Objective To study disfunction of activated T cell from peripheral blood and explore its role on pathogenesis in patients with refractory mycoplasma pneumoniae pneumonia (MPP).Methods T cell subsets and expression of membrane molecule on activated T cell in peripheral blood were measured in 26 refractory MPP patients by using immunofluorescence and flowcytometry,and compared with those of 18 control healthy children.Results The percentage of CD4~+ cell from peripheral blood in refractory MPP children was significantly lower than that in control group(P0.05).Conclusions There are disfunctions of T cell in refractory MPP patients in acute period,and the abnormal T cell activated disfunction may play an important role on pathogenesis.

Objective To investigate the effect of long hairpin RNA ( lhRNA) expression vector targeting HBV X gene ( HBx) on replication of hepatitis B virus ( HBV) and gene expression.Methods Four kinds of small interference RNAs ( siRNAs) were synthesized and lhRNA expression vectors targeting HBx were constructed.Four siRNA oligonucleotides and two lhRNA expression vectors were transfected into HepG2.2.15 cells.HBsAg, HBV DNA in culture supernatants and HBx mRNA in HepG2.2.15 cells were detected by time-resolved immunofluorometric assay, real-time quantitative PCR, and reverse transcription PCR, respectively.Negative sequence group or empty vector group was taken as the control.Independent-samples t test was performed to evaluate the inhibition effect on replication of HBV and gene expression. Results Compared with the negative control, HBsAg, HBV DNA level in culture supernatants and HBx mRNA in HepG2.2.15 cells were significantly decreased after siRNA-1 and siRNA-4 transfected at high concentrations (60 nmol/L or 90 nmol/L) (P<0.05), especially the HBsAg and HBV DNA levels in the siRNA-1 transfection group, which were significantly decreased at 24, 48 and 72 h after transfection ( P<0.05 or P <0.01 ) . Two lhRNA expression vectors ( pMD-HBxlh1 and pMD-HBxlh4 ) were successfully constructed and transfected into HepG2.2.15 cells, HBsAg and HBV DNA level in transfected cells was significantly lower than those in negative control (P<0.05).Conclusion The novel siRNA-1 is confirmed to target HBx gene and lhRNA expression vector targeting HBx can effectively inhibit the replication of HBV and expression of HBV gene.

Objective To evaluate the blood supply features and effectiveness of arterial chemoembolization for pedunculated hepatocellular carcinoma.Methods Angiography and chemoembolization via supplying blood arteries of tumor were performed in five patients with pedunculated hepatocellular carcinoma.Interventional procedure was carried out with tumor vascular infusion of 350 mg hot elemene emulsion and tumor embolization by cisplantin-lipidol emulsion(cisplantin 60-80 mg+lipidol 8-15 ml)and glutin.Results Ten interventional procedures(TACE)were undertaken in 5 patients.Angiography showed that tumor blood supply mainly coming from collateral circulation adjacent to the tumors,but partially from hepatic artery.Tumor sizes decreased from 30% to 50% in 5 cases,and AFP declined in 4 cases after the treatment. Conclusion Pedunculated hepatocellular carcinoma possessing different blood supply features from intrahepatocellular carcinomas.But transarterial ehemoembolization is still an effective method of choice for this treatment.

Objective To explore the effect of uhrasound-phoresis of Chinese herb in the treatment of frac- ture healing on rats.Methods The animal model of femur fracture was established in 36 rats,who were divided in- to 4 groups after operation.The rats in the ultrasound group were given ultrasound treatment daily.The herbal group was given Chinese herb applied on the fracture site.The experimental group was given uhrasound-phoresis of Chinese herb on the site of fracture.The control group was housed without any treatment.All rats were sacrificed at 30 days and the bony callus were harvested and observed with histological anti immunohistoehemical examination.Results The histological examination showed that the appearance of cartilaginous and bony callus in the experimental group were earlier than those in other groups,Immunohistochemical examination showed that the expression of collagenⅠin the experimental group was significantly higher than that in ultrasound group and herbal group(P

OBJECTIVETo observe the TH cell subset function in children with recurrent tonsillitis (RT) at the remission stage and to study the effects of astragalus membranacus (AM) on TH cell subset function.

METHODSThe peripheral blood mononuclear cells (PBMC) from 27 children with RT at the remission stage were stimulated with either phytohemagalutinin (PHA) (RT-PHA group) or PHA together with AM (RT-AM group) and were then cultured in vitro for 48 hrs. The samples from 21 healthy children stimulated with PHA were used as the Control group. The levels of interferon-gamma (IFN-gamma) and interleukin-4 (IL-4) in the supernatants of PBMC were detected using ELISA.

RESULTSThe IFN-gamma level and the ratio of IFN-gamma/IL-4 in the RT-PHA group were statistically lower than those in the Control group (P < 0.01). The level of IFN-gamma and the ratio of IFN-gamma/IL-4 in the RT-AM group were markedly higher than those in the RT-PHA group (P < 0.01), but were significantly lower than those in the Control group (P < 0.05). There were no differences in the IL-4 level among the three groups.

CONCLUSIONSTH1 cell subset dysfunction may exit in RT children at the remission stage, suggesting that TH1 cell subset dysfunction plays an important role in the pathogenesis of RT. AM can improve TH1 cell subset function and therefore shows an important significance in treating RT.

OBJECTIVETo study the effect of vacuum-assisted closure (V.A.C) on the expression of Urokinase-type plasminogen activator (uPA) and Urokinase-type plasminogen activator receptor(uPAR) protein in margin tissue of pigs with acute wounds and patients with chronic wounds.

METHODSAcute wounds were created on the two side of five male pigs' back, the experiment wounds on one side received V. A. C treatment and the control side received traditional treatment. Punch biopsies were taken from margin tissue of the wounds in 0, 1, 3, 6, 9, 12, 18, 25 days after the V.A.C treatment. The uPA and uPAR positive cells were stained with immunohistochemical technique . Six human chronic wounds were also treated with the V. A. C treatment, and the samples of extravasate from those wounds were collected in 0, 1, 3, 5, 7 days after the treatment, and the levels of uPA and uPAR expression were examined by enzyme-linked immunosorbent assay (ELISA).

RESULTSThe expression of uPA and uPAR protein in margin tissues of pigs with acute wounds increased and peaked in 3 days after the treatment with V. A. C, then it presented rapidly downtrend, but the expression and staining in the experiment group were obviously higher than that of the control group. In the six chronic wounds, the high level expression of uPA and uPAR protein was decreased after the treatment with V. A. C.

CONCLUSIONThe V. A. C may increase the expression of uPA and uPAR protein in acute wound keratinocytes and decrease the high expression of uPA and uPAR in chronic wounds.

OBJECTIVE: To study the expression of nerve growth factor (NGF) in diffuse axonal injury (DAI) in rat. METHODS: Eighty SD rats were used and samples were taken at 1 h, 3 h, 6 h, 12 h, 24 h, 48 h, 3 d, and 7 d after brain injury. The expressions of NGF in cerebral cortex, thalamus, cerebellum, and hippocampus were studied with immunohistochemistry and compared with normal group and sham operation group. RESULTS: Low expression of NGF was observed in normal group and sham operation group. The expression of NGF increased 1 h after injury, peaked at 12 h, and returned to basal level at day 7. CONCLUSION NGF is involved in repair of DAI. The changes of NGF expression following DAI may be applied to estimate the post-injury time interval of the brain in forensic medicine.

Antimicrobial peptides, a cluster of small peptides secreted by the majority of creatures, have been demonstrated with activity against a wide range of microorganisms including bacteria, protozoa, yeast, fungi, viruses and even tumor cells. These peptides have some features such as broad spectrum , high effi-cacy and stability, little drug resistance. A lack of new antibiotics combined with emerging multi-drug resis-tance issues demands that new antimicrobial strategies be explored for treating these infections. It has been proposed that the antimicrobial peptides might form the foundation for a new class of clinically useful an-timicrobials. We review the advantages of these molecules in construction features and bioactivity, with the focus on the mechanism and clinical applications.
