Objective:To observe the effects of effects of Xinfeng capsule(XFC) on platelet parameters(platelet count,thrombocytocrit,mean platelet volume,platelet distribution width) and on the serum levels of cytokines(IL-1?,TNF-?,IL-10) in adjuvant arthritis(AA) rats.Methods:40 SD masculine rats were averagely divided into groups of normal control,model control,XFC treatment,methotrexate(MTX) and tripterygium wilfordii polycoride tablet(TPT) alone,with 8 in each.Except for the rats of normal group,the others were intracutaneously injected with 0.1 ml of Freund′s complete adjuvant in the right hindlimb.The changes of platelet parameters (PLT,PCT,PDW,MPV) were observed by Sysmex XT-2000i automatic cytoanalyze and serum cytokines(IL-1?,TNF-?,IL-10)were analyzed with ELISA kit.Results:1.Compared with those of normal control,platelet count,thrombocytocrit,IL-1?、TNF-? of AA rats were obviously increased and IL-10 was obviously decreased in the model groups(P0.05).3.PLT and PCT of the rats were positively correlated with serum contents of IL-1? and TNF-?,as well as voix pedis′swelling and arthritis index(AI).In addition, IL-10 concentration in the serum was negatively correlated with PLT and PCT(P

To optimize the Scutellaria baicalensis extraction process, the filter paper method and the bacteriostatic ratio method were adopted to determine the in vitro bacteriostatic efficacy of water extracts and 60% alcohol extracts from S. baicalensis. The quantitative analysis of multi-components by single-marker (QAMS) was used to determined the contents of four active components, baicalin, wogonoside, baicalein and wogonin. In addition, with the bacteriostatic ratio and the overall desirability of the contents of four active components as indexes, the orthogonal experiment was adopted to detect the effect of water addition, extraction frequency and extraction time. The optimal extraction process was to add 12 times of water for the first time, 10 times of water for the second time, extract for 2 time, 2 h for each time. This optimization process is stable and feasible, with a higher bacteriostatic ratio in extracts.

Objective To understand the current situation of occupational exposure and occupational safety attitudes among nursing students in Kunming Medical University, and to find effective interventions to improve nurses' occupational safety,and to reduce occupational exposure.Methods 854 students in Kunming Medical University were investigated by a questionnaire survey, including Demographic characteristics, Occupational exposure conditions, Occupational safety attitude and Gathers, quit intention . The correlations between occupational safety attitudes and occupational exposure were analyzed with Simple Linear Correlation.Results There were 94.30%female respondents,and only 5.70%male respondents.The average age of respondents was 22.52±2.90 years. The Simple Linear Correlation analysis showed that correlation between Negative attitude and occupational exposure was positive correlated, the occupational exposure and Support from managers, and Optimistic were negative correlation ( <0.05) .Conclusions Occupational safety and attitude are related to occupational exposure. Nursing students should apply standardized operation to improve the professional quality and ability. Schools should strengthen the education of occupational protection for nursing students. Hospital management should regularly organize nurses to study occupational exposure protection knowledge,give full affirmation of the nurse's achievements in the work, and offer certain reward appropriately.

Objective　To study the protocol that induces adult rat bone marrow stromal cells (MSCs) to express neuron-specific enolase (NSE) in vitro. Methods　MSCs were preinduced with β-mercaptoethanol for 24 h, and then induced for 5 h. The positive percentages of NSE protein expression were measured by immunocytochemistry SABC staining. Results　After the induction, MSCs displayed neuronal morphologies, such as pyramidal cell bodies and processes which formed extensive networks. The positive percentage of NSE protein expression was (63.7±4.5)%. Conclusions　β-mercaptoethanol may induce adult rat MSCs to NSE-positive cells in vitro.

OBJECTIVETo explore the experimental conditions for H2O2 to injure astrocytes and the effect of baicalin in protecting neurons and astrocytes from oxidative stress injury.

METHODSNeurons and astrocytes from forebrain of rats were cultured in vitro and treated with H2O2, baicalin and combination of the two, respectively. The cell viability or survival rate was determined using MTT.

RESULTSEffects of H2O2 in different concentrations on survival rate of astrocytes showed significant difference (F = 28.569, P < 0.01) in a dose-dependent manner. Degrees of H2O2 injury, with the same concentration of H2O2, on cells with different seeding density were also significantly different (F = 5.439, P < 0.01), and dose-dependently. Baicalin didn't influence the survival rate of neurons and astrocytes when the concentration was within 2.5-40 mumol/L (for neurons, F = 0.49, P > 0.05; for astrocytes, F = 1.001, P > 0.05), but baicalin showed significant antagonism to the injury of oxidative stress (for neurons, F = 24.384, P < 0.01; for astrocytes, F = 5.000, P < 0.01). The higher the concentration of bainalin, the higher the cell survival rate.

CONCLUSIONA model of astrocytes oxidative injury induced by H2O2 is established. Baicalin shows no toxicity on neurons and astrocytes when the concentration is within 2.5-40 mumol/L, but could antagonize the H2O2 caused oxidative injury on cells in a dose-dependent manner.

Objective To explore whether the transcription factor NF-κB takes part in baicalin-induced differentiation of bone marrow stromal cells (MSCs) into neurons. Methods MSCs from adult rats were induced by baicalin in the serum-free medium for 6 hs. The un-treated cells, as the control group, were only induced in the serum-free medium without baicalin. The expression of neuronal specific markers was evaluated by indirect immunofluorescence cytochemistry staining. The activity of NF-κB was measured by the presence of the NF-κB subunit RelA (p65) translocated into the nucleus in the same way. Results After the induction by baicalin, MSCs displayed neuronal morphologies, such as pyramidal cell bodies and extensive networks of processes, and the expression of neuron-specific markers was detectable 6 ds after the induction. Neuronal specific marker proteins did not express in the control group. Six days after the induction, the P65 positive rate in the cytoplasm of the control group decreased to (18.4±3.0)%, while in the baicalin group, the P65 positive rate in the cytoplasm was (84.8±3.0)%. Conclusions Baicalin may inhibit the activation of NF-κB, which may act in the differentiation of MSCs into neurons.

BACKGROUNDSurgical interventions are the main treatment for primary congenital glaucoma (PCG). This study aimed to compare the efficacy and safety between viscocanalostomy and mitomycin C (MMC)-trabeculectomy in patients with PCG.

METHODSA total number of 43 patients with PCG who underwent either viscocanalostomy (group 1) or MMC- trabeculectomy (group 2) between June 2003 and June 2008 were retrospectively reviewed. The patients' intraocular pressures (IOPs) were examined before surgery and on day 1, week 1, month 1, month 6, and month 12 post-operative. Mean horizontal corneal diameters, success rates, intra- and post-operative complications were compared between the two groups.

RESULTSPre-operative IOPs were (31.96 ± 3.90) mmHg in group 1 and (32.56 ± 4.00) mmHg in group 2. At the last visit, IOPs were (16.78 ± 2.20) mmHg and (15.77 ± 2.60) mmHg, respectively (P < 0.001); the complete success rates of group 1 and group 2 were 45.9% and 67.4%, respectively, and the difference was not statistically significant (P = 0.158). There were no major complications occurred in the two groups.

CONCLUSIONSBoth viscocanalostomy and trabeculectomy can lower IOP in PCG patients effectively. Although there was no major complications occurred in both groups, viscocanlostomy may decrease the probability of postoperative haemorrhage, hypotony, cataract, or choroid effusion.

OBJECTIVETo detect the serum proteomic fingerprints in patients with hypopharyngeal squamous cell carcinoma (HPSCC) by surface enhanced laser desorption/ionization time of flight mass spectrometry (SELDI-TOF-MS) protein chip array technique.

METHODSThe serum samples were obtained from 58 HPSCC patients for protein expression analysis using SELDI-TOF Protein Chip technique and cation-exchange (CM10) protein array. All the spectra were compared and the qualified mass peaks with mass-to-charge ratios (m/z) between 1 and 70 kD were autotimatically detected. The tree analysis pattern was generated using Biomarker Patterns Software.

RESULTSThe protein profiles of HPSCC serum were analyzed according to the clinical and pathological features of the patients and their treatment response. No significant difference was noted in the serum proteins between HPSCC patients with different statuses of cervical lympha node metastasis (P>0.05), and the difference between well differentiated and poorly differentiated HPSCC was only minor. No significant difference was found in the serum proteins between chemotherapy-sensitive patients and the insensitive patients (P>0.05), but 5 proteins were identified to be overexpressed in the sensitive patients (P < / = 0.05). Radiotherapy-sensitive HPSCC patients were segregated from the insensitive group with a sensitivity of 86.67% and specificity of 100%.

CONCLUSIONThe serum protein at the m/z value of 6115.74 is overexpressed in radiotherapy-sensitive HPSCC patients. Serum protein profiling allows the prediction of radiotherapy response in HPSCC patients, and the identified proteins may serve as candidate biomarkers for predicting the radiotherapy sensitivity of HPSCC.
