1.Expression and significance of tyrosine kinase receptor B and brain-derived neurotrophic factor in gastric carcinoma

Objective To investigate the expressions of tyrosine kinase receptor (Trk)B and brain-derived neurotrophic factor (BDNF) protein in human gastric careinoma and compare them with those in epithelial cells of normal mucous, in order to evaluate their clinicopathological significance. Method The expressions of TrkB and BDNF protein in tumor tissues, matched with para-tumor mueosal tissues from 64 cases with gastric carcinoma and normal mucous of 20 cases were observed immunohistochemically and related to some of the clinicopathological parameters. Results The positive rates of TrkB and BDNF protein in tumor tissues were 60.9% and 59.4% respectively, but there was negative in matched para-tumor mueosal tissues and normal mucosal tissues. TrkB and BDNF protein expressions were related to invasive depth,lymph node metastasis and TNM stage of cancer, but not to sex, age and degrees of cancerous differentiation.The positive rates of TrkB and BDNF protein in cases with serosal infiltration, lymph node metastasis and TNM stage Ⅲ - Ⅳ were significantly higher than those in cases without serosal infiltration, lymph node metas-tasis, and TNM stage Ⅰ - Ⅱ (P< 0.01 ). In group with metastasis the positive rates of TrkB and BDNF protein were lower in metastatic foci (76.5%, 26/34, 70.6%, 24134) than those in primary tumor (85.3%, 29134,82.4%, 28/34 ), but statistically there was no significant difference between them (P > 0.05 ). Conclusions The expressions of TrkB and BDNF protein are closely relatedto tumorigenesis and progression of gastric carcinoma. The increased expression of TrkB and BDNF may promote the occurrance of local invasion and metastasis of gastric carcinoma.

3.Efficacy and safety assessment on herbs invigorating spleen and removing dampness, blood stasis and toxin combined with chemotherapy in the treatment of advanced colorectal cancer

Objective To evaluate systematically whether or not the treatment of herbs invigorating spleen and removing dampness,blood stasis and toxin with chemotherapy is better than chemotherapy alone for advanced colorectal cancer.Methods The randomized controlled trails (RCT) involving combined treatment of herbs with chemotherapy,published from January 2000 to October 2015,were searched in CBM,CNKI,Wanfang Data,VIP,PubMed,Embase databases.Stata 14.0 software was used to analyze the data.Results 19 RCT containing 941 patients were included in this reta-analysis.Compared with chemotherapy alone,the combined treatment of Chinese herbal medicine and chemotherapy was obviously better in 1-year survival rate (RR =1.28,95 % CI 1.14-1.45),2-year survival rate (RR =1.52,95 % CI 1.05-2.18),3-year survival rate (RR =2.76,95 % CI 1.56-4.88),objective response (RR =1.11,95 % CI 1.04-1.19),Karnofsky score (RR =1.46,95 % CI 1.27-1.68) and traditional chinese medicine (TCM) symptom score (RR =1.58,95 % CI 1.33-1.75).The adverse effect rate was statistically reduced in the combined treatment group:leukopenia (RR =0.59,95 % CI 0.40-0.8),nausea and vomiting (RR =0.68,95 % CI 0.59-0.79),diarrhea (RR =0.67,95 % CI 0.53-0.85),neurotoxicity (RR =0.79,95 % CI 0.65-0.96).Conclusion Compared with chemotherapy alone,the treatment of herbs invigorating spleen and removing dampness,blood stasis and toxin combined with chemotherapy for advanced colorectal cancer can significantly increase survival rate and objective response rate,improve the quality of life,and meanwhile decrease the adverse effect rate.

5.Study on the transcriptional activation of MTS1 gene beta promoter.

OBJECTIVETo investigate the effect of MTS1 gene beta promoter transcriptional activation in T-cell acute lymphoblastic leukemia (T-ALL) cell lines and identify the fragment with transcriptional activation.
METHODSSeven pGL3 recombinant plasmids with the same 3'-end transcriptional start site but the different 5'sequences were constructed by gene recombinant technique and transfected into Jurkat cell line which is biallelic deletion of MTS1 gene by transient transfection. Luciferase report gene was detected to observe beta promoter transcriptional activation.
RESULTSSeven pGL3 recombinant plasmids containing different fragments of beta promoter were obtained, all of them showed transcriptional activation in Jurkat cell line. Among them, the 0.38 kb fragment cut by SacII-SacI is fundamental in transcription.
CONCLUSIONMTS1 gene beta promoter can be activated in Jurkat cell line.

6.Minimally invasive percutaneous osteosynthesis with intramedullary nail and close reduction by manipulation for the treatment of femoral shaft fractures.

OBJECTIVETo explore the technique and clinical results of close reduction by manipulation and minimally invasive percutaneous osteosynthesis with intramedullary nail for the treatment of femur shaft fractures. methods: A retrospective study was conducted to analyze 96 patients with the femur shaft fractures who had been treated with close reduction by manipulation and minimally invasive percutaneous osteosynthesis with intramedullary nail. There were 67 males and 29 females. The average age of patients was 39 years old (ranging from 16 to 88). According to AO fracture classification for the femur shaft fractures,there were 29 cases of type A,46 type B,21 type C.
RESULTSAll the patients were followed up and the duration ranged from 12 to 24 months (averaged, 15 months). All the fractures showed union. The time required for the bony union ranged from 3 to 10 months (averaged,4 months). The clinical results were evaluated by Thorsen classification system. At the latest follow-up, 87 patients obtained excellent results, 7 good, 2 fair.
CONCLUSIONThis treatment method combines advantages of intramedullary nail with close manipulative reduction, so can get satisfactory clinical results for the treatment of femoral shaft fracture with minimal trauma.

Background:X-linked inhibitor of apoptosis (XIAP) is a vital factor in the anti-apoptosis mechanism of tumors and is highly expressed in renal cell carcinoma (RCC).However,the mechanism through which XIAP regulates DNA damage repair is unknown.This study investigated the regulatory mechanism of XIAP in etoposide-induced apoptosis in two Caki-1 cell lines with high or low XIAP expression.Methods:The two cell lines were established using RNA interference technology.The differentially expressed proteins in the two cell lines were globally analyzed through an isobaric tags for relative and absolute quantitation-based quantitative proteomics approach.Proteomic analysis revealed 255,375,362,and 5 differentially expressed proteins after 0,0.5,3,and 12 h of drug stimulation,respectively,between the two cell lines.The identified differentially expressed proteins were involved in numerous biological processes.In addition,the expression of histone proteins (H1.4,H2AX,H3.1,H3.2,and H3.3) was drastically altered,and the effects of XIAP silencing were accompanied by the marked downregulation of H2AX.Protein-protein interactions were assessed and confirmed through immunofluorescence and Western blot analyses.Results:The results suggested that XIAP may act as a vital cell signal regulator that regulates the expression of DNA repair-related proteins,such as H2AX,and influences the DNA repair process.Conclusions:Given these functions,XIAP may be the decisive factor in determining the sensitivity of RCC cell apoptosis induction in response to chemotherapeutic agents.
8.Effects of hydrocortisone sodium succinate on sodium current in human and guinea pig cardiac myocytes.

AIMTo study the effects of hydrocortisone sodium succinate on sodium current in human atrial myocytes and in guinea pig ventricular myocytes.
METHODSSingle cardiac myocytes were isolated by enzyme. The effects of hydrocortisone sodium succinate on sodium current (INa) were assessed by applying whole-cell patch clamp techniques.
RESULTSHydrocortisone sodium succinate (1, 3, 10 micromol x L(-1)) was shown to inhibit INa of both human atrial myocytes and guinea pig ventricular myocytes in concentration dependent manner and the IC50 were 6.97 and 8.74 micromol x L(-1), respectively. The inhibition effects acted quickly (1-3 min) and the maximal activating voltage of INa was not changed in both human and guinea pig cardiac myocytes.
CONCLUSIONHydrocortisone sodium succinate can exhibit inhibitory effects on INa in both human and guinea pig cardiac myocytes, and its inhibitory effects act rapidly, which are not consistent with genomic effects, so there may be nongenomic effects.

9.Effects of chemical constituent in roots of Achyranthes Bidentata.

The difference of ingredients between the roots from the SP1 plants of Achyranthes Bidentata carried by satellite and the control's were evaluated in this study. The techniques of ultraviolet-visible spectroscopy (UVS), infrared spectroscopy (IR), Xray diffraction (XRD) and SDS-PAGE were used to analyse the chemical constituent in the root of A. bidentata. The results demonstrated that the UVS, IR, XRD and protein fingerprints of the roots from A. bidentata were distinct with special characters. The difference of the IR, XRD and protein fingerprints could be discriminated the satellite plants roots from those of the control, however, there were no difference of the UVS fingerprints between the satellite plants roots and the control. This indicated that the kinds of chemical ingredients were not different between the two groups, but the contents of some chemical ingredients deceased in SP1 plants of A. bidentata carried by satellite.

10.Identification and characterization of a novel gene EOLA1 stimulating ECV304 cell proliferation.

OBJECTIVETo amplify the full-length cDNA and characterize the structure and biological function of a novel expression sequence tag ST55 (GenBank Accession No. BM121646).
METHODSRapid amplification of cDNA ends was used to clone the full-length of cDNA of ST55 in this study. Then, its tissue distribution was checked by Northern blots, and the associated protein was screened by GAL 4-based yeast two-hybrid. The effect of stable transfection of the cDNA on cell proliferation was evaluated in ECV304 cells.
RESULTSA full-length 1404 bp cDNA was cloned, and it was accepted as a novel human mRNA by GenBank (No. AY074889), named endothelial-overexpressed lipopolysaccharide-associated factor 1 (EOLA1). Bioinformatic analysis found that the EOLA1 encoded 158 amino acids, 17.89 kDa protein, and mapped to chromosome Xq27.4 with 5 exons. EOLA1 expressed in different human normal tissues and cancer cell lines. Using the EOLA1 cDNA as bait, we performed a yeast two-hybrid screening of a human liver cDNA library and identified metallothionein 2A (MT2A) as associated protein. The interaction between EOLA1 and MT2A was confirmed by co-immunoprecipitation experiments. Stable transfection of EOLA1 was noted to stimulate ECV304 cell proliferation (P < 0.05).
CONCLUSIONThe findings suggest that EOLA1 is a novel gene and the interaction of EOLA1 and MT2A may play an important role in cell protection in inflammation reaction.
