1.Clinical observation on CO2 laser combined with thymopentin for treating condyloma acuminata in anal canal

Objective ToobservetheefficacyofCO2lasercombinedwiththymopentinfortreatingcondylomaacuminata(CA)in anal canal .Methods 134 patients with CA in anal canal were selected and randomly divided into two groups .The treatment group was treated with CO2 laser for eliminating the wart combined with thymopentin by local injection ,while the control group was trea-ted with CO2 laser .Results The cure rate in the treatment group and the control group were 69 .1% (47 cases) and 45 .4% (30 ca-ses) respectively and the recurrence rate in the treatment group and the control group were 30 .9% (21 cases) and 54 .6% (36 cases) respectively ,the differences between the two groups had statistical significance (P<0 .05) .Conclusion CO2 laser combined with thymopentin has better effect for treating CA in anal canal .
2.The Selection of Double J Tube Internal Drainage or Percutaneous Nephrectomy External Drainage in the Treatment of Upper Urinary Tract Obstruction Complicated with Acute Renal Failure

Objective To explore the selection and clinical effects of internal or external drainage for upper urinary tract obstruction complicated with acute renal failure.Methods A total of 25 cases of upper urinary tract obstruction complicated with acute renal failure were treated with double J(DJ) tube internal drainage(12 cases,15 times) or percutaneous nephrectomy(PCN) external drainage(19 cases,23 times).15 cases of obstruction were induced by tumor and 10 cases of obstruction induced by benign disease.Results The drainage treatments were performed successfully in 23 cases,with the success rate of PCN and DJ drainage being 86.9%(20/23) and 60.0%(9/15),respectively.Postoperative bleeding occurred in 1 patient with PCN.Conclusions PCN is superior to DJ in the treatment of malignant obstruction,but DJ internal drainage may be the first choice for non-malignant obstruction.

Objective To study the immunosuppression of Demethylzeylasteral (T-96) on the lymphocytic blastogenesis in vitro and compare the inhibitory effects on the rejection in renal transplant rats between T-96 with Glucosidorum Tripterygll Totorum (TII) in vivo. Methods Mixed lymphocyte cells were cultured through xenogenous antigen to stimulate C57BL/6 mouse spleen cell blast transformation, then different concentrations of T-96 or TII were added respectively. By 3H-TdR methods, the inhibitory effects of T-96 and TII on the blast transformation of spleen cells were measured. 5.Applications of 3D Reconstruction Dual-source CT Angiography in "Nutcracker" Syndrome

Objective To discuss the 3D reconstruction dual-source CT angiography (DSCT) in diagnosing nutcracker syn-drome,Methods 24 cases with known or suspected "nutcracker" syndrome were underwent with DSCTA and 3D recon-struction, left renal vein (LRV) were demonstrated and measured. Results Among 24 cases, 16 cases were diagnosed with "nutcracker" syndrome; the imaging demonstrated LRV were compressed. The angle between SMA and AO of 24 cases with "nutcracker" syndrome was abnormally acute(<45°). The angle A was 14°~38°, the average angle was 28.12°±7.92°. The angle A' was 11°~21°, the average angle was 16.46°±3.42°. Conclusion The 3D reconstruction DSCTA may be the most useful tool in diagnosing nutcracker phenomenon.

7.Bioinformatics analysis of transcriptome sequencing of early hypoxia damage in photoreceptor 661W cell line

Objective:To analyze the early changes of gene expression levels and signaling pathways in 661W cell line under hypoxic conditions and to find potential functional target genes.Methods:The cultured mouse 661W cells were divided into hypoxia treatment group and normoxia control group. Cells in the hypoxia treatment group were cultured in a three-gas incubator with volume fraction of 1% and 5% CO 2 at 37 ℃. Cells in the normoxia control group were cultured in an incubator at 37 ℃ with volume fraction of 5% CO 2. High-throughput sequencing technology was used to sequence the transcriptome of 661W cell treated with hypoxia and normoxia for 4 hours to screen for differentially expressed genes (DEG). Clustering heat map analysis, gene ontology (GO) functional enrichment analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis and protein-protein interaction network (PPI) analysis were performed. The reverse transcription-polymerase chain reaction (RT-PCR) was used to verify the accuracy of the sequencing results. Results:A total of 506 differentially expressed genes were screened, including 459 up-regulated genes and 47 down-regulated genes. GO functional enrichment analysis showed that the main biological processes of DEG were the cell's response to hypoxia, glycolysis, negative regulation of cell proliferation and apoptosis. hypoxia inducible factor (HIF)-1α pathway, glycolysis, Forkhead box O (FoxO) pathway, Insulin signaling pathway and Adenosine 5′-monophosphate-activated protein kinase (AMPK) pathway were involved in the above process. PPI analysis results showed that hub genes related to hypoxia were Aldoa, Aldoc, Gpi1, Hk2, Hk1, Pfkl, Pfkp, Vhl, Fbxo10 and Fbxo27. The RT-PCR results showed that the relative expression levels of 15 DEG mRNA in the hypoxic treatment group were higher than that of the normoxic control group, and the difference was statistically significant ( P<0.05). The mRNA expression levels of N-myc downstream-regulated gene-1 ( Ndrg1 ), Mt1, and vascular endothelial growth factor A ( VEGFA) were time-dependent on hypoxia. Conclusions:Under hypoxia, DEG is mainly related to glucose metabolism, cell response to hypoxia, regulation of proliferation and apoptosis. HIF-1α pathway, glycolysis, FoxO pathway and AMPK pathway are involved in the early changes of 661W cells under hypoxia. Aldoa, Aldoc, Gpi1, Hk2, Hk1, Pfkl, Pfkp, Vhl, Fbxo10, Fbxo27 may play key roles in the response of 661W cells to hypoxia. Ndrg1, Mt1 and VEGFA could be potential functional target genes for the study of ischemia and hypoxia-related fundus diseases.

Objective: To observe the expression of hMLH1 protein in esophagus squamous cell carcinoma, atypical hyperplasia tissue and normal esophagus tissue, so as to discuss the relationship between hMLH1 expression and esophagus carcinogenesis. Methods: The specimens of esophagus squamous cell carcinoma, atypical hyperplasia and normal esophagus tissue were obtained from 92 esophagus carcinoma patients. Immunohistochemistry (IHC) staining technique was used to detect expression of hMLH1 protein. The relationship between hMLH1 expression with clinical parameters, such as gender, age, cancer differentiation level, infiltration depth, tumor stage, lymphatic metastasis, was analyzed. OBJECTIVETo explore the effect of ouabain on intracellular Ca(2+) concentration ([Ca(2+)]i) in thoracic aorta vascular smooth muscle cells (VSMCs) in vitro.
METHODSPrimary SD rat thoracic aorta VSMCs were cultured by tissue adherent method and identified by immunochemistry. The binding ability between ouabain and VSMCs was detected by autoradiography, and fluo 3-AM (a Ca(2+) fluorescent probe) was employed to investigate whether ouabain affected VSMCs within a short period of time. The effect of a truncated fragment of the sodium pump α2 subunit was assayed in antagonizing the effect of ouabain on [Ca(2+)]i in the VSMCs.
RESULTSWithin the concentration range of 0.1-100 nmol/L, ouabain was found to dose-dependently bind to the VSMCs. Different concentrations of ouabain (0-3200 nmol/L) caused a transient, dose-dependent increase in [Ca(2+)]i in the VSMCs, which was antagonized by the application of the truncated fragment of sodium pump α2 subunit.
CONCLUSIONSElevations in [Ca(2+)]i in the VSMCs can be the cytological basis of high ouabain-induced hypertension. The truncated fragment of the sodium pump α2 subunit can antagonize ouabain-induced increase of [Ca(2+)]i in the VSMCs, which provides a clue for understanding the pathogenesis of and devising a therapeutic strategy for high ouabain-induced hypertension.
