clear. In this paper, the speech features and functional imaging changes of patients with PD are described, and the mechanism of speech disorders is further revealed, and clues for the diagnosis and differential diagnosis of PD are provided.
5.Intensive reduction of blood pressure for the treatment of acute cerebral hemorrhage in elderly patients

Objective To study the application of intensive reduction of blood pressure to the treatment of acute cerebral hemorrhage in elderly patients. Method From January 2006 to December 2007, 40 elderly patients with acute cerebral hemorrhage were enrolled in a randomized and controlled study. Patients were randomly divided into intensive blood pressure reduction group and control group in equal number ( n = 20). The antihyper-tensive agent was used intravenously to reduce the systolic blood pressure to the level lower than 140 mmHg within 1 hour after treatment in patients of intensive blood pressure reduction group, and then the blood pressures was maintained for 7 days.The blood pressure of patients in control group was reduced to that just under 180 mmHg. The size of the haematoma was measured before and 3 days after treatment. The Stroke Scale set by the American National Institutes of Health (NHISS) was assessed before and 7 days and 28 days after treatment,separately. The modified Rankin Scale (mRS) was used to assess the patients 90 days after treatment. The outcomes were statistically analyzed with SPSS version 16.0 software. Measurement data were analyzed with t -test while numeration data were analyzed with chi-square test. Results Before and 3 days after treatment, there was significant difference in size hematoma between two groups (P < 0.05) . Between before and 7 days after treatment, there was significant difference in NHISS scale in intensive blood pressure reduction group (P < 0.05), but there was no significant difference in control group (P > 0.05). Before and 28 days after treatment, there was significant difference in variance of NHISS scale between two groups ( P < 0.05) . Nineth days after treatment, there was significant difference in mild disability and severe disability between two groups ( P < 0.05) . Conclusions Intensive reduction of blood pressure is valid for the treatment of elderly patients with acute cerebral hemorrhage.
6.Clinical features of acute hyperlipidemic pancreatitis

Objective To analyze the clinical features of acute hyperlipidemic panereatitis,and to investigate the therapeutic strategies.Methods In this retrospective study,44 patients with hyperlipidemic pancreatitis admitted to our hospital from January 2003 to December 2007 were enrolled,60 patients with acute pancreatitis of other etiologies were enrolled as the control group.Results The proportions of patients with overweight or obesity,hyperglycemia,fatty liver and hypertension were 81.8%,59.1%,54.5% and 68.2%,respectively;these were significantly higher than those in control group,which were 16.7%,16.7 %,13.3 % and 23.3 %,respectively (P<0.05 or<0.01).The proportion of patients with lithiasis was lower in HLP group than that in control group (13.6% vs 60.0%,P<0.05).There was no difference in the proportions of patients with chronic alcoholism between two groups.The Ranson score,CTSI,complications in HLP group were 3.15±0.07,4.46±2.58 and 3.2±1.7,respectively;these parameters were significantly higher than those in control group,which were 1.62±0.22,2.62±1.90 and 0.9±1.2 (P<0.05 or < 0.01 ).The level of serum amylase in HLP group was 580±222 mmol/L,which was significantly lower than that of control group (1361±472 mmol/L,P < 0.01 ).The triglyceride (TG) level was linearly correlated with Ranson score in HLP group ( r = 0.77,P < 0.05 ),but there was no linear correlation between TG level and Ranson score in the control group.Conclusions There was a close relationship between HLP and metabolic syndrome.Serum TG was positively correlated with the severity of HLP.
7.Histocompatibility of a porous material: Gelatin-bletilla carrying traditional Chinese medicine

BACKGROUNO:Some experiments have proved that some traditional Chinese medicines (TCM) have good biocompatibility when they touch and interact with human body tissues, body fluid or blood.Therefore,the physicochemical properties and functions of biomaterials can be improved by compounding it with some TCM.OBJECTIVE:To evaluate the biocompatibility of gelatin-bletilla porous material carrying TCM.DESIGN:Single sample experiment.SETTING:Department of Acupuncture & Moxibustion and Orthopaedics.Hubei College of Traditional Chinese Medicine. MATERIALS:This experiment was carried out in the Department of Orthopaedics,Tongji Medical College,Huazhong University of Science and Technology between February and April 2005.Twelve mice,male and female in half,weighing 18-24 g, and 6 Japanese big-eared rabbits,weighing 2.8 to 3.0 kg,were chosen for this experiment.They were raised at (18±1)℃ in separate cages.METHODS: Spongy gelatin-bletilla porous materials carrying extract of Coptidis and Radix Salviac Militiorrhizae was prepared by freeze-drying method.The acute systemic toxicity test:The 12 mice were divided into experimental group and control group. Mice in the two groups were intraperitoneally injected with leaching liquor and normal saline,respectively (the dose was 50 mL/Kg),then the general state and body mass of mice were recorded at the 4th,24th,48th and 72nd hours after injection, respectively.The porous materials were implanted in dorsal muscle of rabbits.Two rabbits were sacrificed at the 1st, 2nd and 6th weeks after operation seperately,and tissue reaction of biomaterial in vivo was assessed by gross observation and histological examination. MAIN OUTCOME MEASURES: Gross observation and histological examination of tissue reaction of biomaterials in vivo.RESULTS: Twelve mice and six big-eared rabbits were involved in final analysis.The acute systemic toxicity test:Mice of two groups had good status,normal activity and appetite,stable respiration:and no abdominal irritation,exhaustion, cyanosis or death was found.Body mass of mice of two groups presented tendency of increase at the 24th,48th and 72nd hours after injection,and there were no significant differences between two groups (P>0.05).The implantation test:①Gross observation:The samples were tightly bounded with muscular fibrous tissues at postoperative 1 week.They were connected with muscular fibrous tissues and could not be easily separated at postoperative 4 weeks.Gelatin-bletilla porous materials carrying TCM could hardly been seen at postoperative 8 weeks.②Histological observation:There were a lot of infiammatory cells and mesenchymal stem cells around the samples at 1 week after the implantation of gelatin-bletilla porous materials carrying TCM; Some materials were degraded and infiammatory cell infiltration was reduced around the implanted materials at 4 weeks; The inflammatory cells were further reduced and the materials were absorbed and replaced by muscular tissues at 12 weeks.CONCLUSION:Gelatin-bletilla porous materials carrying TCM is safe and reliable for application in wound healing,skin and muscle tendon tissue engineering.
8.Expressions of CD133 and CD105 in lung cancer tissue and their clinical significance

Objective:To investigate the expressions of tumor stem cell marker CD133 and tumor vascular endothelial cell marker CD105 in lung cancer tissue and their clinical significance.Methods:Streptavidin-biotin-peroxidase complex (SABC)-immunohistochemical staining was used to detect the expression of CD133 and CD105 in 65 cases of lung cancer tissue and 30 specimens of adjacent non-cancerous tissue. The relationship between the expression of CD133/CD105 and tumor size, histological types, differentiation, TNM stage, lymphoid metastasis, and prognosis were analyzed. Results:The positive expression rates of CD133 and CD105 in lung cancer tissues were significantly higher than those in adjacent non cancerous tissues, respectively (69.2% vs 26.7% and 67.7% vs 10.0%, both P<0.05). The positive expression rates of CD133 and CD105 in the group with lymphoid metastasis were higher than those with non lymphoid metastasis, respectively (82.5% vs 48.0% and 80.0% vs 48.0%, both P<0.05). The expressions of CD133 and CD105 were positively correlated with lymphoid metastasis with Pearson coefficient r of 0.35 and 0.32, respectively (P<0.05). No significant correlation was found between the expression of CD133/CD105 and tumor size, histological types, differentiation degree as well as TNM stage(P>0.05). The expression of CD133 was positively correlated with the expression of CD105 in lung cancer tissue with Pearson coefficient r of 0.37 (P<0.05). Postoperative median survival periods of CD133 and CD105-positive group were significantly shorter than the CD133 and CD105-negative groups (37 months vs 66 months,35 months vs 70 months, respectively, both P<0.05). Conclusion:The expression of CD133 and CD105 was associated with lymphoid metastasis and prognosis in patients with lung cancer.Their overexpression implies poor prognosis of lung cancer patients.
9.Polymorphism of HLA-A,B and DRB1 alleles and their haplotypes in Chinese Man bone marrow donors

ObjectiveTo analyze the polymorphism of HLA-A,B,and DRB1 alleles and their haplotypes in Chinese Man population.Methods Frequencies of HLA-A,B and DRB1 alleles and haplotypes were estimated by maximum-likelihood estimation method based on the genotypes of 2183 Chinese Man bone marrow donors.ResultsA total of 18 HLA-A alleles,44 HLA-B alleles and 15 HLA-DRB1 alleles were detected in Man population,and the most frequent alleles were A*02,A*11,A*24,A*30,A*33,B*13,B*35,B*46,B*51,B*40(B60),B*40(B61),B*15(B62),DRB1*04,DRB1*07,DRB1*08,DRB1*09,DRB1*11,DRB1*12,DRB1*13,DRB1*14 and DRB1*15.A*30-B*13,A*02-DRB1*15,B*13-DRB1*07 and A*30-B*13-DRB1*07 were the most frequent haplotypes in Man population for A-B,A-DRB1,B-DRB1 and A-B-DRB1 haplotype,respectively.The number of haplotypes with frequency ≥ 0.01 for A-B,A-DRB1 and B-DRB1 haplotype was 31,24 and 27,respectively,and ≥ 0.005 for A-B-DRB1 haplotype was 32.There were 14 in A-B,3 in A-DRB1,14 in B-DRB1 and 38 in A-B-DRB1 haploypes that showed strong linkage disequilibrium with ALD≥0.40.ConclusionsThe distribution of HLA-A,B and DRB1 alleles and haplotypes in Man population was similar to that in Northern Chinese Han population.
10.Varicella-zoster virus-related glomerulonephritis and encephalitis: case report and literature review

Objective To introduce a case of varicella.zoster virus(VZV)-related glomerulonephritis and encephalitis. Methods The clinical data and renal pathology were analyzed.Associated literatures were reviewed. Results A 15 years old male patient presented nephritic syndrome,nephrotic syndrome and renal dysfunction with reduced serum complement C3 level from the 5th day after he suffered from varicella.The pathological diagnosis of his kidney tissue was endocapillary proliferative glomerulonephritis with podocyte proliferation and severe renal tubular injury by light microscopy.Immunofluorescent and electron microscopic examinations showed "full-house"staining and granular electron-dense deposits in multiple sites.respectively. Furthermore.virus-like particles or/and inclusions could also be seen by electron microscopy and Mann staining light microscopy.Positive varicella-zoster virus (VZV) specific lgM antibody was detected by serum virological test.VZV antigen and RNA transcript were found in glomerular and tubular cells by immunohistochemical staining and in situ hybridization of renal tissues,respectively. The patient had epileptic episodes for many times in his disease course and his brain MRI and electroencephalogram findings accorded with viml encephalitis with secondary epilepsy.So,the diagnosis of VZV-related glomerulonephritis and encephalitis was established. Conclusion This is the first report of VZV-related glomerulonephritis and encephalitis confirmed by serum virology and tissue virology.
